Benoxacor is enantioselectively metabolized by microsomes and cytosol from the human liver

Benoxacor is a safener added to current-use herbicide formulations to protect the target crop from herbicidal toxicity.It is an emerging environmental contaminant that has been detected in surface waters,raising the possibility of human exposure via drinking water.Because it is not subject to the same regulations as active pesticide ingredients,its metabolism and toxicity in humans have not been studied.Here we investigate the enantioselective metabolism of benoxacor in human subcellular fractions.Pooled human liver microsomes(pHLM)and cytosol(pHLC)were incubated with racemic benoxacor for up to 30-min.Gas chromatographic analyses were used to measure the enantioselective depletion of benoxacor.pHLMs with and without nicotinamide adenine dinucleotide phosphate(NADPH,co-factor for cytochrome P450 enzymes[CYPs])and pHLC with glutathione(GSH,co-factor for glutathione Stransferases[GSTs])metabolized benoxacor.These results demonstrate that microsomal CYPs,microsomal carboxylesterase(CESs),and cytosolic GSTs metabolize benoxacor.Females were predicted to have a higher clearance of benoxacor by GSTs than males.Male and female pHLM incubations with NADPH showed enrichment of the first eluting benoxacor enantiomer(E1-benoxacor).pHLM incubations without NADPH and pHLC incubations with GSH showed an enrichment of the second eluting enantiomer of benoxacor(E2-benoxacor).Our results indicate that human hepatic microsomal and cytosolic enzymes enantioselectively metabolize benoxacor,a fact that needs to be considered when investigating human exposures and toxicities of benoxacor. 展开更多

Endophytic bacteria colonizing the shoot-tips of banana cv. Grand Naine were isolated and tested for the antagonistic activity against the Panama wilt pathogen Fusarium oxysporum f. sp. cubense (Foc). Pre-isolation, the suckers were given extensive disinfection treatments and the homogenate from the excised shoot-tip portion was plated on nutrient agar (NA) and trypticase soy agar (TSA). This yielded altogether 47 isolates: 26 on NA and 21 on TSA, respectively, from the 10 suckers collected during August to February. The number of bacterial isolates obtained per sucker varied from one to 15 based on colony characteristics registering up to 10 distinct species per shoot-tip based on 16S rRNA sequence analysis. The 47 isolates belonged to 19 genera and 25 species under the phylogenetic classes of Actinobacteria, α- and γ-Proteobacteria and Firmicutes. Actinobacteria constituted the predominant phylum (55% isolates) with the constituent genera of Arthrobacter, Brevibacterium, Corynebacterium, Curtobacterium, Kocuria, Kytococcus, Micrococcus, Naumanella, Rothia and Tessaracoccus spp. and an unidentified isolate belonging to the family Frankiaceae. Proteobacteria constituted the second major phylum (Brevundimonas, Enterobacter, Klebsiella, Pseudomonas, Serratia and Sphingomonas spp.) followed by Firmicutes (Bacillus and Staphylococcus spp.). Antagonistic activity of the endophytes against Foc was tested through agar plate assays (pit and spot applications on fungal lawn) employing potato dextrose agar and NA. Endophytic Pseudomonas aeruginosa (isolate GNS.13.2a) which was associated with a single sucker showed significant growth inhibition effect on Foc while Klebsiella variicola (GNS.13.3a) and Enterobacter cloacae (GNS13.4a) exhibited moderate inhibition. The study brings out considerable sucker to sucker variation in the associated cultivable endophytic bacteria in “Grand Naine” banana and identifies a few bacterial endophytes with biocontrol potential against the devastating Foc pathogen. 展开更多

The topography, wide rainfall variation and varied temperature conditions enable inhabitancy of several plant species. This plant wealth is being used in various forms by tribes and natives of the area for centuries. These gain further importance due to the fact that ripe fruits serve as a source of food and medicine to tribes throughout the year. These wild fruits are source of minerals and vitamins in the diet of the rural people. The potential productivity and floristic diversity of the area have attracted the experts. There are more than 50 wild edible fruit species are available in Western Ghats region. These belong to 25 families and 38 genera. Out of the 25 families, the important ones as a source of fruits are Apocynaceae, Anacardiaceae, Euphorbiaceae, Moraceae, Sapotaceae and Sapindaceae. The fruiting period of different species ranges between two and six months. The highest number of species mature from April to May. The availability of these fruits reduces after October. Some of these fruits, such as Mangifera indica, Artocarpus heterophyllus, Carissa carandas, Emblica officinalis and Syzygium cuminii, have been given emphasis in order to identify high yield and high fruit quality clones and standardization of production technologies. The collection, conservation and utilization on some of the fruits, such as Garcinia indica and Garcinia gummigutta have started at various research organizations. Some other fruits, namely Chrysophyllum roxburghii (Family Sapotaceae), Canthium parviflora Lamk syn. Plectronia parviflora (Lam.) Bedd. (Family Rubiaceae), Elaeagnus conferta Roxbsyn, Elaeagnus latifolia L. (Family Elaeagnaceae), Securina leucopyrus syn. Flueggea leucopyrus (Family Euphorbiaceae), Elaeocarpus tuberculatus Roxb. In the present study, 2-D large eddy simulations(LES) are conducted for flow past a porous circular array with a solid volume fraction(SVF) of 8.8%, 15.4% and 21.5%. Such simulations are relevant to understanding flow in natural streams and channels containing patches of emerged vegetation. In the simulations discussed in the paper, the porous cylinder of diameter D contains a variable number of identical solid circular cylinders(rigid plant stems) of diameter d= 0.048 D. Most of the simulations are conducted at a Reynolds number of 2 100 based on the diameter D and the velocity of the steady uniform incoming flow. Though in all cases wake billows are shed in the regions where the separated shear layers(SSLs) forming on the sides of the porous cylinder interact, the effect of these wake billows on the mean drag is different. While in the high SVF case(21.5%), the total drag force oscillates quasi-regularly in time, similar to the canonical case of a large solid cylinder, in the cases with a lower SVF the shedding of the wake billows takes place sufficiently far from the cylinder such that the unsteady component of the total drag force is negligible. The mean amplitude of the oscillations of the drag force on the individual cylinders is the largest in a streamwise band centered around the center of the porous cylinder, where the wake to wake interactions are the strongest. In all cases the maximum drag force on the individual cylinders is the largest for the cylinders directly exposed to the flow, but this force is always smaller than the one induced on a small isolated cylinder and the average magnitude of the force on the cylinders directly exposed to the flow decreases monotonically with the increase in the SVF. Predictions of the global drag coefficients, Strouhal numbers associated with the wake vortex shedding and individual forces on the cylinders in the array from the present LES are in very good agreement with those of 2-D direct numerical simulations conducted on finer meshes, which s 展开更多

A one-dimensional non-intrusive Polynomial Chaos (PC) method is applied in Uncertainty Quantification (UQ) studies for CFD-based ship performances simulations. The uncertainty properties of Expected Value (EV) and Standard Deviation (SD) are evaluated by solving the PC coefficients from a linear system of algebraic equations. The one-dimensional PC with the Legendre polynomials is applied to: (1) stochastic input domain and (2) Cumulative Distribution Function (CDF) image domain, allowing for more flexibility. The PC method is validated with the Monte-Carlo benchmark results in several high-fidelity, CFD-based, ship UQ problems, evaluating the geometrical, operational and environmental uncertainties for the Delft Catamaran 372. Convergence is studied versus PC order P for both EV and SD, showing that high order PC is not necessary for present applications. Comparison is carried out for PC with/without the least square minimization when solving the PC coefficients. The least square minimization, using larger number of CFD samples, is recommended for current test cases. The study shows the potentials of PC method in Robust Design Optimization (RDO) and Reliability-Based Design Optimization (RBDO) of ship hydrodynamic performances. 展开更多

This paper describes a novel sharp interface approach for modeling the cavitation phenomena in incompressible viscous flows. A one-field formulation is adopted for the vapor-liquid two-phase flow and the interface is tracked using a volume of fluid(VOF) method. Phase change at the interface is modeled using a simplification of the Rayleigh-Plesset equation. Interface jump conditions in velocity and pressure field are treated using a level set based ghost fluid method. The level set function is constructed from the volume fraction function. A marching cubes method is used to compute the interface area at the interface grid cells. A parallel fast marching method is employed to propagate interface information into the field. A description of the equations and numerical methods is presented. Results for a cavitating hydrofoil are compared with experimental data. 展开更多

Arrays of large immobile boulders,which are often encountered in steep mountain streams,affect the timing and magnitude of sediment transport events through their interactions with the approach flow.Despite their importance in the quantification of the bedload rate,the collective influence of a boulder array on the approach timeaveraged and turbulent flow field has to date been overlooked.The overarching objective is,thus,to assess the collective effects of a boulder array on the time-averaged and turbulent flow fields surrounding an individual boulder within the array,placing particular emphasis on highlighting the bed shear stress spatial variability.The objective of this study is pursued by resolving and comparing the timeaveraged and turbulent flow fields developing around a boulder,with and without an array of isolated boulders being present.The results show that the effects of an individual boulder on the time-averaged streamwise velocity and turbulence intensity were limited to the boulder's immediate vicinity in the streamwise(x/d c < 2-3) and vertical(z/d c < 1) directions.Outside of the boulder's immediate vicinity,the time-averaged streamwise velocity was found to be globally decelerated.This global deceleration was attributed to the form drag generated collectively by the boulder array.More importantly,the boulder array reduced the applied shear stress exerted on theindividual boulders found within the array,by absorbing a portion of the total applied shear.Furthermore,the array was found to have a "homogenizing" effect on the near-bed turbulence thus significantly reducing the turbulence intensity in the near-bed region.The findings of this study suggest that the collective boulder array bears a portion of the total applied bed shear stress as form drag,hence reducing the available bed shear stress for transporting incoming mobile sediment.Thus,the effects of the boulder array should not be ignored in sediment transport predictions.These effects are encapsulated in this study by Equation(6). 展开更多

Hydroelectric facilities impact water temperature; low velocities in a reservoir increase residence time and enhance heat exchange in surface layers. In this study, an unsteady three-dimensional model was developed to predict the temperatm'e dynamics in the McNary Dam forebay. The model is based on the open-source code OpenFOAM. RANS equations with the Boussinesq approximation were used to solve the flow field. A: realizable k-ε model that accounts for the production of wind turbulence was developed. Solar radiation and convective heat transfer at the free surface were included. The result of the model was compared with the field data collected on August 18, 2004. Changes in diurnal stratification were adequately predicted by the model. Observed vertical and lateral temperature distributions were accurately captured. Results indicate that the model can be used as a numerical tool to assess structural and operational alternatives to reduce the forebay temperature. 展开更多

A Diagnostic Approach towards the Causes of Energy Balance Closure Problem 被引量:1

Hydrometeorological models are often evaluated and optimized on the basis of micrometeorological measurements. However, it has been known for more than three decades that surface measurements of sensible and latent heat energy (LE) are systematically underestimated. We studied this problem using six years of eddy-correlation measurements for four fields (corn, soybean, and prairie) in central Iowa, USA. We recorded major components of the energy equation (i.e. net radiation, sensible heat flux, LE, and soil heat flux, photosynthesis), and indirectly estimated most of the minor components of energy balance (namely storage in the soil, canopy and air). Storage in the canopy was related to leaf area index (LAI) acquired from Moderate Resolution Imaging Spectrometer (MODIS). In this paper, a diagnostic approach is investigated where systematic error is identified first. Three dimensional (3D) plots of the residual of energy equation vs. potential variables indicated the imbalance was largest mainly during the cold non-growing season when the soil was dry. Correlations between energy balance residual (EBR) and energy components showed that soil storage was not precisely estimated. Finally, an a-posteriori analysis (constrained linear multiple regression (CMLR)) was conducted to quantify the contribution of major/minor components of the energy equation towards EBR. The result highlights that the contribution of pertinent components of energy to EBR is mainly controlled by prevailing monthly hydrometeorological conditions;however, precise quantification of causes of imbalance is site-specific. A comparison between the a-posteriori analysis technique and the Bowen-ratio method demonstrates that the Bowen-ratio basically presumes a higher level of underestimation in LE. The results obtained in this study suggest that a-posteriori analysis may offer a superior methodology to correct measured eddy-correlation measurements. Delonix elata L.is a Ceasalpinaceae species and is traditionally used in India for treatment of skin diseases,liver diseases and rheumatic problems.However,systematic evaluation of its wound healing activity is lacking.Thus,in the present study,we aimed to assess the wound healing activity of D.elata stem bark extract(DSE) and its isolated constituent quercetin-3-rhamnopyranosyl-(1-6) glucopyranoside(QRPG) in rats.The formulations effects on wound healing were assessed by the wound contraction rate,epithelialization period,tensile strength,content of the hydroxyproline,hexosamine and uronic acid in granulation tissue,histopathological studies and Col 1 α(I) expression level in wound tissue by reverse transcription polymerase chain reaction(RT-PCR) study.The topical application of DSE ointment caused faster epithelialization,significant wound contraction(100%),and better tensile strength(710.5 ± 10.5 g/cm^2),while QRPG showed wound epithelialization with 98.2%contraction,better than that of the control group(78.18%).The biochemical analysis of granulation tissue revealed that DSE and QRPG significantly increased hydroxyproline,hexosamine and uronic acid content.A significant increase in the expression of Col 1 α(I) was observed in the wound tissue of DSE and QRPG treated rats.DSE and QRPG were shown to enhance wound healing by increasing collagen synthesis through upregulation of Col 1 α(I),thus validating ethnomedicinal uses. 展开更多

The incompressible unsteady Reynolds-averaged Navier-Stokes(URANS)simulations are performed for a free-running container ship in maneuvering conditions:the starboard and portside turning circle simulations with 35°rudder deflection.The validation variables include trajectory,motions,and propeller performances,and the prediction shows acceptable agreements against the experimental data.During the steady-state part of the turning,the inertial forces balancing the local forces are reported to quantitatively assess the centrifugal force which appears from the force equilibrium between the rudder,propeller,and the bare-hull.The study on the local flow focuses on finding the correlations between the propeller inflow and the propeller performance to investigate the differences in propeller performances during the portside and starboard turning.The preliminary simulations,performed with the grid triplet,comprise propeller open-water,resistance,and self-propulsion simulations,from which the validation studies and the studies for the local force and the local flow are fulfilled and applied for the main simulations.Both propeller and rudder are fully discretized and controlled,mimicking the experiment.Level-set,overset approach and Mentor’s SST model are employed for the free-surface capturing,large motion prediction,and turbulence closure. 展开更多

Part 2 reports the validation,local force and local flow study results for the free-running added power simulations whose conditions are the same as the self-propulsion test except for the increased propeller rotational speed and the presence of wave.When targeting the same mean Froude number in the wave condition,the propeller requires the increased propeller rotational speed for the operation at the low advance ratio due to the added resistance.The test is performed at five different wavelengths in head waves and four different headings in the oblique waves.For the validation study,the time series of the validation variables is decomposed with discrete Fourier transform to extract the harmonic values.Validation variables are global parameters,including motions,propeller thrust,and torque coefficients,added power variables,and self-propulsion factors which show reasonable agreement against the experiment results and produces a similar error from the self-propulsion simulation.The local force study shows that the added resistance mostly appears at the bow due to the bow plunging during the short head wave and resonance condition.The contributions of the gravitational force and the buoyant force are found to increase as the stern motion exceeds the bow motion during the long head wave condition.The oscillation of the propeller performances shows correlation with the first harmonic amplitude of the propeller inflow.Heave,pitch,and roll decay tests are performed prior to the main test to assess the natural frequencies of the ship.Same as Part 1,a discretized propeller is used. 展开更多

A Time-Based Framework for Evaluating Hydrologic Routing Methodologies Using Wavelet Transform

In this study we explore a method which provides an insight into the effectiveness of various hydrologic models’ routing components based on their ability to accurately represent flood peak times and shapes. The method is based on using Cross-Wavelet Transforms to estimate the phase (time) difference between the time series of the observed and the simulated discharges. In this article we evaluate two routing components, the Routing Application for Parallel Computation of Discharge (RAPID), which is based on the simplified Muskingum routing method, and the routing component of the non-linear Hillslope-Link hydrologic Model (HLM) produced in the Iowa Flood Center (IFC). Both routing components are driven by the same source of runoff and used the same channel network to ensure that the discrepancies between the simulated stream discharges are due to channel routing alone. We also explore the suitability of different wavelet shapes for our application, and how the difference in wavelet shape can affect our evaluation results. Unlike the conventional statistical skill scores used to evaluate model performance (e.g. Root Mean Squared Error, correlation coefficient, and Nash Sutcliff efficiency index), which give an estimate of the overall hydrograph performance, our method conveniently provides time-localized information with higher resolution at peak location. We perform our evaluation at multiple stream gauge locations, covering a wide range of scales (700 to 16,862 km2), located in the eastern part of the state of Iowa. Our results show that the proposed wavelet method is effective in evaluating the performance of the routing components in simulating peak times across spatial scales. Generally, the non-linear routing method employed in the HLM outperformed the Muskingum based method employed in RAPID. In addition, our results suggest that the Paul wavelet is more effective in detecting and separating individual peaks than the Morlet wavelet, which in turn leads to a more accurate evaluation of the routing componen 展开更多
