[Objective] The paper was to study the effect of low temperature stress on physiological and biochemical characteristics of Podocarpus nagi. [Method] Through the determination of physiological indices such as plasma membrane permeability, free proline content, soluble sugar content, malondialdehyde (MDA) content and chlorophyll content, the change law of these indices of P. nagi seedlings under natural conditions and artificially controlled low temperature conditions within the continuous 5 d was studied. [Result] The soluble sugar content, free proline content, MDA content and plasma membrane permeability of P. nagi seedlings were slightly in- creased compared with control; the chlorophyll content gradually decreased with the prolongation of low temperature stress. P. nagi seedlings produced active response to low temperature stress, so the low temperature injury P. nagi seedlings suffered was reduced. [Conclusion] The study provided theoretical basis for winter management in P. nagi cultivation in Hanjiang plain area. 展开更多

Symbiotic nitrogen fixation of rhizobia and leguminous plants is considered as the most important biologic nitrogen fixation system on earth. Symbiotic nodulation of gymnosperm Podocarpus macro-phyllus and rhizobia has never been reported. In this study, 11 endophytic bacteria strains were isolated from root nodules of P. macrophyllus and its variation P. macrophyllus var. maki. The plant infection tests on these strains indicated that the isolated strains could be nodulated on P. macrophyllus plants, and weak nitrogenase activity of nodules was found in acetylene reduction method. According to the physiological and biochemical characteristics of the 11 strains, GXLO 02 was selected as the representative strain. 16S rDNA full-length sequence analysis of GXLO 02 confirmed that the representative strain GXLO 02 belongs to Rhizobium sp. 展开更多

利用扫描电镜对罗汉松属8种2变种植物叶角质层内外表面进行了细致观察。发现罗汉松属植物叶角质层结构具有许多相似特征,表皮细胞较为规则,长方形或多边形,边缘波状弯曲;气孔器排列成带状,长轴均与叶脉一致,气孔器具较为明显的气孔塞和... 利用扫描电镜对罗汉松属8种2变种植物叶角质层内外表面进行了细致观察。发现罗汉松属植物叶角质层结构具有许多相似特征,表皮细胞较为规则,长方形或多边形,边缘波状弯曲;气孔器排列成带状,长轴均与叶脉一致,气孔器具较为明显的气孔塞和伏罗林环,气孔器保卫细胞极延伸明显,通常具有2~4个副卫细胞、不具极副卫细胞。但罗汉松属叶角质层结构也具有明显的种间差异,镰叶罗汉松和洛杉矶罗汉松同其它种类差异最大,这两种植物叶两面均具气孔器,角质层内表面垂周壁直,角质层凸缘不明显;贺氏罗汉松最为显著的特征是近轴面和远轴面表皮细胞的垂周壁角质层厚且凸缘均极其发达;小叶罗汉松近轴面表皮细胞排列较为规则,多数为方形,长轴与叶脉垂直,垂周壁之间的角质层突起较为显著,延伸到皮下层;兰屿罗汉松近轴面表皮细胞排列较不规则,多边形,细胞的角端比较钝,没有棱角;大理罗汉松气孔带间隔较小,有时两条气孔带挤在一起,使副卫细胞紧连,近轴面表皮细胞较短,方形或长方形,垂周壁之间的角质层较不发达;海南罗汉松角质层气孔带间隔较宽,气孔器形状为阔椭圆形,近轴面表皮细胞均为细长方形;变种短叶罗汉松和狭叶罗汉松与罗汉松也具有明显差异,短叶罗汉松近轴面表皮细胞排列不规则,垂周壁深波状弯曲,凸缘极为明显,但原种罗汉松近轴面表皮细胞排列较为规则,垂周壁浅波状弯曲,凸缘不明显,而狭叶罗汉松近轴面表皮细胞方形或长方形,比罗汉松的表皮细胞短,垂周壁直或略弯曲,角质层极厚。这些角质层微形态特征差异可以作为罗汉松属内种类分类鉴定的依据。 展开更多

The leaf development of Podocarpus macrophyllus var.maki Endl.started from the leaf primordium formed by the cell division in shoot apical meristem.During the leaf development,three resin canals from the inferior part of vascular bundles firstly began to differentiate,then protoxylem in vascular bundles differentiated from adaxial side to abaxial side.Meanwhile,a layer of parenchyma cells from the inferior part of protoderm developed into the sclerenchyma,which played an important role in supporting young leaves.When xylem became mature basically,transfusion tissue,phloem and accessory transfusion tissue began to differentiate,so mesophyll tissue finally differentiated into developmental palisade cells and sponge cells.The thicker cuticle was formed in the lateral side of epidermal cells at the same time of differentiation and development for the inner structure of leaves.Therefore,the developmental palisade cells and the cuticle in the leaf structure of Podocarpus macrophyllus var.maki Endl.is greatly significant for photosynthesis and transpiration when it grows in arid and cold regions. 展开更多

The conducting tissue structure of transverse and longitudinal sections was observed on leaves of Podocarpus and Nageia.Results showed:in Podocarpus leaves,there is only one midrib,the xylem tracheid of midrib vascular bundle is multi-form,transfusion tissue belongs to Cycas-type and transfusion tracheids are isodiametric,the accessory transfusion tracheids between palisade tissue and sponge tissue are developed;in Nageia leaves,there are plenty of parallel leaves,the xylem tracheids of each vein are relatively simple,transfusion tissue belongs to Taxus-type and transfusion tracheids are longer in longitudinal section than that in transverse section,the accessory transfusion tissue between palisade tissue and sponge tissue is absent.Considering other differences that in leaves of Podocarpus there are three resin ducts under vascular bundle of midrib,mesophyll cells are differentiated into palisade tissue and sponge tissue;in leaves of Nageia,there is only one resin duct under vascular bundle in each vein and no obvious differentiation in mesophyll cells,palisade tissue can be found on both sides,and sclereids can also be found in mesophyll tissue.The anatomical differences of leaf veins and mesophylls between Nageia and Podocarpus mentioned above support the viewpoint that Nageia and Podocarpus are two independent genera. 展开更多

Effect of Isolation of Hainan Island from the Mainland on the Genetic Variation of Podocarpus imbricatus

Random amplified polymorphic DNA (RAPD) markers were used to analyze genetic variation of Podocarpus imbricatus in Hainan Island and Mainland of China. Two populations of Dacrydium pierrei were used as comparison materials. Both Podocarpus imbricatus and Dacrydium pierrei showed a low level of genetic diversity. However, Podocarpus imbricatus showed higher genetic diversity and higher population differentiation than Dacrydium pierrei. The geographic range may affect the genetic diversity of Podocarpus imbricatus and Dacrydium pierrei significantly. The UPGMA cluster tree showed that populations of Podocarpus imbricatus in Hainan Island and Guangxi Zhuang Autonomous Region were closer than those in Yunnan Province, indicating possible gene flow between Hainan Island and Guangxi Zhuang Autonomous Region. The young geological history of Hainan Island and the three times of unification and separation between Hainan Island and the Mainland may give the two species more possibilities of gene flow. 展开更多

Nonstructural carbon(NSC),which represents the relationship between the carbon source and carbon sink,is an important factor that reflects the functions and performance of a tree.However,little is known regarding the timeseries responses of NSC storage in evergreen species to different nitrogen(N)fertilization regimes.This study,which was based on a pot experiment,examined the response of the NSC(soluble sugars and starch storage to different N addition intensities[light N addition(LN):6.5 g N m^(-2)a^(-1);moderate N addition(MN):13.0 g N m^(-2)a^(-1);and heavy N addition(HN):26.0 g N m^(-2)a^(-1))]in saplings of the evergreen species Podocarpus macrophyllus.Our results showed that the net photosynthetic rate(P_(n))under MN was significantly higher than that under LN,but was comparable to that under HN.Moreover,saplings subject to MN had a significant higher leaf biomass than that to LN and HN.These results indicated that the C supply via photosynthesis under MN was greater than that under LN and HN.The NSCs reserve under MN was considerable with that under LN,which suggested that saplings in MN group consumed higher and stored lower properties of NSCs than those in LN group.However,saplings under HN stored higher properties of NSCs than those under MN considering that no difference in NSCs pools was found between the two treatments.The leaf N concentrations were found in the increasing sequence of LN<MN<HN,whilst the leaf chlorophyll concentration under HN was obviously lower than that under MN.The growth rate under MN was higher than that under LN and HN.We concluded that the NSCs allocation between con-sumption and reserve in P.macrophyllus saplings depended on soil N availability,and an excessive N addition to soil favors the storage rather than the consumption of NSCs by plants. 展开更多

Bioactivity-guided phytochemical investigation of Podocarpus neriifolius D.Don.(Podocarpaceae)has led to the isolation of one new(2)and three known(1,3,and 4)B-type podolactones,along with three totarane-type diterpenes(5-7).Their structures were determined by interpretation of High Resolution ElectroSpray Ionization Mass Spectrometry(HRESIMS)and 1D and 2D NMR data,and comparison with the values reported in the literature.The structure of compound 1,previously identifed as 3-deoxy-2α-hydroxynagilactone E(8),was revised as its 2β-epimer,which has been reported recently as a new compound.All of the isolates were evaluated for their antiproliferative activity against a panel of four human cancer cell lines,namely,ovarian(OVCAR3),breast(MDA-MB-231),colon(HT-29),and melanoma(MDA-MB-435),and compounds 1 and 3 were found to be cytotoxic with IC_(50) values in the low micromolar range for most of the cell lines used.The major compound,inumakilactone A(3),was further tested in vivo using the HT-29,MDA-MB-435,and OVCAR3 cells in a murine hollow fber model,for the frst time. 展开更多
