[Methods] The complete Freund's adjuvant( CFA)-induced AA model of rats was used. The therapeutic effect of K. coccinea on AA rats was evaluated by the articular swelling, arthritis score, immune organ index and hematoxylin-eosin( HE) staining. The content of inflammatory factors TNF-α,IL-6,and IL-1β in the serum of AA rats was detected by enzyme-linked immunosorbent assay( ELISA) respectively. [Results]Compared with the model group, the total extract of K. coccinea( KCTE) could significantly inhibit the primary and secondary articular swelling of AA rats, lower the arthritis score and immune organ index, and improve the pathological state of ankles of AA rats. In the groups with a high and middle dosage of KCTE( KCTE-H and KCTE-M),KCTE could significantly reduce the content of TNF-α and IL-6 in the serum of rats, exhibiting a certain dose-effect relationship. [Conclusions]KCTE had significant therapeutic effect on the AA of rats, and the mechanism might be associated with the modulation of immune function and the decrease in the secretion of inflammatory cytokines TNF-α and IL-6.展开更多

In this paper, we report the synthesis of nanostructured zinc oxide particles by biological method. Highly stable and spherical zinc oxide nanoparticles are produced by using zinc acetate and Ixora coccinea leaf extract. Formation of zinc oxide nanoparticles has been confirmed by UV-Vis absorption spectroscopy, X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), Dynamic light scattering analysis (DLS), zetapotential study and Scanning Electron Microscope with the Energy Dispersive X-ray studies (EDX). Dynamic light scattering analysis shows average particle size of 145.1 nm whereas high zeta potential value confirms the stability of formed zinc oxide nanoparticles. The Scanning Electron Microscope reveals spherical morphology of nanoparticles and Energy Dispersive X-ray analysis confirms the formation of highly pure zinc oxide nanoparticles. The zinc oxide nanoparticles from Ixora coccinea leaves are expected to have applications in biomedical, cosmetic industries, biotechnology, sensors, medical, catalysis, optical device, coatings, drug delivery and water remediation, and also may be applied for electronic and magneto-electric devices. This new eco-friendly approach of synthesis is a novel, cheap, and convenient technique suitable for large scale commercial production.展开更多

The present study focuses on the effect of Ixora coccinea on Triton X-100 induced hyperlipidemia in rats and associated complications.Methodology: In vitro radical scavenging activity of I. coccinea was assessed using DPPH, FRAP and hydrogen peroxide. In vivo antiobesity and antihyperlipidimic activity of I. coccinea was tested in Triton X-100 induced hyperlipidemic rats and assessed for its biochemical parameters in blood and tissue samples. The relationship between physiological responses and regulation of body temperature was investigated by using animal surface temperature images captured with infrared camera. Results: The results of mineral analysis, antioxidant, total flavonoid and phenolic content represented high amount of mineral and had the potential to scavenge free radicals tested with DPPH, FRAP and hydrogen peroxide radicals with dose dependent activity. The highest activity was observed in aqueous extract, DPPH with 71.5% inhibition, FRAP with 56.8%, H2 O2 with 33% activity at 100 μg/mL concentration.Triton X-100 induced hyperlipidemic rats when treated with I. coccinea aqueous extract showed significant activity by regulating the biochemical parameters and maintaining the lipid profile by decreasing TC,LDL-C, VLDL-C, TG and improving HDL-C levels. Similarly, the elevated levels of creatinine, urea, uric acid,AST, ALT, ALP due to induction of hyperlipidemia, were brought back to near normal levels after treatment with I. coccinea. The levels of tissue anti-oxidants enzymes like SOD and CAT were also found to be improved in treated I. coccinea groups. The whole body asymmetrical temperature distribution analysis showed that significant decreases in temperature was observed in obesity induced groups but a gradual increase in temperature(2%–5%) was observed after treatment. Conclusion: Thus, the results indicated that I. coccinea can be a drug of choice to decrea展开更多

It is essential to overcome this problem by the development of new drugs. Marine organisms as corals, sponges, seaweeds, and other are an incredible source of novel pharmacologically active compounds. Herein, the antimicrobial activity (extract/fractions) of the invasive stony coralTubastraea coccinea was evaluated by the disk diffusion method against 21 microbial strains (ATCC and clinical strains). Micro broth dilution was used to determinate the MIC of the fractions that showed antimicrobial activity by the disk diffusion method. Bioautography assay was also performed. Our results showed that the n-butanol (BF) and aqueous fractions (AF) showed activity against ATCC strains Staphylococcus aureus (MIC 31.25 and 250 μg/mL),Salmonella typhimurium (MIC 125 and 500 μg/mL), Escherichia coli (MIC 62.5 and 500 μg/mL) and Pseudomonas aeruginosa (MIC 62.5 and 500 μg/mL), respectively. Moreover, BF fraction was also effective against the clinical strains S. aureus(MIC 62.5 μg/mL), Klebsiella pneumoniae Carbapenemase—KPC (MIC 125 μg/mL), Methicillin-Resistant Staphylococcus aureus—MRSA (MIC 125 μg/mL) and Vancomycin-Resistant Enterococcus faecalis—VRE (MIC 62.5 μg/mL). The ratio MBC/MIC reinforces the bactericidal profile of BF fraction. The bioautography assay of BF fraction showed the presence of antimicrobial components at Rf0.55.展开更多

[Method] The seedlings of Kadsura coccinea growing one year were taken as materials in order to investigate and analyze leaf SPAD value, relative water content (RWC), malonaldehyde (MAD), cell membrane permeability (RP), proline (PRO) and superoxide dismutase activity (SOD). [Result] As drought stress improved, leaf SPAD value and RWC were both diminishing and RWC fell remarkably; MAD, Pro, RP and SOD activity were increasing; MAD and RP grew significantly. [Conclusion] Kadsura coccinea is much sensitive to drought stress and will be affected if water loss is too much high.展开更多

Conclusion: Thus, the results indicated that I. coccinea can be a drug of choice to decrea
文摘Bacterial infection is considered to be one of the most critical health issues of the world. The bioautography assay of BF fraction showed the presence of antimicrobial components at Rf0.55.
文摘Kadsura coccinea(Lem.)is a woody wine plant with a peculiar fruit enriched in important health-promoting compounds. The non-editable part of the fruit,i.e.,the seed and peel, represents more than 60%of the fruit and is considered a biowaste. This significantly restricts the development of the K.coccinea fruit industry.Clarifying the metabolic components of the different fruit parts can help to improve the utilization rate and valorization of K.coccinea.Herein,we evaluated K.coccinea fruit peel, pulp, and seed using widely-targeted metabolomics and quantified a set of 736 bioactive compounds from 11 major metabolite classes. The most prominent metabolite classes included lipids, amino acids, flavonoids, and lignans.Furthermore, our results emphasized a significant accumulation of flavonoids in pulp tissues, while alkaloids and lignans were abundant in peel and seed tissues, respectively.A total of 183 metabolites were differentially accumulated among the three tissues.Procyanidin C2,rutinoside,2-hydroxyoleanolic acid,5-hydroxymethyluracil,nootkatol,isoquercitrin,isohyperoside,quercetin-7-O-glucoside,hyperin,and rutin showed elevated accumulation in the peel.In the seed, kadsuralignan G,kadcoccilactone A,kadsuralignan H,lysoPE 20:5,iso-schisandrin ethyl alcohol, and kadangustin were significantly enriched. Our results highlight the diverse metabolome composition of K.coccinea fruit parts, which can be further exploited for its valorization in various industries.

文摘[Objective] The aim was to research drought resistance and adaptation mechanisms of Kaclsura coccinea under different drought stresses. [Conclusion] Kadsura coccinea is much sensitive to drought stress and will be affected if water loss is too much high.

文摘Terpenoids are the largest class of natural products with complex structures and extensive bioactivities;their scaffolds are generated by diverse terpenoid synthases(TPSs)from a limited number of isoprenoid diphosphate precursors.Promiscuous TPSs play important roles in the evolution of terpenoid chemodiversity, but they remain largely unappreciated. Here,an extremely promiscuous terpenoid synthase(CcTPS1)of the TPS-b subfamily was cloned and functionally characterized from a leaf-specific transcriptome of the Lamiaceae plant Colquhounia coccinea var.mollis.CcTPS1 is the first sester-/di-/sesqui-/mono-TPS identified from the plant kingdom, accepting C_(25)/C_(20)/C_(15)/C_(10) diphosphate substrates to generate a panel of sester-/di-/sesqui-/mono-terpenoids.Engineered Escherichia coli expressing CcTPS1 produced three previously unreported terpenoids(two sesterterpenoids and a diterpenoid)with rare cyclohexanecontaining skeletons, along with four sesquiterpenoids and one monoterpenoid.Their structures were elucidated by extensive nuclear magnetic resonance spectroscopy.Nicotiana benthamiana transiently expressing CcTPS1 also produced the diterpenoid and sesquiterpenoids, demonstrating the enzyme’s promiscuity in planta. Its highly leaf-specific expression pattern combined with detectable terpenoid products in leaves of C.coccinea var.mollis and N.benthamiana expressing CcTPS1 suggested that CcTPS1 was mainly responsible for diterpenoid and sesquiterpenoid biosynthesis in plants.CcTPS1 expression and the terpenoid products could be induced by methyl jasmonate, suggesting their possible role in plant–environment interaction.CcTPS1 was localized to the cytosol and may differ from mono-TPSs in subcellular compartmentalization and substrate tolerance. These findings will greatly aid our understanding of plant TPS evolution and terpenoid chemodiversity;they also highlight the enormous potential of transcriptome mining and heterologous expression for the exploration of unique enzymes and natural products hidden in plants.
