反相高效液相色谱法测定西番莲中的有机酸 被引量:20

采用Waters Atlantis C18色谱柱(150 mm×4.6 mm,5μm),以2%甲醇-98%0.01 mol/L K2HPO4溶液(用H3PO4调节p H2.8)作为流动相,流速为0.6 m L/min,柱温为30℃,进样量为10μL,检测波长为210 nm,建立高效液相色谱同时分析西番莲中10... 采用Waters Atlantis C18色谱柱(150 mm×4.6 mm,5μm),以2%甲醇-98%0.01 mol/L K2HPO4溶液(用H3PO4调节p H2.8)作为流动相,流速为0.6 m L/min,柱温为30℃,进样量为10μL,检测波长为210 nm,建立高效液相色谱同时分析西番莲中10种有机酸的方法。结果表明:该方法相对标准偏差为0.22%-4.00%,平均回收率在88.0%-109.6%,各种酸的线性相关系数r〉0.999 2,检测限为0.001-0.22μg/m L;采用外标法定量。在紫果和黄果西番莲中检测到7种有机酸,其含量趋势呈现为柠檬酸〉L-苹果酸〉L-乳酸〉琥珀酸〉L-抗坏血酸〉酒石酸〉富马酸。该方法具有灵敏度高、测试组分多、速度快等优点,适用于西番莲中多种有机酸的测定。 展开更多

为建立西番莲果肉中叶黄素和β-胡萝卜素的快速提取和HPLC检测方法,以西番莲果肉为试材,对叶黄素和β-胡萝卜素的提取和检测方法进行优化。结果表明:2种类胡萝卜素的最佳提取条件为1 050μL的乙酸乙酯-二氯甲烷-正己烷有机混合溶剂(64... 为建立西番莲果肉中叶黄素和β-胡萝卜素的快速提取和HPLC检测方法,以西番莲果肉为试材,对叶黄素和β-胡萝卜素的提取和检测方法进行优化。结果表明:2种类胡萝卜素的最佳提取条件为1 050μL的乙酸乙酯-二氯甲烷-正己烷有机混合溶剂(64∶16∶4,V/V/V),10min左右可高效提取西番莲果肉中叶黄素和β-胡萝卜素。以C30色谱柱;流动相:甲醇-叔丁基甲醚(80:20,V/V)等度洗脱;流速:0.8 m L/min;进样量:20μL;柱温:30℃;检测波长:450 nm。该色谱条件下,叶黄素和β-胡萝卜素线性关系良好,相关系数(R2)分别为0.999 0和0.999 3;检出限为0.01μg/m L和0.02μg/m L;2种类胡萝卜素的平均回收率分别达到了98.47%、96.43%;样品稳定性和检测方法准确度较好,RSD<5.0%。对3个西番莲品种果实中叶黄素和β-胡萝卜素含量进行测定,其含量呈现双花西番莲>黄果西番莲>紫果西番莲的趋势。该提取和检测方法具有速度快、有机溶剂用量少,灵敏度高等优点,能在短时间内实现大量西番莲样品中叶黄素和β-胡萝卜素的提取和检测。 展开更多

Research on Correlation between Synthesis of Stilbenes and Aflatoxin Accumulation Caused by Aspergillus flavus in Peanut Seeds

In peanuts, a mechanism of resistance to fungal infection is reported on the synthesis of stilbene phytoalexins, which are antibiotic, low molecular weight metabolites. The phytoalexin-associated response of different peanut varieties to the inoculation with A. flavus has not been investigated and might be useful for breed- ing resistant peanut cultivars. In this study, peanut varieties resistant or susceptible to aflatoxin accumulation caused by A. flavus were selected to compare the synthe- sis differences of the four main stilbenes (resveratrol, ε-viniferin, 6-viniferin and pterostilbene) and the activities of their resistant enzymes (PAL, POD, PPO), and explore the relationship between stilbene synthesis and resistance to aflatoxin accu- mulation. The results indicated that the synthesis speed of stilbenes was related to the resistance to aflatoxin accumulation in peanuts seeds. The total content of the four stilbenes reached the highest value of 47.37 μg/g in the resistant cultivars (Qian huasheng 3) after inoculation by A. flavus (54 times of the control) on the 3nd day, while the total content of stilbenes in susceptible cultivars (Huayu 22) was 5.5 μg/g. The content of stilbenes and the activities of their resistant enzymes from the resistant cultivars were both higher than from the susceptible one. The four peanut varieties with relative higher stilbene contents and lower disease index and aflatoxin content were selected by using the stilbene content. Very significant negative corre- lation, with correlation coefficients (r) of -0.789, -0.851 and -0.850, was observed between the stilbene content with disease index, the content of aflatoxin B1 and the total content of aflatoxins, respectively. Therefore, the study suggested that the total content of stilbene compounds might serve as an important chemical index in peanut seeds after inoculation with A. flavus on the 3nd day, during screening and breeding of peanut varieties resistant to aflatoxin accumulation. 展开更多
