Nevertheless, these lesions were categorized as chronic while acute lesions had not been described. This study investigates the histopathological and immuno histochemistry reactions in organs of bucks acutely infected by Brucella melitensis. Results: Only testis and prepuce of acutely infected bucks showed significantly severe histological lesions. Other internal organs had mild to moderate lesions. However, positive immunohistochemistry stainings were observed in organs except the bulbourethral gland. There was a significant positive correlation between the distribution of B. melitensis and IHC intensity but no significant correlation between the IHC intensity and histopathology lesions. Conclusion: The results indicate that acute brucellosis did not lead to clinical presentation, although B. melitensis was well distributed in various organs of bucks.展开更多

In this study we investigated the vulnerability of South African indigenous unimproved buck semen to oxidative stress induced by an oxidative stress inducing agent, namely, hydrogen peroxide (H2O2). Semen ejaculates were collected from four superior South African indigenous unimproved bucks in a total of ten collections and then each duplicate was treated with different concentrations of H2O2 in presence or absence of Dithiothreitol (DTT). Sperm motility and velocities were determined using the computer aided sperm class analyser (CASA). SYBR-14 and propidium iodide (PI) Live/Dead assay kit was used to determine cell viability and Yo-Pro-1 plus PI Apoptosis kit was used to determine apoptosis. Statistical analysis was performed on the data using SPSS version 17.0 for Windows (SPSS Inc., Chicago, IL). South African indigenous unimproved buck raw semen motility was between 97% with 98% viability and 0% apoptotic cells. Comparisons of the untreated controls at 0 and 3 hrs incubations revealed that after 3 hrs there was overall a decrease in the number viable cells with the majority of remaining cells exhibiting circular movements accompanied by high progressive (PM) and rapid (RAP) motilities. In treated South African indigenous unimproved buck semen, H2O2 marginally increased total motility (TM) with few apoptotic sperm cells while retaining high viability. Also, H2O2 increased straight line distances travelled of more than 4 fold as compared to untreated controls with no circularly moving cells. Moreover, inclusion of DTT, an antioxidant, had minimal effects on TM, RAP, curvilinear velocity (VCL), straight line velocity (VSL), linearity (LIN) and wobble (WOB) but positively affected PM, average path velocities (VAP), apoptosis and viability. Methods: A total of 30 bucks of New Zealand White rabbits were randomly divided into three equal groups as follows: Group 1 was treated as control group and fed on balanced commercial ration. Groups 2 and 3 were treated with 10 and 20 mL of dates extract substituting water in the early morning before watering and fed on balanced commercial ration. This schedule was performed daily for 5 days/week, for an experimental period of 5 weeks. Fertility parameters such as reaction time, potential of hydrogen ion (pH), mass motility, individual progressive motility %, percentage of live sperm and abnormal sperm (%) were measured. Blood serum testosterone level, serum glutathione reduced, nitric oxide, ascorbic acid and malondialdehyde were also determined. Results: The daily oral administration of 10 mL DWE significantly increased the pH, the mass motility and individual progressive motility % compared to the control group. Although, the consumption of 20 mL DWE significantly (P<0.0001) increased the live sperm% and decreased the abnormal sperm % compared to the other two treatments. The administration of date extracts (10 and 20 mL) had significantly (P<0.0001) decreased nitric oxide and glutathione reduced levels compared to the control. On the other hand, it increased significantly the lipid peroxidation, ascorbic acid and testosterone level compared to the control. Conclusions:The aqueous extract of date palm (10-20 mL) could enhance the rabbit bucks fertility and its health performance.展开更多
