The promoting molecular mechanism of alphafetoprotein on the growth of human hepatoma Bel7402 cell line 被引量:59

AIM: The goal of this study was to characterize the AFP receptor, its possible signal transduction pathway and its proliferative functions in human hepatoma cell line Bel 7402. METHODS: Cell proliferation enhanced by AFP was detected by MTT assay, 3H-thymidine incorporation and S-stage percentage of cell cycle analysis. With radioactive labeled 125I-AFP for receptor binding assay; cAMP accumulation, protein kinase A activity were detected by radioactive immunosorbent assay and the change of intracellular free calcium (Ca2+i) was monitored by scanning fluorescence intensity under TCS-NT confocal microscope. The expression of oncogenes N- ras, p 53, and p21( ras ) in the cultured cells in vitro were detected by Northern blotting and Western blotting respectively. RESULTS: It was demonstrated that AFP enhanced the proliferation of human hepatoma Bel 7402 cell in a dose dependent fashion as shown in MTT assay, (3)H-thymidine incorporation and S-phase percentage up to 2-fold. Two subtypes of AFP receptors were identified in the cells with Kds of 1.3 x 10(-9)mol.L(-1) and 9.9 x10(-8)mol. (-1)L respectively. Pretreatment of cells with AFP resulted in a significant increase (625%) in cAMP accumulation. The activity of protein kinase A activity were increased up to 37.5, 122.6, 73.7 and 61.2% at treatment time point 2, 6, 12 and 24 hours. The level of intracellular calcium were elevated after the treatment of alpha-fetoprotein and achieved to 204% at 4 min. The results also showed that AFP(20mg.L(-1)) could upregulate the expression of N- ras oncogenes and p 53 and p21( ras ) in Bel 7402 cells. In the later case,the alteration were 81.1%(12h) and 97.3%(12h) respectively compared with control. CONCLUSION: These results demonstrate that AFP is a potential growth factor to promote the proliferation of human hepatoma Bel 7402 cells. Its growth-regulatory effects are mediated by its specific plasma membrane receptors coupled with its transmembrane signaling transduction through the pathway of cAMP-PKA and intracellular calcium 展开更多

AIM: The existence and properties of alpha-fetoprotein (AFP) receptor on the surface of NIH 3T3 cells and the effects of AFP on cellular signal transduction pathway were investigated. METHODS: The effect of AFP on the proliferation of NIH 3T3 cells was measured by incorporation of 3H-TdR. Receptor-binding assay of 125I-AFP was performed to detect the properties of AFP receptor in NIH 3T3 cells. The influences of AFP on the [cAMP]i and the activities of protein kinase A (PKA) were determined. Western blot was used to detect the change of K-ras P21 protein expression. RESULTS: The proliferation of NIH 3T3 cells treated with 0-80 mg/L of AFP was significantly enhanced. The Scatchard analysis indicated that there were two classes of binding sites with KD of 2.722 x 10(-9)M (Bmax=12810 sites per cell) and 8.931 x 10(-8)M (Bmax=119700 sites per cell) respectively. In the presence of AFP (20 mg/L), the content of cAMP and activities of PKA were significantly elevated . The level of K-ras P21 protein was upregulated by AFP at the concentration of 20 mg/L. The monoclonal antibody against AFP could reverse the effects of AFP on the cAMP content, PKA activity and the expression of K-ras p21 gene. CONCLUSION: The effect of AFP on the cell proliferation was achieved by binding its receptor to trigger the signal transduction pathway of cAMP-PKA and alter the expression of K- ras p21 gene. 展开更多

A bench-scale cyclic activated sludge technology (CAST) was operated to study the biological phosphorus removal performance and a series of batch tests was carried out to demonstrate the accumulation of denitrifying polyphosphate-accumulating organisms (DNPAOs) in CAST system. Under all operating conditions, step-feed CAST with enough carbon sources in influent had the highest nitrogen and phosphorus removal efficiency as well as good sludge settling performance. The average removal rate of COD, NH4^+-N, PO4^3--P and total nitrogen (TN) was 88.2%, 98.7%, 97.5% and 92.1%, respectively. The average sludge volume index (SVI) was 133 mL/g. The optimum anaerobic/aerobic/anoxic (AOA) conditions for the cultivation of DNPAOs could be achieved by alternating anoxic/oxic operational strategy, thus a significant denitrifying phosphorus removal occurred in step-feed CAST. The denitrification of NO^x--N completed quickly due to step-feed operation and enough carbon sources, which could enhance phosphorus release and further phosphorus uptake capability of the system. Batch tests also proved that polyphosphate-accumulating organisms (PAOs) in the step-feed process had strong denitrifying phosphorus removal capacity. Both nitrate and nitrite could be used as electron acceptors in denitrifying phosphorus removal. Low COD supply with step-feed operation strategy would favor DNPAOs accumulation. 展开更多

Spraying 1-2 mmol/L solution of NaHSO 3 on rice ( Oryza sativa L.) leaves resulted in the enhancement of net photosynthetic rate for more than three days. It was also observed that NaHSO 3 application caused increases both in ATP content in leaves and the millisecond_delayed light emission of leaves. The increase in net photosynthetic rate caused by NaHSO 3 treatment was similar to that by PMS (phenazine methosulfate) treatment. The grain yield of treated rice was enhanced approximately by 10% after duplicated application of NaHSO 3 in milk_ripening stage. It is suggested that the enhancement of photosynthesis by NaHSO 3 treatment resulted from the effect of increasing ATP supplement. Concomitant with an increase in the photosynthetic rate and ATP content in leaves, the transient increase in chlorophyll fluorescence after the termination of actinic light, which could be used as an index of the cyclic electron flow, was also enhanced by low concentration of NaHSO 3 treatment. Basing on these results it is proposed that the increase in rice photosynthesis caused by low concentrations of NaHSO 3 could be due to the stimulation of the cyclic electron flow around PSⅠ which in turn the enhancement of the coupled photophosphorylation and photosynthesis. 展开更多

Ti/Sb-SnO2 anodes were prepared by thermal decomposition to examine the influence of the amount of Sb dopant on the structure and electrocatalytic capability of the electrodes in the oxidation of 4-chlorophenol. The physicochemical properties of the Sb-SnO2 coating were markedly influenced by different amounts of Sb dopant. The electrodes, which contained 5% Sb dopant in the coating, presented a much more homogenous surface and much smaller mud-cracks, compared with Ti/Sb-SnO2 electrodes containing 10% or 15% Sb dopant, which exibited larger mud cracks and pores on the surface. However, the main microstructure remained unchanged with the addition of the Sb dopant. No new crystal phase was observed by X-ray diffraction (XRD). The electrochemical oxidation of 4-chlorophenol on the Ti/SnO2 electrode with 5% Sb dopant was inclined to electrochemical combustion; while for those containing more Sb dopant, intermediate species were accumulated. The electrodes with 5% Sb dopant showed the highest efficiency in the bulk electrolysis of 4-chlorophenol at a current density of 20 mA/cm^2 for 180 min; and the removal rates of 4-chlorophenol and COD were 51.0% and 48.9%, respectively. 展开更多

In the tests, the influence of the shear-span ratio, axial compression ratio and shear steel plate ratio on the hysteretic behavior, energy dissipation, strength degradation, stiffness degradation, skeleton curve and ductility of the ASCCs is studied. Rock engineering is highly susceptible to cyclic loads resulting from earthquakes,quarrying or rockbursts.Acquiring the fatigue properties and failure mechanism of rocks is pivotal for long-term stability assessment of rock engineering structures.So far,significant progress has been gained on the mechanical characteristics of rocks subjected to cyclic loading.For providing a global insight of typical results and main features of rocks under cyclic loading conditions,this study comprehensively reviews the state-ofthe-art of deformation and failure mechanism and fatigue constitutive relationship of rocks subjected to cyclic loading in the past 60 years.Firstly,cyclic tests on rocks are classified into different types based on loading paths,loading parameters,loading types and environment conditions.Secondly,representative results are summarized and highlighted in terms of the fatigue response of rocks,including the deformation degradation,energy dissipation,damage evolution and failure characteristics;both laboratory testing and numerical results are presented,and various measurement techniques such as X-ray microcomputed tomography(micro-CT)and digital image correlation(DIC)are considered.Thirdly,the influences of cyclic loads on the mechanical characteristics of rocks are discussed,including the cyclic stress,frequency,amplitude and waveform.Subsequently,constitutive relationships for rocks subjected to cyclic loading are outlined,in which typical fatigue constitutive models are compared and analyzed,regarding the elastoplastic model,the internal variable model,the energy-based damage model and the discrete element-based model.Finally,some ambiguous questions and prospective research are interpreted and discussed. 展开更多

Based on a series of cyclic triaxial tests, the effect of cyclic frequency on the undrained behaviors of undisturbed marine clay is investigated. For a given dynamic stress ratio, the accumulated pore water pressure and dynamic strain increase with the number of cycles. There exists a threshold value for both the accumulated pore water pressure and dynamic strain, below which the effect of cyclic frequency is very small, but above which the accumulated pore water pressure and dynamic strain increase intensely with the decrease of cyclic frequency for a given number of cycles. The dynamic strength increases with the increase of cyclic frequency, whereas the effect of cyclic frequency on it gradually diminishes to zero when the number of cycles is large enough, and the dynamic strengths at different frequencies tend to the same limiting minimum dynamic strength. The test results demonstrate that the reasons for the frequency effect on the undrained soil behaviors are both the creep effect induced by the loading rate and the decrease of sample effective confining pressure caused by the accumulated pore water pressure. 展开更多

The results showed that the profound expression of HO-1 protein as well as HO activity was 1.8- and 2.0-fold increased respectively in the transfected cells compared to the non-transfected ones. The treatment of VSMC with different concentrations of H2O2 led to the remarkable cell damage as indicated by survival rate and LDH leakage. However, the resistance of the HO-1 transfected VSMC against H2O2 was significantly raised. This protective effect was dramatically diminished when the transfected VSMC were pretreated with ZnPP-IX, a specific inhibitor of HO, for 24 h. In addition, we found that the growth potential of the transfected cells was significantly inhibited directly by increased activity of HO-1, and this effect might be related to decreased phosphorylation of MAPK. These results suggest that the overexpression of introduced hHO-1 is potentially able to reduce the risk factors of atherosclerosis, partially due to its cellular protection against oxidative injury and to its inhibitory effect on cellular proliferation. 展开更多

Furthermore, a dum my static method for estimating the undrained bearing capacity for offshore soft foundation under wave loads is developed. It can consider the effect of the difference of cyclic stress for different parts of the foundation on both the degradation strength of the foundation soil and the bearing capacity so that the estimated result can better reflect the real condition of foundation under cyclic loading. AIM: To study the effects of aminoguanidine (AG) and two L-arginine analogues N(omega)-nitro-L-arginine methyl ester (L-NAME) and N(omega)-nitro-L-arginine (L-NNA) on nitric oxide (NO) production induced by cytokines (TNF-alpha, IL-1 beta, and IFN-gamma) and bacterial lipopolysaccharide (LPS) mixture (CM) in the cultured rat hepatocytes, and examine their mechanisms action. METHODS: Rat hepatocytes were incubated with AG, L-NAME, L-NNA, Actinomycin D (ActD) and dexamethasone in a medium containing CM (LPS plus TNF-alpha, IL-1 beta, and IFN-gamma) for 24h. NO production in the cultured supernatant was measured with the Griess reaction. Intracellular cGMP level was detected with radioimmunoassy. RESULTS: NO production was markedly blocked by AG and L-NAME in a dose-dependent manner under inflammatory stimuli condition triggered by CM in vitro. The rate of the maximum inhibitory effects of L-NAME (38.9%) was less potent than that obtained with AG(53.7%, P 【 0.05). There was no significant difference between the inhibitory effects of AG and two L-arginine analogues on intracellular cGMP accumulation in rat cultured hepatocytes. Non-specific NOS expression inhibitor dexamethasone (DEX)and iNOS mRNA transcriptional inhibitor ActD also significantly inhibited CM-induced NO production. AG(0.1 mmol x L(-1)) and ActD (0.2 ng x L(-1)) were equipotent in decreasing NO production induced by inflammatory stimuli in vitro, and both effects were more potent than that induced by non-selectivity NOS activity inhibitor L-NAME (0.1 mmol x L(-1)) under similar stimuli conditions (P【0.01). CONCLUSION: AG is a potent selective inhibitor of inducible isoform of NOS,and the mechanism of action may be not only competitive inhibition in the substrate level, but also the gene expression level in rat hepatocytes. 展开更多

High temperature treatment causes thermal damage to rocks in deep mining.To study the thermal effect on the energy dissipation of rocks during the dynamic cyclic loading,cyclic impact loading experiments of heat-treated rocks were carried out using the splitting Hopkinson pressure bar(SHPB)experimental system.The correlations among the energy dissipation,energy dissipation rate,impact times,accumulated absorbed energy per volume,failure mode and temperature were analyzed.The results show that the reflected energy under the first impact increases and finally exceeds the absorbed energy when the temperature increases;however,the total reflected energy decreases above 200℃.The absorbed energy under the first impact and the total absorbed energy all decrease as the temperature increases,the rates of which decrease accordingly.And the same phenomenon appears for the transmitted energy and the rate of the transmitted energy.On the contrary,the rate of the reflected energy increases with the rising temperature.When the temperature increases,the fewer impact times are needed to destroy the sample.In addition,the failure modes are different when the rock is treated at different temperatures;that is,when the temperature is high,even though the absorbed energy is low,the sample breaks into powder after several impacts. 展开更多
