Since the chromatographic fingerprint was introduced, it has been accepted by many countries to assess the quality and authenticity of Chinese herbal medicine (CHM). However, solely using the chromatographic fingerprint to assay numerous chemicals is not suitable for the assessment of the whole internal quality and pharmacodynamics of CHM. Consequently, it is necessary to develop a rational approach to connecting the chromatographic fingerprint with effective components to assess the internal quality of CHM. For this purpose, a spec^tun-effect relationship theory was proposed and accepted as a new method for the assessment of CHM because of its potential use to screen effective components from CHM. In this paper, we systematically reviewed the application of the spectrum-effect relationship theory in the research of CHM, including research mentality, different chromatographic analysis techniques, data processing technologies, and structure determination. 展开更多

目的对酒女贞子特征图谱与抗氧化活性的相关性进行研究,确定酒女贞子抗氧化活性物质基础。方法运用HPLC测定经典恒温加速试验处理的酒女贞子的特征图谱,并通过1,1-二苯基-2-三硝基苯肼(DPPH)、2,2'-联氨-双(3-乙基苯并噻唑啉-6-磺酸... 目的对酒女贞子特征图谱与抗氧化活性的相关性进行研究,确定酒女贞子抗氧化活性物质基础。方法运用HPLC测定经典恒温加速试验处理的酒女贞子的特征图谱,并通过1,1-二苯基-2-三硝基苯肼(DPPH)、2,2'-联氨-双(3-乙基苯并噻唑啉-6-磺酸)二胺盐(ABTS)和FRAP法测定其抗氧化活性,运用偏最小二乘回归分析法进行数据分析,探究酒女贞子量化特征图谱和抗氧化活性的相关性,筛选活性化合物相关色谱峰。结果从17个匹配特征图谱中筛选出峰1、峰3、峰8、峰10、峰14和峰15与酒女贞子清除DPPH自由基、ABTS自由基能力呈显著正相关;峰3、峰8、峰12和峰13与酒女贞子还原Fe^(3+)能力显著正相关。其中峰3和峰8与酒女贞子清除DPPH自由基、ABTS自由基和还原Fe^(3+)能力均正相关。并确定了峰3为红景天苷、峰12为木犀草苷、峰13为特女贞苷、峰14为橄榄苦苷、峰15为女贞苷、峰17为木犀草素。结论酒女贞子药效不是由特女贞苷和红景天苷等个别指标成分的含量而决定。全面分析与药效相关的化合物群组的含量才能真实反映酒女贞子的质量。 展开更多

The traditional Chinese medicine of Radix Hedysari plays an important role in invigorating gas for ascending, benefiting blood for promoting production of fluid, and promoting circulation for removing obstruction in collaterals, which is consistent with the principle of treatment for osteoporosis. This study is designed to investigate the bioactive components on increasing peak bone mass (PBM) by exploring the spectrum-effect relationship between chromatography fingerprints and effect. Multiple indicators are selected to evaluate the pharmacological activity. In fingerprints, 21 common peaks are obtained, five of which are identified. Furthermore, gray relational analysis (GRA) is a quantitative method of gray system theory and is used to describe the correlation degree of common peaks and pharmacological activities with relational value. 21 components are then divided into three different regions, of which ononin and calycosin play an extremely significant role in increasing PBM. In addition, factor analysis and hierarchical cluster analysis (HCA) are used to screen the optimal producing area for Radix Hedysari. This provides a comprehensive and efficient method to improve the quality evaluation of Radix Hedysari, confirming the bioactive components for PBM-enhancement and further develop its medicinal value. 展开更多

Results By analyzing the spectrum- effect relationships between UPLC fingerprints and toxic effects, the active components which had the toxic effects were obtained. Conclusion This work provides a general model of the combination of UPLC and microcalorimetry to study the spectrum-effect relationships of the five Aconitum L. herbal medicines, which could be used to evaluate the toxic effects and analyze the principal toxic components of the five Aconitum L. herbal medicines. Quality control and screening of active substances in traditional Chinese medicines have been performed using fingerprint analysis. The spectrum-effect relationship between chromatography fingerprints and efficacy of herbal drugs is considered as a potentially useful method for determining active ingredients in complex mixtures. The study was designed to develop a method for determining the bioactive components of a compound Chinese medicine called Tiefeng based on spectrum-effect relationships between high-performance liquid chromatography (HPLC) fingerprints and anticoccidial activities. Four peaks of the established HPLC fingerprint indicate the main bioactive components of this medicine. In addition, pharrnacodynamic atlas was defined and used to assess the anticoccidial activity of Tiefeng from different sources for the first time. We found that the level of anticoccidial activity of Tiefeng was consistent with the degree of similarity between the pharmacodynamic atlas and chromatogram of any sample. Furthermore, effect of this medicine was related with the main active constituents, along with the origin and the harvesting time. 展开更多

目的探讨丹参提取物对糖基化终末产物(AGEs)/缺氧条件下视网膜Müller细胞低氧诱导因子-1α(HIF-1α)表达的药理作用与化学成分之间的关系,并以丹参酮IIA为对照,初步探索丹参治疗糖尿病性视网膜病变(DR)的药效物质基础。方法采用HPL... 目的探讨丹参提取物对糖基化终末产物(AGEs)/缺氧条件下视网膜Müller细胞低氧诱导因子-1α(HIF-1α)表达的药理作用与化学成分之间的关系,并以丹参酮IIA为对照,初步探索丹参治疗糖尿病性视网膜病变(DR)的药效物质基础。方法采用HPLC法建立10批丹参提取物的指纹图谱,获得各特征峰的峰面积。在AGEs/缺氧条件下培养视网膜Müller细胞,并测定丹参提取物给药组及丹参酮IIA对照组HIF-1α的表达量。结合灰色关联分析法与偏最小二乘法回归分析成分与药效之间的谱效相关性。结果根据10批样品指纹图谱共标示出17个特征峰。与模型组相比,10批样品及丹参酮IIA对照组均能降低视网膜Müller细胞HIF-1α的表达量。谱效相关性研究显示峰1、5、6、9、10、11、12、14、15、16代表的化学成分对抑制HIF-1α表达具有较大的贡献。结论初步确定15,16-二氢丹参酮I、隐丹参酮、丹参酮I、丹参酮IIA及6个尚未确定结构的成分(峰1、5、6、9、14、15)为丹参治疗DR的可能药效物质。 展开更多

Poria is an important medicine for inducing diuresis to drain dampness from the middle energizer.However,the specific effective components and the potential mechanism of Poria remain largely unknown.To identify the effective components and the mechanism of Poria water extract(PWE)to treat dampness stagnancy due to spleen deficiency syndrome(DSSD),a rat model of DSSD was established through weight-loaded forced swimming,intragastric ice-water stimulation,humid living environment,and alternate-day fasting for 21 days.After 14 days of treatment with PWE,the results indicated that PWE increased fecal moisture percentage,urine output,D-xylose level and weight;amylase,albumin,and total protein levels;and the swimming time of rats with DSSD to different extents.Eleven highly related components were screened out using the spectrum-effect relationship and LC-MS.Mechanistic studies revealed that PWE significantly increased the expression of serum motilin(MTL),gastrin(GAS),ADCY5/6,p-PKAα/β/γcat,and phosphorylated cAMP-response element binding protein in the stomach,and AQP3 expression in the colon.Moreover,it decreased the levels of serum ADH,the expression of AQP3 and AQP4 in the stomach,AQP1 and AQP3 in the duodenum,and AQP4 in the colon.PWE induced diuresis to drain dampness in rats with DSSD.Eleven main effective components were identified in PWE.They exerted therapeutic effect by regulating the AC-cAMP-AQP signaling pathway in the stomach,MTL and GAS levels in the serum,AQP1 and AQP3 expression in the duodenum,and AQP3 and AQP4 expression in the colon. 展开更多

Objective: This study aimed to identify the main medicinal active components of Panax ginseng(P. ginseng) in the compatibility environment of clinical application. For this purpose, the anti-inflammatory ingredients of P. ginseng were investigated based on its therapeutic effect in Sijunzi Decoction(SJD) which is a widely used traditional Chinese formula.Methods: The fingerprints of 10 batches of SJD consisting of different sources of P. ginseng were established by UPLC technique to investigate the chemical components. At the same time, the antiinflammatory effects of these components were evaluated by dextran sulfate sodium-induced ulcerative colitis mouse model. Grey relational analysis was applied to explore the correlation degree between fingerprints and anti-inflammatory effects in SJD. Lipopolysaccharide-stimulated RAW264.7 murine macrophages were established to evaluate the anti-inflammatory action of the screened effective substances of P. ginseng.Results: According to grey relational analysis, notoginsenoside R1, ginsenoside Rg2 and ginsenoside Rb3of P. ginseng were the major anti-inflammatory contributions in SJD. They had been proven to be closely associated with the anti-inflammatory process of SJD and displayed a close effect compared with SJD by LPS-stimulated RAW264.7 murine macrophages.Conclusion: Our work provides a general strategy for exploring the pharmacological ingredients of P. ginseng in traditional Chinese formulas which is beneficial for establishing the quality standards of traditional herbs in traditional Chinese medicine prescription based on their clinical therapeutic effect. 展开更多

Objective:To explore the anaphylaxis effect and anaphylaxis substances of honeysuckle.Methods:Rat peritoneal mast cells(PMC)were separated and purified,the cells were incubated with compound 48/80(0.02 g/L),physiological saline and honeysuckle extract(120 g/L)at 37°C for 0,15,30,45 and 60 min.Degranulation were observed by optical microscope and transmission electron microscope.Annexin V positive cell rate was detected by flow cytometry to reflect the degranulation rate of PMC.SD rats were supplied with honeysuckle extract by intravenous injection at a dose of 2.25 g/L.After administration,different parameters were analyzed,including the symptoms,histamine(HIS)and tryptase(MCT)levels,which were determined to explore the effect of anaphylaxis.Regression analysis was used to calculate the relationships between the peaks and the pharmacological effects to explore potentially anaphylactoid components.Results:The percentage of Annxin V positive cells and the degranulation ratio were markedly elevated in PMC treated with honeysuckle extract for more than 15 min(P<0.05).HIS and MCT level were significantly elevated after injection of honeysuckle extract for more than 15 min.Morphology of PMC and systemic symptoms were also changed compared with the controlled group(P<0.05).Regression analysis was used to calculate the relationship between peaks and pharmacological effects,and to determine peaks 7,10 and 13 as possible anaphylactoid ingredients.Conclusion:This study established a prospective method to clarify the anaphylactoid components of honeysuckle extract,which would provide guidance for screening anaphylactoid components in traditional Chinese medicine injections containing honeysuckle in the prescription. 展开更多

测定了64株赤芝(Ganoderma lucidum)子实体醇提物清除二苯基苦基苯肼(DPPH)自由基的活性和三萜指纹图谱,采用遗传算法筛选其指纹图谱中的26个共有峰,并标定出其中15个特征峰,运用偏最小二乘回归分析方法(partial least squares regressi... 测定了64株赤芝(Ganoderma lucidum)子实体醇提物清除二苯基苦基苯肼(DPPH)自由基的活性和三萜指纹图谱,采用遗传算法筛选其指纹图谱中的26个共有峰,并标定出其中15个特征峰,运用偏最小二乘回归分析方法(partial least squares regression,PLSR)对指纹图谱中26个三萜峰与醇提物清除DPPH自由基的活性进行关联分析,建立了赤芝子实体三萜类化合物的抗氧化谱效关系方程。在赤芝子实体醇提物三萜指纹图谱的26个共有峰中,峰S1、S4、S9、S10和S26的峰面积与醇提物清除DPPH自由基的活性呈显著正相关关系;峰S3、S11、S15、S18、S19、S22和S25的峰面积与醇提物清除DPPH自由基的活性呈显著负相关关系。分别测定鉴定出的15个峰对应的三萜化合物对DPPH自由基的清除活性,发现与清除DPPH自由基活性呈显著正相关的三萜化合物灵芝酸C2(S4)、灵芝酸B(S9)、灵芝酸LM2(S10)、灵芝醛A(S26)对DPPH自由基的清除活性明显高于其他三萜化合物,与清除DPPH自由基活性呈显著负相关的三萜化合物灵芝酮三醇(S18)、灵芝萜酮二醇(S22)无清除DPPH自由基的活性。另外,利用H2O2诱导的PC12细胞损伤模型进一步验证灵芝酸C2、灵芝酸B、灵芝酸LM2、灵芝醛A和灵芝萜酮二醇在细胞水平的抗氧化活性,发现灵芝酸C2、灵芝酸B、灵芝酸LM2、灵芝醛A具有明显的保护神经细胞氧化损伤的活性,而灵芝萜酮二醇对神经细胞的氧化损伤无保护作用。结果表明,本研究建立的赤芝子实体三萜类化合物抗氧化谱效关系方程具有较高的准确性,能够较全面地反应赤芝子实体醇提物中不同三萜化合物与其抗氧化活性的相关性,可快速锁定有效成分以指导活性成分的分离纯化工作。此外,该方法也为赤芝子实体的质量控制提供了更为有效和准确的方法。 展开更多

目的:分析白及的抗氧化谱效关系,初步探索白及的抗氧化活性成分。方法:通过D101大孔树脂对白及提取物进行水、30%、50%、70%、95%乙醇分段洗脱,收集12批白及抗氧化效果最好的洗脱段作为试验材料分别建立高效液相(High performance liqui... 目的:分析白及的抗氧化谱效关系,初步探索白及的抗氧化活性成分。方法:通过D101大孔树脂对白及提取物进行水、30%、50%、70%、95%乙醇分段洗脱,收集12批白及抗氧化效果最好的洗脱段作为试验材料分别建立高效液相(High performance liquid chromatography,HPLC)指纹图谱和进行抗氧化试验,随后通过稀疏偏最小二乘回归法建模进行指纹图谱和抗氧化效果的谱效分析,根据谱效分析结果进行有效峰的二级质谱(MS/MS)初步鉴定。结果:70%乙醇洗脱段的抗氧化效果最好,12批样品1,1-二苯基-2-三硝基苯肼自由基(1,1-diphenyl-2-picrylhydrazyl,DPPH)试验的IC 50范围为(57.883±1.077~135.948±2.096)μg·mL^-1,2,2'-联氮-双-3-乙基苯并噻唑啉-6-磺酸自由基(2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid),ABTS)试验的IC 50为(11.510±0.074~19.180±0.242)μg·mL^-1,其中12批样品的ABTS试验的IC 50均小于维他命E(Vitamin E,VE)。以DPPH为指标时,峰4、12、13、16为主要药效峰,以ABTS为指标时,峰8、13、15为主要药效峰;峰4、8、12、13、15、16经MS/MS初步鉴定为Lusianthridin,3,7-二羟基-2,4-二甲氧基菲,3-[(4-羟苯基)甲基]-4-甲氧基-9,10-二氢菲-2,7-二醇,Bletillatin D,Blestrianol A,3,3'-二羟基-2-(4-羟苄基)-5-甲氧基联苄,其中Bletillatin D与DPPH和ABTS自由基的清除效果均有较好的相关性,是一个潜在的抗氧化剂。结论:经大孔树脂洗脱后,白及的抗氧化效果显著提升,其抗氧化效果可能与其所含化合物Lusianthridin,3,7-二羟基-2,4-二甲氧基菲,3-[(4-羟苯基)甲基]-4-甲氧基-9,10-二氢菲-2,7-二醇,Bletillatin D,Blestrianol A,3,3'-二羟基-2-(4-羟苄基)-5-甲氧基联苄有关。 展开更多

目的:研究川芎不同提取物UPLC指纹图谱与其抗炎药效之间的谱效关系,确定川芎的活性成分群,初步阐述其网络调控机制。方法:采用UPLC-Q-TOF/MS对川芎8组提取物的成分进行分析,以人支气管上皮细胞为研究对象,考察川芎不同提取物的抗炎作... 目的:研究川芎不同提取物UPLC指纹图谱与其抗炎药效之间的谱效关系,确定川芎的活性成分群,初步阐述其网络调控机制。方法:采用UPLC-Q-TOF/MS对川芎8组提取物的成分进行分析,以人支气管上皮细胞为研究对象,考察川芎不同提取物的抗炎作用。联用灰色关联分析(GRA)和偏最小二乘回归分析(PLSR)2种方法建立其谱效关系;运用分子对接技术预测川芎有效成分的作用靶点,并利用京都基因与基因组百科全书(KEGG)对其进行通路分析,阐释川芎的抗炎作用机制。结果:川芎的95%乙醇提取物,75%乙醇提取物,50%乙醇提取物,25%乙醇提取物,结晶,石油醚萃取物和三氯甲烷萃取物对炎症细胞均具有抗炎的作用,其中川芎95%乙醇提取物的抗炎效果最为显著。确定了3种具有抗炎药效的成分(洋川芎内酯A,Z-藁本内酯和新蛇床内酯);Z-藁本内酯可能通过环氧合酶-2(COX-2),细胞外调节蛋白激酶2(ERK2),蛋白激酶C(PKC),Janus激酶1(JAK1),JAK2,JAK3,核转录因子-кB(NF-кB)抑制蛋白激酶β(IKKβ),肿瘤坏死因子-α(TNF-α)阻碍炎症信号传递,影响下游蛋白的表达,发挥抗炎作用;洋川芎内酯A和新蛇床内酯可能通过COX-2,ERK2,PKC,磷酸酰肌醇3-激酶α(PI3K-α),PI3K-γ,JAK1,JAK2,JAK3,IKKβ,TNF-α阻碍炎症信号传递,影响下游蛋白的表达,发挥抗炎作用。结论:川芎中3种具有抗炎药效的成分为洋川芎内酯A,Z-藁本内酯和新蛇床内酯,通过不同的靶点阻碍炎症的信号传递,影响下游蛋白的表达,从而发挥抗炎作用,揭示了川芎抗炎的主要活性成分、靶点及作用机制,为深入探讨其药理学作用提供了参考。 展开更多
