Cerebellar malfunction can lead to sleep disturbance such as excessive daytime sleepiness,suggesting that the cerebellum may be involved in regulating sleep and/or wakefulness.However,understanding the features of cer...Cerebellar malfunction can lead to sleep disturbance such as excessive daytime sleepiness,suggesting that the cerebellum may be involved in regulating sleep and/or wakefulness.However,understanding the features of cerebellar regulation in sleep and wakefulness states requires a detailed characterization of neuronal activity within this area.By performing multiple-unit recordings in mice,we showed that Purkinje cells(PCs)in the cerebellar cortex exhibited increased firing activity prior to the transition from sleep to wakefulness.Notably,the increased PC activity resulted from the inputs of lowfrequency non-PC units in the cerebellar cortex.Moreover,the increased PC activity was accompanied by decreased activity in neurons of the deep cerebellar nuclei at the nonrapid eye-movement sleep-wakefulness transition.Our results provide in vivo electrophysiological evidence that the cerebellum has the potential to actively regulate the sleep-wakefulness transition.展开更多
Objective To determined whether morphine preconditioning induces ischemic tolerance in neron.Methods We used rat cerebellar slices and simulated ischemia by oxygen-glucose deprivation(OGD),Slices were kept in artifica...Objective To determined whether morphine preconditioning induces ischemic tolerance in neron.Methods We used rat cerebellar slices and simulated ischemia by oxygen-glucose deprivation(OGD),Slices were kept in artifical cerebrospinal fluid(aCSF) containing various concentrations of morphine(0.1 μmol/L^1μmol/L),Then they were transferred to aCSF without glucose and oxygen.After 5 h,slices were fixed for morphology examination.To determine the percentage of undamaged Purkinje cell(survival rate).Purkinje cells were recognized based on the morphology and the location.At least 50 Purkinje cells were counted from each of the sections and cells in total three sections were counted for each slice(>150 Purkinje cells).Results The survival rate of Purkinje cells was significantly higher in group preconditioned with morphine(survival rates were 56%) than that of the oxygen-glucose deprivation(survival rates were39%).χ~2=293,P<0.01.The protective effects of morphine preconditioning were concentration dependent and were maximal at 0.3 μmol/L.Conclusion Morphine pretreatment can induces ischemic tolerance in neurons,but also the characteristic were the roof effects.展开更多
