当归四逆汤对BALB/c硬皮病小鼠皮肤组织中CTGF,TGF-β含量的影响 被引量:19

目的:观察当归四逆汤对BALB/c硬皮病小鼠皮肤组织及其结缔组织生长因子(connective tissue growth factor,CTGF)、转化生长因子(transforming growth factor,TGF)β含量的影响。方法:选8周龄近交系BALB/c小鼠60只,剃去背部中央区被毛,... 目的:观察当归四逆汤对BALB/c硬皮病小鼠皮肤组织及其结缔组织生长因子(connective tissue growth factor,CTGF)、转化生长因子(transforming growth factor,TGF)β含量的影响。方法:选8周龄近交系BALB/c小鼠60只,剃去背部中央区被毛,随机分为5组,每组12只。模型组和观察组小鼠背部皮内注射100μL(200 mg·L-1)博来霉素(BLM)溶液,每天1次,连续3周。正常组小鼠用磷酸盐缓冲液(PBS)0.1 mL做背部皮内注射。造模成功后,各组按20 mL·kg-1容量灌胃,连续3周。当归四逆汤高、中、低剂量组(39,23.4,7.8 g·kg-1)。正常组和模型组给等容量生理盐水。实验结束处死动物,切取小鼠背部注射区皮组织,部分剪碎后与生理盐水研磨成2%组织浆,3 000 r·min离心10 min,取部分上清液,用ELISA法测定CTGF,TGF-β的含量。部分组织固定于10%甲醛液,石蜡包埋,苏木素2伊红(HE)染色和Masson三色染色,分别用彩色病理图像分析系统和细胞免疫组织化学定量分析系统测定皮肤厚度和纤维化指数。结果:当归四逆汤高、中、低剂量均能减轻BLM致硬皮病小鼠的真皮厚度高、中剂量组分别为(25.22±2.35),(29.13±2.03)μm,P<0.01;低剂量组为(29.95±2.85)μm,P<0.05);高、中剂量组能降低硬皮病小鼠皮肤纤维化指数,分别为(86 615±8 403),(103 174±18 439),P<0.01;皮肤组织中CTGF分别为(160.2±35.7),(109.5±28.6)ng·L-1,P<0.01,TGF-β的含量分别为(48.48±4.95),(63.30±7.32)ng·L-1,P<0.01),而且存在明显的量效关系。结论:当归四逆汤能使模型小鼠的皮肤硬化得到改善,对其皮肤组织中的CTGF,TGF-β含量有降低作用。 展开更多

UVA1 irradiation inhibits fibroblast proliferation and alleviates pathological changes of scleroderma in a mouse model 被引量:2

The purpose of the present study was to compare the effects of different doses of ultraviolet radiation A1 (UVA1) on human fibroblast proliferation and collagen level in a mouse model of scleroderma, so as to identify appropriate irradiation doses for clinical treatment of scleroderma. Monolayer from human fibroblasts was cultured in vitro, and a mouse model of scleroderma was established by subcutaneous injection of 100 μL of 400 μg/mL bleomycin into the back of BALB/c mice for 4 weeks. The mouse models and human fibroblasts were divided into UVA1- exposed (100, 60 and 20 J/cm2) and UVA-unexposed groups. At 0, 24 and 48 h after exposure, cell proliferation and levels of hydroxyproline and collagen were detected. UVA1 irradiation was performed 3 times weekly for 10 weeks, and the pathological changes of skin tissues, skin thickness and collagen level were observed after phototherapy. Cell proliferation and the levels of hydroxyproline and collagen were inhibited after phototherapy, and there was a significant difference between the UVAl-exposed cells and UVAl-unexposed cells (P 〈 0.001). In addition, UVA1 phototherapy improved dermal sclerosis and softened the skin, and there were significant differences between the high-dose UVA1 group and the model group, and the negative group (P 〈 0.05). It is concluded that UVA1 radiation can reduce cell proliferation, and decrease hydroxyproline and collagen levels in a dose-dependent manner in vitro. High-dose UVA1 phototherapy has marked therapeutic effect on scleroderma in the mouse model. Decreased collagen level may be related to the reduced number and activity of cells, as well as inhibition of collagen synthesis. 展开更多

目的:建立硬皮病小鼠模型,探讨抗硬化复方抗博莱霉素所致小鼠皮肤硬变的作用机理。方法:制作硬皮小鼠动物模型:博莱霉素(BLM)200μg/ml每日于C3H小鼠背部(剃毛后)皮下注射,连续3周。药物作用机理研究:运用免疫组织化学法测定硬皮小鼠模... 目的:建立硬皮病小鼠模型,探讨抗硬化复方抗博莱霉素所致小鼠皮肤硬变的作用机理。方法:制作硬皮小鼠动物模型:博莱霉素(BLM)200μg/ml每日于C3H小鼠背部(剃毛后)皮下注射,连续3周。药物作用机理研究:运用免疫组织化学法测定硬皮小鼠模型皮肤中Ⅰ、Ⅲ型胶原和转化生长因子-β1(TGF-β1)等的表达,观察抗硬化复方对硬皮小鼠皮肤Ⅰ、Ⅲ型胶原和TGF-β1表达的影响。53只C3H鼠分成6组。①空白对照组;②造模对照组;③早服中药组,造模同时服用中药;④中药组,造模成功后服中药;⑤西药组,造模成功后服青霉胺;⑥中西药合用组。比较研究上述指标在各组中的变化,采用SPSS11.0统计软件进行分析。结果:抗硬化复方能对抗博莱霉素所致的小鼠皮肤硬变,早服中药组、中药组、西药组及中、西药组小鼠皮肤COL-Ⅰ、COL-Ⅲ免疫组织化学指数均明显低于造模对照组小鼠,差异有非常显著意义(P<0.01)。早服中药组及中西药合用组效果尤佳。早服中药组、中药组及中、西药合用组小鼠皮肤TGF-β1、COL-Ⅲ免疫组织化学指数又明显低于西药组小鼠,差异有非常显著意义(P<0.01)。结论:抗硬化复方能有效抑制硬皮小鼠皮肤中Ⅰ型胶原、Ⅲ胶原的合成,有抗组织纤维化作用。 展开更多
