ACSLs on endoplasmic reticulum and mitochondrial outer membrance catalyze fatty acids with chain lengths from 12 to 20 carbon atoms to form acyl-Co As, which are lipid metabolic intermediates and involved in fatty acid metabolism, membrane modifications and various physiological processes. Gain- or lossof-function studies have shown that the expression of individual ACSL isoforms can alter the distribution and amount of intracellular fatty acids. Changes in the types and amounts of fatty acids, in turn, can alter the expression of intracellular ACSLs. ACSL family members affect not only the proliferation of normal cells, but the proliferation of malignant tumor cells. They also regulate cell apoptosis through different signaling pathways and molecular mechanisms. ACSL members have individual functions in fatty acid metabolism in different types of cells depending on substrate preferences, subcellular location and tissue specificity, thus contributing to liver diseases and metabolic diseases, such as fatty liver disease, obesity, atherosclerosis and diabetes. They are also linked to neurological disorders and other diseases. However, the mechanisms are unclear. This review addresses new findings in the classification and properties of ACSLs and the fatty acid metabolismassociated effects of ACSLs in diseases.展开更多

According to the similarity of compositions and distributions of the alkenones determined commonly in the sediment section to those of marine sediment, it is reasonably referred that the alkenones might also originate from Prymnesiophyceae. ln addition, the parameter U<sub>37</sub><sup>k</sup>'of alkenone is in positive correlation to δ<sup>18</sup>O values of authigenic calcium and magnesium minerals. Carbon preference index (CPI) of long-chain n-alkanes preserved in surface soil increases gradually from southeastern China to the north margin of Loess Plateau.Along this latitudinal transect,the CPI value correlates...Carbon preference index (CPI) of long-chain n-alkanes preserved in surface soil increases gradually from southeastern China to the north margin of Loess Plateau.Along this latitudinal transect,the CPI value correlates to relative humidity,precipitation,and temperature with a negative linear relationship,respectively,whereas the correlation of CPI to temperature is relatively weak.In the Wuyi,Shennongjia,and Tianshan Mountains,CPI values do not change systemically with altitude increasing (or temperature decreasing).However,mean value of CPI for the individual mountain increases in turn from the humid mountain to the arid.These results jointly suggest that aridity (or humidity) is a dominate climate factor in altering soil CPI value.High CPI values of geological records therefore indicate the arid paleoclimate.Though long-chain n-alkanes in soil are derived mainly from leaf wax of terrestrial vascular plants,the regular latitudinal variations of soil CPI might not be caused by the change of vegetation.We speculate that increased long-chain n-alkanes from microbes and/or enhanced biodegradation in the humid climate lead to the decrease of soil CPI.展开更多

Howe...The hydrogen isotopic composition(δD) of leaf wax long-chain n-alkanes(C27, C29, and C31) from lacustrine sediments has been widely applied to reconstruct terrestrial paleoclimatic and paleohydrological changes. However, few studies have addressed whether the aquatic-derived n-alkanes can affect the δD values of lake sedimentary long-chain n-alkanes, which are usually regarded as a recorder of the terrestrial hydrological signals. Here we systematically investigated δD values of long-chain n-alkanes from modern aquatic plants, both near-shore and off-shore surface sediments, surrounding terrestrial plant litters, as well as river water and lake water in Lake Qinghai and its satellite lakes on the northeastern Qinghai-Tibet Plateau. Our data showed that(i) δD values of long-chain n-alkanes from aquatic plants varied from-184‰ to-132‰ for n-C27, from-183‰ to-138‰ for n-C29, and from-189‰ to-130‰ for n-C31, respectively, with no significant differences among the three n-alkanes homologues;(ii) δD values of long-chain n-alkanes from aquatic plants were generally more positive than those from surrounding terrestrial plants, possibly because that they recorded the D-enrichment of lake water in this semi-arid region;(iii) δD values of long-chain n-alkanes from surface sediments showed significant differences among the three n-alkanes homologues, due to the larger aquatic input of n-C27 to the sedimentary lipid pool than that of n-C31, and(iv) n-C27 δD values of near-shore aquatic plants and near-shore sediments are more negative than those from off-shore as a result of lower δD values of near-shore lake water. Our findings indicate that in this region(i) the offset between sedimentary n-C27 and n-C31 δD values(ΔδDC27-C31) could potentially be used to evaluate if sedimentary long-chain n-alkanes are derived from a single source;(ii) while δD values of n-C27 may be influenced by lake water hydrological changes, sedimentary n-C31 is derived predominantly from terrestrial plants and thus its δD can 展开更多
Human milk is considered to be the optimal source of infant nutrition. Some of the benefits of breastfeeding have been ascribed to human milk oligosaccharides(HMO). For instance, HMO can affect faecal characteristics ...Human milk is considered to be the optimal source of infant nutrition. Some of the benefits of breastfeeding have been ascribed to human milk oligosaccharides(HMO). For instance, HMO can affect faecal characteristics such as stool consistency and stool frequency. Such effects on stool characteristics can be beneficial for young infants as hard stools and even constipation is common in that age group. Prebiotics in infant milk formulas have been introduced to exert similar functionalities. A specific mixture of prebiotics consists of a combination of short chain galacto-oligosaccharides and long-chain fructo-oligosaccharides(scGOS/lcFOS) in a ratio of 9:1. This specific mixture has been developed to closely resemble the molecular size composition of HMO. Many studies have been done with scGOS/lcFOS, and indicators for digestive comfort have often been included as secondary outcomes. This review summarizes the effects of scGOS/lcFOS(9:1) on stool consistency,stool frequency and transit time in healthy term and preterm infants. In several of the studies with scGOS/lcFOS in a ratio of 9:1 in infant milk formulas, positive effects of this mixture on stool characteristics such as stool consistency and stool frequency were observed. As stool consistency was shown to be correlated to whole gut transit time, scGOS/lcFOS can be hypothesised to have a role in reducing the risk of constipation.展开更多
Plant sphingolipids are not only structural components of the plasma membrane and other endomembrane systems but also act as signaling molecules during biotic and abiotic stresses.However,the roles of sphingolipids in...Plant sphingolipids are not only structural components of the plasma membrane and other endomembrane systems but also act as signaling molecules during biotic and abiotic stresses.However,the roles of sphingolipids in plant signal transduction in response to environmental cues are yet to be investigated in detail. In this review,we discuss the signaling roles of sphingolipid metabolites with a focus on plant sphingolipids.We also mention some microbial sphingolipids that initiate signals during their interaction with plants, because of the limited literatures on their plant analogs.The equilibrium of nonphosphorylated and phosphorylated sphingolipid species determine the destiny of plant cells,whereas molecular connections among the enzymes responsible for this equilibrium in a coordinated signaling network are poorly understood.A mechanistic link between the phytohormone-sphingolipid interplay has also not yet been fully understood and many key participants involved in this complex interaction operating under stress conditions await to be identified.Future research is needed to fill these gaps and to better understand the signal pathways of plant sphingolipids and their interplay with other signals in response to environmental stresses.展开更多

Methods Twenty species were collected from the Zhousha...Objective To assess the net health effect caused by the consumption of specific marine species based on Benefit-Risk Analysis for Foods (BRAFO)-tiered approach. Methods Twenty species were collected from the Zhoushan Archipelago, China. Concentrations of n-3 long-chain polyunsaturated fatty acids, methyl mercury (MeHg), and dioxin-like compounds (DLCs) in the samples were analyzed for benefit risk assessment based on BRAFO-tiered approach. Results Based on the BRAFO-tiered approach, reference scenario (no intake) and alternative scenario (intake of specific species of 200 g/week) were determined. The exposure to MeHg/DLCs via alternative scenario of all studied species did not exceed provisional tolerable weekly/monthly intake. However, the adult population with high DLCs exposure in China would significantly exceed the upper limit of DLCs via an additional alternative scenario of some species such as Auxis thazard. The results of deterministic computation showed that alternative scenario of all studied species generated clear net beneficial effects on death prevention and child IQ gain. Conclusion The alternative scenario of all studied species could be recommended to population with average DLCs exposure, and the reference scenario of species with relatively high DLCs concentration could be recommended to population exposed to high DLCs.展开更多
Rationally tailored lipid nanoparticles(LNPs)with efficient and tunable delivery of mRNA in vivo are crucial for mRNA vaccines.Selective expression of antigenic protein in lymphoid tissues/organs could improve the imm...Rationally tailored lipid nanoparticles(LNPs)with efficient and tunable delivery of mRNA in vivo are crucial for mRNA vaccines.Selective expression of antigenic protein in lymphoid tissues/organs could improve the immunostimulatory efficacy and safety of LNPs-based mRNA vaccines.Inspired by the metabolic behavior that long-chain saturated fatty acids tending to enter lymphoid tissue rather than the liver,we developed fatty acid-doped LNPs capable of mediating differential protein expressions in the liver and spleen when administered intravenously.When the molar ratio of saturated fatty acid located 60%–70%,the doped LNPs achieved the spleen selective mRNA translation.The mechanism could be attributed to the different cellular uptake behaviors of saturated fatty acids in hepatocytes.Immunization with a model antigen(ovalbumin)mRNA-loaded spleen selective LNPs,we observed enhanced antigen-specific T cell immune responses,and potent immunotherapeutic and immunoprophylactic efficacy in the mouse lymphoma model.Our natural long-chain saturated fatty acids metabolic characteristics-inspired design of LNPs for spleen-selective mRNA vaccines delivery will provide references for designing mRNA vaccines with high efficacy and safety for tumor immunotherapy.展开更多
BACKGROUND Hepatorenal syndrome(HRS)is the most prevalent form of acute kidney injury in cirrhotic patients.It is characterized by reduced renal blood flow and represents the most severe complication in cirrhotic pati...BACKGROUND Hepatorenal syndrome(HRS)is the most prevalent form of acute kidney injury in cirrhotic patients.It is characterized by reduced renal blood flow and represents the most severe complication in cirrhotic patients with advanced disease.Previous research has indicated that antioxidants can delay the onset of a hyperdynamic circulatory state in cirrhosis and improve renal function in HRS patients.Regular omega-3 supplementation has significantly reduced the risk of liver disease.This supplementation could represent an additional therapy for individuals with HRS.AIM To evaluated the antioxidant effect of omega-3 polyunsaturated fatty acid supplementation on the kidneys of cirrhotic rats.METHODS Secondary biliary cirrhosis was induced in rats by biliary duct ligation(BDL)for 28 d.We used 24 male Wistar rats divided into the following groups:I(control);II(treated with omega-3,1 g/kg of body weight);III(BDL treated with omega-3,1 g/kg of body weight);and IV(BDL without treatment).The animals were killed by overdose of anesthetic;the kidneys were dissected,removed,frozen in liquid nitrogen,and stored in a freezer at-80℃for later analysis.We evaluated oxidative stress,nitric oxide(NO)metabolites,DNA damage by the comet assay,cell viability test,and apoptosis in the kidneys.Data were analyzed by one-way analysis of variance,and means were compared using the Tukey test,with P≤0.05.RESULTS Omega-3 significantly decreased the production of reactive oxygen species(P<0.001)and lipoperoxidation in the kidneys of cirrhotic rats treated with omega-3(P<0.001).The activity of the antioxidant enzymes superoxide dismutase and catalase increased in the BDL+omega-3 group compared to the BDL group(P<0.01).NO production,DNA damage,and caspase-9 cleavage decreased significantly in the omega-3-treated BDL group.There was an increase in mitochondrial electrochemical potential(P<0.001)in BDL treated with omega-3 compared to BDL.No changes in the cell survival index in HRS with omega-3 compared to the control group(P>0.05)were obs展开更多
In order to obtain a uniform and effectively toughened poly(lactic acid)film by blending with low content of poly(ethylene octene)(POE)with high elasticity,the tailored interfacial intermolecular interaction and entan...In order to obtain a uniform and effectively toughened poly(lactic acid)film by blending with low content of poly(ethylene octene)(POE)with high elasticity,the tailored interfacial intermolecular interaction and entanglement between the two phases of the PLA/POE blend was innovatively constructed via the facile reactive melt blending process through the reaction of the epoxy/anhydride groups grafted on the POE chains with the end groups of PLA chains(PLA/GPOE-MPOE).It was observed that POE domains were embedded tightly in PLA matrix with a fuzzy interface and abundant interface transition area,and the impact fractured surface of the blend showed an obvious plastic deformation with less occurrence of fibrillation of PLA matrix or interfacial de-bonding.Compared with neat PLA and directly blended PLA/POE blends,the PLA/GPOE-MPOE blend exhibited much higher complex viscosity/storage modulus,much lower tanδvalues in the terminal region,and obvious strain-hardening behavior.The deviation in viscoelastic behavior of PLA/GPOE-MPOE from linear PLA indicated the enhanced molecular entanglement between the long-branched chains,resulting in an enhancement of the stretching ability during biaxial drawing of the blend.Uniform PLA/GPOE-MPOE films with draw ratio as high as 7×7 were obtained through biaxial stretching,which showed much higher tensile strength and the elongation at break than that of neat PLA and PLA/POE film.This work provides a facile method for fabricating toughening PLA films with application potentials.展开更多

Previous studies have reported upregulation of heme oxygenase-1 in different central nervous system injury models.Heme oxygenase-1 plays a critical anti-inflammatory role and is essential for regulating cellular redox...Previous studies have reported upregulation of heme oxygenase-1 in different central nervous system injury models.Heme oxygenase-1 plays a critical anti-inflammatory role and is essential for regulating cellular redox homeostasis.Metformin is a classic drug used to treat type 2 diabetes that can inhibit ferroptosis.Previous studies have shown that,when used to treat cardiovascular and digestive system diseases,metformin can also upregulate heme oxygenase-1 expression.Therefore,we hypothesized that heme oxygenase-1 plays a significant role in mediating the beneficial effects of metformin on neuronal ferroptosis after spinal cord injury.To test this,we first performed a bioinformatics analysis based on the GEO database and found that heme oxygenase-1 was upregulated in the lesion of rats with spinal cord injury.Next,we confirmed this finding in a rat model of T9 spinal cord compression injury that exhibited spinal cord nerve cell ferroptosis.Continuous intraperitoneal injection of metformin for 14 days was found to both upregulate heme oxygenase-1 expression and reduce neuronal ferroptosis in rats with spinal cord injury.Subsequently,we used a lentivirus vector to knock down heme oxygenase-1 expression in the spinal cord,and found that this significantly reduced the effect of metformin on ferroptosis after spinal cord injury.Taken together,these findings suggest that metformin inhibits neuronal ferroptosis after spinal cord injury,and that this effect is partially dependent on upregulation of heme oxygenase-1.展开更多
This study investigated the effects of the degree of unsaturation(unsaturity) of long-chain fatty acids on microbial protein content and the activities of transaminases and dehydrogenase in vitro using goat rumen fl...This study investigated the effects of the degree of unsaturation(unsaturity) of long-chain fatty acids on microbial protein content and the activities of transaminases and dehydrogenase in vitro using goat rumen fluid as the cultural medium.Six types of fatty acids,stearic acid(C18:0,group A,control group),oleic acid(C18:1,n-9,group B),linoleic acid(C18:2,n-6,group C),α-linolenic acid(C18:3,n-3,group D),arachidonic acid(C20:4,n-6,group E),and eicosapentaenoic acid(C20:5,n-3,group F),were tested,and the inclusion ratio of each fatty acid was 3%(w/w) in total of culture substrate.Samples were taken at 0,3,6,9,12,18 and 24 h,respectively,during culture for analyses.Compared with stearic acid,linoleic acid,a-linolenic acid,and arachidonic acid increased the bacterial protein content,while oleic acid and eicosapentaenoic acid had no significant effects;the protozoal protein content was reduced for all the unsaturated fatty acids,and the magnitude of the reduction appeared to be associated with the degree of unsaturity of fatty acids.The total microbial protein content was dominantly accounted by the protozoal protein content(about 4-9 folds of the bacterial protein),and only increased by linoleic acid,but reduced by oleic acid,arachidonic acid and eicosapentaenoic acid.There were no significant effects in the activities of both glutamic oxaloacetic transaminase(GOT) and glutamic-pyruvic transaminase(GPT) for all the other fatty acids,except for arachidonic acid which enhanced GOT activity and oleic acid which enhanced GPT activity.The total dehydrogenase activity was positively correlated with the degree of unsaturation of fatty acids.In conclusion,the inclusion of 3%of long-chain unsaturated fatty acids increased bacterial protein content,whereas reduced protozoal protein content and enhanced dehydrogenase activity.The fatty acids with more than three double bonds had detrimental effects on the microbial protein content.This work demonstrates for the first tim展开更多

The analysis result indicates that C37:3 methylketone is predominate in the long-chain alkenones from the Chukchi and Bering Sea sediments. The abundance of C37 to C39 unsaturated alkenones changes in an order of C37 > C38 > C39. Based on C37 /C38 ratio, the detected organism precursors of the long-chain alkenones are mainly coccolithophrid (Emiliania huxleyi). By the calibration relationship between U37k and U37k indices, the sea surface paleotemperature in these seas is estimated. The estimated values of U37k vary from 4.147℃ to 5.706℃ , with a mean value of 5.092℃.展开更多

The processing characteristics, mechanical properties and extraction property of the PVC/LP blends in different solvents (xylene, cyclohexane, ethanol) were also studied in detail. All results were compared with that of the PVC plasticized with dioctyl phthalate (DOP). The results show that the molecular weight, molecular structure and loading of LP greatly influence the mechanical properties of the PVC/LP blends. The processability and the mechanical properties of PVC plasticized by LP are comparable to those of the corresponding PVC/DOP blends. However, the PVC/LP blends posses much better migration resistance property than the corresponding PVC/DOP blends, which makes the long-chain linear polyester become a very good plasticizer candidate for PVC industry.展开更多

There have been no previous studies on the genes encoding AS-sphingo...8-Sphingolipid desaturase is the key enzyme that catalyses desaturation at the C8 position of the long-chain base of sphingolipids in higher plants. There have been no previous studies on the genes encoding AS-sphingolipid desaturases in Brassica rapa. In this study, four genes encoding AS-sphingolipid desaturases from B. rapa were isolated and characterised. Phylogenetic analyses indicated that these genes could be divided into two groups: BrD8A, BrD8C and BrD8D in group I, and BrD8B in group II. The two groups of genes diverged before the separation of Arabidopsis and Brassica. Though the four genes shared a high sequence similarity, and their coding desaturases all located in endoplasmic reticulum, they exhibited distinct expression patterns. Heterologous expression in Saccharomyces cerevisiae revealed that BrD8A/B/C/D were functionally diverse AS-sphingolipid desaturases that catalyse different ratios of the two products 8(Z)- and 8(E)-C18-phytosphingenine. The aluminium tolerance of transgenic yeasts expressing BrD8A/B/C/D was enhanced compared with that of control cells. Expression of BrD8A in Arabidopsis changed the ratio of 8(Z):8(E)-C 18-phytosphingenine in transgenic plants. The information reported here provides new insights into the biochemical functional diversity and evolutionary relationship of AS-sphingolipid desaturase in plants and lays a foundation for further investigation of the mechanism of 8(Z)- and 8(E)-C18- phytosphingenine biosynthesis.展开更多
Long-chain acyl-coenzyme A(CoA)synthase 4(ACSL4)is an enzyme that esterifies CoA into specific polyunsaturated fatty acids,such as arachidonic acid and adrenic acid.Based on accumulated evidence,the ACSL4-catalyzed bi...Long-chain acyl-coenzyme A(CoA)synthase 4(ACSL4)is an enzyme that esterifies CoA into specific polyunsaturated fatty acids,such as arachidonic acid and adrenic acid.Based on accumulated evidence,the ACSL4-catalyzed biosynthesis of arachidonoyl-CoA contributes to the execution of ferroptosis by triggering phospholipid peroxidation.Ferroptosis is a type of programmed cell death caused by iron-dependent peroxidation of lipids;ACSL4 and glutathione peroxidase 4 positively and negatively regulate ferroptosis,respectively.In addition,ACSL4 is an essential regulator of fatty acid(FA)metabolism.ACSL4 remodels the phospholipid composition of cell membranes,regulates steroidogenesis,and balances eicosanoid biosynthesis.In addition,ACSL4-mediated metabolic reprogramming and antitumor immunity have attracted much attention in cancer biology.Because it facilitates the cross-talk between ferroptosis and FA metabolism,ACSL4 is also a research hotspot in metabolic diseases and ischemia/reperfusion injuries.In this review,we focus on the structure,biological function,and unique role of ASCL4 in various human diseases.Finally,we propose that ACSL4 might be a potential therapeutic target.展开更多

According to the similarity of compositions and distributions of the alkenones determined commonly in the sediment section to those of marine sediment, it is reasonably referred that the alkenones might also originate from Prymnesiophyceae. ln addition, the parameter U<sub>37</sub><sup>k</sup>'of alkenone is in positive correlation to δ<sup>18</sup>O values of authigenic calcium and magnesium minerals. This indicates that the parameter U<sub>37</sub><sup>k</sup>'of long-chain alkenone is also available to indicate paleoclimate of high-salinity alkaline lake.

文摘Carbon preference index (CPI) of long-chain n-alkanes preserved in surface soil increases gradually from southeastern China to the north margin of Loess Plateau.Along this latitudinal transect,the CPI value correlates to relative humidity,precipitation,and temperature with a negative linear relationship,respectively,whereas the correlation of CPI to temperature is relatively weak.In the Wuyi,Shennongjia,and Tianshan Mountains,CPI values do not change systemically with altitude increasing (or temperature decreasing).However,mean value of CPI for the individual mountain increases in turn from the humid mountain to the arid.These results jointly suggest that aridity (or humidity) is a dominate climate factor in altering soil CPI value.High CPI values of geological records therefore indicate the arid paleoclimate.Though long-chain n-alkanes in soil are derived mainly from leaf wax of terrestrial vascular plants,the regular latitudinal variations of soil CPI might not be caused by the change of vegetation.We speculate that increased long-chain n-alkanes from microbes and/or enhanced biodegradation in the humid climate lead to the decrease of soil CPI.

文摘The hydrogen isotopic composition(δD) of leaf wax long-chain n-alkanes(C27, C29, and C31) from lacustrine sediments has been widely applied to reconstruct terrestrial paleoclimatic and paleohydrological changes. However, few studies have addressed whether the aquatic-derived n-alkanes can affect the δD values of lake sedimentary long-chain n-alkanes, which are usually regarded as a recorder of the terrestrial hydrological signals. Here we systematically investigated δD values of long-chain n-alkanes from modern aquatic plants, both near-shore and off-shore surface sediments, surrounding terrestrial plant litters, as well as river water and lake water in Lake Qinghai and its satellite lakes on the northeastern Qinghai-Tibet Plateau. Our data showed that(i) δD values of long-chain n-alkanes from aquatic plants varied from-184‰ to-132‰ for n-C27, from-183‰ to-138‰ for n-C29, and from-189‰ to-130‰ for n-C31, respectively, with no significant differences among the three n-alkanes homologues;(ii) δD values of long-chain n-alkanes from aquatic plants were generally more positive than those from surrounding terrestrial plants, possibly because that they recorded the D-enrichment of lake water in this semi-arid region;(iii) δD values of long-chain n-alkanes from surface sediments showed significant differences among the three n-alkanes homologues, due to the larger aquatic input of n-C27 to the sedimentary lipid pool than that of n-C31, and(iv) n-C27 δD values of near-shore aquatic plants and near-shore sediments are more negative than those from off-shore as a result of lower δD values of near-shore lake water. Our findings indicate that in this region(i) the offset between sedimentary n-C27 and n-C31 δD values(ΔδDC27-C31) could potentially be used to evaluate if sedimentary long-chain n-alkanes are derived from a single source;(ii) while δD values of n-C27 may be influenced by lake water hydrological changes, sedimentary n-C31 is derived predominantly from terrestrial plants and thus its δD can
文摘Human milk is considered to be the optimal source of infant nutrition. Some of the benefits of breastfeeding have been ascribed to human milk oligosaccharides(HMO). For instance, HMO can affect faecal characteristics such as stool consistency and stool frequency. Such effects on stool characteristics can be beneficial for young infants as hard stools and even constipation is common in that age group. Prebiotics in infant milk formulas have been introduced to exert similar functionalities. A specific mixture of prebiotics consists of a combination of short chain galacto-oligosaccharides and long-chain fructo-oligosaccharides(scGOS/lcFOS) in a ratio of 9:1. This specific mixture has been developed to closely resemble the molecular size composition of HMO. Many studies have been done with scGOS/lcFOS, and indicators for digestive comfort have often been included as secondary outcomes. This review summarizes the effects of scGOS/lcFOS(9:1) on stool consistency,stool frequency and transit time in healthy term and preterm infants. In several of the studies with scGOS/lcFOS in a ratio of 9:1 in infant milk formulas, positive effects of this mixture on stool characteristics such as stool consistency and stool frequency were observed. As stool consistency was shown to be correlated to whole gut transit time, scGOS/lcFOS can be hypothesised to have a role in reducing the risk of constipation.

文摘Plant sphingolipids are not only structural components of the plasma membrane and other endomembrane systems but also act as signaling molecules during biotic and abiotic stresses.However,the roles of sphingolipids in plant signal transduction in response to environmental cues are yet to be investigated in detail. In this review,we discuss the signaling roles of sphingolipid metabolites with a focus on plant sphingolipids.We also mention some microbial sphingolipids that initiate signals during their interaction with plants, because of the limited literatures on their plant analogs.The equilibrium of nonphosphorylated and phosphorylated sphingolipid species determine the destiny of plant cells,whereas molecular connections among the enzymes responsible for this equilibrium in a coordinated signaling network are poorly understood.A mechanistic link between the phytohormone-sphingolipid interplay has also not yet been fully understood and many key participants involved in this complex interaction operating under stress conditions await to be identified.Future research is needed to fill these gaps and to better understand the signal pathways of plant sphingolipids and their interplay with other signals in response to environmental stresses.

文摘Objective To assess the net health effect caused by the consumption of specific marine species based on Benefit-Risk Analysis for Foods (BRAFO)-tiered approach. Methods Twenty species were collected from the Zhoushan Archipelago, China. Concentrations of n-3 long-chain polyunsaturated fatty acids, methyl mercury (MeHg), and dioxin-like compounds (DLCs) in the samples were analyzed for benefit risk assessment based on BRAFO-tiered approach. Results Based on the BRAFO-tiered approach, reference scenario (no intake) and alternative scenario (intake of specific species of 200 g/week) were determined. The exposure to MeHg/DLCs via alternative scenario of all studied species did not exceed provisional tolerable weekly/monthly intake. However, the adult population with high DLCs exposure in China would significantly exceed the upper limit of DLCs via an additional alternative scenario of some species such as Auxis thazard. The results of deterministic computation showed that alternative scenario of all studied species generated clear net beneficial effects on death prevention and child IQ gain. Conclusion The alternative scenario of all studied species could be recommended to population with average DLCs exposure, and the reference scenario of species with relatively high DLCs concentration could be recommended to population exposed to high DLCs.

文摘Rationally tailored lipid nanoparticles(LNPs)with efficient and tunable delivery of mRNA in vivo are crucial for mRNA vaccines.Selective expression of antigenic protein in lymphoid tissues/organs could improve the immunostimulatory efficacy and safety of LNPs-based mRNA vaccines.Inspired by the metabolic behavior that long-chain saturated fatty acids tending to enter lymphoid tissue rather than the liver,we developed fatty acid-doped LNPs capable of mediating differential protein expressions in the liver and spleen when administered intravenously.When the molar ratio of saturated fatty acid located 60%–70%,the doped LNPs achieved the spleen selective mRNA translation.The mechanism could be attributed to the different cellular uptake behaviors of saturated fatty acids in hepatocytes.Immunization with a model antigen(ovalbumin)mRNA-loaded spleen selective LNPs,we observed enhanced antigen-specific T cell immune responses,and potent immunotherapeutic and immunoprophylactic efficacy in the mouse lymphoma model.Our natural long-chain saturated fatty acids metabolic characteristics-inspired design of LNPs for spleen-selective mRNA vaccines delivery will provide references for designing mRNA vaccines with high efficacy and safety for tumor immunotherapy.

文摘BACKGROUND Hepatorenal syndrome(HRS)is the most prevalent form of acute kidney injury in cirrhotic patients.It is characterized by reduced renal blood flow and represents the most severe complication in cirrhotic patients with advanced disease.Previous research has indicated that antioxidants can delay the onset of a hyperdynamic circulatory state in cirrhosis and improve renal function in HRS patients.Regular omega-3 supplementation has significantly reduced the risk of liver disease.This supplementation could represent an additional therapy for individuals with HRS.AIM To evaluated the antioxidant effect of omega-3 polyunsaturated fatty acid supplementation on the kidneys of cirrhotic rats.METHODS Secondary biliary cirrhosis was induced in rats by biliary duct ligation(BDL)for 28 d.We used 24 male Wistar rats divided into the following groups:I(control);II(treated with omega-3,1 g/kg of body weight);III(BDL treated with omega-3,1 g/kg of body weight);and IV(BDL without treatment).The animals were killed by overdose of anesthetic;the kidneys were dissected,removed,frozen in liquid nitrogen,and stored in a freezer at-80℃for later analysis.We evaluated oxidative stress,nitric oxide(NO)metabolites,DNA damage by the comet assay,cell viability test,and apoptosis in the kidneys.Data were analyzed by one-way analysis of variance,and means were compared using the Tukey test,with P≤0.05.RESULTS Omega-3 significantly decreased the production of reactive oxygen species(P<0.001)and lipoperoxidation in the kidneys of cirrhotic rats treated with omega-3(P<0.001).The activity of the antioxidant enzymes superoxide dismutase and catalase increased in the BDL+omega-3 group compared to the BDL group(P<0.01).NO production,DNA damage,and caspase-9 cleavage decreased significantly in the omega-3-treated BDL group.There was an increase in mitochondrial electrochemical potential(P<0.001)in BDL treated with omega-3 compared to BDL.No changes in the cell survival index in HRS with omega-3 compared to the control group(P>0.05)were obs

文摘In order to obtain a uniform and effectively toughened poly(lactic acid)film by blending with low content of poly(ethylene octene)(POE)with high elasticity,the tailored interfacial intermolecular interaction and entanglement between the two phases of the PLA/POE blend was innovatively constructed via the facile reactive melt blending process through the reaction of the epoxy/anhydride groups grafted on the POE chains with the end groups of PLA chains(PLA/GPOE-MPOE).It was observed that POE domains were embedded tightly in PLA matrix with a fuzzy interface and abundant interface transition area,and the impact fractured surface of the blend showed an obvious plastic deformation with less occurrence of fibrillation of PLA matrix or interfacial de-bonding.Compared with neat PLA and directly blended PLA/POE blends,the PLA/GPOE-MPOE blend exhibited much higher complex viscosity/storage modulus,much lower tanδvalues in the terminal region,and obvious strain-hardening behavior.The deviation in viscoelastic behavior of PLA/GPOE-MPOE from linear PLA indicated the enhanced molecular entanglement between the long-branched chains,resulting in an enhancement of the stretching ability during biaxial drawing of the blend.Uniform PLA/GPOE-MPOE films with draw ratio as high as 7×7 were obtained through biaxial stretching,which showed much higher tensile strength and the elongation at break than that of neat PLA and PLA/POE film.This work provides a facile method for fabricating toughening PLA films with application potentials.

文摘Ce-encapsulated Beta zeolite was synthesized by a one-pot hydrothermal method with citric acid complexing Ce in the absence of Na species.Additional citric acid can effectively prevent the deposition of Ce species during the hydrothermal synthesis of zeolites,leading to uniform distribution of Ce cluster in the framework of Beta zeolites.Moreover,the sodium-free synthesis system resulted that the Brønsted acid sites were mainly located on the straight channels and external surface of Beta zeolites,improving the utilization of Brønsted acid sites.In addition,Ce encapsulated Beta zeolites showed enhanced activity and robust stability in the alkylation of benzene with 1-dodecene based on the synergistic effect between Ce species and Brønsted acid sites,which pave the way for its practical application in the production of alkylbenzene.
文摘Previous studies have reported upregulation of heme oxygenase-1 in different central nervous system injury models.Heme oxygenase-1 plays a critical anti-inflammatory role and is essential for regulating cellular redox homeostasis.Metformin is a classic drug used to treat type 2 diabetes that can inhibit ferroptosis.Previous studies have shown that,when used to treat cardiovascular and digestive system diseases,metformin can also upregulate heme oxygenase-1 expression.Therefore,we hypothesized that heme oxygenase-1 plays a significant role in mediating the beneficial effects of metformin on neuronal ferroptosis after spinal cord injury.To test this,we first performed a bioinformatics analysis based on the GEO database and found that heme oxygenase-1 was upregulated in the lesion of rats with spinal cord injury.Next,we confirmed this finding in a rat model of T9 spinal cord compression injury that exhibited spinal cord nerve cell ferroptosis.Continuous intraperitoneal injection of metformin for 14 days was found to both upregulate heme oxygenase-1 expression and reduce neuronal ferroptosis in rats with spinal cord injury.Subsequently,we used a lentivirus vector to knock down heme oxygenase-1 expression in the spinal cord,and found that this significantly reduced the effect of metformin on ferroptosis after spinal cord injury.Taken together,these findings suggest that metformin inhibits neuronal ferroptosis after spinal cord injury,and that this effect is partially dependent on upregulation of heme oxygenase-1.

文摘This study investigated the effects of the degree of unsaturation(unsaturity) of long-chain fatty acids on microbial protein content and the activities of transaminases and dehydrogenase in vitro using goat rumen fluid as the cultural medium.Six types of fatty acids,stearic acid(C18:0,group A,control group),oleic acid(C18:1,n-9,group B),linoleic acid(C18:2,n-6,group C),α-linolenic acid(C18:3,n-3,group D),arachidonic acid(C20:4,n-6,group E),and eicosapentaenoic acid(C20:5,n-3,group F),were tested,and the inclusion ratio of each fatty acid was 3%(w/w) in total of culture substrate.Samples were taken at 0,3,6,9,12,18 and 24 h,respectively,during culture for analyses.Compared with stearic acid,linoleic acid,a-linolenic acid,and arachidonic acid increased the bacterial protein content,while oleic acid and eicosapentaenoic acid had no significant effects;the protozoal protein content was reduced for all the unsaturated fatty acids,and the magnitude of the reduction appeared to be associated with the degree of unsaturity of fatty acids.The total microbial protein content was dominantly accounted by the protozoal protein content(about 4-9 folds of the bacterial protein),and only increased by linoleic acid,but reduced by oleic acid,arachidonic acid and eicosapentaenoic acid.There were no significant effects in the activities of both glutamic oxaloacetic transaminase(GOT) and glutamic-pyruvic transaminase(GPT) for all the other fatty acids,except for arachidonic acid which enhanced GOT activity and oleic acid which enhanced GPT activity.The total dehydrogenase activity was positively correlated with the degree of unsaturation of fatty acids.In conclusion,the inclusion of 3%of long-chain unsaturated fatty acids increased bacterial protein content,whereas reduced protozoal protein content and enhanced dehydrogenase activity.The fatty acids with more than three double bonds had detrimental effects on the microbial protein content.This work demonstrates for the first tim

文摘Long-chain alkenones were detected in samples of sea surface sediments from the Chukchi Sea and the Bering Sea areas, the Arctic Pole. The analysis result indicates that C37:3 methylketone is predominate in the long-chain alkenones from the Chukchi and Bering Sea sediments. The abundance of C37 to C39 unsaturated alkenones changes in an order of C37 > C38 > C39. Based on C37 /C38 ratio, the detected organism precursors of the long-chain alkenones are mainly coccolithophrid (Emiliania huxleyi). By the calibration relationship between U37k and U37k indices, the sea surface paleotemperature in these seas is estimated. The estimated values of U37k vary from 4.147℃ to 5.706℃ , with a mean value of 5.092℃.

文摘The plastification modification of poly (vinyl chloride) (PVC) with the long-chain linear polyester (LP) synthesized by linear dodecanedioic acid (DC12) or tetradecanedioic acid (DC14) with different diols is investigated. The processing characteristics, mechanical properties and extraction property of the PVC/LP blends in different solvents (xylene, cyclohexane, ethanol) were also studied in detail. All results were compared with that of the PVC plasticized with dioctyl phthalate (DOP). The results show that the molecular weight, molecular structure and loading of LP greatly influence the mechanical properties of the PVC/LP blends. The processability and the mechanical properties of PVC plasticized by LP are comparable to those of the corresponding PVC/DOP blends. However, the PVC/LP blends posses much better migration resistance property than the corresponding PVC/DOP blends, which makes the long-chain linear polyester become a very good plasticizer candidate for PVC industry.

文摘8-Sphingolipid desaturase is the key enzyme that catalyses desaturation at the C8 position of the long-chain base of sphingolipids in higher plants. There have been no previous studies on the genes encoding AS-sphingolipid desaturases in Brassica rapa. In this study, four genes encoding AS-sphingolipid desaturases from B. rapa were isolated and characterised. Phylogenetic analyses indicated that these genes could be divided into two groups: BrD8A, BrD8C and BrD8D in group I, and BrD8B in group II. The two groups of genes diverged before the separation of Arabidopsis and Brassica. Though the four genes shared a high sequence similarity, and their coding desaturases all located in endoplasmic reticulum, they exhibited distinct expression patterns. Heterologous expression in Saccharomyces cerevisiae revealed that BrD8A/B/C/D were functionally diverse AS-sphingolipid desaturases that catalyse different ratios of the two products 8(Z)- and 8(E)-C18-phytosphingenine. The aluminium tolerance of transgenic yeasts expressing BrD8A/B/C/D was enhanced compared with that of control cells. Expression of BrD8A in Arabidopsis changed the ratio of 8(Z):8(E)-C 18-phytosphingenine in transgenic plants. The information reported here provides new insights into the biochemical functional diversity and evolutionary relationship of AS-sphingolipid desaturase in plants and lays a foundation for further investigation of the mechanism of 8(Z)- and 8(E)-C18- phytosphingenine biosynthesis.

文摘Long-chain acyl-coenzyme A(CoA)synthase 4(ACSL4)is an enzyme that esterifies CoA into specific polyunsaturated fatty acids,such as arachidonic acid and adrenic acid.Based on accumulated evidence,the ACSL4-catalyzed biosynthesis of arachidonoyl-CoA contributes to the execution of ferroptosis by triggering phospholipid peroxidation.Ferroptosis is a type of programmed cell death caused by iron-dependent peroxidation of lipids;ACSL4 and glutathione peroxidase 4 positively and negatively regulate ferroptosis,respectively.In addition,ACSL4 is an essential regulator of fatty acid(FA)metabolism.ACSL4 remodels the phospholipid composition of cell membranes,regulates steroidogenesis,and balances eicosanoid biosynthesis.In addition,ACSL4-mediated metabolic reprogramming and antitumor immunity have attracted much attention in cancer biology.Because it facilitates the cross-talk between ferroptosis and FA metabolism,ACSL4 is also a research hotspot in metabolic diseases and ischemia/reperfusion injuries.In this review,we focus on the structure,biological function,and unique role of ASCL4 in various human diseases.Finally,we propose that ACSL4 might be a potential therapeutic target.
