The results show that the microenvironment of tryptophan residues of protein molecules in various solutions can be directly indicated and TDFS is an effective tool for studying protein conformation in solution. 以酸水解4 h的卵清蛋白(Acid-hydrolyzed egg albumin for 4 h,AHEA4)作为乳化剂制备O/W乳液,采用表观稳定性、微观结构、粒径分布、平均粒径和ξ-电位等指标探究pH值对AHEA4乳化活性及稳定性的影响,以及AHEA4溶解性、表面疏水性及界面...以酸水解4 h的卵清蛋白(Acid-hydrolyzed egg albumin for 4 h,AHEA4)作为乳化剂制备O/W乳液,采用表观稳定性、微观结构、粒径分布、平均粒径和ξ-电位等指标探究pH值对AHEA4乳化活性及稳定性的影响,以及AHEA4溶解性、表面疏水性及界面张力与AHEA4乳化特性的相关性。结果表明,pH值对AHEA4乳化活性和稳定性有显著影响;pH值为8.0的AHEA4乳液稳定性最好,表观稳定性、粒度分布、平均粒径等在50℃贮藏7 d后无明显变化,电位变化相对较小。采用激光共聚焦扫描显微镜(CLSM)观察新鲜乳液微观结构,pH5.0和pH6.0的新鲜乳液有明显聚集,而其他各pH值乳液的油滴微小且分布均匀;pH8.0以上时,AHEA4的溶解性、表面疏水性和油水界面吸附和稳定能力均较好,因此乳化活性和稳定性好。研究结果可为卵清蛋白的高值化应用提供依据。展开更多
