Objective:To measure the effect of doxycycline on inflammatory marker[IL-6,TNF-α,ferritin and C reactive protein(CRP)]levels in patients with dengue infection.Methods:A single-centre,open-label,parallel-group randomi...Objective:To measure the effect of doxycycline on inflammatory marker[IL-6,TNF-α,ferritin and C reactive protein(CRP)]levels in patients with dengue infection.Methods:A single-centre,open-label,parallel-group randomized controlled trial was done in PGIMER Chandigarh from June 2021 to October 2022.Patients were randomized using a simple randomization process into two groups:the doxycycline treatment group(n=35)and the control group(n=34).Patients in the treatment group were given oral doxycycline 100 mg twice daily for five days along with standard treatment,whereas patients in the control group received only standard treatment.The objective was to measure the effect of doxycycline on inflammatory markers in dengue infection.Results:On comparative analysis at day 5,there was a statistically significant reduction in the median values of ferritin and CRP in cases compared to the control group(ferritin:P=0.006 and CRP:P=0.006).No significant reduction was noted in the levels of IL-6 and TNF-α.Conclusions:Doxycycline treatment led to a reduction of inflammatory markers in dengue infection.展开更多

Methods Microarrayer was used to print both anti-SF antibodi...Objective To establish and evaluate a protein serum ferritin (SF) and soluble transferrin receptor microarray method for combined measurement of (sTfR). Methods Microarrayer was used to print both anti-SF antibodies I and anti-sTfR antibodies I on each protein microarray. Anti-SF antibodies II and anti-sTfR antibodies II were used as detection antibodies and goat antibodies coupled to Cy3 were used as antibodies Ill. The detection conditions of the quantitative analysis method for simultaneous measurement of SF and sTfR with protein microarray were optimized and evaluated. The protein microarray was compared with commercially available traditional tests with 26 serum samples. Results By comparison experiment, mouse monoclonal antibodies were chosen as the probes and contact printing was chosen as the printing method. The concentrations of SF and sTfR probes were 0.5 mg/mL and 0.5 mg/mL respectively, while those of SF and sTfR detection antibodies were 5 μg/mL and 0.36 μg/mL respectively. Intra- and inter-assay variability was between 3.26% and 18.38% for all tests. The regression coefficients comparing protein microarray with traditional test assays were better than 0.81 for SF and sTfR. Conclusion The present study has established a protein microarray method for combined measurement of SF and sTfR.展开更多
Ferritin stores and releases iron ions in mammals.It is globally important as a drug nanocarrier.This is because of its unique hollow-spherical structure,desirable stability and biological properties.Novel drug-loadin...Ferritin stores and releases iron ions in mammals.It is globally important as a drug nanocarrier.This is because of its unique hollow-spherical structure,desirable stability and biological properties.Novel drug-loading approaches plus various functionalization approaches have been developed to improve ferritin in response to differing demands in disease treatments.Here,we critically review ferritin drug delivery and evaluate its diverse drug-loading and functionalization approaches,we:(1)Introduce basic structural and property information related to ferritin as a drug nanocarrier;(2)Contrast in detail the different means to load drugs and the selection of drug loading means;(3)Discuss multiple ferritin functionalization approaches,together with related advantages and potential risks;and,(4)Compare ferritin with alternative,commonly-used drug nanocarriers.We conclude that despite that no drugs based on ferritin are commercially available,the market potential for it is significant,and evaluate future research directions.Findings from this work will be of immediate benefit and interest to a wide range of researchers and manufacturers for drug delivery using ferritin.展开更多

Accumulation of lipid-laden macrophages(foam cells)is characteristic of atherosclerosis developme nt in the arterial walls.Ferritin nano cages have been found to passively accumulate in the atherosclerotic plaque.Ferr...Accumulation of lipid-laden macrophages(foam cells)is characteristic of atherosclerosis developme nt in the arterial walls.Ferritin nano cages have been found to passively accumulate in the atherosclerotic plaque.Ferritin has been actively investigated as a carrier for contrast agents in atherosclerosis diag no sis.We dem on strate the pote ntial of ferritin as a carrier for therapeutic molecules to mediate cholesterol reducti on from foam cells.Cyclodextrin molecules are chemically conjugated to the ferritin anocages surface or encapsulated within the nanocages using metal co-loadi ng methods.The cyclodextrin-conjugated ferriti n has nano molar affinity to cholesterol molecules.Treatment of foam cells with the conjugates shows decreased levels of intracellular accumulated cholesterol.The preferential localization of ferritin to foam cells is due to transferrin receptor-mediated endocytosis process.These findings show that ferritin nanocages as carriers localize cyclodextrin molecules to foam cells which mediate intracellular cholesterol reductio n,thus highlight!ng its potential use as a therapeutic agent.展开更多
In a recent article on World J Clin Cases 2019;7:3859-3865,Sun et al reported a case of 36-year-old female with macrophage activity syndrome as an onset of systemic lupus erythematosus.Although this is a very interest...In a recent article on World J Clin Cases 2019;7:3859-3865,Sun et al reported a case of 36-year-old female with macrophage activity syndrome as an onset of systemic lupus erythematosus.Although this is a very interesting case,some concerns still need to be addressed.First,the patient had an extremely elevated serum ferritin but a normal C-reactive protein level,which was unparallel with the inflammatory condition before she received any treatments.Second,the diagnosis of systemic lupus erythematosus seemed to be insufficient according to the patient’s medical information presented,most of which were not specific to lupus but could be explained by macrophage activity syndrome.Hence,more medical information on the patient should be provided,and a profound discussion needs to be addressed.展开更多
