However, currently available probe for image tumor based on GRP78 has not been reported, owning to no obvious strategy in probe design towards this protein. In this paper, a hairpin-shaped peptidyl probe(pep FAM) conjugated with a 5-FAM fluorophore and a dabcyl quencher at both ends was developed, respectively. The probe was designed by performing a traditional fluorescence resonance energy transfer mechanism and employing a GRP78 specifically-binding peptide. Furthermore, the probe was used to specifically image cancer cells,and accurately image xenograft tumors in mice models. The novel fluorescent probe is expected to be a useful tool for the diagnostics of cancer.展开更多
The localized surface plasmon resonance of gold nanoparticles enables them to be excellent light scattering sensing reagents and efficient lignt-heat convertors for potential diagnostics and therapeutics.In this work,...The localized surface plasmon resonance of gold nanoparticles enables them to be excellent light scattering sensing reagents and efficient lignt-heat convertors for potential diagnostics and therapeutics.In this work,gold nanoparticles of 15 nm in size were synthesized and conjugated with a kind of arginine-glycine-aspartic acid peptide(RGD)to target the cancer cells.Under a conventional dark field microscope,the scattering images from normal and cancer cells are very different.Only a few of gold nanoparticles bind to the membrane of normal cells due to nonspecific interaction.Yet for cancer cells,the concentration of gold nanoparticles inside cancer cells is much higher than that inside normal cells,showing a strong light scattering signal.By exposing these cells to an external laser irradiation,cancer cells were selectively destructed because of the heat conversion of gold nanoparticles inside them,but normal cells still kept alive due to the absence of gold nanoparticles in them.These results incidated that gold nanoparticles might be potential contrast reagents for targeted cancer imaging and laser therapy.展开更多
目的应用磁共振弥散峰度成像(diffusional kurtosis imaging,DKI)研究阻塞性睡眠呼吸暂停低通气综合征(OSAHS)患者的脑白质变化情况。方法选取2017年11月~2021年6月行多导睡眠监测(PSG)确诊为OSAHS的患者38例(OSAHS组),另选取性别、年...目的应用磁共振弥散峰度成像(diffusional kurtosis imaging,DKI)研究阻塞性睡眠呼吸暂停低通气综合征(OSAHS)患者的脑白质变化情况。方法选取2017年11月~2021年6月行多导睡眠监测(PSG)确诊为OSAHS的患者38例(OSAHS组),另选取性别、年龄相匹配的非OSAHS者32例(对照组);所有受试者均行DKI扫描,通过比较两组各脑区的峰度各向异性分数(kurtosis fractional anisotropy,KFA)找出差异性脑区并分析OSAHS组患者差异性脑区的KFA值与PSG结果中呼吸暂停低通气指数(AHI)、最低血氧饱和度(lowest saturation oxygen,LSaO_(2))、血氧饱和度小于90%时间占监测总时间的百分比(the percent of the total record time spent below 90%oxygen saturation,TS90%)的相关性。结果OSAHS组在胼胝体、双侧放射冠、扣带回以及右侧小脑上脚、下脚区域白质纤维束的KFA值均低于对照组(P<0.05)。在控制年龄和体重指数(BMI)两个指标的前提下行偏相关分析得出:OSAHS组患者的AHI与胼胝体、左侧后放射冠、右侧前放射冠、扣带回的KFA值呈负相关(r=-0.344、-0.380、-0.406、-0.537,P<0.05);LSaO_(2)与胼胝体、左侧后放射冠、右侧前放射冠、扣带回、右侧小脑上脚的KFA值呈正相关(r=0.366、0.406、0.446、0.404、0.342,P<0.05);TS90%与胼胝体、左侧后放射冠、右侧前放射冠、扣带回、右侧小脑上脚、右侧小脑下脚的KFA值呈负相关(r=-0.414、-0.352、-0.355、-0.336、-0.456、-0.360,P<0.05)。结论OSAHS患者夜间的反复呼吸暂停和低氧血症会导致部分脑区白质纤维受损,从而引发对应的脑功能障碍。展开更多
