目的探讨支气管哮喘患者外周血CD4~^+、CD8^+T细胞、CD4^+CD25^+调节性T细胞表达水平与肺功能的关系。方法收集2012年3月~2015年6月北京市房山区中医医院呼吸科收治的支气管哮喘患者106例,其中急性发作(急性发作组)患者59例,非急性发作... 目的探讨支气管哮喘患者外周血CD4~^+、CD8^+T细胞、CD4^+CD25^+调节性T细胞表达水平与肺功能的关系。方法收集2012年3月~2015年6月北京市房山区中医医院呼吸科收治的支气管哮喘患者106例,其中急性发作(急性发作组)患者59例,非急性发作(非急性发作组)患者47例,同时以72例健康者作为对照组。对所有入组研究对象进行肺活量(VC),一秒用力呼气容积占用力肺活量的百分比(FEV1/FVC%)及呼气流量峰值(PEF)检查,采用流式细胞仪检测外周血CD4^+、CD8^+T细胞及CD4^+CD25^+调节性T细胞水平,并对急性发作期患者CD4^+、CD8^+T细胞、CD4^+CD25^+Treg水平与肺功能的相关性进行分析。结果 1非急性发作组及急性发作组的肺功能指标与对照组比较均显著降低(P<0.05);与非急性发作组比较,急性发作组患者肺功能指标均明显偏低(P<0.05)。2非急性发作组及急性发作组患者CD4^+T细胞水平、CD4^+CD25^+Treg水平及CD4^+/CD8^+比值明显低于对照组,CD8^+T细胞水平、CD4^+CD25^+Treg/CD4^+比值显著高于对照组(P<0.05);与非急性发作组比较,急性发作组CD4^+T细胞水平、CD4^+CD25^+Treg水平及CD4^+/CD8^+比值显著下降,CD8^+T细胞水平、CD4^+CD25^+Treg/CD4^+比值显著上升(P<0.05)。3急性发作期患者外周血CD4^+T细胞水平与肺功能指标均呈正相关(r=0.47、0.50、0.45,P<0.05);CD8^+T细胞水平与肺功能指标均呈负相关(r=-0.54、-0.49、-0.56,P<0.05);CD4^+CD25^+Treg水平与肺功能指标均呈正相关(r=0.39、0.42、0.40,P<0.05)。结论 CD4^+、CD8^+T细胞及CD4^+CD25^+调节性T细胞在支气管哮喘疾病的发生、发展过程中发挥着重要作用,其水平可用来评估支气管哮喘病情的严重程度。 展开更多

目的探讨标本放置时间及年龄对外周血CD4+CD25+调节性T细胞的影响,明确标本可放置的时间界限,建立不同年龄段的正常参考值范围。方法以171例不同年龄健康人的外周血为标本,采用双色直接荧光素标记法和多参数流式细胞术检测和分析。结果1... 目的探讨标本放置时间及年龄对外周血CD4+CD25+调节性T细胞的影响,明确标本可放置的时间界限,建立不同年龄段的正常参考值范围。方法以171例不同年龄健康人的外周血为标本,采用双色直接荧光素标记法和多参数流式细胞术检测和分析。结果10份标本均分别在室温下放置0、1、2、3、4、5和6h检测时,CD4+CD25+Treg及CD4+CD25highT细胞分别为(12.00±1.12)%、(11.56±1.19)%、(10.72±1.29)%、(10.29±1.57)%、(8.11±1.28)%、(7.97±1.07)%、(7.70±0.93)%及(1.41±0.26)%、(1.45±0.26)%、(1.32±0.26)%、(1.27±0.28)%、(1.09±0.24)%、(0.95±0.18)%、(0.91±0.21)%。4~6岁组(n=34)、7~14岁组(n=24)、20~59岁组(n=95)、60岁以上组(n=18)健康人群CD4+CD25+Treg参考值分别是(6.55±1.28)%、(9.41±2.08)%、(11.67±2.14)%和(9.73±2.79)%,CD4+CD25high分别是(0.92±0.30)%、(1.21±0.45)%、(1.44±0.52)%及(1.51±0.55)%。结论外周血CD4+CD25+Treg和CD4+CD25highT细胞的检测结果随标本放置时间延长呈逐渐降低趋势,因此为真实反映体内CD4+CD25+Treg水平,标本放置时间应限制在3h以内。不同年龄段健康人群外周血中的CD4+CD25+Treg和CD4+CD25highT数值有明显差异,在探讨各种免疫相关性疾病与CD4+CD25+Treg之间的关系时,应该以与其年龄段相匹配的正常参考值为依据。 展开更多

The changes of CD4^+CD25^+ regulatory T cells (CD4^+CD25^+ Treg) and Foxp3 mRNA in peripheral blood mononuclear cells (PBMCs) from patients with asthma were investigated in order to elucidate the possible roles of CD4^+CD25^+ Treg in the development of asthma. The peripheral blood samples were collected from 29 healthy controls (normal control group) and 78 patients with asthma which included 30 patients in exacerbation group, 25 patients in persistent group, and 23 patients in remission group. By using flow cytometry and RT-PCR, the CD4^+CD25^+ Treg ratio and Foxp3 mRNA in PBMCs were detected. The CD4^+CD25^+ Treg ratio and Foxp3 mRNA in PBMCs of exacerbation and persistent groups were lower than that of remission and normal control groups (P〈0.05). Although the CD4^+CD25^+ Treg ratio and Foxp3 mRNA of remission group were also lower than that of normal control group, there was no significant difference between them (P〉0.05). As compared with persistent group, exacerbation group had lower CD4^+CD25^+ Treg ratio and Foxp3 mRNA (P〈0.05). It was indicated that the decrease of CD4^+CD25^+ Treg ratio and its function in PBMCs may be responsible for pathogenesis of asthma. 展开更多

目的:观察艾灸对胃荷瘤大鼠外周血中CD4^+、CD4^+CD25^+T淋巴细胞亚群的影响。方法:将50只健康SPF级SD大鼠,雌雄各半,随机分为空白组、假手术组、模型组、艾灸组和红外组5组,每组10只。利用Walker-256细胞来源的实体瘤组织移植胃部制备... 目的:观察艾灸对胃荷瘤大鼠外周血中CD4^+、CD4^+CD25^+T淋巴细胞亚群的影响。方法:将50只健康SPF级SD大鼠,雌雄各半,随机分为空白组、假手术组、模型组、艾灸组和红外组5组,每组10只。利用Walker-256细胞来源的实体瘤组织移植胃部制备模型。模型成功后,艾灸组予以悬灸中脘、胃俞等穴位;红外组予以短波红外线照射腹部胃脘及背部胃脘投影区;其余组捆绑束缚。干预20 min/次,1次/d,持续21 d。干预结束后,眼眶采血,流式细胞技术测定外周血中CD4^+、CD4^+CD25^+T淋巴比例。处死动物,测算胸腺指数及肿瘤体积生长抑制率。结果:与空白组比较,模型组动物生存状态变差,胸腺指数增加,外周血中CD4^+CD25^+T淋巴细胞比例显著增加(P <0. 05);与模型组比较,艾灸组动物艾灸组动物生存状态改善,胸腺指数进一步增加,瘤体内坏死灶较多,血管较少,外周血中CD4^+T淋巴细胞比例显著增加(P <0. 01),CD4^+CD25^+T淋巴细胞比例减少(P <0. 01);与红外组比较,艾灸组瘤体内坏死灶更多,血管较少,外周血中CD4^+T淋巴细胞比例增加(P <0. 01)。结论:艾灸可一定程度改善荷瘤动物生存状态,减少肿瘤细胞转移、促进肿瘤细胞坏死,减少血管增生。这种影响可能与增加CD4^+T淋巴细胞,减少CD4^+CD25^+T淋巴细胞有关。 展开更多

The function of CD4+CD25+ regulatory T lymphocytes (Treg) in patients with acute coronary syndrome (ACS) and the effects of atorvastatin were investigated. Forty-eight patients with ACS were randomly divided into two groups: group C receiving conventional therapy (n=24), and group C+A receiving conventional therapy+atorvastatin (10 mg/day, n=24). T lymphocytes from ACS patients (before and 2 weeks after the treatment) or 18 healthy subjects were separated and the flow cytometry was used to measure the percentage of Treg. The inhibitory ability of Treg on effector T cells was determined by mixed lymphocyte reaction (MLR). ELISA was used to measure the serum levels of cytokines (IL-10, TGF-β1 and IFN-γ) before and after treatment. The results showed that as compared with normal control group, Treg percentage was decreased significantly (P〈0.01), the inhibitory ability of Treg on the T lymphocytes proliferation was reduced (P〈0.01), IFN-γ levels were increased and IL-10 and TGF-β1 levels were lowered in ACS patients. After treatment with atorvastatin, Treg percentage and the inhibitory ability of Treg on T lymphocytes proliferation were significantly increased in ACS patients. Serum IFN-γ was decreased significantly, while IL-10 and TGF-β1 were elevated significantly as compared with the non-atorvastatin group. The number of Treg was positively correlated with serum TGF-β1, but negatively with serum IFN-γ and CRP. It was concluded that ACS was associated with decreased number and defected function of Treg, which may play an important role in initiating immune-inflammatory response in ACS. The inhibitory effects of atorvastatin on inflammation in ACS may be due to its beneficial effects on Treg and restoration of immune homeostasis. 展开更多

目的通过检测鼻咽癌患者肿瘤组织及外周血中CD4+T、CD8+T、CD4+CD25-T、CD4+CD25+T细胞的频数,寻找客观、全面评价鼻咽癌患者免疫状态的临床指标。方法采用流式细胞术检测40例初诊鼻咽癌患者及10例正常对照鼻咽部组织和外周血CD4+T、CD... 目的通过检测鼻咽癌患者肿瘤组织及外周血中CD4+T、CD8+T、CD4+CD25-T、CD4+CD25+T细胞的频数,寻找客观、全面评价鼻咽癌患者免疫状态的临床指标。方法采用流式细胞术检测40例初诊鼻咽癌患者及10例正常对照鼻咽部组织和外周血CD4+T、CD8+T、CD4+CD25-T、CD4+CD25+T细胞比例。结果鼻咽癌患者CD4+T细胞比例及CD4+/CD8+T比值均低于对照组(P<0.05),而CD8+T细胞两组间差异无统计学意义(P>0.05),但是CD4+/CD8+T比值在鼻咽癌组织与外周血间差异无统计学意义(P>0.05)。鼻咽癌组织及外周血中CD4+CD25+T细胞比例都高于对照组(P<0.05),同时癌组织中该细胞比例远远高于外周血(P<0.05)。在鼻咽癌组织中CD4+CD25+T细胞与CD8+T细胞、CD4+CD25-T细胞呈负相关(r分别为-0.70、-0.675,P<0.05),而在外周血中没有相关关系(P>0.05)。在不同T(原发肿瘤大小)组间,T4组的鼻咽癌组织中CD4+CD25+T细胞分别高于T1、T2、T3各组(P<0.05),而在T1、T2、T3各组间差异无统计学意义(P>0.05);鼻咽癌中CD4+CD25+T细胞比例与患者有无淋巴结转移并无关系(P>0.05);鼻咽癌组织中Ⅲ+Ⅳ期组CD4+CD25+T细胞比例高于Ⅰ+Ⅱ期组(P<0.05),而在外周血中两组间差异无统计学意义(P>0.05)。结论CD4+CD25+T细胞与鼻咽癌病程进展无相关性,但是联合检测患者肿瘤组织及外周血中CD4+CD25+T细胞的频数并结合既往CD4+/CD8+T比值会全面反应患者免疫状态,为临床治疗提供依据。 展开更多

Objective Age-related increment of the prevalence of CD4^+CD25^+ regulatory T (Treg) cells were described controversially, and whether such changes explain immune dysfunction in the elderly is still unclear. The aim of this systematic review is to evaluate the role of the Tregs in immunosenescence. Methods Medline and manual searches were performed to identify all published epidemiological and animal studies investigating the efficacy of the association between immunosenescence and Treg cells. Results It was founded that the frequency, phenotypic characteristics, and number/function of Tregs were altered significantly with aging. Medical conditions in individuals with advanced ageas well as apoptosis intensity of Treg cells had an impact on the accumulation of Tregs which in turn could deteriorate cytotoxic activity of CD8+ T and NK cells and production of IL-2. The range of immune cells that could be suppressed by Treg cells was quite wide and covered CD4^+CD25^+ T cells, NK cells, dendritic cells and even monocytes. These changes were observed both in humans and experimental animals. Besides, it was believed that frequency of Tregs increased with age and was accompanied by intensified suppressive activity for Tregs in patients, for example, with Alzheimer disease (AD) and Parkinson disease (PD). The impaired condition of CD4+ T cells, so-called immunosenescence, rendered transplant recipients less responsive to an allogeneic kidney graft, an effect that was limited to transplant recipients who were aged over 60 years. Conclusions Treg cells are associated with immunosenescence. All these changes contribute to the aging-related decline of immune responses and lead to the higher risk of immune-mediated diseases, cancer or infections in aged individuals. 展开更多

CD4+CD25+FoxP3 + regulatory T ceils (Tregs) are increased in patients with chronic hepatitis C, which may contribute to the sustained suppression of hepatitis C virus (HCV)-specific T-cell responses and viral persistence in HCV-infected individuals. We postulated that HCV core protein (HCVc) directly contributes to the expansion of Tregs in HCV-infected patients, and we provide evidence to support this hypothesis in the report. Peripheral blood mononuclear cells (PBMCs) and sera were collected from 87 treatment-naive chronic HCV-infected patients, CD4+CD25+ Tregs were measured by flow cytometry, and HCV RNA and HCVc levels were detected using qPCR and enzyme-linked immunosorbent assay (ELISA), respectively. CD4+, CD8+, CD4+CD25+ and CD4+CD25- T cells were purified from healthy donors and cultured with recombinant HCVc and Toll-like receptor (TLR) ligands. Flow cytometry was used to analyze cell proliferation, and ELISA was performed to measure cytokine production. In the 87 chronic HCV-infected patients, HCVc showed a significant correlation with HCV RNA and CD4+CD25+Tregs. Mechanistic studies showed that HCVc, together with anti-CD3 antibody, augmented CD4+CD25+ Treg proliferation, but inhibited CD4+CD25- T-cell proliferation and IFN-γ production, in a dose-dependent and Treg-dependent manner. Moreover, unlike the TLR3 ligand (poly hC) and the TLR4 ligand (lipopolysaccharide, LPS), the TLR2 ligand (lipoteichoic acid, LTA) and HCVc both inhibited TCR-induced CD4+ T-cell proliferation and IFN-γ secretion in a Treg-dependent manner. These data indicate that HCVc, like other TLR2 ligands, triggers CD4+CD25+ Treg activation and expansion to inhibit host immune responses, which may play a critical role in viral persistence in HCV-infected patients. 展开更多

Background Regulatory T cell populations, particularly CD4+CD25+ T regulatory cells, have been implicated in the persistence of hepatitis B virus (HBV) infection. However, no clear relationship has been established between the frequency of CD4~CD25~ T regulatory cells in the peripheral blood and either the disease phases in the natural history of chronic HBV infection or in the response to interferon-a therapy. Methods In the present study, three different common markers of CD4+CD25+ T regulatory cells were used to determine the numbers of T regulatory cells in healthy controls and in patients with chronic HBV infection. Results No significant difference was found when samples were gated for CD25hi and CD25+FoxP3+ T cells. A significant correlation was found between the number of CD4+ Treg cells that gated with CD25+FoxP3+ and CD25+CD127low/- in healthy controls and in patients with chronic hepatitis B (CHB) (r=0.67, 0.59; P 〈0.01). The percentages of Treg cells were (8.56±2.01)% in asymptomatic carriers (Asc), (8.74±3.04)% in inactive HBsAg carriers, (10.7±2.93)% in CHB and (7.42±1.28)% in healthy controls (F=-11.1, P 〈0.001). The percentage of Treg cells in patients with CHB was higher than in asymptomatic HBV patients, inactive HBsAg carriers, or healthy controls (P 〈0.01). The proportion of CD4+CD25+CD127 low/T cells in patients who responded to interferon-(] was (11.9±3.3)%, (9.1±2.4)% and (9.0±2.9)% at baseline, week 12 and week 24 after treatment, respectively (Z=2.42, P〈0.05; Z=2.67, P〈0.01). Conclusions These results suggest that the proportion of the CD4+CD25+ regulatory T cells might be affected by the application of different markers in process to detect T regulatory cells. The frequency of Treg cells was increased in patients with CHB, which might be associated with the disease activity of these patients and contribute to prevention of extensive liver damage. This study was conducted to analyze whether the CD4^+CD25^+ regulatory T cells exist and function normally in tuberculous pleural effusion. Methods The percentages of CD4^+CD25^+ T cells in pleural effusion and peripheral blood from patients with tuberculous pleurisy and peripheral blood from healthy control subjects were determined by flow cytometry. The expression of forkhead transcription factor Foxp3 was also examined. CD4^+CD25^+ and CD4^+CD25^-T cells from pleural effusion and blood were isolated, and were cultured to observe the effects of CD4^+CD25^+ T cells on proliferation response of CD4^+CD25^- T cells in vitro. Results There were increased numbers of CD4^+CD25^+ T cells in tuberculous pleural effusion compared with peripheral blood from both patients with tuberculous pleurisy and normal subjects, and these cells demonstrated a constitutive high-level expression of Foxp3. Moreover, CD4^+CD25^+ T cells mediated potent inhibition of proliferation response of CD4^+CD25^- T cells. Conclusion The increased CD4^+CD25^+ T cells in tuberculous pleural effusion express a high level of Foxp3 transcription factor, while potently suppressing the proliferation of CD4^+CD25^- T cells. 展开更多

AIM: To elucidate the distribution of CD4^+CD25^+ regulatory T cells (Tregs) in different lymphoid tissues and its local enhancement on tumor growth before and after depletion of CD4^+CD25^+ Tregs. METHODS: Female ICR mice were garaged with benzo[a]pyrene (BaP) to induce forestomach carcinoma. CD4^+CD25^+ Tregs were intraperitoneally depleted with monoclonal antibody PC61. These mice were divided into BaP-only, BaP + IgG, BaP + PC61, and control groups. The forestomach of mice was dissected for histological analysis, and tunnel test was performed for apoptosis of tumor cells. CD4^+CD25^+ Tregs were sorted from different lymphoid tissues and expression of Foxp3, IL-10, and chemokine receptors was analyzed by flow cytometry, semi-quantitative and veal-time polymerase chain reaction. RESULTS: The mice gavaged with only BaP showed increased forestomach papilloma and carcinoma at wk 16 and 32. The proportion of CD4^+CD25^+ Tregs was significantly higher in peri-stomach regional lymph nodes than in other lymphoid tissues. These CD4^+CD25^+ Tregs in regional lymph nodes expressed higher levels of Foxp3 and IL-10, enriched in the CD62L-subset, and CCR1 and CCR5 chemokine receptors. In mice gavaged with BaP + PC61, the number of tumor nodules and tumor volume decreased significantly with massive infiltrating cells and apoptosis of tumor cells. In the draining regional lymph nodes, the number of CD4^+CD25^+ Tregs also decreased significantly. CONCLUSION: Inducible and activated CD4^+CD25^+ Tregs in the draining regional lymph nodes suppress host local immunity during tumor growth. Depletion of CD4^+CD25^+ Tregs can promote host local immunity to suppress tumor growth. 展开更多
