However, the effect of coexisting polymorphisms of other genes has not yet been reported in the Chinese population. This study investigated the effect of coexisting polymorphisms of CYP2C19 and P2Y12 on clopidogrel responsiveness and adverse clinical events in Chinese patients. Methods In 577 Han Chinese patients undergoing stent placement because of acute coronary syndrome had platelet reactivity assessed by thromboelastography, and the CYP2C19 G681A and P2Y12 C34T polymorphisms were detected by the ligase detection reaction. Primary clinical endpoints included cardiovascular death, nonfatal myocardial infarction, target vessel revascularization, and stent thrombosis. The secondary clinical endpoints were thrombolysis in myocardial infarction bleeding. The follow-up period was 12 months. Results Genotyping revealed 194 carriers of the wild type GG genotype of CYP2C19 and the wild type CC genotype of P2Y12 (group 1), 102 carriers of the wild type GG genotype of CYP2C19 and the mutational T allele of P2Y12 (group 2), 163 carriers of the mutational A allele of CYP2C19 and the wild type CC genotype of P2Y12 (group 3), and 118 carriers of the mutational A allele of CYP2C19 and the mutational T allele of P2Y12 (group 4). Group 4 had the lowest ADP-inhibition (49.74+32.61) and the highest prevalence of clopidogrel low response (29.7%) of the four groups. The rate of the composite of primary clinical endpoints increased more in group 4 (8.5%) than in the other three groups; the rate of composite primary endpoints in group 2 (2.9%) and group 3 (3.7%) were not significantly different than that of group 1 (1.5%). Conclusion Coexisting polymorphisms of different genes affected clopidogrel responsiveness and clinical outcome more than single polymorphism in Chinese patients with acute coronary syndrome undergoing percutaneous coronary intervention.展开更多

Sequence analyses indicated that the agent was closely related to the ...A novel agent (hence termed as P2) was isolated from pig sera in China, which contained covalently bound circular genomic DNAs of 993 nucleotides. Sequence analyses indicated that the agent was closely related to the porcine circovirus (PCV). The molecular clone of P2 was constructed subsequently and used for the following studies. Intracytoplasmic inclusions and intranuclear inclusions were only found in PK-15 cells transfected with the tandem dimer of P2 molecular DNA clone. Intracytoplasmic inclusions were round or irregular in shape and 0.1-0.4 μm in diameter, and intranuclear inclusions were electronically denser than intracytoplasmic inclusions and had two general shapes: round/small (0.1 μm in diameter) and hexagonal/large (0.5―1.4 μm in diameter). The inclusions were not membranously bound. The cells transfected with the tandem dimer of P2 molecular DNA clone were tested positive for P2 DNA at passages 5. The P2 antigen could be detected in both transfected and passaged PK-15 cells. This is the first report regarding the complete nucleotide sequence of a small DNA genome in a circovirus-like infectious agent in vitro.展开更多
A biosorbent prepared from powdered mycelial biomass of Ceriporia lacerata (CLB), a basidiomycetous fungus, was applied for the uptake of Crystal Violet from aqueous solution. A batch adsorption experiment was used ...A biosorbent prepared from powdered mycelial biomass of Ceriporia lacerata (CLB), a basidiomycetous fungus, was applied for the uptake of Crystal Violet from aqueous solution. A batch adsorption experiment was used for the biosorption process, involving effect of experimental factors and biosorption kinetics and equilibrium. Biosorption process showed that the removal of Crystal Violet by CLB was effective over wide pH range, and meanwhile was independent on ionic strength. Biosorption capacities of CLB increased with the initial dye concentration increasing, due to an increase in the driving force of the concentration gradient. The adsorbed Crystal Violet amount per unit biomass weight decreased with increasing biosorbent dosage, due to the splitting effect of flux (concentration gradient) between sorbate and biosorbent. A maximum sorption capacity of 239.25 mg/g was observed. Biosorption kinetics was found to be best represented by the pseudo second-order kinetic model. The equilibrium adsorption data was well described by the Koble-Corrigan model. FT-IR (Fourier Transform Infrared Spetroscopy) spectrum showed the presence of O–H, COOH, C=O, C–N, C–H, –NH2 and P–OH in the surface of CLB as functional groups. This study showed CLB can effectively remove CV from dye wastewater.展开更多
