Embryonic stem cells(ESCs), i...The relevance of retinal diseases, both in society's economy and in the quality of people's life who suffer with them, has made stem cell therapy an interesting topic forresearch. Embryonic stem cells(ESCs), induced pluripotent stem cells(i PSCs) and adipose derived mesenchymal stem cells(ADMSCs) are the focus in current endeavors as a source of different retinal cells, such as photoreceptors and retinal pigment epithelial cells. The aim is to apply them for cell replacement as an option for treating retinal diseases which so far are untreatable in their advanced stage. ESCs, despite the great potential for differentiation, have the dangerous risk of teratoma formation as well as ethical issues, which must be resolved before starting a clinical trial. i PSCs, like ESCs, are able to differentiate in to several types of retinal cells. However, the process to get them for personalized cell therapy has a high cost in terms of time and money. Researchers are working to resolve this since i PSCs seem to be a realistic option for treating retinal diseases. ADMSCs have the advantage that the procedures to obtain them are easier. Despite advancements in stem cell application, there are still several challenges that need to be overcome before transferring the research results to clinical application. This paper reviews recent research achievements of the applications of these three types of stem cells as well as clinical trials currently based on them.展开更多
目的:探讨姜黄素联合脂肪间充质干细胞(ADSCs)促进大鼠皮肤创面愈合的效应及相关信号通路的研究。方法:用酶消化法分离和培养大鼠ADSCs,流式细胞仪鉴定干细胞表面抗原。取活性良好的第3代细胞,不同浓度(0、2.5、5、10、20、40μmol/L)...目的:探讨姜黄素联合脂肪间充质干细胞(ADSCs)促进大鼠皮肤创面愈合的效应及相关信号通路的研究。方法:用酶消化法分离和培养大鼠ADSCs,流式细胞仪鉴定干细胞表面抗原。取活性良好的第3代细胞,不同浓度(0、2.5、5、10、20、40μmol/L)的姜黄素作用于ADSCs,通过CCK8法测定干细胞的存活率,筛选姜黄素最适宜浓度。40只雄性SD大鼠切除背部直径1 cm全层皮肤,建立皮肤创面模型,按照不同治疗方法分为模型对照组、干细胞组、姜黄素20μmol/L组、姜黄素20μmol/L联合干细胞组,测量记录各组0、3、7、14 d的皮肤创面愈合率。治疗后7 d ELISA法检测大鼠血清成纤维细胞生长因子(FGF)、血管内皮生长因子(VEGF)、转化生长因子-β(TGF-β)、白细胞介素-1β(IL-1β)、IL-6、肿瘤坏死因子α(TNF-α)含量。治疗后14 d HE染色观察各组大鼠皮肤创面情况,Masson染色观察胶原纤维含量,免疫荧光观察干细胞数量及分布情况。荧光定量PCR检测各组大鼠皮肤创面组织内Wnt4、β-catenin mRNA表达。结果:流式细胞仪鉴定ADSCs表面抗原CD44、CD90阳性,CD34阴性。CCK8法检测姜黄素在20μmol/L时对干细胞增殖无明显毒性作用。与模型对照组比较,干细胞组、姜黄素20μmol/L组、姜黄素20μmol/L联合干细胞组的皮肤创面愈合率、胶原纤维面积百分比、血清生长因子FGF、VEGF、TGF-β明显升高(P<0.05),血清炎症因子IL-1β、IL-6、TNF-α明显降低(P<0.05),Wnt4、β-catenin mRNA表达明显上调(P<0.05),与姜黄素20μmol/L联合干细胞组比较,姜黄素组FGF、VEGF、TGF-β含量、胶原纤维面积明显降低,IL-1β、IL-6、TNF-α含量明显升高,Wnt4、β-catenin mRNA明显下调(P<0.05或P<0.01)。姜黄素20μmol/L联合干细胞组皮肤创面中的ADSCs荧光表达明显高于干细胞组(P<0.05),ADSCs主要分布在真皮层和表皮基底层。结论:姜黄素联合ADSCs可促进大鼠皮肤创面愈合,提高移植后干细胞的展开更多
