Objective To explore the regulating effect of moxibustion on the enteric nervous system of rats with Crohn's disease. Methods Ten SD rats were selected randomly from 40 rats as normal control(group A), and the other 30 rats were established into Crohn's disease rat models by adopting clysis method with TNBS. On the basis of modeling successfully, the model rats were randomly divided into model group(group B), herbs-partitioned moxibustion group(group C) and mild moxibustion group(group D) with 8 rats in each group(4 rats were dead during modeling. After modeling, 2 rats were selected from group A, and 2 rats were selected from models for determination,at last, 8 rats were included in each group). In group C and group D, herbs-partitioned moxibustion or mild moxibustion was applied on "Tiānshū(天枢 ST 25)" bilaterally, and the rats in group A and group B were fixed as in treatment groups. HE stain was conducted, and morphological observation was performed on the colonic tissue of rats; the expression levels of proteins of S-100, SP, NPY and their receptors were observed by adopting immunohistochemical method. Results Compared with group A, the expression levels of proteins of S-100, SP and its receptor NK1R, NPY and its receptors NPY1R and NPY2R in the intestinal tract of rats in model groups increased obviously, and the differences were statistically significant.(P_(S-100)0.01, P_(SP)0.05, P_(NK1R)0.01, P_(NPY)0.05, P_(NPY1R)0.05, P_(NPY2R)0.01), after treatment with drug-paste interposed moxibustion and mild moxibustion, the levels reduced significantly(P_(S-100)0.05, P_(SP)0.05, P_(NK1R)0.01, P_(NPY)0.05, P_(NPY1R)0.05, P_(NPY2R)0.01). Conclusion Moxibustion treatment may regulate the expression levels of proteins of S-100, SP, NK1R, NPY, NPY1R and NPY2R through warm stimulation, alleviate inflammatory response of colonic tissue, and repair impaired colonic mucosa, thus achieving the goal of treating Crohn's disease. 展开更多

The orange-spotted grouper, Epinephelus coioides, is one of the most popular fish in China and Southeast Asian countries because of its important economic value. However, molecular mechanism underlying the growth of orange-spotted grouper has never been fully understood. Herein, we performed a genome-wide association study (GWAS) on a natural population of 198 individuals aiming to screen the whole genome of orange-spotted grouper for identification of growth-related loci by restriction-site associated DNA sequencing. In this research, 261,366 single nucleotide polymorphisms (SNPs) were developed, in which 110 SNPs were identified to be correlated with growth and 20 SNPs were further confirmed to be associated with both body weight and total length. From these identified SNPs, we annotated a total of 34 genes, including adgrb2, csnkzal, cers5, co122al, creb5, dndl, dzankl, dnail, npy2r, fat3, lrrk2, lrp5, map3k9, and so on. Among these candidate genes, npy2r (neuropeptide Y receptor Y2) was reported to play a critical role in growth of the orange-spotted grouper. In addition, population structure, principal component analysis, kinship matrix and linkage disequilibrium were examined to verify the accuracy and reliability of our GWAS results. Our data will also provide a valuable genetic resource for further marker-assisted selection program to improve growth quality in groupers. 展开更多

Noxious mechanical information is transmitted through molecularly distinct nociceptors,with pinprickevoked sharp sensitivity via A-fiber nociceptors marked by developmental expression of the neuropeptide Y receptor 2(Npy2 r)and von Frey filament-evoked punctate pressure information via unmyelinated C fiber nociceptors marked by MrgprD.However,the molecular programs controlling their development are only beginning to be understood.Here we demonstrate that Npy2 r-expressing sensory neurons are in fact divided into two groups,based on transient or persistent Npy2 r expression.Npy2 r-transient neurons are myelinated,likely including A-fiber nociceptors,whereas Npy2 r-persistent ones belong to unmyelinated pruriceptors that co-express Nppb.We then showed that the transcription factors NFIA and Runx1 are necessary for the development of Npy2 r-transient A-fiber nociceptors and MrgprD^+C-fiber nociceptors,respectively.Behaviorally,mice with conditional knockout of Nfia,but not Runx1 showed a marked attenuation of pinprick-evoked nocifensive responses.Our studies therefore identify a transcription factor controlling the development of myelinated nociceptors. 展开更多

目的克隆与肥胖相关的人神经肽Y受体Y2(NPY2R)基因,并鉴定该克隆基因序列的正确性。方法从人的脂肪组织中提取总RNA并进行反转录,以此为模板用PCR扩增NPY2R基因的eDNA,然后与pET28a+载体重组,筛选阳性克隆和DNA序列分析鉴定,测... 目的克隆与肥胖相关的人神经肽Y受体Y2(NPY2R)基因,并鉴定该克隆基因序列的正确性。方法从人的脂肪组织中提取总RNA并进行反转录,以此为模板用PCR扩增NPY2R基因的eDNA,然后与pET28a+载体重组,筛选阳性克隆和DNA序列分析鉴定,测序后与GeneBank中NPY2R基因进行同源性比较和序列分析。结果PCR扩增出一个1100bp左右的DNA片段,与载体重组和DNA序列分析鉴定,DNA序列分析显示克隆的DNA片段是人NPY2R基因,且所克隆的基因共编码381个氨基酸,分子量为4.2kD,与GeneBank中NPY2R基因序列同源性达100%。结论克隆的人NPY2R基因与GeneBank中NPY2R序列完全一致,为进一步应用分子生物学技术深入研究NPY2R与人体肥胖发生、发展及转化等相关作用机制及应用该基因进行其基因蛋白表达奠定了基础。 展开更多

目的:研究NPY2R基因5'调控区rs6851222位点与云南彝族原发性高血压发生的关系。方法:研究对象为400例彝族人群,原发性高血压患者和正常对照个体各200例。运用直接测序的方法,对NPY2R基因5'调控区rs6851222多态性进行检测,分析... 目的:研究NPY2R基因5'调控区rs6851222位点与云南彝族原发性高血压发生的关系。方法:研究对象为400例彝族人群,原发性高血压患者和正常对照个体各200例。运用直接测序的方法,对NPY2R基因5'调控区rs6851222多态性进行检测,分析其与云南彝族人群原发性高血压的关系。结果:本研究共检测到AA、AG和GG这3种基因型,并且rs6851222位点基因型分布在原发性高血压组和正常对照组中均符合HardyWeinberg平衡。rs6851222等位基因和基因型频率在原发性高血压组和对照组间的分布均具有显著性差异(P>0.05),回归分析结果显示rs6851222A等位基因携带者的患病风险显著升高(OR=1.77,95%CI 1.32~2.38,P=0.000 1)。结论:本研究结果提示NPY2R基因5'调控区rs6851222位点与云南彝族原发性高血压发生相关,该位点A等位基因可增加原发性高血压发病风险。 展开更多
