Results show that RGM significantly decreased after 40 days at 21 ℃ and markedly decreased over the whole experiment period at 28℃; however, no significant effect of duration was observed at 7 or 14℃. At 14℃, trypsin activity significantly decreased over 10 and 20 days, then increased; amylase and trypsin activity significantly decreased after 40 days at 28℃. However, no significant effect of duration was found on amylase, pepsin or trypsin activities in the other temperature treatment groups. At 28℃, lipase activity peaked in 20 days and then markedly decreased to a minimum at the end of the experiment. On the other hand, pepsin activity at 28℃ continuously increased over the whole experimental period. Principle component analysis showed that sea cucumbers on day 40 in the 21℃ group and in the previous 20 days in the 28℃ group were in the prophase of aestivation. At 28℃, sea cucumbers aestivated at 30-40 days after the start of the experiment. It is concluded that the effect of temperature on the digestion ofA. japonicus is comparatively weak within a specific range of water temperatures and aestivation behavior is accompanied by significant changes in RGM and digestive enzyme activities.展开更多
On the basis of the data of oceanographic survey in the East China Sea in four seasons during 1997-2000 (23°30'~33°00'N, 118°30'-128°E), the variation of total biomass and diet biomass ...On the basis of the data of oceanographic survey in the East China Sea in four seasons during 1997-2000 (23°30'~33°00'N, 118°30'-128°E), the variation of total biomass and diet biomass of zooplankton and their spatial-temporal distribution and relationship with the fishing ground of Engraulis japonicus are approached and analyzed. The results show that the average biomass is 65.32 mg/m3 in four seasons, autumn (86.18 mg/m3) being greater than summer (69.18 mg/m3) greater than spring (55.67 mg/m3) greater than winter (50.33 mg/m3). The average value of diet zooplankton biomass is 40.9 mg/m3. The trends of horizontal distribution both in the total biomass and the diet biomass of zooplankton are similar. The high biomass region (250-500 mg/m3) is very limited, only accounting for 1% of the investigation area. Seasonal variation of the biomass is very remarkable in the west and north parts of East China Sea coastal waters ( 29°30'N,125°E). The horizontal distribution of diet zooplankton depends on the abundance distribution of crustacean. The distribution of diet zooplankton is related to the fishing ground of Engraulis japonicus and the high-density area of young fish and larval. In spring, the central fishing ground of Engraulis japonicus (>100 kg/h) and the high-density area of young fish and larval (>100 individuals per net) are located at the same place of high-density (100-250 mg/m3)area of diet zooplankton in the middle-southern part of East China Sea or the edge of its waters.展开更多

MAb AB10 was derived from an immunogen in which human serum albumin (HSA) was linked to G...The production and characterization of a monoclonal antibody (MAb AB10) against GA 3 glucoside as well as GA 3 is described. MAb AB10 was derived from an immunogen in which human serum albumin (HSA) was linked to GA 3 at carbon 3. This antibody showed high affinity for GA 3 glucoside as well as for 13 hydroxy gibberellins (GA 1, GA 3, GA 5, etc). The affinity of MAb AB10 for 13 hydroxy GAs was significantly reduced by methylation of the 7 oic acid but not by glycosylation of 3 hydroxyl group. Based on this antibody, both of competitive enzyme linked immunosorbent assays (ELISAs) for GA 3 glucoside and for GA 3 were developed. These two ELISAs displayed linear detection ranges from 0 2 pmol to 20 pmol. Using these assays, the fluctuation of GA 3 like and GA 3 glucoside like substances in the leaves of Rumex japonicus was investigated. The results indicated that the glycosylation of free GAs was connected with leaf senescence and that the function of 6 benzyl amino purine in retarding the leaf senescence was probably related to delaying the process of glycosylation of free GAs.展开更多
Activities of hexokinase (HK), pyruvate kinase (PK), superoxide dismutase (SOD) and catalase (CAT) and Hsp70 level were measured to evaluate the response of the commercially important sea cucumber (Apostichop...Activities of hexokinase (HK), pyruvate kinase (PK), superoxide dismutase (SOD) and catalase (CAT) and Hsp70 level were measured to evaluate the response of the commercially important sea cucumber (Apostichopus japonicus Selenka) to rapid temperature changes in laboratory. Animals were subjected to a higher temperature (from 10 to 20℃) (Tinc treatment) or to a lower temperature (from 20 to 10℃) (Tddec treatment) for 72 h. At 1, 3, 12, 24, 72 h of exposure, animals were removed and prepared for further analysis. Results showed that the effect of acute temperature changes on enzyme activities was significant. In Tinc treatment, activities of SOD and CAT increased immediately. The significant enhancement of SOD and CAT activities suggested that oxidative stress increases significantly when ambient temperature increasing from 10 to 20℃. The up-regulation of Flsp70 in Tinc and Tdec treatments indicated that Hsp70 was a bioindicator of thermal stress in the sea cucumber, and the expression pattern depended on the thermal treatment.展开更多
