AIM: To investigate the association of plasma levels of interleukin(IL)-6 and-8 with Wilms' tumor 1(WT1)-specific immune responses and clinical outcomes in patients with pancreatic ductal adenocarcinoma(PDA) treated with dendritic cells(DCs) pulsed with three types of major histocompatibility complex classⅠand Ⅱ-restricted WT1 peptides combined with chemotherapy.METHODS: During the entire treatment period, plasma levels of IL-6 and-8 were analyzed by ELISA. The induction of WT1-specific immune responses was assessed using the WT1 peptide-specific delayed-type hypersensitivity(DTH) test.RESULTS: Three of 7 patients displayed strong WT1-DTH reactions throughout long-term vaccination with significantly decreased levels of IL-6/-8 after vaccinations compared with the levels prior to treatment. Moreover, overall survival(OS) was significantly longer in PDA patients with low plasma IL-6 levels(< 2 pg/m L) after 5 vaccinations than in patients with high plasma IL-6 levels(≥ 2 pg/m L)(P = 0.025). After disease progression, WT1-DTH reactions decreased severely and were ultimately negative at the terminal stage of cancer. The decreased levels of IL-6/-8 observed throughout long-term vaccination were associated with WT1-specific DTH reactions and long-term OS.CONCLUSION: Prolonged low levels of plasma IL-6/-8 in PDA patients may be a prognostic marker for the clinical outcomes of chemoimmunotherapy. 展开更多

Objective:To investigate Phlebotomus(P.)sergenti Parrot,1917(Diptera:Psychodidae)salivary gland antigens and their immune response in human.Methods:Human volunteers were exposed to sand flies’bites in the laboratory,and following each exposure the size of induration was recorded.The mean protein concentration of salivary gland lysate and specific anti-P.sergenti saliva IgG was measured.Sand fly salivary proteins were separated by SDS-PAGE and their immunoreactivity was examined by Western blotting assays.Results:Individuals exposed to P.sergenti salivary gland lysate for 8 months showed both antibody and delayed type hypersensitivity responses,although exposure for one month did not provoke any immune responses.The trend of antibody fluctuated during the exposure time and dropped by the end of antigen loading.The mean protein content was(0.36±0.08)μg in each pair salivary glands.Salivary gland lysate showed 11 to 12 major protein bands and 3 to 6 of them were immunoreactive.Conclusions:Our study showed that the salivary gland components of P.sergenti provoked both cellular and humoral immune responses in human.Furthermore,there are some immunogenic proteins in P.sergenti saliva which could be subjected for further investigation as vector-based vaccine candidate/s against anthroponotic cutaneous leishmaniasis. 展开更多

The tuberculin Purified Protein Derivative (PPD) skin test is widely used;however, the results are often inaccurate. Positive results can be observed in patients with active tuberculosis (TB) as well as in BCG-vaccinated persons and individuals who are infected with mycobacteria but have not developed the disease. MPT64, an antigen secreted from actively growing Mycobacterium tuberculosis and some strains of M. bovis BCG such as BCG Tokyo and BCG Russia, is immunogenic and elicits delayed-type hypersensitivity (DTH) in guinea pigs and humans. This antigen has been used to develop a new skin test for the diagnosis of active TB infection. Two of the antigens encoded by the M. tuberculosis-specific region of difference 1 (RD1, deleted in M. bovis BCG strains), CFP10 (culture filtrate protein 10) and ESAT6 (early secreted antigenic target-6), also induce M. tuberculosis-specific DTH responses. The aim of this study was to investigate the DTH responses in guinea pigs infected with M. tuberculosis or M. bovis BCG Tokyo elicited by three purified recombinant proteins (rMPT64, rCFP10 and rESAT6) compared to those elicited by PPD. In this study genes encoding MPT64, CFP10, and ESAT6 were cloned and expressed as recombinant proteins with the addition of a C-terminal His6 tag for ease of purification by Immobilized Metal ion Affinity Chromatography (IMAC). The recombinant proteins (rMPT64, rCFP10, and rESAT6) were purified to homogeneity and were used to elicit DTH responses in guinea pigs infected with M. tuberculosis or M. bovis BCG Tokyo. The results showed that rMPT64 elicits a DTH response comparable to that of PPD in M. bovis BCG Tokyo-vaccinated animals. However, M. tuberculosis-infected animals show less reactivity to rMPT64 than they do to PPD. Although single rCFP10 or rESAT6 did not readily elicit a DTH response in M. tuberculosis-infected animals, combining these antigens with rMPT64 led to an increased DTH response, thus enabling the detection of TB infection. 展开更多

It is well documented that dietary restriction can prevent many different diseases and extend the life spans of different rodent species. In the previous study, we reported that intermittent fasting (IF) as well as moderate dietary restriction ameliorated the allergic dermatitis in ICR mice. In the present study, we demonstrated the ameliorative effects of IF on allergic dermatitis in NC/Nga mice, a strain known as a model for atopic dermatitis. Interestingly, the total number of CD4+CD8+ double positive thymocyte in mice after IF significantly decreased in comparison to that in mice fed ad libitum. Although it was reported that an immunosuppressive compound inhibited the contact allergic response by inducing the CD4+CD25+ regulatory T-cells, IF did not affect regulatory T cells in the present study. These results suggested that CD4+CD8+ double positive thymocytes play an important role in the regulation of allergy by IF in NC/Nga mice. 展开更多

We reported an 8-year-old boy with panscleritis in left eye and right epididymitis after falling on the ground.Etiologic diagnosis played a key role in this case.Systemic examinations ruled out systemic autoimmune diseases,tumors,and infections as the cause of scleritis and suggested that the disease was caused by a local delayedtype hypersensitivity(DTH)induced by ocular trauma and was non-infectious.Still,the right epididymitis was infectious.Both conditions were treated successfully using steroids and antibiotics,respectively.Thus,early etiologic diagnosis and reasonable treatment are crucial to prevent visual loss. 展开更多

Methods: T-lymphocyte subtypes of the peripheral blood and cutaneous delayed-type hypersensitivity (DTH) to two recall antigens, tetanus toxoid (TT) and purified protein derivative(PPD), were measured in 5 patients with HIE and 15 healthy controls, respectively. Results: The CD4 + cell counts in HIE group were significantly lower than those in the control group (P<0.01). In contrast, CD8 + cells were significantly higher in HIE group than those in the controls. The induration sizes of DTH to two recall antigens were smaller in HIE group than those in controls (P<0.01). Conclusion: There is an immunologic dysfunction of T lymphocytes in the patients with HIE and T cells play an important role in the pathogenesis. 展开更多

目的初步探讨青蒿素(artemisinin,Art)体内外发挥免疫抑制作用的调控机制。方法体外研究检测Art对小鼠淋巴细胞增殖及细胞毒性的影响。体内研究中,通过迟发型超敏反应(delayed-type hypersensitivity,DTH)小鼠模型,分析不同剂量Art对小... 目的初步探讨青蒿素(artemisinin,Art)体内外发挥免疫抑制作用的调控机制。方法体外研究检测Art对小鼠淋巴细胞增殖及细胞毒性的影响。体内研究中,通过迟发型超敏反应(delayed-type hypersensitivity,DTH)小鼠模型,分析不同剂量Art对小鼠DTH的效应;RT-PCR技术检测Th1和Th2细胞转录因子T-bet及GATA-3的基因表达;ELISA法检测转化生长因子β1(transforming growth factorβ1,TGF-β1)的含量;Western blot方法检测p38丝裂酶原激活蛋白激酶(p38 MAPK)蛋白的活性表达水平。结果 Art明显抑制刀豆蛋白A(concanavalin A,ConA)诱导的T淋巴细胞增殖;与地塞米松相比,具有更高的安全性。体内给药后,Art能够减轻DTH小鼠耳肿胀;降低小鼠的脾指数和胸腺指数;调节Th1/Th2细胞分化过程中特异性转录因子T-bet和GATA-3的mRNA表达水平;增加TGF-β1水平;抑制p38 MAPK的磷酸化活性表达。结论提取自传统中草药的Art是一种高效低毒的新型免疫调节剂,通过多种途径发挥免疫抑制作用。 展开更多

茶树花富含多糖,是茶树生长过程中的产物,然而目前茶树花多糖(tea plant flow polysaccharide,TFP)尚未被充分利用.因此,认为茶树花多糖有着和茶叶中的多糖相似的功能效果.研究目的是通过系列的体内和体外实验系统来评价茶树花多糖的抗... 茶树花富含多糖,是茶树生长过程中的产物,然而目前茶树花多糖(tea plant flow polysaccharide,TFP)尚未被充分利用.因此,认为茶树花多糖有着和茶叶中的多糖相似的功能效果.研究目的是通过系列的体内和体外实验系统来评价茶树花多糖的抗肿瘤以及免疫调节的活性.利用S180(sarcoma180)荷瘤小鼠模型,系统地研究了灌胃不同剂量茶树花多糖对荷瘤小鼠的肉瘤抑制率、存活率以及细胞免疫的影响.结果显示,连续灌胃10天茶树花多糖,能显著抑制S180肉瘤的生长,延长荷瘤小鼠的存活时间,增强迟发型超敏作用(delayed-type hypersensitivity,DTH),促进血液白介素-2(IL-2)、γ-干扰素(IFN-γ)的分泌,增强巨噬细胞的吞噬作用,改善T淋巴细胞亚群CD4+数量以及CD4+/CD8+的比值,表明茶树花多糖能增强机体对肿瘤的防御,在一定程度上归功于其对免疫调节作用. 展开更多

[目的]验证黄芩对皮肤过敏的治疗作用。[方法]通过灌胃给予中药黄芩有效成分黄芩素,进行了2,4-二硝基氯苯致小鼠迟发型变态反应、豚鼠Forssman皮肤血管炎症反应及小鼠同种被动皮肤过敏反应实验;低分子右旋糖酐致小鼠瘙痒、组织胺致豚鼠... [目的]验证黄芩对皮肤过敏的治疗作用。[方法]通过灌胃给予中药黄芩有效成分黄芩素,进行了2,4-二硝基氯苯致小鼠迟发型变态反应、豚鼠Forssman皮肤血管炎症反应及小鼠同种被动皮肤过敏反应实验;低分子右旋糖酐致小鼠瘙痒、组织胺致豚鼠足痒实验,豚鼠离体回肠收缩实验和二甲苯致小鼠耳肿实验。[结果]黄芩素对2,4-二硝基氯苯所致小鼠迟发型变态反应性耳肿、对豚鼠Forssman皮肤血管炎症反应以及小鼠同种被动皮肤过敏反应均有明显抑制作用。同时对低分子右旋糖酐所致小鼠搔痒反应、组胺所致豚鼠足痒反应亦有明显抑制作用,并可抑制组胺引起的豚鼠离体回肠收缩,还可抑制二甲苯所致小鼠耳肿反应。[结论]实验结果表明黄芩具有明显的抗过敏作用,并可明显拮抗组胺,同时具有一定的抗炎作用。 展开更多

目的 阐明金铁锁总甙 (TGP)对小鼠细胞免疫功能的影响。方法 以 80和 2 5 0mg/kg环磷酰胺分别诱导小鼠迟发性超敏反应 (DTH)增强和抑制模型 ,观察不同剂量的TGP对小鼠DTH的影响。以 0 6 %巯基乙醇酸钠诱导、制备和培养小鼠腹腔单层... 目的 阐明金铁锁总甙 (TGP)对小鼠细胞免疫功能的影响。方法 以 80和 2 5 0mg/kg环磷酰胺分别诱导小鼠迟发性超敏反应 (DTH)增强和抑制模型 ,观察不同剂量的TGP对小鼠DTH的影响。以 0 6 %巯基乙醇酸钠诱导、制备和培养小鼠腹腔单层巨噬细胞 ,观察不同浓度的TGP对巨噬细胞产生IL - 1的影响。以ConA诱导小鼠脾淋巴细胞和胸腺淋巴细胞增殖反应 ,观察不同剂量TGP在给药 (ig)的第 5、10、15天对小鼠脾淋巴细胞的增殖反应和生成IL - 2的影响。结果  6 0~ 10 0mg/(kg·d)的TGP均能显著提高细胞免疫抑制小鼠的DTH反应 (P <0 0 1)。 2 0~ 10 0mg/(kg·d)能显著下调细胞免疫增强小鼠的DTH(P <0 0 1)。 3μg/ml的TGP能显著提升小鼠巨噬细胞产生IL - 1的水平。 2 0~ 80mg/(kg·d)的TGP对小鼠脾淋巴细胞增殖反应均有明显的促进作用 ,80mg/(kg·d)的促进作用最为显著 (P <0 0 1) ,10 0mg/(kg·d)时 ,这种促进作用减弱。 2 0~ 80mg/(kg·d)的TGP在给药的第 15天均能显著提升小鼠脾淋巴细胞产生IL - 2的水平 ,以 6 0mg/(kg·d)最为显著 ,80mg/(kg·d)以上 ,这种提升作用减弱。结论 适当剂量的TGP既是小鼠细胞免疫的增强剂 。 展开更多
