Antimicrobial activity of Sapindus mukorossi and Rheum emodi extracts against H pylori: In vitro and in vivo studies 被引量:47

METHODS: Powders of S. mukorossi and R. emodi were extracted successively with petroleum ether, benzen... AIM: To evaluate the antibacterial activity of Sapindus mukorossi (S. mukorossl) and Rheum emodi (R. emodl). METHODS: Powders of S. mukorossi and R. emodi were extracted successively with petroleum ether, benzene, chloroform and ethanol and were concentrated in vacuum. The disk diffusion method was used for in vitro studies and in vivo studies were performed on male Wister rats. Thirty resistant clinical isolates of H pylon, as determined by their antibiotic sensitivity patterns by E-test, along with two Gram +ve (S. aureus, B. subtilis) and two Gram -ve (E. coli, P. vugaris) organisms were screened for their susceptibility patterns against these extracts. RESULTS: In our screening, all 30 resistant isolates and the other four organisms (two Gram +ve S. aureus, B. subtilis and two Gram -ve, E. coli, P. vugaris) were sensitive to the test compounds. It was found that ethanol and chloroform extracts of S. mukorossi and ethanol and benzene extracts of R. emodi inhibited Hpylori at very low concentrations, In the in vitro study, the isolates showed a considerable zone of inhibition at very low concentrations (10 μg/mL) for both the extracts. In the in vivo study, the H pylori infection was cleared with minimal doses of extracts of S. mukorossi (2.5 mg/mL) and R. emodi (3.0 mg/mL) given orally for seven days. CONCLUSION: We can conclude from this study that the extracts of S. mukorossi and R. emodi inhibited the growth of pylori in vitro and, in in vivo studies, the H pylori infection cleared within seven days at very low concentrations. We also found that H pylori did not acquire resistance against these herbal extracts even after 10 consecutive passages. 展开更多

Objective:To investigate the antihyperglycemic and antihyperlipidemie properties of hydroalcoholic extract of fruits of Sapindus mukorossi Caerlen and its beneficial effect on haeniatological parameters with histopathological analysis in streptpzotocin induced diabetic rats.Methods:Sapindus mukorossi fruits extract(250 and 500 mg/kg body weight) and standard drug glybenclamide(0.5 mg/kg body weight) were administered to diabetic rats.Effect of extract on hyperglycemia,hyperlipidemia and hematological parameters was studied in diabetic rats.Histopathological changes in diabetic rat pancreas were also observed after extract and glybenclamide treatment.Results:Daily oral administration of Sapindus mukorossi fruits extract(250 and 500 mg/kg body weight) and glybenclamide for 20 days showed beneficial effects on blood glucose level(P【0.01) and lipid level.The extract has a favorable effect on the histopathological changes of the pancreas in streptozotocin induced diabetes.Conclusions: These findings reveal that the hydroalcoholic extract of Sapindus mukorossi fruits extract possesses antihyperglycemic and antihyperlipidemie properties.In addition.the extract can prevent various complications of diabetes and improve some haeniatological parameters. 展开更多

Aim: To observe the alterations in the biochemical and biophysical changes in the sperm membrane during sperm maturation in male rats treated with the water extract of the fruit pericarp of S. mukorossi. Methods: Adult male Sprague-Dawley rats were gavaged the aqueous extract of the fruit pericarp of S. mukorossi at a dose of 50 mg/kg/d for 45 days. On day 46, the sperm parameters were observed in different sections of the epididymis and the sperm superoxide dismutase and the lipid peroxidation was determined and compared with the controls. The testis and epididymis were routinely prepared for histological examination under the light microscope. Results: No significant differences in the sperm number and morphology were observed between the control and treated groups. However, a significant inhibition (P<0.05-0.01) of sperm motility in the caput, corpus and cauda regions of the epididymis was seen in the treated group. No significant histopathological changes were found in the testis and epididymis. The important finding was that in the treated animals, the spermatozoa showed an abnormal distribution of the superoxide dismutase activity, being minimum in the caput and maximum in the corpus, which was just opposite to that of the controls. Conclusion: The study provides a unique observation where the plant extract alters the sperm membrane physiology without change the testicular and epididymal morphology. 展开更多

This study explores the possible application of a biodegradable plant based surfactant, obtained from Sapindus mukorossi, for washing low levels of arsenic (As) from an iron (Fe) rich soil. Natural association of As(Ⅴ) with Fe(Ⅲ) makes the process difficult. Soapnut solution was compared to anionic surfactant sodium dodecyl sulfate (SDS) in down-flow and a newly introduced suction mode for soil column washing. It was observed that soapnut attained up to 86% efficiency with respect to SDS in removing As. Full factorial design of experiment revealed a very good fit of data. The suction mode generated up to 83 kPa pressure inside column whilst down-flow mode generated a much higher pressure of 214 kPa, thus making the suction mode more efficient. Micellar solubilisation was found to be responsible for As desorption from the soil and it followed 1st order kinetics. Desorption rate coefficient of suction mode was found to be in the range of 0.005 to 0.01, much higher than down-flow mode values. Analysis of the PT-IR data suggested that the soapnut solution did not interact chemically with As, offering an option for reusing the surfactant. Soapnut can be considered as a soil washing agent for removing As even from soil with high Fe content. 展开更多

The study has been carried out to investigate the effects of single factors such as solvents extraction temperature, times, solid-liquid ration, and the time of extractions on the yields of saponins present in the pulp of Sapindus mukorossi Gaerth. On this basis, an L9 orthogonal design of experiment was adopted to determine the optimal conditions for the extraction of saponins. The factors that influence the extraction of saponins are put in the order of extraction times, extraction time, solid-liquid ration, and the best combination is that the powder of the pulp is extracted with EtOH (solid-to-solvent ratio = 1:8, w/v) for three times at 60°C for 3 hours. Under these conditions, about 1.63 g saponins will be extracted from 10 g raw material. The stability test showed that the Sapindus mukuross saponins can maintain surface activity at water temperature (25°C - 40°C), pH (6.3 - 7.7) and water hardness (50 - 250 mg.L-1). It is proved that Sapindus mukuross saponins are quality non-ionic active agent. 展开更多

To explore differences in leaf morphology between Sapindus mukorossi and Sapindus delavayi,and how the environment might drive these differences,80 germplasm samples from the Sapindus germplasm nursery in Fujian Province were selected.The study revealed a wide variation and diversity in 16 germplasm traits,both within and between species grown under the same conditions.On average,the relative contribution of intraspecific variability to total variability was more important(83%) than the relative contribution of interspecific variability(17%).PERMANOVA analysis showed differences in leaflet thickness,length,perimeter,length to width ratio,and leaf hairs or trichome density.Correlation analyses between leaf morphological traits and environmental variables indicated that leaves tended to be larger,longer,and thicker in wetter,warmer,and low-altitude conditions.Our analysis of the relationship between climate and leaf morphology revealed that S.mukorossi had a greater sensitivity to climate variation,particularly in response to mean temperatures of the coldest and warmest seasons,which led to differences in leaf traits and the distribution of the two species.These findings contribute to the understanding of leaf morphology variations in S.mukorossi and S.delavayi,and provide a basis for the collection of Sapindus germplasm resources,their cultivation and use to help address climate change. 展开更多

[Objectives]To investigate the antimicrobial activity and mechanism for saponin from Sapindus mukorossi against Escherichia coli.[Methods]E.coli was used as the test bacteria,and the antimicrobial effect of Sapindus saponin was studied by the minimum inhibitory concentration method.The antimicrobial mechanism of Sapindus saponin was studied in terms of growth curves,membrane potential,activity of cells,cell surface morphology and cell internal structure.[Results]The results indicate that Sapindus saponin can inhibit the growth of E.coli,the minimum inhibitory concentration is 5 mg/mL and has a significant inhibitory effect on the growth of E.coli.After treated with Sapindus saponin,the membrane potential of E.coli increased by 16.7,17.3 and 19.1 times after exposition to 0.5,1,2 MIC Sapindus saponin respectively.PI staining results show that cell viability decreased and permeability of cell membranes increased.The results of SEM and TEM further corroborate the membrane damage,the cell was damaged after exposition to Sapindus saponin.[Conclusions]The antimicrobial mechanism of Sapindus saponin were to changed membrane potential of cell,and damage the cell membrane structures. 展开更多

The results showed that the rooting rate of cuttings of the ten clones ranged from 30.9% to 83.0%. Among them, the rooting rate of cuttings of clones SM01 and SM06 was higher than that of other clones, and there were significant differences between different clones in the rooting rate of cuttings. The rooting rate of the semi-lignified shoots having two leaves could be improved significantly. The treatment of the cuttings by GGR6 solutions had significant effects on their rooting rate. 展开更多

Human pulp tissue dissolution ability of different extracts of Sapindus mukorossi: An in vitro study

Objective:Due to the many negative properties of sodium hypochlorite used in current root canal treatment,interest in biocompatible natural agents is increasing day by day.The aim of this study was to evaluate whether various extract solutions of Sapindus mukorossi have dissolution effects on human pulp tissues.Methods:Primarily powder extracts were obtained by extracting fruit shells of S.mukorossi in different solvents(ethanol,methanol,buthanol and distilled water).The test solutions were prepared and randomly separated into six groups with 10 samples in each group:ethanol extract,methanol extract,butanol extract,distilled water extract of S.mukorossi,sodium hypochlorite(Na OCl)and the control group.Among these,S.mukorossi extracts were separated into two subgroups,depending on their concentration level(50μg/m L and 100μg/m L).The pulp tissues of freshly extracted human molars were used for dissolution test.The weights of the pulpal tissues were measured and recorded for two times after the samples were placed in the solutions.Statistical analysis for all descriptive statistics was performed using SPSS 22(P<0.05).Results:Our results showed that maximum percent yield of preparation was obtained in methanol extract of S.mukorossi.Among all of the groups,the best dissolution capacity was seen in the Na OCl group(positive control group).Among S.mukorossi groups,the best tissue solvent solution was found in SMM group at 50μg/m L and SMB group at 100μg/m L.Conclusion:The different extracts of S.mukorossi had a capacity to dissolve pulp tissue but this capacity was less than Na OCl.Therefore,further studies will enable the creation of a commercial solution for clinical use by increasing the effectiveness of S.mukorossi while combining it with other endodontic irrigation solutions. 展开更多

无患子(Sapindus mukorossi Gaertn.)是我国长江以南地区传统的重要绿化树种,其果皮富含皂苷,种仁富含油脂,是国家林业局审定的新型木本油料树种之一。为揭示无患子群体间和群体内种实表型性状变异式样,采用单因素方差分析、巢式方差分... 无患子(Sapindus mukorossi Gaertn.)是我国长江以南地区传统的重要绿化树种,其果皮富含皂苷,种仁富含油脂,是国家林业局审定的新型木本油料树种之一。为揭示无患子群体间和群体内种实表型性状变异式样,采用单因素方差分析、巢式方差分析、相关分析和主成分分析等方法,对无患子13个天然群体的10个种实表型性状进行比较分析,研究其群体间和群体内种实表型多样性以及表型变异与地理生态因子间的相关性。结果表明:无患子种实表型性状变异系数平均为7.34%,在群体间和群体内均存在丰富的表型变异。表型性状分化系数平均为62.21%,群体间变异(39.93%)大于群体内(27.46%),是无患子种实表型性状变异的主要来源。多数性状在群体间差异显著,不同程度的表现出边缘群体易于分化的特点,但地理变化规律不连续,而在群体内不同性状的差异性亦不一致。种子形态受群体地理生态的影响较果实形态大,西北部群体种子趋于椭圆形,东部、南部则趋于圆球形;地理纬度、年平均气温与多数种实性状间呈显著相关,是无患子种实表型性状变异的主要地理生态影响因子。可见,无患子种实表型性状在群体间、群体内变异都较为丰富,这些变异是由遗传和环境因素共同作用的结果。群体间和群体内多层次的变异为无患子优良种质资源保育和利用提供了物质基础。 展开更多

对我国无患子分布区7个产地的种实形态性状及种仁含油率进行取样测定。方差分析结果表明,种实各性状在产地内单株间的变异程度不同,但多数性状在多数产地内均存在丰富的变异,在产地内进行单株选择是可行的。在不同产地间,无患子种实各... 对我国无患子分布区7个产地的种实形态性状及种仁含油率进行取样测定。方差分析结果表明,种实各性状在产地内单株间的变异程度不同,但多数性状在多数产地内均存在丰富的变异,在产地内进行单株选择是可行的。在不同产地间,无患子种实各性状均存在显著差异,河南西峡产地果实和种子较大,与其它产地间差异显著。无患子种仁含油率产地间变异系数最小,与地理气候因子间不存在明显的相关性,而种子质量、种子出仁率在不同产地间变异系数相对较大,且与种实大小显著正相关,可以通过选择较大的果实、种子来提高产油量。聚类及相关性分析表明,在一定程度上,无患子种实大小和种子出仁率有从低纬度产地向高纬度产地变大的变异趋势,因此无患子高产油单株的选择应重点在较高纬度产地果实、种子较大的个体中进行。 展开更多

研究了无患子油脂的提取条件、理化性质和以该油制备生物柴油的工艺。结果表明:油脂提取最佳条件:石油醚体积与种籽质量比5∶1,提取时间为2 h,提取温度为70℃,无患子得油率为42.0%;无患子油脂主要由棕榈酸、硬脂酸、油酸、亚油酸、亚麻... 研究了无患子油脂的提取条件、理化性质和以该油制备生物柴油的工艺。结果表明:油脂提取最佳条件:石油醚体积与种籽质量比5∶1,提取时间为2 h,提取温度为70℃,无患子得油率为42.0%;无患子油脂主要由棕榈酸、硬脂酸、油酸、亚油酸、亚麻酸、花生酸、二十碳烯酸、山嵛酸、芥子酸等脂肪酸组成,其中不饱和脂肪酸质量分数91.85%,其主要理化性质为酸值:4.1 mg KOH/g、皂化值:184.83 mg KOH/g、碘值:110.16 gI2/100 g、折光率:1.480 8,脂肪酸组成、质量分数及理化性质等符合生物柴油标准。应用L9(34)正交试验得出无患子油酯交换反应制备生物柴油的最佳条件为:油醇物质的量比1∶6、催化剂用量为油质量的1.2%、反应时间2 h、反应温度60℃,转化率为93.6%。并对无患子生物柴油性能进行了检测,它与0#柴油、国标GB/T 20828—2007《柴油机燃料调和用生物柴油》的主要性能指标相接近,它是一种理想的0#柴油的替代品。 展开更多
