Pentachlorophenol(PCP), a priority pollutant due to its persistence and high toxicity, has been used worldwide as a pesticide and biocide. To understand the adverse effects of PCP, adult male white-rumped munias(Lonchura striata) were orally administrated commercial PCP mixed with corn oil at dosages of 0, 0.05, 0.5, and 5 mg/(kg·day) for 42 day. Gas chromatography–mass spectrometry(GC–MS) analysis found that PCP was preferentially accumulated in the kidney rather than in the liver and muscle in all exposure groups. To examine the function of CYP1A in pollutant metabolism, we isolated two full-length cDNA fragments(designated as CYP1A4 and CYP1A5) from L. striata liver using reverse transcription-polymerase chain reaction and rapid amplification of cDNA ends. PCP induced the expression of CYP1A5, although no obvious change was observed in CYP1A4 expression. Furthermore, PCP significantly elevated the activities of ethoxyresorufin O-deethylase and methoxyresorufin O-demethylase and decreased the activity of benzyloxy-trifluoromethyl-coumarin, with no significant responses observed in benzyloxyresorufin O-debenzylase. PCP induced significant changes in antioxidant enzyme(superoxide dismutase and catalase) activities and malondialdehyde content, but decreased glutathione peroxidase(GSH-Px) and glutathione S-transferase activities and GSH content in the liver of L. striata. The present study demonstrated that PCP had hepatic toxic effects by affecting CYP1As and anti-oxidative status. 展开更多

CYP2E1 immunoglobulin G4 subclass antibodies after desflurane anesthesia

AIM: To investigate CYP2E1 IgG4 autoantibody levels and liver biochemical markers in adult patients after anesthesia with desflurane. METHODS: Forty patients who were > 18 years old and undergoing elective surgery under general anes-thesia with desflurane were studied. Alpha-glutathione-S-transferase(aGST) and IgG4 antibodies againstCYP2E1 were measured preoperatively and 96 h post-operatively, as well as complete blood count, prothrom-bin time(PT), activated partial thromboplastin time(aPTT), international normalized ratio(INR), aspartate aminotransferase(SGOT), alanine aminotransferase(SGPT), g-glutamyl-transpeptidase(gGT), alkaline phosphatase, total serum proteins, albumin and bili-rubin. A separate group of 8 patients who received re-gional anesthesia was also studied for calibration of the methodology used for CYP2E1 IgG4 and aGST measure-ments. Student's t-test and the Mann-Whitney U test were used for comparison of the continuous variables, and Fisher's exact test was used for the categorical variables. All tests were two-tailed, with statistical sig-nificance set as P < 0.05.RESULTS: None of the patients developed postopera-tive liver dysfunction, and all patients were successfully discharged from the hospital. No statistically significant difference was observed regarding liver function tests(SGOT, SGPT, γGT, bilirubin, INR), aGST and CYP2E1 IgG4, before and after exposure to desflurane. After dividing patients into two subgroups based on whether or not they had received general anesthesia in the past, no significant difference in the levels of CYP2E1 IgG4 was observed at baseline or 96 h after desflurane administration(P = 0.099 and P = 0.051, respectively). Alpha-GST baseline levels and levels after the interven-tion also did not differ significantly between these two subgroups(P > 0.1). 目的:使用"鸡尾酒"(cocktail)探针药物法研究苦参对大鼠CYP1A2、CYP2C9、CYP2E1和CYP3A4体内代谢活性的影响。方法:将大鼠随机分为4组,空白对照组、实验组、诱导剂组和抑制剂组。实验组大鼠每日灌胃给予苦参溶液100mg·kg... 目的:使用"鸡尾酒"(cocktail)探针药物法研究苦参对大鼠CYP1A2、CYP2C9、CYP2E1和CYP3A4体内代谢活性的影响。方法:将大鼠随机分为4组,空白对照组、实验组、诱导剂组和抑制剂组。实验组大鼠每日灌胃给予苦参溶液100mg·kg-1,空白对照组灌胃给予与实验组体积相同的生理盐水;诱导剂组腹腔注射苯巴比妥注射液50 mg·kg-1;抑制剂组腹腔注射西咪替丁注射液50 mg·kg-1,各组均为每日1次,连续5 d。各组分别于第6天注射"鸡尾酒"探针药物溶液,于给药前及给药后不同时间点取血,使用HPLC检测各探针药物的血药浓度,计算其药动学参数,评价相应的CYP450亚型的体内代谢活性。结果:实验组咖啡因和甲苯磺丁脲的AUC0-t和AUC0-∞明显低于空白对照组和抑制剂组(P<0.05),高于诱导剂组(P<0.05);而CL明显高于空白对照组和抑制剂组(P<0.05),低于诱导剂组(P<0.05)。实验组氯唑沙宗的AUC0-t和AUC0-∞明显低于空白对照组和抑制剂组(P<0.05),CL明显高于空白对照组和抑制剂组(P<0.05);与诱导剂组无显著差异(P>0.05)。咪达唑仑的主要药动学参数与空白对照组比较均没有显著差异(P>0.05)。结论:苦参可诱导大鼠体内CYP1A2和CYP2C9,其诱导强度低于苯巴比妥;苦参可诱导大鼠体内CYP2E1,其诱导强度与苯巴比妥相似;苦参对大鼠CYP3A4体内活性未见显著影响。 展开更多
