Objective:To observe the effects of electroacupuncture on urodynamics and intramedullary apoptosis related factor B cell lymphoma gene-2(Bcl-2),Bcl-2 related protein X(Bax)and brain-derived neurotrophic factor(BDNF)in neurogenic bladder rats after supersacral spinal cord injury.Methods:A total of 60 female SD rats,SPF grade,were randomly selected,12 rats in the blank group and 12 rats in the sham operation group,and the remaining 36 rats were made models.Of these 36 rats,24 rats meeting the model standard were randomized into the model group and electroacupuncture group,with 12 rats in each group.The neurogenic bladder models after supracacral spinal cord injury were prepared by the Hassan Shaker spinal cord transection method,via combining the previous model study experience.At the end of spinal shock stage,"Dazhui(大椎GV14)""Zhongji(中极CV 3)""Ciliao(次髎BL 32)",and"Sanyinjiao(三阴交SP 6)"were selected for electroacupuncture.Observation on the histomorphological changes of bladder detrusor and spinal cord in rats was conducted by HE staining.The apoptosis rate of spinal cord cells was measured by TUNEL assay.The maximum bladder volume and compliance were measured by urodynamics test.The protein expressions of Bcl-2,Bax,and BDNF in spinal cord tissues were measured by Western blot assay.The expressions of Bcl-2,Bax,and BDNF mRNA were measured by RT-PCR assay.Results:(1)The results of HE staining showed that compared with the blank group and sham operation group,in the model group the bladder epithelial cell structure was destroyed,the detrusor muscle fibers proliferated,and there were severe bleeding changes,the spinal cord tissue was disordered,the structure was destroyed,the cell space was enlarged,and accompanied by a large number of inflammatory cell infiltration and cystic cavity formation.Compared with the model group,in the electroacupuncture group,the intact bladder epithelial cells increased,the detrusor muscle fiber proliferation decreased,the bleeding changes decreased,the complete structural 展开更多

Brain functional impairment after stroke is common;however,the molecular mechanisms of post-stroke recovery remain unclear.It is well-recognized that age is the most important independent predictor of poor outcomes after stroke as older patients show poorer functional outcomes following stroke.Mounting evidence suggests that axonal regeneration and angiogenesis,the major forms of brain plasticity responsible for post-stroke recovery,diminished with advanced age.Previous studies suggest that Ras-related C3 botulinum toxin substrate(Rac)1 enhances stroke recovery as activation of Rac1 improved behavior recovery in a young mice stroke model.Here,we investigated the role of Rac1 signaling in long-term functional recovery and brain plasticity in an aged(male,18 to 22 months old C57BL/6J)brain after ischemic stroke.We found that as mice aged,Rac1 expression declined in the brain.Delayed overexpression of Rac1,using lentivirus encoding Rac1 injected day 1 after ischemic stroke,promoted cognitive(assessed using novel object recognition test)and sensorimotor(assessed using adhesive removal tests)recovery on days 14–28.This was accompanied by the increase of neurite and proliferative endothelial cells in the periinfarct zone assessed by immunostaining.In a reverse approach,pharmacological inhibition of Rac1 by intraperitoneal injection of Rac1 inhibitor NSC23766 for 14 successive days after ischemic stroke worsened the outcome with the reduction of neurite and proliferative endothelial cells.Furthermore,Rac1 inhibition reduced the activation of p21-activated kinase 1,the protein level of brain-derived neurotrophic factor,and increased the protein level of glial fibrillary acidic protein in the ischemic brain on day 28 after stroke.Our work provided insight into the mechanisms behind the diminished plasticity after cerebral ischemia in aged brains and identified Rac1 as a potential therapeutic target for improving functional recovery in the older adults after stroke. 展开更多

Objective:to explore brain-derived neu-rotrophic factor(BDNF)and matrix metalloproteinase-9(MMP9)messenger in peripheral blood lymphocytes of patients with depressive episodes before and after anti-depressant treatment changes in RNA(mRNA)expres-sion levels.Methods:fifteen patients hospitalized with depression in the Afiliated Mental Hospital of Kunming Medical University from December 2021 to June 2022 were collected as research objects.After 4 weeks of anti-depressant treatment,BDNF mRNA and MMP9 mRNA in patients with depression were detected before and af-ter treatment,and statistically analyzed.Results:after 4 weeks of antidepressant treatment,there was no signif-icant difference in BDNF mRNA expression level after treatment compared with before treatment(P>0.05).The mRNA level of MMP9 was higher than that before treatment,and the difference was statistically significant(P<0.05).Conclusion:antidepressant therapy increases the expression of MMP9 mRNA,increasing the conver-sion of BDNF precursor(proBDNF)to mature for BDNF(mBDNF)to relieve depressive symptoms,and the total content of BDNF did not change significantly. 展开更多

目的探讨氟西汀联合重复经颅磁刺激对急性脑卒中后抑郁患者的认知功能、神经功能及血清脑源性神经营养因子(BDNF)、C反应蛋白(CRP)的影响。方法选择2016年2月至2018年11月空军军医大学第二附属医院神经内科收治的110例急性脑卒中后抑郁... 目的探讨氟西汀联合重复经颅磁刺激对急性脑卒中后抑郁患者的认知功能、神经功能及血清脑源性神经营养因子(BDNF)、C反应蛋白(CRP)的影响。方法选择2016年2月至2018年11月空军军医大学第二附属医院神经内科收治的110例急性脑卒中后抑郁患者进行研究,采用随机数表法将患者分为观察组和对照组,每组55例。对照组给予氟西汀治疗,观察组在此基础上联合重复经颅磁刺激治疗,均连续治疗8周。比较两组患者治疗前后的汉密尔顿抑郁量表(HAMD)、蒙特利尔认知评估量表(MoCA)、美国国立卫生研究院卒中量表(NIHSS)的评分,以及血清BDNF、CRP水平和不良反应。结果治疗后,观察组患者的HAMD评分和NIHSS评分分别为(9.05±1.86)分、(9.33±1.75)分,明显低于对照组的(13.07±2.21)分、(11.28±2.04)分,MoCA评分为(26.74±1.49)分,明显高于对照组的(24.05±1.52)分,差异均具有统计学意义(P<0.05);治疗后,观察组患者的血清BDNF为(13.12±2.49)μg/L,明显高于对照组的(10.06±2.08)μg/L,CRP为(10.06±2.08)mg/L,明显低于对照组的(9.86±1.73)mg/L,差异均具有统计学意义(P<0.05);观察组和对照组患者的不良反应总发生率分别为1.82%和3.64%,差异无统计学意义(P>0.05)。结论氟西汀联合重复经颅磁刺激治疗急性脑卒中后抑郁患者疗效显著,其不仅能明显改善患者的认知功能和神经功能,还可提高患者的血清BDNF,降低CRP水平,且安全性好。 展开更多

目的:基于脑源性神经营养因子(BDNF)/络氨酸激酶B(TrkB)/环磷腺苷效应元件结合蛋白(CREB)途径探讨柴胡加龙骨牡蛎汤对慢性应激抑郁大鼠海马的影响。方法:取60只SD大鼠,其中10只作为空白组,余50只建立慢性应激抑郁大鼠模型后,随机分为5组... 目的:基于脑源性神经营养因子(BDNF)/络氨酸激酶B(TrkB)/环磷腺苷效应元件结合蛋白(CREB)途径探讨柴胡加龙骨牡蛎汤对慢性应激抑郁大鼠海马的影响。方法:取60只SD大鼠,其中10只作为空白组,余50只建立慢性应激抑郁大鼠模型后,随机分为5组,即模型组,盐酸氟西汀组(0.003 g·kg^(-1)),柴胡加龙骨牡蛎汤低、中、高剂量组(1.625,3.25,6.5 g·kg^(-1))。连续灌胃给药8周,1次/d,分别于造模前、造模后及给药后进行行为学实验评估大鼠抑郁状态。苏木素-伊红(HE)染色观察大鼠海马形态学变化,免疫组织化学法(IHC)检测大鼠海马组织中BDNF蛋白表达,实时荧光定量聚合酶链式反应(Real-time PCR)检测大鼠海马组织中BDNF,TrkB,CREB mRNA表达,蛋白免疫印迹法(Western blot)定量检测大鼠海马组织中BDNF,TrkB,CREB蛋白表达。结果:与空白组比较,模型组大鼠糖水偏嗜率明显降低(P<0.05),水平得分、垂直得分明显降低(P<0.05),不动时间明显增加(P<0.05),漂浮时间显著增加(P<0.05),HE染色结果显示海马神经元结构损伤,免疫组织化学染色中BDNF表达明显降低(P<0.05),BDNF,TrkB,CREB mRNA及蛋白表达明显降低(P<0.05);与模型组比较,盐酸氟西汀组及柴胡加龙骨牡蛎汤低、中、高剂量组大鼠糖水偏嗜率明显升高(P<0.05),水平得分、垂直得分明显增加(P<0.05),不动时间明显减少(P<0.05),漂浮时间明显减少(P<0.05),海马神经元结构明显复原,免疫组织化学染色中BDNF表达明显增加(P<0.05),BDNF,TrkB,CREB mRNA及蛋白表达明显增加(P<0.05)。结论:柴胡加龙骨牡蛎汤能显著改善经慢性应激刺激后大鼠的抑郁样行为,增强海马组织神经元的再生与修复,其机制可能与上调大鼠海马BDNF/TrkB/CREB信号通路有关。 展开更多

目的:观察酸枣仁汤联合氟西汀治疗肝郁血虚型抑郁症伴失眠患者的临床疗效。方法:将120例肝郁血虚型抑郁伴失眠患者随机平均分为观察组与对照组,每组60例,对照组口服盐酸氟西汀胶囊(百忧解),观察组用酸枣仁汤联合盐酸氟西汀,两组疗程均为... 目的:观察酸枣仁汤联合氟西汀治疗肝郁血虚型抑郁症伴失眠患者的临床疗效。方法:将120例肝郁血虚型抑郁伴失眠患者随机平均分为观察组与对照组,每组60例,对照组口服盐酸氟西汀胶囊(百忧解),观察组用酸枣仁汤联合盐酸氟西汀,两组疗程均为8周。用汉密尔顿抑郁量表(HAMD),日常生活能力量表(ADL)和匹兹堡睡眠质量指数量表(PSQI)评估临床疗效;ELISA法测定血浆中5-羟色胺(5-HT),去甲肾上腺素(NE),脑源性神经营养因子(BDNF),胶质细胞源性神经营养因子(GDNF),神经元特异性烯醇化酶(NSE)和S100β水平。结果:治疗8周后,与同组治疗前比较,观察组的HAMD和PSQI评分均显著下降(P<0.01);ADL显著提高(P<0.01);血浆5-HT,NE,GDNF和BDNF均显著增高(P<0.01)。治疗后,与对照组比较,观察组总有效率显著升高(P<0.01),HAMD评分和PSQI均显著降低(P<0.01),ADL评分显著升高(P<0.01);血浆5-HT,NE,BDNF和GDNF水平均显著增高(P<0.01),而NSE和S100β水平均显著降低(P<0.01)。结论:酸枣仁汤联合氟西汀治疗肝郁血虚型抑郁症伴失眠的疗效优于单用氟西汀。其机制可能酸枣仁汤对氟西汀增加单胺类神经递质有协同作用,并与促进BDNF和GDNF的分泌,增加对神经细胞的保护有关。 展开更多

长程经颅磁刺激对脑梗死大鼠皮质脑源性神经营养因子表达及神经功能恢复的影响 被引量:25

目的探索长程经颅磁刺激(TMS)对脑梗死大鼠梗死灶周围皮质脑源性神经营养因子(BD-NF)表达和脑损伤体积、神经功能恢复的影响及作用机制,为经颅磁刺激在脑梗死治疗及康复中的应用提供理论依据。方法TMS组与假刺激组大鼠各48只,于大脑中... 目的探索长程经颅磁刺激(TMS)对脑梗死大鼠梗死灶周围皮质脑源性神经营养因子(BD-NF)表达和脑损伤体积、神经功能恢复的影响及作用机制,为经颅磁刺激在脑梗死治疗及康复中的应用提供理论依据。方法TMS组与假刺激组大鼠各48只,于大脑中动脉阻塞/再灌注(MCAO/R)90min后的3周内每日接受1次TMS(200脉冲)与假刺激治疗。检测2组大鼠神经功能恢复情况、梗死灶周围皮质BDNF免疫阳性细胞表达及脑损伤体积,对所得资料进行统计学分析。结果在治疗2周和3周时,TMS组神经功能缺损评分与假刺激组相比均明显降低(P<0.01)。TMS组在治疗3d、7d、14d、21d时梗死灶周围皮质BDNF阳性细胞计数与假刺激组各对应时间点相比,差异均有统计学意义(P<0.01),且2组治疗21d时梗死灶周围皮质BDNF阳性细胞计数与大鼠神经功能缺损评分呈显著负相关(r=-0.877,P<0.01)。治疗3周后,TMS组脑损伤体积明显小于假刺激组(P<0.05),2组脑损伤体积与最终神经功能缺损评分明显相关(r=0.859,P<0.01)。结论长程TMS有促进脑梗死大鼠神经功能缺损恢复的作用,其作用通过持续上调梗死灶周围皮质BDNF阳性细胞表达、减小梗死后脑损伤体积等作用而实现。 展开更多

目的探讨抑郁症患者执行功能、血清脑源性神经营养因子(BDNF)水平治疗前后的动态变化及其与抑郁严重程度三者之间的关系。方法采用威斯康星卡片分类试验(WCST)和汉密尔顿抑郁量表(HAMD)分别评定77例抑郁症患者抗抑郁药物治疗4周前后及7... 目的探讨抑郁症患者执行功能、血清脑源性神经营养因子(BDNF)水平治疗前后的动态变化及其与抑郁严重程度三者之间的关系。方法采用威斯康星卡片分类试验(WCST)和汉密尔顿抑郁量表(HAMD)分别评定77例抑郁症患者抗抑郁药物治疗4周前后及74名正常对照组的执行功能和抑郁严重程度;采用酶联免疫吸附法(ELISA)测定受试者血清BDNF水平。结果与对照组比较,患者组治疗前WCST的总应答数、持续性错误数、随机错误数均增加,完成分类数减少,血清BDNF水平降低[(36.34±15.16)ng/mLvs(23.09±12.13)ng/mL],上述差异均有统计学意义(P<0.01),而4周治疗后两组间的上述指标的差异均无统计学意义(P>0.05)。与治疗前比较,患者组治疗后完成分类数增加、血清BDNF水平[(37.37±21.04)ng/mL]升高,总应答数、持续错误数、随机错误数减少和HAMD总分降低,上述差异均有统计学意义(P<0.01)。治疗前后血清BDNF水平与相应HAMD总分值均呈负相关(r=-0.28,P=0.01;r=-0.28,P=0.01),而治疗前WCST5个指标及其治疗后变化值分别与相应血清BDNF水平、HAMD总分值及疗后变化值均无相关(P>0.05)。结论抑郁症患者存在执行功能受损和血清BDNF水平的下降,后者与抑郁严重程度密切相关,且抗抑郁治疗后执行功能和抑郁症状均改善,血清BDNF水平升高。 展开更多

目的:验证电针合药物治疗帕金森病伴发抑郁症的疗效并探讨其作用机制。方法:将60例帕金森病伴发抑郁症患者随机分为针药组和西药组,每组30例。两组均采用常规治疗,口服美多巴、氟西汀,针药组在此基础上行电针疗法,穴取百会、印堂、四神... 目的:验证电针合药物治疗帕金森病伴发抑郁症的疗效并探讨其作用机制。方法:将60例帕金森病伴发抑郁症患者随机分为针药组和西药组,每组30例。两组均采用常规治疗,口服美多巴、氟西汀,针药组在此基础上行电针疗法,穴取百会、印堂、四神聪、太冲、三阴交等。对比观察两组治疗前与治疗3个月后血清脑源性神经营养因子(BDNF)水平和汉密尔顿抑郁量表(HAMD)评分,并评定两组疗效。结果:两组治疗后血清BDNF水平较治疗前均显著提高(均P<0.05),且针药组优于西药组(P<0.05);两组治疗后HAMD评分较治疗前显著降低(均P<0.05),且针药组优于西药组(P<0.05)。针药组总有效率为90.0%(27/30),优于西药组的83.3%(25/30,P<0.05)。结论:电针合药物治疗帕金森病伴发抑郁症疗效显著,能有效调节患者血清中BDNF水平,从而缓解抑郁症状,其疗效优于单纯药物治疗。 展开更多

目的:该研究基于环磷酸腺苷(cAMP)/蛋白激酶A(PKA)/cAMP反应元件结合蛋白(CREB)/脑源性神经营养因子(BDNF)信号通路探究柴胡加龙骨牡蛎汤对慢性不可预见性温和应激(CUMS)法制备的大鼠抑郁模型的干预作用。方法:随机数字表法将60只SD大... 目的:该研究基于环磷酸腺苷(cAMP)/蛋白激酶A(PKA)/cAMP反应元件结合蛋白(CREB)/脑源性神经营养因子(BDNF)信号通路探究柴胡加龙骨牡蛎汤对慢性不可预见性温和应激(CUMS)法制备的大鼠抑郁模型的干预作用。方法:随机数字表法将60只SD大鼠分为正常组、模型组、柴胡加龙骨牡蛎汤低、中、高剂量组和氟西汀组。除正常组外,其他各组进行49 d的CUMS制备大鼠抑郁模型。第29天开始给药,柴胡加龙骨牡蛎汤组低、中、高剂量组分别给予柴胡龙骨牡蛎汤颗粒剂2.89、5.78、11.56 g·kg^(-1),氟西汀组给予盐酸氟西汀胶囊2.06 mg·kg^(-1)。旷场实验和强迫游泳实验观察大鼠行为学表现,酶联免疫吸附测定法(ELISA)测定大鼠海马组织5-羟色胺(5-HT)、去甲肾上腺素(NE)、cAMP水平;实时荧光定量聚合酶链式反应(Real-time PCR)检测大鼠海马组织PKA、CREB、BDNF mRNA表达;蛋白免疫印迹法(Western blot)检测大鼠海马组织PKA、BDNF蛋白表达水平;免疫组化法(IHC)检测CREB定位表达;苏木素-伊红(HE)染色和尼氏染色观察海马组织形态学的变化。结果:与正常组比较,模型组大鼠强迫游泳不动时间显著增加(P<0.01);运动总距离、中央区停留时间、中央区进入总次数、中央区运动距离均显著降低(P<0.01),海马5-HT、NE、cAMP含量显著降低(P<0.01),PKA蛋白表达、BDNF蛋白表达和mRNA水平、CREB蛋白表达和mRNA水平显著降低(P<0.01);与模型组比较,柴胡加龙骨牡蛎汤给药组大鼠强迫游泳不动时间均显著降低(P<0.01),柴胡加龙骨牡蛎汤给药组大鼠运动总距离、中央区停留时间、中央区运动距离显著、央区进入次数明显增加(P<0.05,P<0.01),柴胡加龙骨牡蛎汤给药组海马5-HT含量、NE含量、cAMP含量明显增高(P<0.05,P<0.01),柴胡加龙骨牡蛎汤给药组海马PKA蛋白表达、CERB和BDNF的mRNA及蛋白表达都显著增加(P<0.01)。HE和尼氏染色显示海马神经元结构恢复� 展开更多
