The dynamics of soil inorganic nitrogen (NH4^+ -N and NO3^- -N) and microbial biomass carbon (Cmic) and nitrogen (Nmic) under 30-year-old fenced Pinus sylvestris L. var. mongolica Litvin (SF), unfenced P. sylvestris L. var. mongolica Litvin (SUF), and unfenced Pinus densiflora Siebold et Zucc. (DUF) plantations in the Zhanggutai sandy soil of China were studied during Apr. to Oct. 2004 by the in situ closed-top core incubation method. All mentioned C and N indices in each stand type fluctuated over time. The ranges of inorganic N, Cmic, and Nmic contents in the three stand types were 0.7-2.6, 40.0-128.9, and 5.4-15.2 μg g^-1, respectively. The average contents of soil NH4^+ -N and Cmic under the three 30-year-old pine plantations were not different. However, soil NO3^ -N and total inorganic N contents decreased in the order of SUF ≥ SF ≥ DUF, the Nmic content was in the order of SF = SUF 〉 DUF, and the Cmic:Nmic ratio was in the order of SUF = DUF 〉 SF. Seasonal variations were observed in soil inorganic N, microbial biomass, and plant growth. These seasonal variations had certain correlations with microbe and plant N use in the soil, and their competition for NH4^+ -N was mostly regulated by soil N availability. The influence of tree species on inorganic N and Nmic were mainly because of differences in litter quality. Lack of gazing decreased the Cmic:Nmic ratio owing to decreased carbon output and increased the ability of soil to supply N. The soil N supply under the P. sylvestris var. mongolica plantation was lower than under the P. densiflora plantation. 展开更多

BACKGROUND Acute ischemic stroke(AIS)is one of the major causes of the continuous increasing rate of global mortality due to the lack of timely diagnosis,prognosis,and management.This study provides a primitive platform for non-invasive and cost-effective diagnosis and prognosis of patients with AIS using circulating cellfree mitochondrial DNA(cf-mtDNA)quantification and validation.AIM To evaluate the role of cf-mtDNA as s non-invasive,and affordable tool for realtime monitoring and prognosticating AIS patients at disease onset and during treatment.METHODS This study enrolled 88 participants including 44 patients with AIS and 44 healthy controls with almost similar mean age group at stroke onset,and at 24 h and 72 h of treatment.Peripheral blood samples were collected from each study participant and plasma was separated using centrifugation.The cf-mtDNA concentration was quantified using nanodrop reading and validated through real-time quantitative polymerase chain reaction(RT-qPCR)of NADH-ubiquinone oxidoreductase chain 1(ND1)relative transcript expression levels.RESULTS Comparative analysis of cf-mtDNA concentration in patients at disease onset showed significantly increased levels compared to control individuals for both nanodrop reading,as well as ND1 relative expression levels(P<0.0001).Intergroup analysis of cf-mtDNA concentration using nanodrop showed significantly reduced levels in patients at 72 h of treatment compared to onset(P<0.01).However,RT-qPCR analysis showed a significant reduction at 24 h and 72 h of treatment compared to the disease onset(P<0.001).The sensitivity and specificity were relatively higher for RT-qPCR than nanodrop-based cfmtDNA quantification.Correlation analysis of both cf-mtDNA concentration as well as ND1 relative expression with National Institute of Health Stroke Scale score at baseline showed a positive trend.CONCLUSION In summary,quantitative estimation of highly pure cf-mtDNA provides a simple,highly sensitive and specific,non-invasive,and affordable approach for real-time 展开更多

Immediate Respiratory Response to Electronic Cigarette Use

Background: Electronic cigarettes were originally designed to reduce adult dependency on normal cigarettes and as a tobacco cessation tool to substitute traditional cigarettes. But it has become the most popular among teenagers. Rationale: To investigate the immediate adverse respiratory effect of short-term electronic cigarette vapor inhalation. Method: Twenty-five subjects were randomly selected and used in this study. The respiratory resistance values were evaluated and used for comparison. The subjects were asked to breathe into the Airflow Perturbation Device (APD) for evaluation of their respiratory resistance before vaping (in triplicate). The same subjects, a minute later, were then asked to use one poke (3 seconds) of the e-Cigarette device to inhale e-Cigarette vapor with nicotine from a pod with 59 mg/ml nicotine. Immediately following the e-Cigarette use, their respiratory resistance was measured again (in triplicate). Results: Comparing the respiratory resistance values before and immediately after exposure to e-Cigarette vapor showed that their respiratory resistance increased almost immediately. Conclusion: Although there are long-term studies showing that the e-Cigarette is as harmful as regular cigarettes, this study showed a nearly immediate effect of using the e-Cigarette that significantly increased the respiratory resistance of the user. Very short exposure time to e-Cigarette vapor (3 seconds only), caused an immediate adverse physiologic effect in respiratory resistance. 展开更多

Early diagnosis and prognosis of ischemic stroke remains a critical challenge in clinical settings.A blood biomarker can be a promising quantitative tool to represent the clinical manifestations in ischemic stroke.Cell-free DNA(cfDNA)has recently turned out to be a popular circulating biomarker due to its potential relevance for diagnostic applications in a variety of disorders.Despite bright outlook of cfDNA in clinical applications,very less is known about its origin,composition,or function.Several recent studies have identified cell-derived mitochondrial components including mitochondrial DNA(mtDNA)in the extracellular spaces including blood and cerebrospinal fluid.However,the time course of alterations in plasma mtDNA concentrations in patients after an ischemic stroke is poorly understood.DNA is thought to be freed into the plasma shortly after the commencement of an ischemic stroke and then gradually decreased.However,the importance of cell-free mtDNA(cf-mtDNA)in ischemic stroke is still unknown.This review summarizes about the utility of biomarkers which has been standardized in clinical settings and role of cfDNA including cfmtDNA as a non-invasive potential biomarker of ischemic stroke. 展开更多

Assessment of Bronchodilator Response in Children with Asthma Exacerbation Using the Respiratory Resistance Values

Eleven of the children could not perform spirometry. The APD re</span><span style="font-family:Verdana;">sults demonstrated that respiratory resistance of the patients decreased by a</span><span style="font-family:Verdana;">bout 20%, indicating that the APD could detect the expected response to bronchodilator. However, no similar conclusion could be made with the spirometry parameters (</span><span style="font-family:Verdana;">FVC, FEV1, FEV1/FVC, and FEF25% - 75%) performed on the same patients. The differences on the spirometry parameters did not change </span><span style="font-family:Verdana;">significantly before and after bronchodilator administration. Furthermore, th</span><span style="font-family:Verdana;">ese differences were negligibly increased or decreased for some with no consistency between the FVC, FEV1, FEV1/FVC, and FEF25% - 75%. Even though all the children were clinically improved after albuterol administration and discharged home, this could not be demonstrated by spirometry data. This study validates previous reports that spirometry is not a reliable pulmonary diagnostic tool for young children, as spirometry is highly effort-dependent and requires a substantial degree of patient cooperation. An efficient and compact double-pass optical fiber amplifier is demonstrated using a newly developed hafnia bismuth erbium co-doped fiber(HBEDF) as a gain medium. The HBEDF is fabricated using a modified chemical vapor deposition in conjunction with solution doping. The fiber has an erbium ion concentration of 12500 ppm.At the optimum length of 0.5 m, the HBEDF amplifier(HBEDFA) achieves a flat gain of 26 d B with a gain variation of less than 1.5 d B within a wavelength region from 1530 to 1560 nm when the input signal and pump power are fixed at-30 d Bm and 140 m W, respectively. On the other hand, at the input signal power of-10 d Bm,the HBEDFA also achieves a flat gain of 14.2 d B with a gain variation of less than 2.5 d B within a wide wavelength region from 1525 to 1570 nm. Compared with the conventional zirconia erbium co-doped fiber based amplifier,the proposed HBEDFA obtains a more efficient gain and lower noise figure. For an input signal of-30 d Bm, the gain improvements of 6.2 d B and 4.8 d B are obtained at 1525 nm and 1540 nm, respectively. 展开更多
