Without any apparent extracellular domain, RLCKs relay intracellular signaling often via RLK complex-mediated transphosphorylation events. Recent advances have suggested essential roles of diverse RLCKs in concert with RLKs in regulating various cellular and physiological responses. We summarize here the complex roles of RLCKs in mediating plant immune responses and growth regulation, and discuss specific and overlapping functions of RLCKs in transducing diverse signaling pathways.展开更多

Hence, assembly factors are essential for chloroplast development and the transition from heterotrophic to phototrophic growth. Snowy cotyledon 2 (SCO2) is a DNAJ-like protein involved in thylakoid membrane biogenesis and interacts with the light-harvesting chlorophyll-binding protein LHCBI. In Arabidopsis thaliana, SCO2 function was previ- ously reported to be restricted to cotyledons. Here we show that disruption of SC02 in Lotus japonicus results not only in paler cotyledons but also in variegated true leaves. Furthermore, smaller and pale- green true leaves can also be observed in A. thaliana sco2 (atsco2) mutants under short-day conditions. In both species, SCO2 is required for proper accumulation of PSlI-LHCll complexes. In contrast to other variegated mutants, inhibition of chloroplastic translation strongly affects L. japonicus sco2 mutant devel- opment and fails to suppress their variegated phenotype. Moreover, inactivation of the suppressor of variegation AtClpR1 in the atsco2 background results in an additive double-mutant phenotype with variegated true leaves. Taken together, our results indicate that SCO2 plays a distinct role in PSll assembly or repair and constitutes a novel factor involved in leaf variegation.展开更多

To explore the relationship between Insulin-like growth factor (IGF)- Ⅰ, -Ⅱ and lung development in neonatal rats. 80 timed pregnant Sprague-Dawley ( SD) rats were randomly divided into 4 groups (n=20): group A (Con...To explore the relationship between Insulin-like growth factor (IGF)- Ⅰ, -Ⅱ and lung development in neonatal rats. 80 timed pregnant Sprague-Dawley ( SD) rats were randomly divided into 4 groups (n=20): group A (Control group), group B (Dexamethasone (DEX) 1 group), group C (DEX 2 group), group D (retinoic acid (RA) group). 20 pregnant rats in group A, B and D were injected subcutaneously or intraperitoneally with vehicle (NS), DEX, or RA respectively during gestational day 16 to 18. All newborn rats in group C were subcutaneously injected with DEX at day 1 to 3 after birth. The lung tissue was obtained at the following times: fetuses at gestational ages of 18, 20 and 21 days, and 1, 3, 5, 7, 10, 14 and 21 days after birth. Lung tissues were used for histopathological study, the polypeptides analysis of IGF-Ⅰ, -Ⅱ (immunohistochemistry and Western blot) and mRNA analysis ( RT- PCR). The results showed that the strongest expression of IGF-Ⅰ in group A and D occurred at ages of 5-7 days (alveolar stage). The stronger their expressions, the better the alveolar develop. The peak stage of expression in group B occurred earlier, on the day 3 after birth. Compared with group A, the expression of IGF-Ⅰ during gestation age of 18 days to age of 3 days in group B were significantly higher (P<0.01), but significantly lower at other time points (P<0.01). The expression of IGF-Ⅰ was lower in group C all the time and always higher in group D than those in group A (P<0.01). The peak expression of IGF-Ⅱ took place at the gestation age of 18 days, then gradually dropped to trace. During 18 days of gestation to age of 3 days, the expression of IGF-Ⅱ in group B was significantly higher than that in group A (P<0.01). No difference was found among all other groups. The change in the expression of IGF-Ⅰ, -Ⅱ mRNA in all 4 groups was similar to that of their polypeptides. The growing variety of RNA classes,such as mRNAs,lncRNAs,and circRNAs,plays pivotal roles in both developmental processes and various pathophysiological conditions.Nonetheless,our comprehension of RNA functions in liv...The growing variety of RNA classes,such as mRNAs,lncRNAs,and circRNAs,plays pivotal roles in both developmental processes and various pathophysiological conditions.Nonetheless,our comprehension of RNA functions in live organisms remains limited due to the absence of durable and effective strategies for directly influencing RNA levels.In this study,we combined the CRISPR-RfxCas13d system with spermlike stem cell-mediated semi-cloning techniques,which enabled the suppressed expression of different RNA species.This approach was employed to interfere with the expression of three types of RNA molecules:Sfmbt2 mRNA,Fendrr lncRNA,and circMan1a2(2,3,4,5,6).The results confirmed the critical roles of these RNAs in embryonic development,as their loss led to observable phenotypes,including embryonic lethality,delayed embryonic development,and embryo resorption.In summary,our methodology offers a potent toolkit for silencing specific RNA targets in living organisms without introducing genetic alterations.展开更多
Neuropeptides are widely distributed in vertebrates and invertebrates,regulating a variety of physiological activities in the organisms,such as metabolism,feeding and reproduction.In this study,to explore the function...Neuropeptides are widely distributed in vertebrates and invertebrates,regulating a variety of physiological activities in the organisms,such as metabolism,feeding and reproduction.In this study,to explore the function of neuropeptide LFRFamide in Sepia pharaonis,the full-length cDNA of LFRFamide-like gene(named SpLFRFL,MG869822.1)was identified with rapid amplification of cDNA ends(RACE)method.The sequence of SpLFRFL was 860 bp in length and encoded 188 amino acids containing 4 different mature peptides:1 copy of PHTPFRFamide,NSLFRFamide,TIFRFamide,and 3 copies of GNLFRFamide.Multiple alignment and phylogenetic analysis results showed that SpLFRFL shared high identity with LFRFamides of Sepia officinalis and Sepiella japonica and had the closest relationship with them.Through quantitative Real-time PCR(qRT-PCR),it was found that the SpLFRFL gene was highly expressed in the optic lobe and brain at three different stages during gonad development in both genders.Moreover,the four mature peptides at a concentration of 0.01μmol L^(−1) could inhibit the protein synthesis in the Chinese hamster ovary cell strain-K1(CHOK1)induced by SpGnRH.These data suggest that SpLFRFL might be involved in the development and reproduction of S.pharaonis.The results can contribute to future studies on neuropeptide evolution and function and benefit the cuttlefish farming.展开更多

The ovary generally undergoes tissue remodeling during larval to pupal transition,which includes membrane degeneration and ovariole growth.At the same time,the hormones produced by insects significantly change during ...The ovary generally undergoes tissue remodeling during larval to pupal transition,which includes membrane degeneration and ovariole growth.At the same time,the hormones produced by insects significantly change during metamorphosis.However,the regulatory mechanism for ovarian development and hormones is not fully understood in insects.Herein,we found that matrix metalloproteinase 2(MMP2)was highly expressed in the ovarian capsules and ovarioles,and the development was abnormal after knocking out MMP2 in Bombyx mori.The process of abnormal degradation of collagen I due to MMP2 deletion,which resulted in abnormal development of ovarioles and eggs,was analyzed in detail.The proteomics of ovaries in the MMP2-knock out and wild type strains showed a critically significant difference in the expression of a protein,insulin-like peptide(ILP).Additional analysis revealed significant alteration of ILP during ovarian development,and abnormal expression of ILP significantly affected ovarian development in vivo and MMP2 expression in vitro and in vivo.These results showed that MMP2 regulation of ovarian tissue remodeling is closely related to ILP expression.Our study provides new insights into the regulatory mechanism of MMP2 and ovarian development in B.mori.展开更多

Reproduction is a crucial process in the life span of flowering plants,and directly affects human basic requirements in agriculture,such as grain yield and quality.Typical receptor-like protein kinases(RLKs)are a larg...Reproduction is a crucial process in the life span of flowering plants,and directly affects human basic requirements in agriculture,such as grain yield and quality.Typical receptor-like protein kinases(RLKs)are a large family of membrane proteins sensing extracellular signals to regulate plant growth,development,and stress responses.In Arabidopsis thaliana and other plant species,RLK-mediated signaling pathways play essential roles in regulating the reproductive process by sensing different ligand signals.Molecular understanding of the reproductive process is vital from the perspective of controlling male and female fertility.Here,we summarize the roles of RLKs during plant reproduction at the genetic and molecular levels,including RLK-mediated floral organ development,ovule and anther development,and embryogenesis.In addition,the possible molecular regulatory patterns of those RLKs with unrevealed mechanisms during reproductive development are discussed.We also point out the thought-provoking questions raised by the research on these plant RLKs during reproduction for future investigation.展开更多

Grape pistil has an important influence on fruit size and quality.However,there were few studies on grape ovary,and the development process of the ovary is still unclear.Therefore,in this paper,four different grape va...Grape pistil has an important influence on fruit size and quality.However,there were few studies on grape ovary,and the development process of the ovary is still unclear.Therefore,in this paper,four different grape varieties with different lengths of small inflorescences,namely‘Musct Hambourg’grape(Vitis vinifera),‘Concord’grape(Vitis labrusca),‘ShanPuTao’grape(Vitis amurensis)and‘GongNiang2Hao’grape(Vitis amurensis×Vitis vinifera)were used as test materials.Four varieties ovary were significant differences by means of stereomicroscope,paraffin section.The expression of ovary determining gene VvAGAMOUS(VvAG)and its development related genes VvCRABS CLAW(VvCRC)and VvAGAMOUS-LIKE 11(VvAGL11)with similar functions during the development of different grape varieties were preliminarily explored using fluorescence quantitative test.The relationship between VvAG and VvCRC,VvAG and VvAGL11 were analyzed using Y1H assay.Our results showed that there were obvious abdominal sutures on the surface of expect for‘Musct Hambourg’grape,and existing poly carpels.The ovary development of‘ShanPuTao’and‘GongNiang2Hao’grape was completed when the inflorescence length was less than 1 cm,while the‘Concord’and‘Musct Hambourg’grape were fully developed when the length of inflorescence was 3–4 and 4–5 cm,respectively.VvAG and VvCRC began to express in large quantities after the formation of stamen primordia,while VvAGL11 during the forming of ovule primordia.Therefore,VvAG and VvCRC mainly regulated the development of stamens and carpels and also promote the development of ovules,while VvAGL11 major regulated the development of ovules.The promoters of VvCRC and VvAGL11 were bound by VvAG.This study provides an important theoretical basis for further research on the molecular mechanism of grape ovary development.展开更多

文摘Human stem cells are scalable cell populations capable of cellular differentiation.This makes them a very attractive in vitro cellular resource and in theory provides unlimited amounts of primary cells.Such an approach has the potential to improve our understanding of human biology and treating disease.In the future it may be possible to deploy novel stem cell-based approaches to treat human liver diseases.In recent years,eff icient hepatic differentiation from human stem cells has been achieved by several research groups including our own.In this review we provide an overview of the f ield and discuss the future potential and limitations of stem cell technology.

文摘Reproduction is a crucial process in the life span of flowering plants,and directly affects human basic requirements in agriculture,such as grain yield and quality.Typical receptor-like protein kinases(RLKs)are a large family of membrane proteins sensing extracellular signals to regulate plant growth,development,and stress responses.In Arabidopsis thaliana and other plant species,RLK-mediated signaling pathways play essential roles in regulating the reproductive process by sensing different ligand signals.Molecular understanding of the reproductive process is vital from the perspective of controlling male and female fertility.Here,we summarize the roles of RLKs during plant reproduction at the genetic and molecular levels,including RLK-mediated floral organ development,ovule and anther development,and embryogenesis.In addition,the possible molecular regulatory patterns of those RLKs with unrevealed mechanisms during reproductive development are discussed.We also point out the thought-provoking questions raised by the research on these plant RLKs during reproduction for future investigation.
文摘Polo-like kinase 1(Plk1),a well-characterized member of serine/threonine kinases Plk family,has been shown to play pivotal roles in mitosis and cytokinesis in eu-karyotic cells.Recent studies suggest that Plk1 not only controls the process of mitosis and cytokinesis,but also,going beyond those previously described functions,plays critical roles in DNA replication and Pten null prostate cancer initiation.In this review,we briefly summarize the functions of Plk1 in mitosis and cytokinesis,and then mainly focus on newly discov-ered functions of Plk1 in DNA replication and in Pten-null prostate cancer initiation.Furthermore,we briefly introduce the architectures of human and mouse pros-tate glands and the possible roles of Plk1 in human prostate cancer development.And finally,the newly chemotherapeutic development of small-molecule Plk1 inhibitors to target Plk1 in cancer treatment and their translational studies are also briefly reviewed.

文摘Cell-to-cell and cell-to-environment communications are critical to the growth and development of plants.Cell surface-localized receptor-like kinases(RLKs)are mainly involved in sensing various extracellular signals to initiate their corresponding cellular responses.As important vegetative organs for higher plants to adapt to a terrestrial living situation,roots play a critical role for the survival of plants.It has been demonstrated that RLKs control many biological processes during root growth and development.In this review,we summarize several key regulatory processes during Arabidopsis root developm ent in which RLKs play critical roles.We also put forward a number of relevant questions that are required to be explored in future studies.

文摘Grape pistil has an important influence on fruit size and quality.However,there were few studies on grape ovary,and the development process of the ovary is still unclear.Therefore,in this paper,four different grape varieties with different lengths of small inflorescences,namely‘Musct Hambourg’grape(Vitis vinifera),‘Concord’grape(Vitis labrusca),‘ShanPuTao’grape(Vitis amurensis)and‘GongNiang2Hao’grape(Vitis amurensis×Vitis vinifera)were used as test materials.Four varieties ovary were significant differences by means of stereomicroscope,paraffin section.The expression of ovary determining gene VvAGAMOUS(VvAG)and its development related genes VvCRABS CLAW(VvCRC)and VvAGAMOUS-LIKE 11(VvAGL11)with similar functions during the development of different grape varieties were preliminarily explored using fluorescence quantitative test.The relationship between VvAG and VvCRC,VvAG and VvAGL11 were analyzed using Y1H assay.Our results showed that there were obvious abdominal sutures on the surface of expect for‘Musct Hambourg’grape,and existing poly carpels.The ovary development of‘ShanPuTao’and‘GongNiang2Hao’grape was completed when the inflorescence length was less than 1 cm,while the‘Concord’and‘Musct Hambourg’grape were fully developed when the length of inflorescence was 3–4 and 4–5 cm,respectively.VvAG and VvCRC began to express in large quantities after the formation of stamen primordia,while VvAGL11 during the forming of ovule primordia.Therefore,VvAG and VvCRC mainly regulated the development of stamens and carpels and also promote the development of ovules,while VvAGL11 major regulated the development of ovules.The promoters of VvCRC and VvAGL11 were bound by VvAG.This study provides an important theoretical basis for further research on the molecular mechanism of grape ovary development.
