以海鞘(Ciona savignyi)为研究对象,考察了海鞘血细胞原代培养方法及培养中细菌污染的鉴定和控制。血细胞在培养1 h后贴壁,细胞基本以圆形细胞为主,变形细胞一般呈不规则形。变形细胞存活时间较短,而圆形细胞存活时间较长。对于培养过... 以海鞘(Ciona savignyi)为研究对象,考察了海鞘血细胞原代培养方法及培养中细菌污染的鉴定和控制。血细胞在培养1 h后贴壁,细胞基本以圆形细胞为主,变形细胞一般呈不规则形。变形细胞存活时间较短,而圆形细胞存活时间较长。对于培养过程中的细菌污染,通过细菌分离、培养和纯培养发现两类菌株检出率较高,均为革兰氏阴性菌。经PCR扩增16SrDNA基因序列片断,结果显示这两类菌株分别属于弧菌属(Vibrio)和施万氏菌属(Shewanella)。药敏试验结果显示,弧菌属对氯霉素和环丙沙星等具有较强的敏感性;对施万氏菌属较敏感的药物依次为亚胺培南和氯霉素等。最后比较了几种抗生素组合控制血细胞培养中细菌污染的使用效果,其中氯霉素、亚胺培南加双抗的组合有较好的抑菌效果并对培养细胞的贴壁和生长没有影响。 展开更多

Siphon-Specific Expression of an Actin Encoding Gene Is Regulated by Six1/2 in Ciona savignyi

Actin is a ubiquitous protein and plays essential roles on cellular structure maintenance and cellular motility in both muscle and non-muscle tissues.Multiple genes encoding muscle actin have been identified from the ascidians,including those expressed in the larval tail muscle,the adult body-wall muscle,and adult heart muscle.In this study,a novel striated non-tail muscle actin gene was identified from the RNA-seq data of Ciona savignyi embryos.Phylogenetic analysis,alignment of the N-terminal amino acid sequences and comparation of diagnostic residues provided evidence that it had high similarity with vertebrate cardiac and skeletal muscle actin.In situ hybridization and promoter-driven GFP reporter assay revealed that it was specifically expressed in the primordia of the oral and atrial siphon.We hereby defined it as siphon-specific muscle actin coding gene(Cs-SMA).A 201 bp(−1350 bp to−1150 bp)sequence containing T-box and Six1/2 binding motif within the upstream region of Cs-SMA confined the expression of GFP in the siphons of electroporated embryos.Six1/2 binding motif was experimentally confirmed to play indispensable role in controlling the siphon-specific expression of Cs-SMA.The tissue-specific expression of Cs-SMA in the siphon primordia indicated its potential crucial roles in Ciona embryogenesis and organogenesis. 展开更多

Changes in Tree Frog (<i>Hyla savignyi</i>) Coloration in Unstable Habitats at the Southern Border of Its Distribution

The results show that tree frog color is affected by the dominant colors in its habitat, which vary seasonally, especially between winter and summer. Tree frog colors were various shades of green, white, brown, and black. No genetic marker was found to characterize the color. The ability of a small frog to infer its own time with the help of color changes occurring in the habitat on the southern border of its distribution, which are relatively broad, gives this species an advantage.</span> 展开更多
