Leptin is the protein product encoded by the obese (ob)gene. Leptin is the protein product encoded by the obese (ob)gene. It is a circulating hormone produced primarily by the adipose tissue. ob/ob mice with mutations of the gene encoding leptin become morbidly obese, infertile, hyperphagic, hypothermic,and diabetic. Since the cloning of leptin in 1994, our knowledge in body weight regulation and the role played by leptin has increased substantially. We now know that leptin signals through its receptor, OB-R, which is a member of the cytokine receptor superfamily. Leptin serves as an adiposity signal to inform the brain the adipose tissue mass in a negative feedback loop regulating food intake and energy expenditure. Leptin also plays important roles in angiogenesis, immune function, fertility and bone formation. Humans with mutations in the gene encoding leptin are also morbidly obese and respond to leptin treatment,demonstrating that enhancing or inhibiting leptin’s activities in vivo may have potential therapeutic benefits. 展开更多

目的 调查几种细胞因子及其受体在放射性肺纤维化 (RPF)发生中的作用。方法采用免疫组织化学、细胞免疫化学和原位杂交的方法对RPF大鼠肺组织细胞进行了研究。结果正常大鼠肺组织有TGFβ1和TGFβR的弱表达 ,照射动物肺脏TGFβ1的表达... 目的 调查几种细胞因子及其受体在放射性肺纤维化 (RPF)发生中的作用。方法采用免疫组织化学、细胞免疫化学和原位杂交的方法对RPF大鼠肺组织细胞进行了研究。结果正常大鼠肺组织有TGFβ1和TGFβR的弱表达 ,照射动物肺脏TGFβ1的表达于照射后 2周开始增高 ,从第 8周直到 3月表达水平最高。肺脏表达TGFβ1的部位主要是支气管上皮细胞、肺泡巨噬细胞、肺泡上皮细胞、支气管和血管平滑肌细胞以及成纤维细胞等。TGFβ2的表达变化规律除了比TGFβ1稍弱外类似于TGFβ1。TGFβR表达增强的时间比TGFβ1稍晚 ,于第 8周开始增强 ,直到 1年也保持较高水平的表达。bFGF和PDGF在照射后 2~ 3个月表达水平明显增高。对支气管肺泡灌洗液中的细胞TGFβ1免疫细胞化学研究表明 :其中的细胞 ,特别是巨噬细胞表达时间出现得较早 ,于照射后 1周即有许多巨噬细胞呈现TGFβ1较强的表达。从肺组织培养的细胞研究表明 ,来源于照射后 3月肺组织的成纤维细胞呈现较强的TGFβ1阳性。用地高辛标记的TGFβ1探针和α1(I)寡核苷酸探针原位杂交试验表明 ,TGFβ1mRNA表达水平于 2~ 8周表达最高 ,而α1(I)前胶原mR NA则从 6周开始增强 ,到 3月表达出现高峰 ,稍滞后于TGFβ1的峰值期 ,与TGFβ1的表达呈现明显的时空关联和交叠现象。 展开更多

Prolactin and interleukin-6 are described in detail as the best studied examples. AIM: To determine whether a specific adenosine A2A receptor agonist (ATL-146e) can ameliorate aspirin-induced gastric mucosal lesions in rats, and reduce neutrophil accumulation and production of pro-inflammatory cytokines. METHODS: Gastric lesions were produced by oral gavage of aspirin (200 mg/kg) and HCI (0.15 mol/L, 8.0 mL/kg). 4-{3-[6-Amino-9-(5-ethylcarbamoyl-3,4- dihydroxy-tetrahydro-furan-2-yl)-9H-purin-2-yl]-prop-2- ynyl}-cyclohexanecarboxylic acid methyl ester (ATL-146e, 2.5-5μg/kg, IP) was injected 30 min before the administration of aspirin. Tissue myeloperoxidase (MPO) concentration in gastric mucosa was measured as an index of neutrophil infiltration. Gastric mucosal concentrations of tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) were determined by ELISA. Also, we examined the effect of ATL-146e on tissue prostaglandin E2 (PGE2) production and gastric secretion. RESULTS: Intragastric administration of aspirin induced multiple hemorrhagic erosions in rat gastric mucosa. The total length of gastric erosions (ulcer index) in control rats was 29.8±7.75 mm and was reduced to 3.8±1.42 mm alter pretreatment with 5.0 g/kg ATL-146e (P〈 0.01). The gastric contents of MPO and pro-inflammatory cytokines were all increased after the administration of aspirin and reduced to nearly normal levels by ATL-146e. Gastric mucosal PGE2 concentration was not affected by intraperitoneal injection of ATL-146e. CONCLUSION: The specific adenosine A2A receptor agonist, ATL-146e, has potent anti-ulcer effects presumably mediated by its anti-inflammatory properties. 展开更多

AIM: To evaluate the effect of Chinese traditional medicinal prescription, JIANPI HUOXUE decoction (JHD) on cytokine secretion pathway in rat liver induced by lipopolysaccharide (LPS). METHODS: Twenty-four male SD rats were divided into normal group (n = 4), model group (n = 10) and JHD group (n = 10) randomly. Rats in model group and JHD group were administrated with normal saline or JHD via gastrogavage respectively twice a day for 3 d. One hour after the last administration, rats were injected with LPS via tail vein, 50 μg/kg. Simultaneously, rats in normal group were injected with equivalent normal saline. After LPS stimulation for 1.5 h, serum and liver tissue were collected. Pathological change of liver tissues was observed through hematoxylineosin (H.E.) staining. Tumor necrosis factor alpha (TNF-α) in serum were assayed by enzyme linked immunosorbent assay (ELISA). The protein expression of TNF-α, phosphorylated inhibit-κB (p-κB) and CD68 in liver were assayed by Western blot. The distribution of CD68 protein in liver was observed through immunohistochemical staining. The mRNA expression of TNF-α, interleukin-6 (IL-6), CD14, toll-like receptor 2 (TLR2) and TLR4 in liver were assayed by real-time RT-PCR.RESULTS: Predominant microvesicular change, hepatocyte tumefaction and cytoplasm dilution were observed in liver tissues after LPS administration as well as obvious CD68 positive staining in hepatic sinusoidal. After LPS stimulation, serum TNF-α (31.35 ± 6.06 vs 12225.40 ± 9007.03, P 〈 0.05), protein expression of CD68 (1.13 ± 0.49 vs 3.36 ±1.69, P 〈 0.05), p-IκB (0.01 ±0.01 vs 2.07 + 0.83, P 〈 0.01) and TNF-α (0.27 ± 0.13 vs 1.29 ± 0.37, P 〈 0.01) in liver and mRNA expression of TNF-α (1.96 ± 2.23 vs 21.45 ±6.00, P 〈 0.01), IL-6 (4.80 ± 6.42 vs 193.50 ± 36.36, P 〈 0.01) and TLR2 (1.44 ± 0.62 vs 4.16 ± 0.08, P 〈 0.01) in liver were also increased significantly. 目的探讨多项血清中细胞因子及可溶性细胞因子受体蛋白在类风湿关节炎临床诊断中的应用价值。方法选择2015年~2019年在本院确诊为类风湿关节炎(RA)的患者59例作为RA组,并以在我院接受定期体检的年龄和性别匹配的健康受试者46例作为健康... 目的探讨多项血清中细胞因子及可溶性细胞因子受体蛋白在类风湿关节炎临床诊断中的应用价值。方法选择2015年~2019年在本院确诊为类风湿关节炎(RA)的患者59例作为RA组,并以在我院接受定期体检的年龄和性别匹配的健康受试者46例作为健康对照组。通过人细胞因子蛋白芯片检测其中5例RA组及5例健康对照组血清样本,选取4种差异表达细胞因子LIMPII、ROBO3、Periostin、IGFBP-4及可溶性受体蛋白2B4、Tie-2,于另外54例RA组及41例健康对照组血清样本中,对其六者血清水平进行ELISA验证。Spearman相关分析检测细胞因子及可溶性细胞因子受体蛋白与临床指标(包括:类风湿因子(RF)、红细胞沉降率(ESR)、C反应蛋白(CRP)、疾病活动评分(DAS28)及健康评估问卷(HAQ))的关系;受试者工作特征曲线(ROC)分析评估其诊断价值。结果与健康对照相比,2B4、LIMPII、Tie-2、ROBO3、Periostin和IGFBP-4在RA血清中的表达显著升高(P<0.01)。同时,RA血清中LIMPII、ROBO3和Periostin血清水平与类风湿因子、C反应蛋白、疾病活动评分及健康评估问卷等反映疾病活动程度评价指标正相关(P<0.01)。血清2B4、LIMPⅡ、Periostin、ROBO3、IGFBP-4和Tie-2表达水平均对RA的诊断有显著意义(P<0.05),其中2B4曲线下面积为最大(AUC:0.861,P<0.001),其次依顺序LIMPII、ROBO3、Periostin、Tie-2、和IGFBP-4。结论血清LIMPII、ROBO3和Periostin的表达水平可反映RA的疾病活动度,此外血清2B4、LIMPⅡ、Periostin、ROBO3、IGFBP-4和Tie-2可能作为RA的诊断的生物标记物。 展开更多

Gastric cancer,a prevalent malignancy worldwide,ranks sixth in terms of frequency and third in fatality,causing over a million new cases and 769000 annual deaths.Predominant in Eastern Europe and Eastern Asia,risk factors include family medical history,dietary habits,tobacco use,Helicobacter pylori,and Epstein-Barr virus infections.Unfortunately,gastric cancer is often diagnosed at an advanced stage,leading to a grim prognosis,with a 5-year overall survival rate below 5%.Surgical intervention,particularly with D2 Lymphadenectomy,is the mainstay for early-stage cases but offers limited success.For advanced cases,the National Comprehensive Cancer Network recommends chemotherapy,radiation,and targeted therapy.Emerging immunotherapy presents promise,especially for unresectable or metastatic cases,with strategies like immune checkpoint inhibitors,tumor vaccines,adoptive immunotherapy,and nonspecific immunomodulators.In this Editorial,with regards to the article“Advances and key focus areas in gastric cancer immunotherapy:A comprehensive scientometric and clinical trial review”,we address the advances in the field of immunotherapy in gastric cancer and its future prospects. 展开更多

The counter receptor was presented. Membrane bound macrophage colony-stimulating factor (m-M-CSF) and its receptor (M-CSF-R) were shown in human leukemic cell line J6-1 as autojuxtacrine mechanism. Soluble M-CSF receptor (sM-CSF-R), which was isolated from J6-1 cells membrane, was added into J6-1 cell culture. It was observed inhibition of J6-1 cell proliferation, decreasing of mitosis index and ratio of multinuclear cells, enlargement of cell diameter and volume, down regulation of numerous surface antigens. Dramatic change of intracellular pH was shown between several min to 20 min after treatment of sM-CSF-R. It suggested that some information was transmitted via m-M-CSF from sM-CSF-R. This counter signaling was not influenced by saccharification of m-M-CSF. 展开更多

Objective To study the significance of scavenger receptor class A(SR A)in mediating human peripheral blood monocyte to uptake oxidized low density lipoprotein(OxLDL) and promoting the atherosclerotic immuno pathological lesion in the local blood vessel. Methods With the Digoxenin labeled Oligonucleotide probes In situ Hybridization, this research investigated the effects of OxLDL on the mRNA expression of proinflammatory cytokines including MCP 1, bFGF, PDGF and IL 10 in the human peripheral blood monocyte and whether fucoidin, a peculiarly inhibitory ligand for SR A, would influence this process. Results Monocyte was significantly increased the mRNA expression of MCP 1, bFGF, PDGF and IL 10 in a dose dependent manner after incubating with OxLDL (10,15,20,25,30?mg·L -1 , respectively)for 24 hours( P < 0.001 ). Fucoidin(50,100,150,200,250?mg·mL -1 , respectively)completely inhibited OxLDL(20?mg·L -1 )from inducing monocyte the mRNA expression of above proinflammatory cytokines( P < 0.001 ). Conclusion OxLDL can stimulate human peripheral blood monocyte to give expression to proinflammatory cytokines mRNA in a dose dependent manner, while a peculiarly inhibitory ligand for SR A fucoidin has an obviously opposed role. 展开更多

In the pathogenesis of major depressive disorder, chronic stress-related neuroinflammation hinders favorable prognosis and antidepressant response. Mitochondrial DNA may be an inflammatory trigger, after its release from stress-induced dysfunctional central nervous system mitochondria into peripheral circulation. This evidence supports the potential use of peripheral mitochondrial DNA as a neuroinflammatory biomarker for the diagnosis and treatment of major depressive disorder. Herein, we critically review the neuroinflammation theory in major depressive disorder, providing compelling evidence that mitochondrial DNA release acts as a critical biological substrate, and that it constitutes the neuroinflammatory disease pathway. After its release, mitochondrial DNA can be carried in the exosomes and transported to extracellular spaces in the central nervous system and peripheral circulation. Detectable exosomes render encaged mitochondrial DNA relatively stable. This mitochondrial DNA in peripheral circulation can thus be directly detected in clinical practice. These characteristics illustrate the potential for mitochondrial DNA to serve as an innovative clinical biomarker and molecular treatment target for major depressive disorder. This review also highlights the future potential value of clinical applications combining mitochondrial DNA with a panel of other biomarkers, to improve diagnostic precision in major depressive disorder. 展开更多
