Customized molecular tools to strengthen metabolic engineering of cyanobacteria

Cyanobacteria are promising oxygenic phototrophs for the production of various compounds.For their(photo)biotechnological exploitation,molecular tools are required,such as,for the introduction and expression of heterologous genes,or the modulation of enzyme activities or entire pathways.Concepts and strategies for the development of photosynthetic biomanufacturing technologies based on cyanobacteria have been extensively reviewed,as well as certain specialized aspects of their genetic manipulation.However,options for metabolic engineering of specific cyanobacterial cells are still less developed than those for other bacteria of biotechnological relevance.In addition to the standard genetic toolbox for“classical”metabolic engineering,we emphasize certain aspects,including recently developed vector systems for the extrachromosomal maintenance of genes and approaches based on clustered regularly interspaced short palindromic repeats(CRISPR)interference.We highlight the development of custom molecular tools for specific strains or products,discuss the emerging use of small regulatory proteins that appear promising for advanced metabolic engineering approaches to promote specific product formation,and provide an overview of suitable online resources.Furthermore,we discuss the current trends in this field and indicate their potential,such as using suitable product sensors that enable systematic screening,and optimization approaches. 展开更多

Sirtuins(SIRTs) are NAD+-dependent histone deacetylases and play a role in virtually all cell biological processes. As SIRTs functions vary according to their subtypes, they can either activate or inhibit signaling pathways upon different conditions or tissues. Recent studies have focused on metabolic effects performed by SIRTs in several cell types since specific metabolic pathways(e.g., aerobic glycolysis, oxidative phosphorylation, β-oxidation, glutaminolysis) are used to determine the cell fate. However, few efforts have been made to understand the role of SIRTs on B lymphocytes metabolism and function. These cells are associated with humoral immune responses by secreting larger amounts of antibodies after differentiating into antibody-secreting cells. Besides, both the SIRTs and B lymphocytes are potential targets to treat several immune-mediated disorders, including cancer. Here, we provide an outlook of recent studies regarding the role of SIRTs in general cellular metabolism and B lymphocytes functions, pointing out the future perspectives of this field. 展开更多

Reactive Arrays of Colorimetric Sensors for Metabolite and Steroid Identification

The work described herein examines a rapid mix-and-measure method called DETECHIP suitable for screening of steroids and metabolites. The addition of steroids and metabolites to reactive arrays of colorimetric sensors generated characteristic color “fingerprints” that were used to identify the analyte. A color analysis tool was used to identify the analyte pool that now includes biologically relevant analytes. The mix-and-measure arrays allowed the detection of disease metabolites, orotic acid and argininosuccinic acid;and the steroids androsterone, 1,4-androstadiene, testosterone, stanozolol, and estrone. The steroid 1,4-androstadiene was also detected by this method while dissolved in synthetic urine. Some of the steroids, such as androstadiene, stanozolol, and androsterone were co-dissolved with (2-hydroxypropyl)-β-cyclodextrin in order to increase solubility in aqueous buffered solutions. The colorimetric arrays do not intend to eliminate ELISA or mass spectroscopy based screening, but to possibly provide an alternative analytical detection method for steroids and metabolites. 展开更多
