Morphological responses to water-level fluctuations of two submerged macrophytes, Myriophyllum spicatum and Hydrilla verticillata 被引量:14

Aims Myriophyllum spicatum and Hydrilla verticillata are common submerged macrophytes in the Yangtze River basin.To investigate their tolerances and adaptations to water-level fluctuations,an experiment was conducted in a pond.Methods We designed five different amplitudes of water-level fluctuations as static,615,630,660 and 690 cm during the 74 days of the experiment.In each amplitude treatment,two cultivation methods were examined as monoculture and mixed culture.Important Findings The results showed that M.spicatum had greater morphological responses to water-level fluctuations than H.verticillata.Fluctuating amplitude had significant effects on branch number,shoot length and root dry weight(DW)of M.spicatum,whereas it only had significant effect on branch number of H.verticillata.Both fluctuating amplitude and cultivation method had significant effects on total DW of M.spicatum,which was higher in monoculture than mixed culture.The total DW of H.verticillata was affected by fluctuating amplitude only,and the largest biomass was in the amplitude of 630 cm.Fruit DW of M.spicatum was largest in the amplitude of 630 cm,high amplitude of water-level fluctuations would inhibit flowering and seed production.Root DW proportion was significantly affected by fluctuating amplitude and cultivation method in both species,and the root DW proportion of M.spicatum was significantly higher in the amplitude-of 690 cm.We conclude that moderate amplitude of water-level fluctuations can promote the distribution and growth of both species,and in order to accelerate the restoration of both species in natural habitats,the optimum amplitude should be keep at 615 to 630 cm. 展开更多

Plants have been used as good bio-indicators and genetic toxicity of environmental pollution in recent years. In this study, aquatic plants Hydrilla verticillata and Ceratophyllum demersum treated with 10umol/L Cd, 5 umol/L Hg, and 20 umol/L Cu for 96 h, showed changes in chlorophyll, protein content, and in DNA profiles. The changes in DNA profiles included variation in band intensity, presence or absence of certain bands and even appearance of new bands. Genomic template stability test performed for the qualitative measurement of changes in randomly amplified polymorphic DNA (RAPD) profiles, showed significant effect at the given concentration of metals. Cloning and sequencing of bands suggested that these markers although may not be homologous to any known gene but its conversion as a sequence characterized amplified region (SCAR) marker is useful in detecting the effects of genotoxin agents. 展开更多

研究了压条日期、环剥宽度、植物生长调节剂浓度和压条基质对3-4年生的盆栽北美冬青(Ilex verticillata)"奥斯特"结果枝高空压条成活率和生根率的影响。结果表明:4种不同浓度(原液、3/4原液、1/2原液、1/4原液、)的植物生长调节... 研究了压条日期、环剥宽度、植物生长调节剂浓度和压条基质对3-4年生的盆栽北美冬青(Ilex verticillata)"奥斯特"结果枝高空压条成活率和生根率的影响。结果表明:4种不同浓度(原液、3/4原液、1/2原液、1/4原液、)的植物生长调节剂(NAA+IBA)处理能够提高压条的生根率,生根率可达100%;不同环剥宽度影响压条的愈伤率和成活率,环剥宽度过小(0.2cm),愈伤化严重,生根率降低,环剥宽度过大(1.5cm),成活率降低;3种压条基质以V_(泥炭)∶V_(珍珠岩)=3∶1混配基质效果最好,平均生根条数达10.73条,平均根长7.3cm。从综合指标看,压条时间选择在6-7月,环剥宽度0.5-1.0cm,以1/4-1/2原液浓度的植物生长调节剂处理最适宜。 展开更多

为探究乙酸提高北美冬青抗旱能力的分子机制,通过对CK (清水)、T1 (20 mmol/L乙酸溶液)、T2(40 mmol/L乙酸溶液)处理后的北美冬青叶片进行转录组测序及分析。通过组装共得到120 421条Unigene序列,共计73.75%的序列在相关数据库中得到注... 为探究乙酸提高北美冬青抗旱能力的分子机制,通过对CK (清水)、T1 (20 mmol/L乙酸溶液)、T2(40 mmol/L乙酸溶液)处理后的北美冬青叶片进行转录组测序及分析。通过组装共得到120 421条Unigene序列,共计73.75%的序列在相关数据库中得到注释;通过3个组差异表达基因比较得到以下结果:与CK相比,T1处理后的北美冬青叶片有10 229个基因表达量上调,559个基因表达量下调;T2分别有10 959个上调和938个下调;T1相比较T2有3 524个上调,4 287个下调。GO富集分析显示差异基因主要涉及结合、催化活性、转运活性、细胞成分的组织与生物发生、细胞定位、生物调节细胞、细胞分离及细胞器等。KEGG分析表明在使用乙酸溶液处理后,北美冬青植株内的乙烯、水杨酸、油菜素内酯等植物激素信号增强。综合以上分析结果,北美冬青可能通过代谢调节、物质能量准备及有害自由基的清除等途径提高处理植株应对干旱胁迫的能力,其中激素通路信号提高倍数最强的乙烯途径值得深入研究。 展开更多

Royle is a submerged plant for phytoremediation on Taihu Lake, China. The planted submerged plant should be harvested in every autumn to reduce the nutrition burden of the lake. Five compounds were firstly isolated by column chromatography and purified from the extractions of Hydrilla verticillata (Linn. f.) Royle. 1-(5'-Hydroxy-4'-hydroxymethyl-1'-methyl-1H-pyrrol-2'-yl)-henicosa-2,12,15-trien-1-one (2) and thymidine (3) were identified by EIMS, 1H NMR, laC NMR and IR spectra. Crystal loliolide (1) and sulfur-gamma (4) were identified by X-ray diffraction. Octadecanedioic acid (5) was also elucidated. Among them, compound 2 is a new natural compound, loliolide (1) and thymidine (3) are known bioactive substances. The results provided basic research data for exploring their application as medical materials. 展开更多

[Objective] The research aimed to study the influence of temperature on the growth of Hydrilla verticillata in the wastewater treatment process.[Method] By using the single factor experiment design,four temperatures which were 5,15,25 and 35 ℃ were set.The growth situation of H.verticillata under the different temperatures was observed in the treatment process of domestic sewerage.[Result] H.verticillata grew luxuriantly at 35 and 25 ℃.Via the cultivation of 15 days,the plant length,branch number,root number and biomass of H.verticillata respectively increased 2.8 and 1.2 cm,7.0 and 5.9 branches,6.2 and 3.7 roots,21% and 25%,which were all significantly or extremely significant higher than that at 5 ℃.Under the cultivation condition of 15 ℃,H.verticillata grew slowly.The increment of plant length,branch number and root number were higher than that at 5 ℃,but they had no significant difference.[Conclusion] During 5-35 ℃,the higher temperature could promote the growth of H.verticillata in wastewater treatment process. 展开更多

The acute toxicity of Sodium dodecylbenzene sulfonate (SDBS) to subcamersed macrophyte Hydrilla verticillata (L.f) Royle was studied. Chlorophyll contents and the activities of 3 antioxidant enzymes: superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), peroxidase (POD, EC 1.11.1.7) in the leaves were investigated under different concentrations of SDBS, The chlorophyll contents in leaves of experimental plants decreased gradually, while the electrical conductivity of culture solution increased gradually with the increased dose of SDBS. Under higher concentrations of SDBS (32 mg/L and 128 mg/L) treatment, SOD lost its activities completely after 24 h. The native PAGE analysis indicated the diversity of POD isoenzymes was obvious under different concentrations of SDBS stress. 展开更多

Cloning and Characterization of the Strictosidine-β-D-glucosidase (SGD) Gene from Rauvolfia verticillata

[Objective] This study aimed to clone Scrictosidine-β-D-glucosidase (SGD) gene from Rauvolfia verticillata and analyze its characteristics. [Method] The full-length cDNA of SGD was cloned from R. verticillata with RACE technique. Then the expression levels in different tissues were analyzed with quantitative RT-PCR and the bioinformatic characteristics were also predicted. [Result] The full-length cDNA of RvSGD was 1 856 bp, containing a 1 608 bp CDS that encoded 536 amino acids with a calculated molecular mass of 61.0 kDa and an isoelectric point of 6.16. Bioinformatic analysis revealed that RvSGD shared high similarity with SGDs from Cantharanthus roseus and Rauvolfia serpentina at the amino acids; three conserved catalytic sites His-161, Glu-207 and Glu-419 were also presented in RvSGD. Quantitative RT-PCR showed that expression level of RvSGD was the highest in barks, followed by old leaves, roots, tender leaves and tender stems. [Conclusion] The present study helps to understand more about the functions of the SGD gene at the level of molecular genetics, and provides new targets for molecular regulation of TIAs biosynthesis. 展开更多

Baill. var. hainanensis Tsiang extract on mice with colitis induced by Dextran sulphate sodium (DSS). Methods: 80 BALB/C mice were randomly divided into (1) normal control group, (2) DSS group, (3) DSS + SASP treatment group, (4) DSS + PP treatment group. After the experiment, the mice in each group were observed daily general conditions, Immunohistochemistry was used to detect the expression of ERK, p38 MAPK and JNK protein in colonic mucosa of mice in each group. The expression of TNF-_and IL-6 cytokines in colonic tissue of mice was detected by ELASA. Results: The general situation of SASP group and PP group was better than DSS group;Compared with DSS group, the expression of ERK, p38 MAPK and JNK protein was significantly decreased. The levels of TNF-a and IL-6 in colon tissue of SASP group and PP group were significantly decreased. Conclusion: The results suggest that pectin polysaccharide from Rauvolfia hainanensis extract has a significant therapeutic effect on colitis in DSS mice, which may via regulating the MAPK signal pathway pathways. 展开更多

Briq. is an Algerian endemic species; the phytochemical screening (methods of Harborne, 1973) of the aerials parts wealth in polyphenol compounds: flavonoids, saponins, leucoanthocyanes, terpens and steroids and tannins; there is no alkaloids. There is an important quantity of essential oils in the flowers; the interest of this study is that this species remains until then it is not very known. The results of Aromatogram method by incorporation of Mtiller-Hinton on solid medium, showed a significant antimicrobial activity (method of Duraffourd, 1987) of the infusion and the ethanolic extract; the infusion of stems and flowers is indeed much more active on Echerichia coli, Citrobacter, Enterobacter, Staph aureus and Staph epidermidis, such as flowers extracts demonstrate an important antimicrobial activity on Staph aureus, Staph epidermidis, Streptococcus, Pseudomonas and Acinetobacter recognized as antibiotic resistant. This could give opportunities for using this species in the treatment of diverse infections and as a disinfecting additive on nosocomial area. 研究了不同Cd、Cu、Zn处理浓度对黑藻体内活性氧()产生及对抗氧化酶(SOD、POD、CAT)活性的分子毒理学效应以探讨高等水生植物抗氧化酶对重金属胁迫的反应。结果表明,三种重金属都不同程度地加快了产生速率;Cu使SOD、POD、CAT活性下降;C... 研究了不同Cd、Cu、Zn处理浓度对黑藻体内活性氧()产生及对抗氧化酶(SOD、POD、CAT)活性的分子毒理学效应以探讨高等水生植物抗氧化酶对重金属胁迫的反应。结果表明,三种重金属都不同程度地加快了产生速率;Cu使SOD、POD、CAT活性下降;Cd也都减弱了SOD和POD活性,而CAT活性在0.5—5mg/L处理浓度时增加;Zn对SOD活性也为抑制作用,当浓度为0.5—5mg/L时POD和CAT活性都上升。关联度分析发现Cd、Cu和Zn胁迫下黑藻起主要保护作用的分别为SOD、POD和CAT,而SOD最易受到影响。Cd、Cu处理下的叶绿素含量也都呈下降趋势,而0.5—5mg/L的Zn浓度刺激了叶绿素合成。所有Zn处理、0.5mg/L的Cu处理和0.5—1mg/L的Cd处理的叶绿素a/b值都大于对照值。除了Cu使可溶性蛋白含量减少外,0.5—5mg/L的Zn和0.5—1mg/L的Cd都使其含量增加。综合起来,Cu的毒性最强,其次为Cd,Zn最弱。致死阈浓度分别为:Cu:0.5—1mg/L;Cd:1—2mg/L;Zn:5—6mg/L。SOD是评价重金属对沉水植物毒性效应的灵敏指标。黑藻对水环境Cu污染反应敏感。 展开更多

在实验室条件下,比较研究了不同浓度(0.5、1、2、4、8、16mg/L)的氨氮(NH4+-N)对沉水植物黑藻(Hydrilla verticillata)的生理生化影响,测定了黑藻生物量、叶绿素、可溶性糖、蛋白质含量和过氧化物酶(POD)、超氧化物歧化酶(SOD)、谷氨酰... 在实验室条件下,比较研究了不同浓度(0.5、1、2、4、8、16mg/L)的氨氮(NH4+-N)对沉水植物黑藻(Hydrilla verticillata)的生理生化影响,测定了黑藻生物量、叶绿素、可溶性糖、蛋白质含量和过氧化物酶(POD)、超氧化物歧化酶(SOD)、谷氨酰氨合成酶(GS)的活性变化。实验结果表明低浓度的氨氮(0.5、1、2mg/L)对轮叶黑藻的生长稍有促进作用,但培养液中氨氮浓度超过4mg/L时,黑藻的相对生长率(R)明显下降,当浓度达到16mg/L时,黑藻在20多天内全部死亡。在低浓度氨氮条件下,黑藻叶绿素和可溶性糖含量随氨氮浓度增加呈上升趋势,当培养液中氨氮浓度超过4mg/L时,叶绿素和可溶性糖含量在第24、32、40天取样时较对照组明显降低。低浓度氨氮处理组(0.5、1、2mg/L)的蛋白质含量先下降后又上升,而对照组的蛋白质含量一直在上升,高浓度氨氮处理组(8、16mg/L)的蛋白质含量则呈明显下降趋势。POD、SOD和GS活性变化趋势基本一致,在高浓度氨氮条件下增加显著,并表现为先上升再下降的格局,在第16天或第24天达到最大值。研究结果提示在富营养化条件下氨氮浓度的升高将影响其生理功能,过高浓度的氨氮对轮叶黑藻是一种逆境胁迫,可抑制其生长甚至导致植物死亡。轮叶黑藻对氨氮浓度变化虽有一定耐性,但耐性会随时间延长而变弱。 展开更多
