METHODS: Twenty patients with persistent HBV infection were included in this study, 1...AIM: To identify the property of dendritic cells (DCs) of peripheral blood monocytes (PBMC) in patients with chronic HBV infection. METHODS: Twenty patients with persistent HBV infection were included in this study, 10 healthy subjects being used as a control group. The peripheral blood mononuclear cells (PBMC) of T cell-depleted populations were incubated and induced into mature dendritic cells in the RPMI-1640 medium in the presence of cytokines GM-CSF, IL-4, FLt-3,TNF-alpha and 100mL.L(-1 )of fetal calf serum for a total of 10-12 days. The expressions of surface markers on DCs were evaluated using flow cytometric analysis. ELISA method was used to determine the cytokine levels of interleukin-12 (IL-12) and IL-10 in the supernatant produced by DCs. For detection of the stimulatory capacity of DCs to T cell proliferation, mytomycin C-treated DC were incubated with allogenic T cells. RESULTS: A typical morphology of mature DCs from healthy subjects and HBV-infected patients was induced in in vitro incubation, but the proliferation ability and cellular number of DCs from HBV-infected patients significantly decreased compared with healthy individuals. In particular, the expression levels of HLA-DR, CD80 (B7-1) and CD86 (B7-2) on DC surface from patients were also lower than that from healthy individuals (0.46 vs 0.92 for HLA-DR, 0.44 vs 0.88 for CD80 and 0.44 vs 0.84 for CD86,P【0.05). The stimulatory capacity and production of IL-12 of DCs from patients in allogenic mixed lymphocyte reaction (AMLR) significantly decreased, but the production level of nitric oxide (NO) by DCs simultaneously increased compared with healthy subjects (86 +/- 15 vs 170 +/- 22 micromol.L(-1), P 【0.05). CONCLUSION: The patients with chronic HBV infection have the defective function and immature phenotype of dendritic cells, which may be associated with the inability of efficient presentation of HBV antigens to host immune system for the clearance of HBV.展开更多

METHODS: Cloning procedure was performed on parental hepatocellular carcinoma (HCC) cell line MHCC97, and biological characteristics of the target clones selected by in vivo screening were studied. RESULTS: Two clones with high (MHCC97-H) and low (MHCC97-L) metastatic potential were isolated from the parent cell line. Compared with MHCC97-L, MHCC97-H had smaller cell size (average cell diameter 43 microm vs 50 microm) and faster in vitro and in vivo growth rate (tumor cell doubling time was 34.2h vs 60.0h). The main ranges of chromosomes were 55-58 in MHCC97-H and 57-62 in MHCC97-L. Boyden chamber in vitro invasion assay demonstrated that the number of penetrating cells through the artificial basement membrane was (37.5 +/- 11.0) cells/field for MHCC97-H vs (17.7 +/- 6.3)/field for MHCC97-L. The proportions of cells in G0-G1 phase, S phase, and G2-M phase for MHCC97-H/MHCC97-L were 0.56/0.65, 0.28/0.25 and 0.16/0.10, respectively, as measured by flow cytometry. The serum AFP levels in nude mice 5wk after orthotopic implantation of tumor tissue were (246 +/- 66) microg.L(-1) for MHCC97-H and (91 +/- 66) microg.L(-1) for MHCC97-L. The pulmonary metastatic rate was 100% (10/10) vs 40% (4/10). CONCLUSION: Two clones of the same genetic background but with different biological behaviors were established, which could be valuable models for investigation on HCC metastasis.展开更多

Especially, the newly developed numerical methods for simulating cavitating turbulent flows and the achievements with regard to the complicated flow features revealed by using advanced optical techniques as well as cavitation simulation are introduced so as to make a better understanding of the cavitating flow mechanism for hydraulic machinery. Since cavitation instabilities are also vital issue and rather harmful for the operation safety of hydro machines, we present the 1-D analysis method, which is identified to be very useful for engineering applications regarding the cavitating flows in inducers, turbine draft tubes, etc. Though both cavitation and hydraulic machinery are extensively discussed in literatures, one should be aware that a few problems still remains and are open for solution, such as the comprehensive understanding of cavitating turbulent flows especially inside hydro turbines, the unneglectable discrepancies between the numerical and experimental data, etc.. To further promote the study of cavitation in hydraulic machinery, some advanced tooics such as a Density-Based solver suitable for highly comoressible cavitating turbulent flows, a virtual cavitation tunnel, etc. are addressed for the future works.展开更多
Ischemic colitis is the most common form of ischemic injury of the gastrointestinal tract and can present either as an occlusive or a non-occlusive form. It accounts for 1 in 1000 hospitalizations but its incidence is...Ischemic colitis is the most common form of ischemic injury of the gastrointestinal tract and can present either as an occlusive or a non-occlusive form. It accounts for 1 in 1000 hospitalizations but its incidence is underesti- mated because it often has a mild and transient nature. The etiology of ischemic colitis is multifactorial and the clinical presentation variable. The diagnosis is based on a combination of clinical suspicion, radiographic, endo- scopic and histological findings. Therapy and outcome depends on the severity of the disease. Most cases of the non-gangrenous form are transient and resolve spontaneously without complications. On the other hand, high morbidity and mortality and urgent operative intervention are the hallmarks of gangrenous ischemic colitis.展开更多
Computer vision techniques, in conjunction with acquisition through remote cameras and unmanned aerial vehicles (UAVs), offer promising non-contact solutions to civil infrastructure condition assessment. The ultimate ...Computer vision techniques, in conjunction with acquisition through remote cameras and unmanned aerial vehicles (UAVs), offer promising non-contact solutions to civil infrastructure condition assessment. The ultimate goal of such a system is to automatically and robustly convert the image or video data into actionable information. This paper provides an overview of recent advances in computer vision techniques as they apply to the problem of civil infrastructure condition assessment. In particular, relevant research in the fields of computer vision, machine learning, and structural engineering is presented. The work reviewed is classified into two types: inspection applications and monitoring applications. The inspection applications reviewed include identifying context such as structural components, characterizing local and global visible damage, and detecting changes from a reference image. The monitoring applications discussed include static measurement of strain and displacement, as well as dynamic measurement of displacement for modal analysis. Subsequently, some of the key challenges that persist toward the goal of automated vision-based civil infrastructure and monitoring are presented. The paper concludes with ongoing work aimed at addressing some of these stated challenges.展开更多

The liver is indeed essential to life,not only due to its complex functions in biosynthesis,metabolism and...The interest in the liver dates back to ancient times when it was considered to be the seat of life processes. The liver is indeed essential to life,not only due to its complex functions in biosynthesis,metabolism and clearance,but also its dramatic role as the blood volume reservoir. Among parenchymal organs,blood flow to the liver is unique due to the dual supply from the portal vein and the hepatic artery. Knowledge of the mutual communication of both the hepatic artery and the portal vein is essential to understand hepatic physiology and pathophysiology. To distinguish the individual importance of each of these inflows in normal and abnormal states is still a challenging task and the subject of on-going research. A central mechanism that controls and allows constancy of hepatic blood flow is the hepatic arterial buffer response. The current paper reviews the relevance of this intimate hepatic blood flow regulatory system in health and disease. We exclusively focus on the endogenous interrelationship between the hepatic arterial and portal venous inflow circuits in liver resection and transplantation,as well as inflammatory and chronic liver diseases. We do not consider the hepatic microvascular anatomy,as this has been the subject of another recent review.展开更多
This paper analyzes the monitored data of the 4 times of stream water conveyances to the river section where the stream flow was cut-off, of 9 groundwater-monitoring sections and 18 vegetation plots in the lower reach...This paper analyzes the monitored data of the 4 times of stream water conveyances to the river section where the stream flow was cut-off, of 9 groundwater-monitoring sections and 18 vegetation plots in the lower reaches of Tarim River. The results show that the groundwater depth in the lower reaches of Tarim River rose from 9.87 m before the conveyances to 7.74 m and 3.79 m after the first and second conveyances, 3.61 and 3.16 m after the 2 phases of the third conveyance, and 2.66 m after the fourth conveyance. The transverse response scope of groundwater level was gradually enlarged along both sides of the channel of conveyances, i.e., from 450 m in width after the first conveyance to 1050 m after the fourth conveyance, but the response degree of groundwater level was reduced with the increase of the distance away from the channel of conveyances. The composition, distribution and growth status of the natural vegetation are directly related to the groundwater depth. The indexes of Simpson’s biodiversity, McIntosh’s evenness and Margalef’s richness, which reflect the change of the quantity of species and the degree of biodiversity, are reduced from 0.70, 0.48 and 0.90 to 0.26, 0.17 and 0.37 re- spectively along with the drawdown of groundwater level from the upper reaches to the lower reaches. After the stream water conveyances, the natural vegetation in the lower reaches is saved and restored along with the rise of groundwater level, the response scope of vegetation is gradually enlarged, i.e., from 200— 250 m in width after the first conveyance to 800 m after the fourth conveyance. However, there is still a great disparity to the objective of protecting the “Green Corridor”in the lower reaches of Tarim River. Thus, it is suggested to convey the stream water in double-channel way, combine the conveyance with water supply in surface scope, or construct the modern pipe-conveyance network systems so as to save the natural vegetation in an intensive way, achieve the efficient water consumption and speed u展开更多

Test pump has a positive slope of a head-flow performance curve at 65% flow rate of BEP (Best Efficiency Point) because of a rotating stall. Dynamic Particle Image Velocimetry (PIV) and pressure fluctuation measurements are used for investigating the propagation mechanism of a rotating stall. It was found that unstable performance was caused by periodical large scale abrupt backflow generated from the vaned diffuser to the outlet of impeller. Further, the relation between the static pressure at the inlet of diffuser vane and the internal flow condition was clarified. From these experimental results, in order to improve the positive slope of a head-flow performance curve, to suppress the growth of strong vortex toward the inlet of diffuser vane was proved to be a key point.展开更多

Dynamic Particle Image Velocimetry (PIV) and pressure fluctuation measurements are used for investigating the propagation mechanism of a rotating stall. It was found that unstable performance was caused by periodical large scale abrupt backflow generated from the vaned diffuser to the outlet of impeller. Further, the relation between the static pressure at the inlet of diffuser vane and the internal flow condition was clarified. From these experimental results, in order to improve the positive slope of a head-flow performance curve, to suppress the growth of strong vortex toward the inlet of diffuser vane was proved to be a key point.
