The parameters of fluorescence induction kinetics and the maximal light-saturated net CO2 assimilation rate (P-sat) of the flag leaves of four cultivars of winter wheat (Triticum aestivum L.) were compared at three different developing stages for the first time. From the blooming stage to the milky stage, the quantum efficiency of PS II photochemistry (F-v/F-m) declined slightly only at the milk stage. The photochemical quenching co-efficient (qP), actual quantum yield of photosystem II (PS II)electron transport (Phi (PSII)) and P-sat decreased substantially (> 15%), while the non-photochemical quenching co-efficient (qN) increased significantly (> 100%). There existed a linear correlation between the Phi (PSII) and the P-sat (r = 0.918). The results indicate that with the senescence of the flag leaves of winter wheat the photosynthetic efficiency including that of the energy transport and the CO2 assimilation significantly decreased. 展开更多

The aim of the present study was to map quantitative trait loci (QTL) for post-anthesis dry matter accumulation (... Post-anthesis photoassimilation is very important for wheat (Triticum aestivum L.) grain filling. The aim of the present study was to map quantitative trait loci (QTL) for post-anthesis dry matter accumulation (DMA). A set of 120 doubled haploid (DH) lines, derived from winter wheat varieties Hanxuan 10 and Lumai 14, was grown under field conditions in two consecutive growing seasons during 2002-2004 in Beijing. Post-anthesis DMA per culm and related traits, including flag leaf greenness (FLG) and flag leaf weight (FLW; dry weight per flag leaf) at flowering, and grain weight per ear (GWE) were investigated. All traits segregated continuously in the DH population in both trials. The DMA was significantly and positively correlated with GWE, with the correlation coefficients being 0.79 and 0.66 in the 2002-2003 and 2003-2004 growing seasons (both P〈0.01), suggesting the importance of DMA in grain filling. Further correlation analysis showed that FLW was more closely correlated with DMA and GWE than FLG in both growing seasons, indicating that FLW was more important than FLG in influencing DMA and GWE. In total, 30 QTLs for these four traits were mapped and distributed on 10 chromosomes. Phenotypic variations explained by an individual QTL were in the range 5.8%-21.3%, 5.9%-17.2%, 5.1%-18.1%, and 5.6%-16.2% for FLG, FLW, DMA, and GWE, respectively. Eight QTLs for DMA were detected, of which four (on chromosome arms 2AS, 4BL, 5AS, and 7AS) were linked with QTLs for GWE; two (on chromosome arms 5BL and 7BL) coincided with QTLs for FLW. These results may provide useful information for developing marker-assisted selection for the improvement of DMA. 展开更多

背景与目的:长春新碱耐药胃癌细胞SGC7901/VCR是具有P-糖蛋白(P-gp)高表达的典型多药耐药细胞,但P-gp抑制剂维拉帕米(VRP)不能完全逆转其耐药,说明还存在其它耐药机制。我们运用差异蛋白质组学技术研究发现,可溶性耐药相关钙结合蛋白(So... 背景与目的:长春新碱耐药胃癌细胞SGC7901/VCR是具有P-糖蛋白(P-gp)高表达的典型多药耐药细胞,但P-gp抑制剂维拉帕米(VRP)不能完全逆转其耐药,说明还存在其它耐药机制。我们运用差异蛋白质组学技术研究发现,可溶性耐药相关钙结合蛋白(Sorcin)在SGC7901/VCR中表达较SGC7901细胞明显上调。为进一步阐明Sorcin与胃癌细胞耐药的关系,本研究构建了FLAG-Sorcin融合表达载体,探讨Sorcin在胃癌多药耐药中的作用。方法:采用逆转录聚合酶链反应法(reverse transcription-polymerase chain reaction,RT-PCR)扩增Sorcin cDNA片段编码区序列全长,DNA重组技术构建FLAG-Sorcin融合表达载体,经脂质体介导稳定转染SGC7901细胞,RT-PCR及Western blot分别检测转染细胞Sorcin mRNA表达的变化;噻唑蓝(MTT)法测定转染细胞及对照细胞对化疗药物的敏感性。用Sorcin反义核苷酸转染SGC7901-F-SOR细胞,RT-PCR检测Sorcin表达水平的变化,MTT法测定转染后细胞对VCR敏感性的变化。结果:RT-PCR法扩增出616bp全长Sorcin cDNA片段,成功构建了FLAG-Sorcin融合表达载体;RT-PCR及Western blot证实,Sorcin在SGC7901转染细胞中高表达;Sorcin高表达的SGC7901细胞对长春新碱(VCR)、阿霉素(ADM)、紫杉醇(Taxol)、氟尿嘧啶(5-FU)分别产生了8.87倍、6.13倍、6.67倍和2.80倍耐药。Sorcin反义核苷酸转染SGC7901-F-SOR细胞后,RT-PCR验证Sorcin表达被抑制,MTT结果显示细胞对VCR敏感性增加。结论:Sorcin高表达可致SGC7901细胞产生低倍数的多药耐药,说明Sorcin与SGC7901细胞多药耐药密切相关,有望成为逆转胃癌多药耐药的靶点。 展开更多

The ear, together with the flag leaf, is believed to play a major role as a source of assimilates during grain filling in C3cereals. However, the intrusive nature of most of the available methodologies prevents reaching conclusive results in this regard. This study compares the carbon isotope composition(d13C) in its natural abundance in the water‐soluble fractions of the flag leaf blade and the ear with the d13C of mature kernels to assess the relative contribution of both organs to grain filling in durum wheat(Triticum turgidum L. var.durum). The relative contribution of the ear was higher in landraces compared to modern cultivars, as well as in response to nitrogen fertilization and water stress. Such genotypic and environmentally driven differences were associated with changes in harvest index(HI), with the relative contribution of the ear being negatively associated with HI. In the case of the genotypic differences, the lower relative contribution of the ear in modern cultivars compared with landraces is probably associated with the appearance in the former of a certain amount of source limitation driven by a higher HI. In fact, the relative contribution of the ear was far more responsive to changes in HI in modern cultivars compared with landraces. 展开更多

目的:系统评价氟达拉滨+阿糖胞苷+重组人粒细胞集落刺激因子(FLAG)和米托蒽醌+依托泊苷+阿糖胞苷(MEA)方案治疗难治复发性急性髓系白血病(RRAML)的疗效及不良反应。方法:计算机检索Pub Med、Cochrane Library、Web of Science、VIP、Wan... 目的:系统评价氟达拉滨+阿糖胞苷+重组人粒细胞集落刺激因子(FLAG)和米托蒽醌+依托泊苷+阿糖胞苷(MEA)方案治疗难治复发性急性髓系白血病(RRAML)的疗效及不良反应。方法:计算机检索Pub Med、Cochrane Library、Web of Science、VIP、Wan Fang Data、CNKI、CBM等数据库发表的关于FLAG和MEA方案治疗RRAML的随机对照试验(RCT)。由两位研究者独立进行文献筛选、资料提取和质量评价后,采用Rev Man 5. 0软件进行Meta分析。结果:共纳入6个研究,299例患者。Meta分析结果显示:FLAG方案和MEA方案治疗RRAML时,两者的CR[RR=1. 49,95%CI(0. 92,2. 43),P=0. 11]及PR[RR=1. 38,95%CI(0. 82,2. 32),P=0. 23]无明显差异,但FLAG方案治疗组的总有效率高于MEA方案治疗组,两组的差异有统计学意义[RR=1. 98,95%CI(1. 21,3. 24),P=0. 006],其真菌感染率亦高于MEA组,两组的差异有统计学意义[RR=2. 17,95%CI(1. 09,4. 31),P=0. 03]。结论:FLAG方案治疗RRAML的总有效率高于MEA方案,但其真菌感染率亦高。受纳入研究的数量及质量限制,本系统评价结论仍需进一步开展更多大样本、严格设计的随机对照试验加以验证。 展开更多

Male sterility induced by a chemical hybridization agent (CHA) is an important tool for utilizing crop heterosis. Leaves, especially the flag leaves, as CHA initial recipients play a decisive role in inducing male sterility. To investigate effects of different treatment times of CHA-SQ-1 used, morphological, biochemical and physiological responses of wheat flag leaves were detected in thistudy. CHA induced programmed cell death (PCD) as shown in terminal deoxynucleotidyl transferase-mediated dUTP nick end-labelling (TUNEL) and DNA laddering analysis. In the early phase, CHA-SQ-1 trig- gered organelle changes arid PCD in wheat leaves accompanied by excess production of reactive oxygen species (O2- and H202) and down-regulation of the activities of superoxide dismutase (SOD), catalase (CAT) and guaiacol peroxidase (POD). Meanwhile, leaf cell DNAs showed ladder-like patterns on agarose gel, indicating that CHA-SQ-1 led to the activation of the responsible endonuclease. The oxidative stress assays showed that lipid peroxidation was strongly activated and photosynthesis was obviously inhibited in SQ-l-induced leaves. However, CHA contents in wheat leaves gradually reduced along with the time CHA-SQ-1 applied. Young flags returned to an oxidative/antioxidative balance and ultimately developed into mature green leaves. These results provide explanation of the relations between PCD and anther abortion and practical application of CHA for hybrid breeding. 展开更多

Photosynthesis and chlorophyll a fluorescence parameters, photochemical efficiency of PS II (Fv/Fm), photochemical quenching of PS II (qP), nonphotochemical quenching of PS II (NPQ), maximum activity of PS II (Fv/Fo) as well as electron transport rate (ETR), and quantum yield of PS II (ФPS II) were measured on flag leaves of the winter wheat treated by methanol at different concentrations. The results revealed that photosynthesis was greatly improved by methanol, as indicated by higher photosynthetic rates and stomatal conductance. The enhancement effect of methanol on photosynthesis was maintained for 3-4 days. Different methanol concentration treatments also increased intercellular CO2 concentration and transpiration rates. No significant decline was found in Fv/Fm, Fv/Fo, and ФPS II, which revealed no photoinhibition during methanol application in different methanol concentrations. Methanol showing no apparent inhibitory effects indicated higher potential photosynthetic capacity of flag leaves of winter wheat. However, the increase in photosynthesis was not followed by an increase in the photosynthetic activity (Fv/Fm), and fluorescence parameters did not indicate an improvement in intercellular CO2 concentration and PS II photochemical efficiency compared with the control, thereby encouraging us to propose that lower leaf temperatures caused by applied methanol would reduce both dark respiration and photorespiration (most importantly), thus, increasing net CO2 uptake and photosynthetic rates. 展开更多

[Method] Changbai9 (CB9) and Jijing81 (JJ81) were used as experimental materials for pot experiment, and five levels of phosphorus nutrient ( Po - P4 ) were set for each variety, the net photosynthesis rate, Chlorophyll Meter Readings (SPAD Readings), stomatal conductance, transpiration rate, intercellular CO2 concentration, and stomatal limitation were observed and compared between five different phosphorus levels at filling stage. [ Result] The net photosynthesis rate of CB9 reached the highest at P3, and was significantly different from other treatments (P 〈0.05);the net photosynthesis rate of groups with phosphorus of J J81 were higher than that of the control group,and reached the highest at P,. The SPAD Readings of CB9 and JJ81 all achieved the highest at P3, and stomatal conductance and transpiration rate were all at first increased then decreased with the phosphor- us increased in CB9 and JJ81. With the phosphorus increased, intercellular CO2 concentration of CB9 decreased at first and then increased, while in JJ81 the trend was inversed,but the tendency of intercellular CO2 concentration were opposite to that of stomatal limitation in both culti- vars. [ Conclusion] Appropriate adding phosphorus could improve photosynthetic characteristic of rice flag-leaf,but displayed significant genotypic difference. 展开更多

Five QTLs (qLL-2, qLL.4, qLL-6, qLL-IO and qLL-12) controlling the ligule length (LL) were detected on chromosomes 2, 4, 6, 10 and 12, with the variances explained 11.4%, 13.6%, 27.8%, 22.1% and 11.0%, respectively. Using four known genes of ZmGL1, ZmGL2, ZmGL3 and ZmGL4 in maize from the MaizeGDB, their homologs in rice were aligned and integrated into the existing simple sequence repeats linkage map by in silico mapping. A ZmLG1 homolog gene, OsLG1 encoding a squamosa promoter binding protein, was located between the markers RM255 and RM280, which is just identical to the interval of qLL.4 on the long arm of chromosome 4. The results are beneficial to dissection of the ligule molecular mechanism and the study of cereal evolution. 展开更多

On the basis of the phytotron, the effects of high temperature (daily average temperature 25, 30, 35 and 40℃, respectively) on antioxidant enzymes and lipid peroxidation in flag leaves of wheat at 50% relative air moisture during grain fastest filling stage [19-21 days after anthesis (DAA)] were studied. The wheat cultivars tested were Yangmai 9 with weak-gluten and Yangmai 12 with medium-gluten. Compared with 25℃, the higher the temperature was, the higher was the MDA content in flag leaves, while lower were the SOD, POD, and CAT activities. SOD and CAT activities in Yangmai 12 appeared to be more sensitive to high temperature than that in Yangmai 9. But POD activity in Yangmai 12 was less sensitive to high temperature. MDA content in Yangmai 12 was higher than that in Yangmai 9. The 1000-grain weight declined with increase in temperature. 展开更多

To understand genetic patterns of the morphological and physiological traits in flag leaf of barley, a double haploid (DH) population derived from the parents Yerong and Franklin was used to determine quantitative trait loci (QTL) controlling length, width, length/width, and chlorophyll content of flag leaves. A total of 9 QTLs showing significantly additive effect were detected in 8 intervals on 5 chromosomes. The variation of individual QTL ranged from 1.9% to 20.2%. For chlorophyll content expressed as SPAD value, 4 QTLs were identified on chromosomes 2H, 3H and 6H; for leaf length and width, 2 QTLs located on chromosomes 5H and 7H, and 2 QTLs located on chromosome 5H were detected; and for length/width, I QTL was detected on chromosome 7H. The identification of these QTLs associated with the properties of flag leaf is useful for barley improvement in breeding programs. 展开更多
