Objective:To assess the antimicrobial and cytotoxic effects of Methylobacterium sp.isolated from soil sample of Doddabetta forest,Nilgiris,Western Ghats of Tamil Nadu.Methods:Isolation of Methylobacterium was performe...Objective:To assess the antimicrobial and cytotoxic effects of Methylobacterium sp.isolated from soil sample of Doddabetta forest,Nilgiris,Western Ghats of Tamil Nadu.Methods:Isolation of Methylobacterium was performed from soils by serial dilution plate technique.The strain was grown in modified nutrient gulucose agar(MNGA)medium to study the morphology and biochemical characteristics.Methylobacterium sp.was screened for its antimicrobial activity against pathogenic bacteria and fungi.The strain was subjected to 16S rRNA analysis and was identified as Methylohacterium sp.The nucleotide sequence of the 16S rRNA gene of the isolate exhibited close similarity with other Methylobacterium sp.and has been submitted to Genbank.The antibacterial substances were extracted using chloroform and ethyl acetate from MNGA medium in which ERI-135 had grown for 5 d at 30℃.Cytotoxic effect was also studied.GC-MS analysis was carried out.The antimicrobial activity was assessed using broth micro dilution technique.Results:Ethyl acetate extract showed activity against bacteria such as Bacillus subtilis,Klebsiella pneumoniae(K.pneumoniae),Pseudomonas aeruginosa,Salmonella typkimurium,Shigella flexneri,Enterobacter aerogenes,Staphylococcus aureu and Staphylococcus epidermidis(S.epidermidis)and fungi such as,Candida albicans and Trichophyton rubrum.The lowest minimum inhibitory concentrations were:250μg/mL against 5.epidermidis and 250μg/mL against K.pneumonia.The isolate had the ability to produce enzymes such as protease.The exyract showed cytotoxic effect in human adenocarcinoma cancer cell line(A549).GC-MS analysis showed the presence of isovaleric acid(3.64%),2-Methylbulanoic acid(5.03%),isobutyramide(5.05%),N,Noimethylformamide-di-t-butylacetal(9.79%),benzeneacetamide(15.56%),octyl butyl phthalate(3.59%)and diisooctyl phthalate(5.79)in the extract.Conclusions:Methylobaeterium sp.(ERI-135)showed promising antibacterial and cytotoxic activity.This is the first report in the antimicrobial and cytotoxic effect of Methylobaeterium展开更多
The present study was undertaken to evaluate the influence of the methanolic fruit extract of Momordica cymbalaria (MFMC) on PPARγ, (Peroxisome Proliferator Activated Receptor gamma) and GLUT-4 (Glucose transpor...The present study was undertaken to evaluate the influence of the methanolic fruit extract of Momordica cymbalaria (MFMC) on PPARγ, (Peroxisome Proliferator Activated Receptor gamma) and GLUT-4 (Glucose transporter-4) with respect to glucose transport. Various concentrations of MFMC ranging from 62.5 to 500 μg·mL^-1 were evaluated for glucose uptake activity in vitro using L6 myotubes, rosiglitazone was used as a reference standard. The MFMC showed significant and dose-dependent increase in glucose uptake at the tested concentrations, further, the glucose uptake activity of MFMC (500 μg·mL^-1) was comparable with rosigilitazone. Furthermore, MFMC has shown up-regulation of GLUT-4 and PPARy gene expressions in L6 myotubes. In addition, the MFMC when incubated along with cycloheximide (CHX), which is a protein synthesis inhibitor, has shown complete blockade of glucose uptake. This indicates that new protein synthesis is required for increased GLUT-4 translocation. In conclusion, these findings suggest that MFMC is enhancing the glucose uptake significantly and dose dependently through the enhanced expression of PPARμ and GLUT-4 in vitro.展开更多

In clonally selected cultures, 2-deoxyglucose uptake into L6-GLUT1myc myoblasts and myotubes was linear within the time of study. In L6-GLUT1myc and L6-GLUT4myc myoblasts, 100 nmol/L insulin treatment increased the GLUT1 content of the plasma membrane by 1.58±0.01 fold and the GLUT4 content 1.96±0.11 fold, as well as the 2-deoxyglucose uptake 1.53±0.09 and 1.86±0.17 fold respectively, all by a wortmannin-inhibitable manner. The phosphorylation of Akt in these two cell lines was increased by insulin. L6-GLUT1myc myoblasts showed a dose-dependent stimulation of glucose uptake by insulin, with unaltered sensitiv- ity and maximal responsiveness compared with wild type cells. By contrast, the improved insulin re- sponsiveness and sensitivity of glucose uptake were observed in L6-GLUT4myc myoblasts. Earlier studies indicated that forskolin might affect insulin-stimulated GLUT4 translocation. A 65% decrease of insulin-stimulated 2-deoxyglucose uptake in GLUT4myc cells was not due to an effect on GLUT4 mobi- lization to the plasma membrane, but instead on direct inhibition of GLUT4. Forskolin and dipyridamole are more potent inhibitors of GLUT4 than GLUT1. Alternatively, pentobarbital inhibits GLUT1 more than GLUT4. The use of these inhibitors confirmed that the overexpressed GLUT1 or GLUT4 are the major functional glucose transporters in unstimulated and insulin-stimulated L6 myoblasts. Therefore, L6-GLUT1myc and L6-GLUT4myc cells provide a platform to screen compounds that may have differ- ential effects on GLUT isoform activity or may influence GLUT isoform mobilization to the cell surface of muscle cells.展开更多
In this paper we propose and analyze a backward differentiation formula(BDF)type numerical scheme for the Cahn-Hilliard equation with third order temporal accuracy.The Fourier pseudo-spectral method is used to discret...In this paper we propose and analyze a backward differentiation formula(BDF)type numerical scheme for the Cahn-Hilliard equation with third order temporal accuracy.The Fourier pseudo-spectral method is used to discretize space.The surface diffusion and the nonlinear chemical potential terms are treated implicitly,while the expansive term is approximated by a third order explicit extrapolation formula for the sake of solvability.In addition,a third order accurate Douglas-Dupont regularization term,in the form of−A_(0)△t^(2)△_( N)(φ^(n+1)−φ^(n)),is added in the numerical scheme.In particular,the energy stability is carefully derived in a modified version,so that a uniform bound for the original energy functional is available,and a theoretical justification of the coefficient A becomes available.As a result of this energy stability analysis,a uniform-in-time L_(N)^(6)bound of the numerical solution is obtained.And also,the optimal rate convergence analysis and error estimate are provided,in the L_(△t)^(∞)(0,T;L_(N)^(2))∩L^(2)_(△ t)(0,T;H_(h)^(2))norm,with the help of the L_(N)^(6)bound for the numerical solution.A few numerical simulation results are presented to demonstrate the efficiency of the numerical scheme and the third order convergence.展开更多
Objective:Wild musk melon(Cucumis melo var.agrestis,CMA)is one of the edible plants form Tamil Nadu.Traditionally,this plant was used as diabetic diet(leaves of CMA with Momordica charantia leaves),but there is no sci...Objective:Wild musk melon(Cucumis melo var.agrestis,CMA)is one of the edible plants form Tamil Nadu.Traditionally,this plant was used as diabetic diet(leaves of CMA with Momordica charantia leaves),but there is no scientific report on antidiabetic action of this plant material.Hence,the current research work was designed to evaluate the antihyperglycemic and antihyperlipidemic effect of hydroalcoholic extract of CMA leaves(HALEC)in streptozotocin(STZ)-nicotinamide(NIC)-induced diabetic rats.Methods:Diabetes was induced by administration of STZ(60 mg/kg,i.p.)after 15 min of NIC(120 mg/kg i.p.)administration.The diabetic rats were treated with HALEC(300 and 600 mg/kg,p.o.,respectively)for 21 d.Results:After the management with HALEC,blood glucose,HbA1c levels,total cholesterol,LDL cholesterol,triglycerides levels,glycogen phosphorylase and glucose-6-phosphatase levels were significantly diminished in diabetic rats.However,haemoglobin level,HDL cholesterol,liver glycogen,total protein,hexokinase,glucose-6-phosphate dehydrogenase levels were significantly increased in HALEC treated diabetic rats.The histopathological studies of the pancreas in HALEC-treated diabetic rats showed almost normal appearance.L6 cell line study revealed the increased glucose uptake activity of HALEC.High performance thin layer chromatography(HPTLC)analysis confirms the presence of active principles such as rutin,gallic acid and quercetin in HALEC.Conclusion:The results indicated that HALEC possess significant antihyperglycemic and antihyperlipidemic activity in STZ-NIC-induced typeⅡdiabetic rats with protective effect.This research work will be useful for the isolation of active principles and development of herbal formulation in phytopharmaceuticals.展开更多

Moreover, the tracking structure does not change the ...A tracking structure suitable for L6 signal of quasi-zenith satellite system(QZSS) was proposed in order to track the L6 signal without other frequency assistance. Moreover, the tracking structure does not change the receiver’s hardware structure. The main difference between the proposed and the traditional tracking structure lies in the generation of local codes of E,P and L branches. The method of local code generation is designed in a two-stage manner. The first stage is the generation of the P branch local code with fast fouriertransform(FFT). In the second stage, the local codes of the E and L branches are obtained with the code-chip interval. The tracking structure can track and decode L6 signal separately, and track code shift keying(CSK)modulated signal as well. The structure was verified using both simulation data, generated in different conditions, and actual data obtained from QZSS satellites respectively. The results show that the improved tracking loop is able to track L6 signal without other frequency assitance. Furthermore, the biterror ratio(BER) of L6 tracking algorithm is lower than that of L1C/A assist L6 algorithm, when the Doppler remains a constant and reed solomn(RS)encode are applied. To be more specific, with the proposed structure the BER decreased by 11.40%, 17.07%, 15.00%, 11.15%, 5.19% when carrier to noise ratio(CNR) is 36-40 dB·Hz.展开更多
