Complex 3 was structurally characterized by X-ray diffraction method, which crystallizes in orthorhornbic space group P21212 in a unit cell of dimensions a=3.0567(4) nm, b=0.74079(9) nm, c= 1.06198(13) nm, V=2.4047(5) nm^3, Z=4,μ=0.1084 cm^-1. The SOD-like activities of catalytic disrnutation of superoxide anions (O2^-· ) by the complexes were determined by means of modified nitroblue tetrazolium (NBT) photoreduction. The IC50 values of complexes 1, 2, 3 and 4 are 0.072, 0.147, 0.429 and 0.264 μmol·L^-1, respectively展开更多

Liver metastasis is a normal result in Ehrlich Ascites Carcinoma Cell (EAC) infected mice. A partially purified protein with Superoxide Dismutase (SOD)-like activity extracted from leaves of Raphnus sativus was tested...Liver metastasis is a normal result in Ehrlich Ascites Carcinoma Cell (EAC) infected mice. A partially purified protein with Superoxide Dismutase (SOD)-like activity extracted from leaves of Raphnus sativus was tested for its ability to down regulate liver metastasis of Ehrlich Ascites Carcinoma Cell (EAC) intraperitoneally implanted in albino mice. Sixty albino mice were divided equally into 4 groups. Group I mice (G1 control) were treated with physiologic saline solution for 7 days. Group 2 mice (G2) were injected with the partially purified protein. Group 3 mice (G3) were intraperitoneally inoculated with EAC cells and treated with the partially purified protein. Group 4 mice (G4) were inoculated EAC cells and treated with physiologic saline solution. The results showed a disturbance in liver functions. This disturbance was observed as a reduction in the values of serum albumin, hemoglobin, total counts of erythrocytes and platelets, total lipids in liver tissues, erythrocytes SOD activity, serum glucose, reduced glutathione (GSH) and the trace elements (Fe, Mn, Zn, and Cu) contents in liver and spleen tissues as well as the elevation of serum transaminases activities, the levels of malondialdehyde in blood, DNA and RNA of liver tissues in the infected mice (G4). On the other hand, in G3, after treatment with the partial purified protein all these parameters restored their normal values and became near from the normal group (GI). Moreover, the protein showed no toxicity towards the mice as shown in G2. It could be concluded that a partially purified protein with Superoxide Dismutase (SOD)-like activity extracted from leaves of Raphnus sativus as a nature source has a promised future in controlling liver metastasis.展开更多
Aim: In order to decipher the mechanisms underlying the known protective effects of the thermal water from Uriage-les-Bains (TWFULB) on the skin barrier function, we studied its antioxidant properties as well as its e...Aim: In order to decipher the mechanisms underlying the known protective effects of the thermal water from Uriage-les-Bains (TWFULB) on the skin barrier function, we studied its antioxidant properties as well as its effect on the expression of the tight-junctional protein claudin-6, a candidate tumor suppressor factor. Study Design/Methods: In a first step, TBARS and SOD activity assays were performed in an in vitro model of human dermal fibroblasts treated by hypoxanthine/xanthine oxidase (HO/XO) mixture, in order to evaluate the own antioxidant effect of the thermal water. In a second step, human keratinocytes irradiated or not by UVB were used to evaluate the protective role of TWFULB on nuclear DNA damage using a comet assay. In a third step, an ex vivo model of human skin explants irradiated or not by UVA and UVB was used to evaluate the effect of TWFULB on the intracellular catalase activity and on the cutaneous claudin-6 expression. Results: TWFULB showed significant protective effects against oxidative stress induced by HO/XO: the cell viability was improved and the lipid peroxidation was reduced. The tested thermal water also showed significant SOD-like activity and protective effect on the UVB-stressed DNA. Considering the ex vivo models of skin explants, TWFULB was able to counterbalance the “negative” effect of UVB on the intracellular catalase activity and on the cutaneous claudin-6 expression.Conclusion: This multiparametric approach shows the antioxidant activity of TWFULB and emphasizes its role in the DNA protection of the cutaneous tissue in front of the UV irradiations, and finally suggests that some effects could involve the candidate suppressor functions of claudin-6.展开更多

Two novel complexes [Cu(TATP)(L-Leu)(H2O)]NO3·H2O and [Cu(TATP)L-Met)(H2O)]NO3·0.5H2O(TATP=1,4,8,9-tetraazatriphenylene,L-Leu=L-Leucinate,L-Met=L-Methioninate)were synthesized and characterized b...Two novel complexes [Cu(TATP)(L-Leu)(H2O)]NO3·H2O and [Cu(TATP)L-Met)(H2O)]NO3·0.5H2O(TATP=1,4,8,9-tetraazatriphenylene,L-Leu=L-Leucinate,L-Met=L-Methioninate)were synthesized and characterized by elemental analysis,molar conductivity,infrared absorption spectrum,electronic absorption spectrum and DTA-TGA.The results showed that both of the complexes had a distorted square-pyramidal geometry with two nitrogen atoms of TATP and one amino nitrogen and one carboxylate oxygen atoms of L-Leu/L-Met at the equatorial positions,and one water oxygen at the axial position.The SOD-Like activity of the complexes was determined by NBT-illumination method.The result indicated that the complexes possessed excellent SOD-like activity and showed 50% inhibition ration in the concentration of 0.355 μmol·L-1 and 0.185 μmol·L-1,respectively.The greater SOD-like activity of the complex[Cu(TATP)(L-Leu)(H2O)]NO3·H2O may be attributed to the smaller spatial hindrance of the side-chain group of L-Met.展开更多

文摘乳酸菌发酵果蔬汁是最受关注的益生菌饮品之一,不同果蔬汁经乳酸菌发酵后其抗氧化性等品质变化不一,影响着消费者的选择。用植物乳杆菌分别发酵脐橙、蜜柚、芒果、苹果、梨、西瓜、番茄和青瓜。各果蔬汁经48 h发酵后,活菌数均能达9.0 lg (CFU/mL)以上;苹果、番茄、脐橙、蜜梨的DPPH自由基清除率基本持平,蜜柚、芒果和西瓜的显著上升,且果蔬汁发酵后的总酚含量和DPPH自由基清除率两者呈极显著的正相关关系(p<0.01);除青瓜汁外,其余果蔬汁的类超氧化物歧化酶(SOD-like)活性增加14%~55%不等,其中芒果、蜜柚的增幅最大。在感官上,乳酸菌发酵能提升果蔬汁风味、保护色泽,脐橙、蜜柚、芒果、苹果和梨发酵后感官总体接受度提高。发酵过程合成或释放的酚类及其他SOD-like成分,活菌数的增加及产酸都有利于果蔬汁的总酚含量及相关抗氧化性的保持及增加。
