AIM: To investigate the impact of minor abdominal surgery on the caecal microbial population and on markers of gut inflammation.METHODS: Four week old piglets were randomly allocated to a no-surgery "control" group(n = 6) or a "transection surgery" group(n = 5).During the transection surgery procedure, a conventional midline incision of the lower abdominal wall was made and the small intestine was transected at a site 225 cm proximal to the ileocaecal valve, a 2 cm segment was removed and the intestine was re-anastomosed.Piglets received a polymeric infant formula diet throughout the study period and were sacrificed at two weeks post-surgery.Clinical outcomes including weight, stool consistency and presence of stool fat globules were monitored.High throughput DNA sequencing of colonic content was used to detect surgery-relateddisturbances in microbial composition at phylum, family and genus level.Diversity and richness estimates were calculated for the control and minor surgery groups.As disturbances in the gut microbial community are linked to inflammation we compared the gene expression of key inflammatory cytokines(TNF, IL1 B, IL18, IL12, IL8, IL6 and IL10) in ileum, terminal ileum and colon mucosal extracts obtained from control and abdominal surgery groups at two weeks post-surgery.RESULTS: Changes in the relative abundance of bacterial species at family and genus level were confined to bacterial members of the Proteobacteria and Bacteroidetes phyla.Family level compositional shifts included a reduction in the relative abundance of Enterobacteriaceae(22.95 ± 5.27 vs 2.07 ± 0.72, P < 0.01), Bacteroidaceae(2.54 ± 0.56 vs 0.86 ± 0.43, P < 0.05) and Rhodospirillaceae(0.40 ± 0.14 vs 0.00 ± 0.00, P < 0.05) following transection surgery.Similarly, at the genus level, changes associated with transection surgery were restricted to members of the Proteobacteria and Bacteroidetes phyla and included decreased relative abundance of Enterobacteriaceae(29.20 ± 6.74 vs 2.88 ± 1.08, P < 0.01), Alistipes(4.82 ± 1.73 展开更多

目的探讨大黄素缓解类风湿关节炎(RA)痛的作用及机制。方法将30只C57BL/6小鼠随机分为对照组、模型组和大黄素给药组;采用左后膝关节腔注射完全弗式佐剂构建小鼠RA痛模型;腹腔注射大黄素进行给药;记录小鼠痛觉行为及运动能力改变;免疫... 目的探讨大黄素缓解类风湿关节炎(RA)痛的作用及机制。方法将30只C57BL/6小鼠随机分为对照组、模型组和大黄素给药组;采用左后膝关节腔注射完全弗式佐剂构建小鼠RA痛模型;腹腔注射大黄素进行给药;记录小鼠痛觉行为及运动能力改变;免疫印迹法(Western blot)检测各组动物脊髓组织氧化及炎症相关蛋白变化。结果完全弗式佐剂注射14 d后,与对照组小鼠相比,模型组小鼠自发缩足次数增加,机械痛阈值下降,运动能力减弱,脊髓组织核因子κB(NF-κB)、白介素1β(IL-1β)、NOD样受体热蛋白结构域相关蛋白3(NLRP3)、小胶质细胞标记蛋白游离钙结合适配器蛋白1(Iba1)、环氧化酶2(COX-2)蛋白水平均增加;核因子E2相关因子2(Nrf2)蛋白表达降低,差异均有统计学意义(P<0.05)。大黄素腹腔给药后,与模型组小鼠相比,小鼠自发缩足次数减少,机械痛阈值增加,改善小鼠的运动能力,降低NF-κB、IL-1β、NLRP3、Iba1和COX-2蛋白表达水平,升高Nrf2蛋白表达水平,差异均有统计学意义(P<0.05)。结论大黄素具有显著缓解RA痛的作用效果,其机制是通过抑制脊髓炎症和氧化应激反应。 展开更多

[Objectives]To observe the effects of daphnetin on mastitis induced by Staphylococcus aureus in mice.[Methods]18 postpartum ICR female mice were used to establish mastitis animal model,and were randomly divided into three groups(A,B,and C)with 6 mice in each group.Group A:blank control group;group B:S.Aureus model group;group C:S.Aureus model+daphnetin group.The experimental groups were injected 1 mL of 1.0×104 CFU/100μL of S.aureus of along the nipple catheter.The suspension was placed in the 3 rd and 4 th pairs of mammary glands,and the control group was injected with the same dose of normal saline.On the second day after infection,the rats in group A,B and C were given drugs by gavage,while the rats in group A and B were given normal saline and the rats in group C were given daphnetin once a day for 6 consecutive days.Blood samples were collected from living eyeballs,and blood cells were analyzed by automatic flow cytometer after anticoagulation.[Results]The NLR and Systemie Immune Inflammati-on Index(SII)in the blood of mastitis mice induced by S.aureus were significantly higher than those in the control group(P<0.01),suggesting that neutrophil to lymphocyte ratio(NLR)and SII can be used as diagnostic indicators of mastitis,and the levels of NLR and SII decreased significantly after daphnetin intervention.[Conclusions]NLR and SII showed high levels in mastitis mice,which are valuable for the diagnosis of mastitis and the evaluation of its prognosis.After the intervention of daphnetin,both of them decreased significantly,indicating that daphnetin has a good prognosis trend in mastitis mice induced by S.aureus. 展开更多
