To improve the amylose content(AC)and resistant starch content(RSC)of maize kernel starch,we employed the CRISPR/Cas9 system to create mutants of starch branching enzyme I(SBEI)and starch branching enzyme IIb(SBEIIb).A frameshift mutation in SBEI(E1,a nucleotide insertion in exon 6)led to plants with higher RSC(1.07%),lower hundred-kernel weight(HKW,24.71±0.14 g),and lower plant height(PH,218.50±9.42 cm)compared to the wild type(WT).Like the WT,E1 kernel starch had irregular,polygonal shapes with sharp edges.A frameshift mutation in SBEIIb(E2,a four-nucleotide deletion in exon 8)led to higher AC(53.48%)and higher RSC(26.93%)than that for the WT.E2 kernel starch was significantly different from the WT regarding granule morphology,chain length distribution pattern,X-ray diffraction pattern,and thermal characteristics;the starch granules were more irregular in shape and comprised typical B-type crystals.Mutating SBEI and SBEIIb(E12)had a synergistic effect on RSC,HKW,PH,starch properties,and starch biosynthesis-associated gene expression.SBEIIa,SS1,SSIIa,SSIIIa,and SSIIIb were upregulated in E12 endosperm compared to WT endosperm.This study lays the foundation for rapidly improving the starch properties of elite maize lines. 展开更多

目的探讨老年(≥75岁)急性ST段抬高心肌梗死患者急诊经皮冠状动脉介入治疗联合应用替罗非班抗血小板治疗的安全性。方法分析2009年12月至2012年3月因急性ST段抬高性心肌梗死行急诊冠状动脉造影术的老年患者56例,根据是否应用替罗非... 目的探讨老年(≥75岁)急性ST段抬高心肌梗死患者急诊经皮冠状动脉介入治疗联合应用替罗非班抗血小板治疗的安全性。方法分析2009年12月至2012年3月因急性ST段抬高性心肌梗死行急诊冠状动脉造影术的老年患者56例,根据是否应用替罗非班分为替罗非班组(18例)及双联抗血小板组(38例)。观察患者住院期间死亡、非致死性再梗死、急性或亚急性血栓形成、出血及消化道不良症状的发生率。结果替罗非班组与双联抗血小板组患者住院期间病死率分别为5.56%(1/18)和7.89%(13/38),差异无统计学意义(P〉0.05),两组患者均未发生严重出血事件,总出血事件替罗非班组高于双联抗血小板组,分别为22.22%(4/18)和2.63%(1/38)(χ2=5.765,P〈0.05);两组消化道不良症状发生率分别为38.89%(7/18)和42.11%(16/38),差异均无统计学意义(P均〉0.05)。结论老年急性心肌梗死患者应用替罗非班抗血小板治疗不增加严重出血发生率,但需严密观察。 展开更多

Background Suppression of myostatin (MSTN) has been associated with skeletal muscle atrophy and insulin resistance (IR).However,few studies link MSTN suppression by ladder-climbing training (LCT) and IR.Therefore,we intended to identify the correlation with IR between LCT and to analyze the signaling pathways through which MSTN suppression by LCT regulates IR.Methods The rats were randomly assigned to two types of diet:normal pellet diet (NPD,n=8) and high-fat diet (HFD,n=16).After 8 weeks,the HFD rats were randomly re-assigned to two groups (n=8 for each group):HFD sedentary (HFD-S) and high-fat diet ladder-climbing training (HFD-LCT).HFD-LCT rats were assigned to LCT for 8 weeks.Western blotting,immunohistochemistry and enzyme assays were used to measure expression levels and activities of MSTN,GLUT4,PI3K,Akt and Akt-activated targets (mTOR,FoxO1 and GSK-3β).Results The LCT significantly improved IR and whole-body insulin sensitivity in HDF-fed rats.MSTN protein levels decreased in matching serum (42%,P=0.007) and muscle samples (25%,P=0.035) and its receptor mRNA expression also decreased (16%,P=0.041) from obese rats after LCT.But the mRNA expression of insulin receptor had no obvious changes in LCT group compared with NPD and HFD-S groups (P=0.074).The ladder-climbing training significantly enhanced PI3K activity (1.7-fold,P=0.024) and Akt phosphorylation (83.3%,P=0.022) in HFD-fed rats,significantly increased GLUT4 protein expression (84.5%,P=-0.036),enhanced phosphorylation of mTOR (4.8-fold,P <0.001) and inhibited phosphorylation of FoxO1 (57.7%,P=0.020),but did not affect the phosphorylation of GSK-3β.Conclusions The LCT significantly reduced IR in diet-induced obese rats.MSTN may play an important role in regulating IR and fat accumulation by LCT via PI3K/Akt/mTOR and PI3K/Akt/FoxO1 signaling pathway in HFD-fed rats. 展开更多

BACKGROUND Mannosyl-oligosaccharide glucosidase(MOGS)deficiency is an extremely rare type of congenital disorder of glycosylation(CDG),with only 12 reported cases.Its clinical,genetic,and glycomic features are still expanding.Our aim is to update the novel clinical and glycosylation features of 2 previously reported patients with MOGS-CDG.CASE SUMMARY We collected comprehensive clinical information,and conducted the immunoglobulin G1 glycosylation assay using nano-electrospray ionization source quadruple time-of-flight mass spectrometry.Novel dysmorphic features included an enlarged tongue,forwardly rotated earlobes,a birth mark,overlapped toes,and abnormal fat distribution.Novel imaging findings included pericardial effusion,a deep interarytenoid groove,mild congenital subglottic stenosis,and laryngomalacia.Novel laboratory findings included peripheral leukocytosis with neutrophil predominance,elevated C-reactive protein and creatine kinase,dyslipidemia,coagulopathy,complement 3 and complement 4 deficiencies,decreased proportions of T lymphocytes and natural killer cells,and increased serum interleukin 6.Glycosylation studies showed a significant increase of hypermannosylated glycopeptides(Glc3Man7GlcNAc2/N2H10 and Man5GlcNAc2/N2H5)and hypersialylated glycopeptides.A compensatory glycosylation pathway leading to an increase in Man5GlcNAc2/N2H5 was indicated with the glycosylation profile.CONCLUSION We confirmed abnormal glycomics in 1 patient,expanding the clinical and glycomic spectrum of MOGS-CDG.We also postulated a compensatory glycosylation pathway,leading to a possible serum biomarker for future diagnosis. 展开更多

A typical case confirmed by au-topsy is here reported and discussed briefly.CASE REPORTA 27-year-old male patient complained of fre-quent epigastric pain and diarrhea for 12 years. Atthe age of 21, he had a biopsy of a papillary lesionin his eyelids, and histological examination con-firmed the diagnosis of mucosal neuromas. Twoyears later, subtotal gastrectomy was done for a sus-pected perforated peptic ulcer at a local hospital. Ex-ploration showed an ulcer at the gastric antrum andanother at the duodenal bulb and hypertrophicgastritis and proliferation of the parietal cells werefound histologically. Postoperatively, diarrhea per-sisted and epigastric distention appeared. On Septem-ber 12,1990, the patient was referred to ourhospital. 展开更多

Shu Xin Yi Mai Capsules (舒心益脉胶囊) combined with western medicine was used in the routine treatment of 22 cases who successfully received coronary artery introducing therapy (Chinese-western medicine group),and the results was compared with the 26 cases treated routinely with simple western medicine in the control group (western medicine group).It was found that both the recurrence rate of angina pectoris and the incidence rate of recurrent stricture in the Chinese-western medicine group were significantly lower than that in the control group (both P<0.05).There was no significant difference between the two groups in expression of platelet activating molecules CD62P (a-granular membrane protein),CD63 (lysosome intact membrane protein) and CD41 (glucoprotein IIb) before the treatment,but with a significant difference after the treatment (P<0.05). 展开更多

Platelets are small anucleate cells generated from megakaryocytes in the bone marrow. Although platelet genera- tion, maturation, and clearance are still not fully understood, significant progress has been made in the last 1-2 dec- ades. In blood circulation, platelets can quickly adhere and aggregate at sites of vascular injury, forming the platelet plug (i.e. the first wave of hemostasis). Activated platelets can also provide negatively charged phosphatidylserine- rich membrane surface that enhances cell-based thrombin generation, which facilitates blood coagulation (i.e. the second wave of hemostasis). Platelets therefore play central roles in hemostasis. However, the same process of hemostasis may also cause thrombosis and vessel occlusion, which are the most common mechanisms leading to heart attack and stroke following ruptured atherosclerotic lesions. In this review, we will introduce the classical mechanisms and newly discovered pathways of platelets in hemostasis and thrombosis, including fibrinogen-inde- pendent platelet aggregation and thrombosis, and the plasma fibronectin-mediated "protein wave" of hemostasis that precedes the classical first wave of hemostasis. Furthermore, we briefly discuss the roles of platelets in inflam- marion and atherosclerosis and the potential strategies to control atherothrombosis. 展开更多

目的 :观察外源性IGF -I对SD大鼠骨骼肌钝挫伤后修复过程中IIb型肌球蛋白重链(myosinheavychain ,MHC)及Ⅰ、Ⅲ型胶原(collagen-Ⅰ和Ⅲ )mRNA表达的影响。方法 :采用自制的肌肉损伤打击装置对 84只雄性SD大鼠后下肢肌群造成钝挫伤 ,然... 目的 :观察外源性IGF -I对SD大鼠骨骼肌钝挫伤后修复过程中IIb型肌球蛋白重链(myosinheavychain ,MHC)及Ⅰ、Ⅲ型胶原(collagen-Ⅰ和Ⅲ )mRNA表达的影响。方法 :采用自制的肌肉损伤打击装置对 84只雄性SD大鼠后下肢肌群造成钝挫伤 ,然后将动物随机分为两组 ,分别于损伤局部注射外源性IGF-I及生理盐水 ,然后在损伤后第 1、3、7、1 0、1 4、2 1和 2 8天分别处死大鼠并取损伤处肌肉 ,采用半定量逆转录 -聚合酶链式反应 (semi-quantitativeRT -PCR)分析IIb型MHC、Ⅰ型及Ⅲ型胶原mRNA的表达。结果 :(1 )急性钝挫伤修复期 ,两组动物骨骼肌IIb型MHC及Ⅰ、Ⅲ型胶原的mRNA表达量均显著增高 ,第 7~1 0天达到高峰 ,然后呈下降趋势。 (2 )在骨骼肌修复过程中 ,外源性IGF-I治疗组大鼠IIb型MHCmRNA表达水平的升幅显著高于生理盐水对照组 ,且达到峰值的时程早于生理盐水组 3天。 (3 )在骨骼肌修复过程中 ,外源性IGF -I治疗组大鼠Ⅲ型与Ⅰ型胶原mRNA表达水平的改变明显低于生理盐水对照组 ,但峰值时程与生理盐水对照组相同。结论 :(1 )急性骨骼肌钝挫伤修复过程中 ,外源性IGF -I能促进骨骼肌急性钝挫伤后IIb型MHCmRNA表达。 (2 )急性骨骼肌钝挫伤修复期 ,外源性IGF -I能抑制Ⅰ。 展开更多

目的:观察比伐芦定在急性ST段抬高型心肌梗死(STEMI)患者急诊经皮冠状动脉(冠脉)介入治疗(PCI)术中应用的有效性及安全性。方法:选择2011-09至2014-01期间因急性STEMI就诊于我院并行急诊PCI术的患者159例,根据术中是否使用比伐芦定随机... 目的:观察比伐芦定在急性ST段抬高型心肌梗死(STEMI)患者急诊经皮冠状动脉(冠脉)介入治疗(PCI)术中应用的有效性及安全性。方法:选择2011-09至2014-01期间因急性STEMI就诊于我院并行急诊PCI术的患者159例,根据术中是否使用比伐芦定随机分为两组:比伐芦定组及肝素组。术中血小板糖蛋白Ⅱb/Ⅲa受体拮抗剂(GPI)即替罗非班的应用由术者评估患者病情特点决定是否合并应用。分析两组一般情况、冠脉影像特点等一系列基线资料,并对两组围手术期及术后30 d内的出血及主要心血管不良事件(MACE)的发生情况进行随访统计,分析比较其中差异。结果:在最终完成研究的153例样本中,比伐芦定组(n=72)与肝素组(n=81)比较,围手术期出血事件发生率6.5%比11.0%、支架内血栓事件发生率0%比1.2%以及术后30 d内的出血事件发生率9.7%比13.5%、MACE发生率1.4%比7.4%均有所下降,但两组差异均无统计学意义(P均>0.05)。结论:比伐芦定应用于急性STEMI患者急诊PCI术中是安全有效的,且其具有降低出血及MACE发生率的趋势。 展开更多

主要组织相容性复合体(Major histocompatibility complex,MHC)是脊椎动物体内重要的非特异性免疫基因之一。为了研究赤点石斑鱼的MHC基因的结构与表达特性,本研究运用RACE,Realtime PCR及SSCP等技术,首次成功克隆获得赤点石斑鱼... 主要组织相容性复合体(Major histocompatibility complex,MHC)是脊椎动物体内重要的非特异性免疫基因之一。为了研究赤点石斑鱼的MHC基因的结构与表达特性,本研究运用RACE,Realtime PCR及SSCP等技术,首次成功克隆获得赤点石斑鱼(Epinephelus akaara)非特异性免疫因子MHC IIB基因的4个cDNA全序列,序列全长为1195~1355bp,含有3’UTR、启动子、多肽结合区(β1)、IGC区(β2)、跨膜区、胞质区和5’UTR区,编码区大小为750bp。氨基酸序列分析发现赤点石斑鱼MHC IIB分子具有经典MHC蛋白分子的空间结构,在多肽结合区存在极其丰富的变异,β1区由1个α螺旋与4个反向平行的β折叠构成多肽结合区,β2区由多个β折叠构成2个反向平行的三明治结构。利用SSCP技术研究了赤点石斑鱼MHC IIB的表达多态性,发现其在头肾、心、肝等12个组织器官中均能有效表达,表达多态性也异常丰富。此外,根据MHC IIB分子中相对保守的IGC区氨基酸序列构建了脊椎动物的系统进化树,也表明了该分子的IGC区氨基酸序列可以作为研究鱼类物种间进化关系的良好标记。 展开更多

目的:比较根治性子宫切除术联合辅助放疗和同步放化疗对FIGOIIB期宫颈癌患者的疗效和安全性差异。方法:回顾性分析收治的219例FIGOIIB宫颈癌患者的病例资料,其中74例接受根治性子宫切除术联合术后辅助放疗(手术组),145例接受同步... 目的:比较根治性子宫切除术联合辅助放疗和同步放化疗对FIGOIIB期宫颈癌患者的疗效和安全性差异。方法:回顾性分析收治的219例FIGOIIB宫颈癌患者的病例资料,其中74例接受根治性子宫切除术联合术后辅助放疗(手术组),145例接受同步放化疗(放疗组),比较两组复发率、病死率、无进展生存期、总生存期和治疗相关并发症发生率的差异。结果:两组复发率、无进展生存期和总生存期无明显差异(均P〉0.05)。手术组患者3。4级急性胃肠道反应和3~4级下肢淋巴水肿的发生率明显高于放疗组(均P〈0.05)。多因素分析证实肿瘤直径、病理类型是总生存期的独立预后因素。结论:根治性手术联合辅助放疗与同步放化疗对FIGOIIB宫颈癌患者的疗效相当,但同步放化疗相关的不良反应发生率相对较低。 展开更多
