A phylogenetic analysis of Trametes and related taxa is proposed,based on a wide sampling of temperate and tropical species.Concatenate sequences of ribosomal DNA(ITS1-5.8S-ITS2)and RPB2 gene from mycelia cultures wer...A phylogenetic analysis of Trametes and related taxa is proposed,based on a wide sampling of temperate and tropical species.Concatenate sequences of ribosomal DNA(ITS1-5.8S-ITS2)and RPB2 gene from mycelia cultures were analyzed by Maximum Likelihood and Bayesian methods,whilst morphological features were documented from the corresponding herbarium vouchers.Congruent results were obtained from analyses of ribosomal LSU sequences downloaded from Genbank.The Bayesian analysis of ITS+RPB2 sequences gave the best resolution for the phylogenetic reconstructions and revealed the existence of three main lineages in the Trametes-clade:1)a monospecific lineage represented by Artolenzites elegans 2)a lineage including the genus Pycnoporus in its traditional sense and several species usually classified in the genus Trametes(T.cingulata,T.lactinea,T.ljubarskyi,T.menziesii)&3)a lineage corresponding to the core genus Trametes,including type species of Trametes,Coriolopsis and Lenzites.The presence of a pseudostipe,aspect and structure of the abhymenial surface,colour change with 5%aqueous solutions of potassium hydroxide and topography of pigments on skeletal hyphae gave relevant morphological support to these clades.When the structure of the hymenial surface,presence of a black line below the tomentum and color of context(except for genus Pycnoporus)usually used in traditional polypore-classifications did not reveal any phylogenetic significance.A partial systematic arrangement of the Trametes clade is proposed,with the introduction of a new genus:Leiotrametes Welti&Courtec.gen.nov..Two new combinations:Leiotrametes lactinea(Berk.)Welti&Courtec.comb.nov.and L.menziesii(Berk.)Welti&Courtec.comb.nov.are proposed.展开更多
Objective: To examine the anti-hyperglycemic effects of aqueous Lenzites betulina(L. betulina) extracts on normoglycemic glucose-loaded mice.Methods: Different doses of aqueous extract from L. betulina were administer...Objective: To examine the anti-hyperglycemic effects of aqueous Lenzites betulina(L. betulina) extracts on normoglycemic glucose-loaded mice.Methods: Different doses of aqueous extract from L. betulina were administered to 45 ICR mice(Mus musculus) to determine whether there was an effect of L. betulina extracts on the blood glucose level of the ICR mice. Aqueous extracts of L. betulina were orally gavaged to mice using oral glucose tolerance test. A total of five groups were used to determine the effect of the fungi on blood glucose of the mice. Group A(positive control)was given 16.7 mg/kg glimepiride; Group B(negative control) was given distilled water;Group C(low dosage) was given 200 mg/kg aqueous extract; Group D(mid dosage) was given 400 mg/kg aqueous extract and Group E(high dosage) was given 800 mg/kg aqueous extract. Baseline blood glucose value was firstly acquired before induction of hyperglycemia through D-glucose, after which another check on blood glucose was made after 0.5 h. Immediately, after the acquisition of hyperglycemic blood glucose level, the individual administration of treatments were done. After that, three blood collections were done spanning 3 h with 1 h interval.Results: The low dose(200 mg/kg) and the mid dose(400 mg/kg) of L. betulina extracts were significantly different(P < 0.05) from their respective baseline values throughout the whole experiment with the latter surpassing its baseline value during the 3rd hour. On the other hand, the high dose(800 mg/kg) during the 1st hour after administration was not significantly different(P > 0.05) from its corresponding baseline value, acting faster than the positive control(glimepiride), which only became significantly different(P < 0.05) at the 2nd hour.Conclusions: Aqueous L. betulina extract is able to produce hypoglycemic effects on the mice with all doses, which are able to normalize blood glucose levels at varying times.展开更多
Zinc finger transcription factors play significant roles in the growth and development of plant and animal,but their function remains obscure in fungi.Lenzites gibbosa mycelia were extracted and sequenced by transcrip...Zinc finger transcription factors play significant roles in the growth and development of plant and animal,but their function remains obscure in fungi.Lenzites gibbosa mycelia were extracted and sequenced by transcriptome analysis after growing on sawdust at different times to support mycelial growth of L.gibbosa in a nutrient matrix.Data bases used for analysis were the Kyoto encyclopedia of genes and genomes(KEGG)annotation,the cluster of orthologous groups of proteins(COG)and gene ontology(GO)annotation.Zinc finger class genes related to the growth and development of L.gibbosa were screened.GO annotation and enrichment analysis of diff erentially expressed genes were carried out.A total of 114.55 Gb Clean Data were obtained from the L.gibbosa transcriptome.The average Clean Data in each sample was 6.16 Gb.The relative efficiency of reads between each sample and the reference genome was 88.5%to 91.4%.The COG analysis showed that most zinc finger protein genes were related to replication,recombination and repair function.GO enrichment analysis showed that the expressed genes involved in cellular process,cell part and binding.We identifi ed seventy-two expressed genes including seven up-regulated genes and sixty-five down-regulated genes by applying DESeq2 data analysis software.By comparing the significantly expressed genes with KEGG database,66 annotated sequences were obtained,and 35 primary metabolic pathways were annotated.Pathway enrichment analysis showed that differentially expressed genes were signifi cantly enriched in protein processing in endoplasmic reticulum and ubiquitin-mediated proteolysis pathways.Gene_11750 and gene_5266 are highly correlated with the growth and development of L.gibbosa and are closely related to protein processing in endoplasmic reticulum and ubiquitin-mediated proteolysis pathway.According to gene functional analysis,seven important differentially expressed genes related to the growth and development of L.gibbosa were identified.展开更多
