Background and Aims:Nonbiological artificial liver(NBAL)is frequently used as a first-line treatment for hepatitis B virus-associated acute-on-chronic liver failure(HBV-ACLF).This study aimed to compare the therapeuti...Background and Aims:Nonbiological artificial liver(NBAL)is frequently used as a first-line treatment for hepatitis B virus-associated acute-on-chronic liver failure(HBV-ACLF).This study aimed to compare the therapeutic efficacy and cost-effectiveness ratio(CER)of comprehensive medical treatment,plasma exchange(PE),and double plasma molecular adsorption system(DPMAS)plus half-dose PE(DPMAS+PE)in patients with HBV-ACLF.Methods:A total of 186 patients with HBV-ACLF randomly received comprehensive medical treatment,PE,or DPMAS+PE and were prospectively evaluated.Patients were divided into four subgroups based on the pretreatment prothrombin activity(PTA):Group I(PTA>40%),group II(PTA 30–40%),group III(PTA 20–30%),and group IV(PTA<20%).The main outcome measures were 28 day effectiveness;90 day liver transplantation-free survival;change of biochemical parameters;and CER.Results:DPMAS+PE treatment was associated with significantly higher 28 day effectiveness and 90 day liver transplantation-free survival compared with PE treatment in patients with group I liver failure.Clearance of serum total bilirubin(TBIL),AST,and creatinine(Cr)were significantly higher in the DPMAS+PE group than in the PE group.For subjects with group I liver failure,DPMAS+PE treatment had advantages of lower CER values and better cost-effectiveness.Conclusions:Compared with comprehensive medical treatment and PE alone,DPMAS with halfdose sequential PE treatment more effectively improved TBIL,AST,and Cr in HBV-ACLF patients,improved 28 day effectiveness and 90 day survival rates in patients with group I liver failure,and was more cost effective.DPMAS+PE is a viable NBAL approach for treatment of HBV-ACLF.展开更多
Co_(3)O_(4)spinel oxides have manifested promising activity toward the oxygen evolution reaction(OER)through effective modifications.For them to become top electrocatalysts,however,accurate accounts of the catalytic k...Co_(3)O_(4)spinel oxides have manifested promising activity toward the oxygen evolution reaction(OER)through effective modifications.For them to become top electrocatalysts,however,accurate accounts of the catalytic kinetics are essential to gain a deep understanding of the activity promotion mechanisms.Herein,we use a newly proposed kinetic model based on energetic span as the rate-determining term for the electrocatalytic reaction to throw light on the promotion mechanism of Co_(3)O_(4)interfaced with nickel hydroxides(NiO_(x)H_(y))for the OER.We find that depending on the electrode potential,the OER kinetics at the designed interface between Co_(3)O_(4)and NiO_(x)H_(y)are boosted in entirely different ways.As a result,the OER can occur at a lower onset potential as well as a low Tafel slope.This work emphasizes the benefit of using rational theoretical models for electrocatalyst design.展开更多
Inducible expression systems are indispensable for precise regulation and in-depth analysis of biological process.Binary Tet-On system has been widely employed to regulate transgenic expression by doxycycline.Previous...Inducible expression systems are indispensable for precise regulation and in-depth analysis of biological process.Binary Tet-On system has been widely employed to regulate transgenic expression by doxycycline.Previous pig models with tetracycline regulatory elements were generated through random integration.This process often resulted in uncertain expression and unpredictable phenotypes,thus hindering their applications.Here,by precise knock-in of binary Tet-On 3G elements into Rosa26 and Hipp11 locus,respectively,a double knock-in reporter pig model was generated.We characterized excellent properties of this system for controllable transgenic expression both in vitro and in vivo.Two att P sites were arranged to flank the td Tomato to switch reporter gene.Single or multiple gene replacement was efficiently and faithfully achieved in fetal fibroblasts and nuclear transfer embryos.To display the flexible application of this system,we generated a pig strain with Dox-inducing h KRASexpression through phiC31 integrase-mediated cassette exchange.After eight months of Dox administration,squamous cell carcinoma developed in the nose,mouth,and scrotum,which indicated this pig strain could serve as an ideal large animal model to study tumorigenesis.Overall,the established pig models with controllable and switchable transgene expression system will provide a facilitating platform for transgenic and biomedical research.展开更多
NiFe_(2)O_(4) nanoparticles(<10 nm)embedded in a NiO matrix have been fabricated by calcining the corresponding Ni^(Ⅱ)Fe^(Ⅲ)-layered double hydroxide(LDH)precursors at high temperature(500℃).Compared with the Ni...NiFe_(2)O_(4) nanoparticles(<10 nm)embedded in a NiO matrix have been fabricated by calcining the corresponding Ni^(Ⅱ)Fe^(Ⅲ)-layered double hydroxide(LDH)precursors at high temperature(500℃).Compared with the NiFe_(2)O_(4)/NiO nanocomposite obtained by calcination of a precursor prepared by a traditional chemical coprecipitation method,those derived from NiFe-LDH precursors show much higher blocking temperatures(TB)(~380 K).The enhanced magnetic stability can be ascribed to the much stronger interfacial interaction between NiFe_(2)O_(4) and NiO phases due to the topotactic nature of the transformation of the LDH precursor to the NiFe_(2)O_(4)/NiO composite material.Through tuning the Ni^(Ⅱ)/Fe^(Ⅲ) molar ratio of the NiFe-LDH precursor,the NiFe_(2)O_(4) concentration can be precisely controlled,and the TB value as well as the magnetic properties of the final material can also be regulated.This work represents a successful example of the fabrication of ferro(ferri)magnetic(FM)/antiferrimagnetic(AFM)systems with high magnetic stability from LDH precursors.This method is general and may be readily extended to other FM/AFM systems due to the wide range of available LDH precursors.展开更多

Absorption peak splitting corresponding to the stretching oscillation of the Mn-O bond, together with a shift of peak position, is observed below the Curie temperature. These features weaken and even disappear as the samples are warmed up to the Curie temperature, which indicates that this anomaly may be a result of phase separation in the compounds.展开更多
