目的:通过响应面法优化复合酶提取半夏多糖的工艺,并评价其抗氧化活性。方法:以半夏多糖得率为响应值,以液料比、酶解温度、酶解时间、复合酶(木瓜蛋白酶、纤维素酶、果胶酶的质量比为2∶2∶1)添加量为试验因素,采用响应面法建立数学模... 目的:通过响应面法优化复合酶提取半夏多糖的工艺,并评价其抗氧化活性。方法:以半夏多糖得率为响应值,以液料比、酶解温度、酶解时间、复合酶(木瓜蛋白酶、纤维素酶、果胶酶的质量比为2∶2∶1)添加量为试验因素,采用响应面法建立数学模型,优化提取条件;体外抗氧化活性考察半夏多糖对DPPH和O_2^-·自由基的清除能力。结果:通过二次回归模型响应面分析,酶解温度、时间、复合酶添加量、液料比4因素对半夏多糖得率的影响依次减弱;酶解温度与时间优化为54℃和57分钟,复合酶添加量为7.2 mg/mL,液料比例为27∶1 mL/g,在此最佳工艺条件下半夏多糖得率为27.98%,模型方程理论预测值为28.32%,两者相对误差小于5%。半夏多糖具有较强的抗氧化活性,对DPPH和O_2^-·自由基的半数抑制浓度分别为0.987 mg/m L、1.309mg/m L,但与维生素C比较,抗氧化活性较弱。结论:采用响应面法优化得到了半夏多糖的最佳复合酶提取条件,且半夏多糖有一定的体外抗氧化作用。 展开更多

目的探讨附子、半夏反药同方配伍的应用情况和配伍特点。方法收集中国知识资源总库(CNKI)、万方医学期刊全文库、维普中文科技期刊全文数据库中附子、半夏同方配伍应用的文献,建立数据库,提取纳入文献中病例数、治疗疾病、药物等信息,... 目的探讨附子、半夏反药同方配伍的应用情况和配伍特点。方法收集中国知识资源总库(CNKI)、万方医学期刊全文库、维普中文科技期刊全文数据库中附子、半夏同方配伍应用的文献,建立数据库,提取纳入文献中病例数、治疗疾病、药物等信息,采用描述性分析对附子、半夏反药同方应用的疾病系统、主要病症进行统计,并对其同方配伍药对及核心药组进行关联规则分析。结果纳入文献共1295篇,涉及患者30042例。纳入文献中附子、半夏同方配伍临床主要应用于咳喘(176篇,占13.59%)、胸痹(86篇,占6.64%)、胃炎胃痛(77篇,占5.95%),所治疗病症主要归属于消化系统(290篇,占22.39%)、心血管疾病(260篇,占20.07%)、呼吸系统(208篇,占16.06%);附子、半夏同方应用治疗疾病的以咳喘、慢性胃炎、胸痹为多见。关联分析显示,附子、半夏同方配伍的核心药物为甘草、茯苓、干姜、白术、桂枝、陈皮、党参、细辛等,核心药对为附子-半夏、附子-甘草、半夏-甘草、附子-干姜、半夏-干姜、半夏-白术等。3篇文献报道附子、半夏同方配伍有不良反应。结论附子、半夏反药同方配伍多治疗咳喘、慢性胃炎、胸痹等呼吸、消化系统及心血管疾病,其同方应用时多与补气、健脾、温中等药物配伍;二者配伍应谨慎使用。 展开更多

目的:运用网络药理学方法和技术研究"半夏-苍术-香附"组药治疗多囊卵巢综合征(PCOS)的主要活性成分、靶点和通路。方法:通过数据挖掘获得PCOS痰湿型核心组药半夏-苍术-香附;检索中药系统药理学分析平台(TCMSP),筛选出半夏、... 目的:运用网络药理学方法和技术研究"半夏-苍术-香附"组药治疗多囊卵巢综合征(PCOS)的主要活性成分、靶点和通路。方法:通过数据挖掘获得PCOS痰湿型核心组药半夏-苍术-香附;检索中药系统药理学分析平台(TCMSP),筛选出半夏、苍术和香附的主要活性成分和潜在靶点;查询NCBI数据库获得PCOS疾病靶点,找到药物和疾病的共靶点;通过David数据库和Omicshare数据库对共靶点进行基因功能GO富集分析和KEGG富集分析;采用Cytoscape软件构建中药-成分-靶标网络。结果:从"半夏-苍术-香附"组药中筛选得到40个候选活性分子,作用于681个靶点,其中与PCOS重合的23个疾病靶点主要涉及癌症通路、流体剪切应力与动脉粥样硬化通路、HIF-1信号通路、糖尿病并发症中的AGE-RAGE信号通路等生物学过程中的多条信号通路。结论:"半夏-苍术-香附"组药通过槲皮素、山奈酚、β-谷甾醇、木犀草素、汉黄芩素等活性物质形成多靶点、多通路调控网络,增强免疫机制,改善胰岛素抵抗,降低炎症细胞因子水平,达到系统治疗PCOS的目的。 展开更多

OBJECTIVE:To investigate the effectiveness of Banxia Xiexin decoction(BXD)in a rat model of chronic atrophic gastritis(CAG).METHODS:Sixty 6-week-old healthy Wistar rats(30 males,30 females)were used in the present study.A rat model of CAG was successfully established using the combined active immunization/ethanol/sodium deoxycholate method.BXD was prepared from a mixture of seven Chinese herbs,and was intragastrically administered to CAG rats at three different doses(6,12,and 24 g·kg^-1·d^-1).Af-ter 24 weeks,the rats were euthanized,and gastric tissue specimens were collected.Gastric mucosal specimens were stained with hematoxylin and eosin for histological examination to evaluate the degree of inflammation and morphological changes.Immunohistochemical staining was performed to examine the mucosal expression of proliferating cell nuclear antigen.Serum gastrin levels were measured using radioimmunoassay.The expression of Notch signaling-associated genes was examined by quantitative polymerase chain reaction and Western blot assay.RESULTS:BXD at all three doses significantly reversed the adverse effects generated by CAG in rats.Compared with control rats,the CAG rats who were administered BXD had an accelerated growth rate,reduced inflammatory cell infiltration,improved gastric mucosal morphology,augmented thickness of the gastric mucosa,increased number of gastric glands,enhanced mucosal expression of proliferating cell nuclear antigen,and elevated serum gastrin levels. 展开更多

Objective: To evaluate the myocardial protective effect of Gualou Xiebai Banxia decoction (瓜蒌薤白半夏汤GXBD) and explore the mechanisms of inhibition of NF-kappa B activation and blockade of inflammatory responses induced by ischemia-reperfusion in rats. Methods: Twenty-four Sprague Dawley (SD) rats were randomly divided into three groups. Rats in the treatment group received GXBD (13 g crude drug/kg) for three weeks, while rats in the model control and normal control groups received equal volumes of distilled water. On the 22nd day, rats in the ischemia-reperfusion (I/R) control and GXBD-treated groups underwent 30 min occlusion of the left anterior descending (LAD) coronary artery, followed by 120 min reperfusion. Electrocardiogram was recorded, and the activities of cardiac enzymes, cytokines, and NF-кB were assessed after I/R. Results: Compared with the I/R control group, GXBD treatment restored the activity of the specific myocardial-injury marker creatine kinase (CK) and lactate dehydrogenase (LDH), and inhibited the inflammatory response involving the nuclear factor-кB (NF-кB) pathway, including down-regulation of interleukin (IL)-1β and IL-6, and up-regulation of IL-10 gene expression. Conclusion: GXBD strongly reduced myocardial impairment in our I/R model, including inhibition of NF-кB activation and inflammatory cytokine responses. 展开更多

Objective: To examinie the synergistic effects of Banxia Xiexin Decoction (半夏泻心汤, Known as Banhasasim-tang in Korean) extract (BXDE) on cisplatin-induced cytotoxicity in the A549 human lung cancer ceil lines. Methods: A549 cells were treated with varying concentrations (50-200μg/mL) of cisplatin and BXDE alone or in combination for 96 h. We used 1-(4,5-dimethylthiazol-2-yl)-3,5-diphenylformazan assay and flow cytometry to analyze cell viability and apoptosis, respectively. Results: The exposure of cells to cispiatin and BXDE alone or in combination decreased ceil viability dose- and time-dependently (P〈0.05), which was found to be mediated by the apoptotic pathway as confirmed by the increase in the annexin V^+/propidium iodide stained cell population and a ladder pattern of discontinuous DNA fragments. Furthermore, the apoptosis was inhibited by the pan-caspase inhibitor, benzyloxycarbonyI-Val-Ala-Asp (OMe) fluoromethylketone (z-VAD-FMK). Conclusions: BXDE significantly potentiated apoptotic effects of cisplatin in A549 cells. Moreover, apoptosis induced by BXDE might be the pivotal mechanism mediating its chemopreventative action against cancer. 展开更多

目的:统计分析现代临床中药汤剂中半夏的用量,为半夏的临床安全使用剂量提供依据。方法:以“半夏”+“中医”、“半夏”+“临床”、“半夏”+“中毒”等为关建词,从中国知网检索现代临床应用半夏的文献共5380篇,经筛选排除,纳入病案信... 目的:统计分析现代临床中药汤剂中半夏的用量,为半夏的临床安全使用剂量提供依据。方法:以“半夏”+“中医”、“半夏”+“临床”、“半夏”+“中毒”等为关建词,从中国知网检索现代临床应用半夏的文献共5380篇,经筛选排除,纳入病案信息完整的合格文献832篇,建立半夏的用量数据库、不同炮制品频次数据库、半夏中毒情况数据库,用SPSS软件分析其临床运用规律。结果:半夏临床用量范围较广,从1.5~90.0 g不等,10.0~12.0 g使用频率最高;在临床处方中半夏名称繁杂多样,大多使用制半夏,其中又以法半夏使用频率较高,占44.4%;有明确半夏中毒剂量的报告中,使用生半夏中毒者较多,剂量以9.0~12.0 g中毒者最多。结论:半夏作为有毒中药,临床用量不可过大过猛,不同的炮制方法均可减轻其毒性,临床以应用制半夏为主,但是“有故无殒,亦无殒也”,临床运用时又不能拘泥,应根据病情需要确定应用剂量,并合理选用生半夏或不同的炮制品。 展开更多

目的:研究酸枣仁、延胡索、百合、地黄、半夏、夏枯草6味安神中药对戊巴比妥钠阈下剂量小鼠的睡眠只数,阈上剂量小鼠的睡眠潜伏期和睡眠持续时间以及对对氯苯丙氨酸(PCPA)失眠小鼠脑神经递质含量的影响,探讨其镇静催眠的作用及机制。方... 目的:研究酸枣仁、延胡索、百合、地黄、半夏、夏枯草6味安神中药对戊巴比妥钠阈下剂量小鼠的睡眠只数,阈上剂量小鼠的睡眠潜伏期和睡眠持续时间以及对对氯苯丙氨酸(PCPA)失眠小鼠脑神经递质含量的影响,探讨其镇静催眠的作用及机制。方法:168只昆明小鼠,随机分为7组:正常组、酸枣仁组(2.25 g/kg)、延胡索组(1.50 g/kg)、百合组(2.25 g/kg)、地黄组(2.25 g/kg)、半夏组(1.35 g/kg)、夏枯草组(2.25 g/kg)。连续灌胃给药10 d,通过阈下剂量戊巴比妥钠协同睡眠实验,观察各组小鼠睡眠只数;通过阈上剂量戊巴比妥钠协同睡眠实验,观察各组小鼠睡眠潜伏期及睡眠持续时间。96只昆明小鼠,随机分为8组:正常组、模型组、酸枣仁组、延胡索组、百合组、地黄组、半夏组、夏枯草组。采用腹腔注射PCPA法复制失眠小鼠模型,药物治疗14 d。运用酶联免疫吸附(ELISA)法检测血清去甲肾上腺素(NE)、多巴胺(DA)、五-羟色胺(5-HT)的含量,同法检测垂体促肾上腺皮质激素(ACTH)和下丘脑促肾上腺皮质激素释放素(CRH)的含量。结果:与正常组比较,各给药组小鼠睡眠只数增加,睡眠潜伏期缩短,睡眠持续时间延长。与正常组比较,模型组小鼠血清NE、DA、ACTH和CRH含量升高,5-HT含量降低。与模型组比较,半夏组小鼠血清NE含量升高,延胡索组5-HT含量升高,其余各给药组小鼠血清NE、DA、ACTH和CRH含量降低,5-HT含量降低。结论:酸枣仁、延胡索、百合、地黄、半夏、夏枯草6味安神中药具有改善小鼠睡眠的作用,酸枣仁、延胡索、百合、地黄适用于睡后易醒的患者,半夏、夏枯草可以广泛用于失眠患者。其作用机制可能与降低脑内DA、NE、ACTH、CRH水平有关。 展开更多

Objective:To compare the effectiveness and safety of Banxia Xiexin Decoction (BXD) as alternative therapy versus standard triple therapy or quadruple therapy for patients with peptic ulcer or chronic gastritis infected with Helicobacter pylori (H.pylori).Methods:Databases including China National Knowledge Infrastructure,Chongqing VIP,Wanfang Database,PubMed,the Cochrane Library and clinicaltrials.gov were searched in December 2018 for relevant randomized controlled trials (RCTs).Two authors independently screened and selected studies,extracted data and checked data extraction.Methodological quality was evaluated using the Cochrane Risk of Bias tool.Meta-analysis was performed by using RevMan 5.3.5 software.Results:Fourteen RCTs were included in our analysis involving 1300 patients.Thirteen RCTs compared the effects of BXD alone versus standard therapy,11 involving triple therapy and 2 in quadruple therapy.The cure rate for both diseases were higher in the BXD alone group than in the standard therapy group (RR and 95% Cl:1.85 [1.07,3.17] and 1.48 [1.24,1.75],respectively).And also,the same result with effectiveness rate (RR and 95% CI:1.18 [1.08,1.29] and 1.14 [1.08,1.20],respectively).However,there was no significant difference in the clearance of H.pylori one month after treatment,neither compared with quadruple therapy nor triple therapy (RR and 95% CI:1.10 [1.00,1.22] and 1.04 [0.97,1.12],respectively).Adverse events appeared in 3 participants in the BXD group and 26 participants in the conventional therapy group.The quality of the trials included in this review was not very good.Conclusion:BXD has a superior effect to standard conventional therapy in improving clinical symptoms and repair of the mucosal lesion,and a similar effect to standard conventional therapy in clearing H.pylori.We still need good quality trials,especially placebo-controlled trials,in the future to confirm this result. 展开更多

Objective To test the effect of Banxia Xiexin Decoction(半夏泻心汤,BXD)on the contraction and relaxation of gastric smooth muscle(SM)in diabetic gastroparesis(DGP)model rats,and to explore the mechanism of BXD in the prevention and treatment of DGP through experiments of signal pathway both in vivo and in vitro.Methods Sixty Sprague-Dawley rats were divided into 6 groups according to a random number table:control group,model group,high-,medium-and low-dose BXD groups(9.2,4.6 and 1.8 g/(kg·d),respectively),and domperidone group(10 mg/(kg·d)),10 rats per group.DGP model was established initially by a single intraperitoneal injection of streptozotocin(STZ),and was confirmed by recording gastric emptying,intestinal transport velocity and gastric myoelectric activity of rats after 2 months.Each group was treated with a corresponding drug for 4 weeks.The mRNA and protein expressions of phospholipase C(PLC),inositol triphosphate(IP3),neuronal nitric oxide synthase(nNOS),and cyclic guanosine monophosphate(cGMP)dependent protein kinase G(PKG)were detected by reverse transcription-polymerase chain reaction and Western blot,respectively,while nitric oxide(NO)and cGMP expressions were detected by enzyme-linked immunosorbent assay.Gastric tissues were obtained from rats for primary cell culture preparation.Gastric SM cells were treated with 0.8µmol/L of STZ or STZ plus 1,000,500 and 200µg/mL of BXD or STZ plus 2.5µmol/mL of domperidone for 24,48,72 or 96 h,respectively.The length of gastric SM cells and intracellular Ca^2+concentration([Ca^2+]i)before and after BXD treatment was measured.Results Compared with the model group,high-and medium-dose BXD and domperidone significantly increased the expressions of PLC,IP3,NO,nNOS,cGMP and PKG in rat’s gastric tissue(P<0.01).Gastric SM cells treated with BXD showed a time-and dose-dependent increase in cell viability(P<0.01).The treatment with high-and medium-dose BXD and domperidone inhibited the increase in gastric SM cells length and increased[Ca^2+]i compared with the 展开更多

OBJECTIVE: To investigate the effect of a modified Banxia Xiexin decoction(MBXD) plus chemotherapy on postoperative adverse reaction in patients with stage Ⅲ colon cancer in patients whose symptoms were identified as cold-heat complicated pattern in terms of the theory of Traditional Chinese Medicine(TCM).METHODS: A prospective non-randomized control study of patients with stage Ⅲ colon cancer in Beijing Shijitan Hospital and Guang'anmen Hospital between January 2012 and December 2013. A total of 80 patients were divided into experimental group(MBXD + chemotherapy) and control group(chemotherapy). The adverse reactions, life quality and disease-free survival(DFS) were compared between the two groups.RESULTS: The cold-heat complex pattern was released in 94.3% of patients in experimental group whereas 62.2% in control group(P < 0.05). Life quality was improved in 24 vs 14 cases, stabilized in 9 vs14 cases whereas decreased in 2 vs 15 cases in experimental group and control group, respectively(P < 0.05). The average DFS was 22.371 months in experimental group, while 13.932 months in control group(P < 0.05).CONCLUSION: MBXD combined with chemotherapy may significantly relieve clinical symptoms, reduce chemotherapy associated adverse effects, improve life quality, and prolong DFS of patients with stage Ⅲ colon cancer(cold-heat complicated pattern). 展开更多

OBJECTIVE: To investigate the effect of Banxia Xiexin decoction(BXD) on Helicobacter pylori(Hp)-related peptic ulcers(PUs) and the possible mecha-nism underlying BXD actions via the transforming growth factor-β/small mothers against decapentaplegic(TGF-β/Smad) signaling pathway.METHODS: PU patients with cold-heat complex syndrome were randomly assigned to groups that received Chinese or Western medicines with 20 patients in each group. Serum was collected after 7 d of treatment. The healthy group included 20 individuals. Gastric mucosal epithelial cell line GES-1 was cultured in vitro and randomly divided into the following seven groups: control, model, healthy,Western Medicine, prior treatment, low dosage,and high dosage. After 72 h of treatment with the corresponding serum, the m RNA and protein expression levels of TGF-β1, Smad3, and Smad7 were measured by reverse transcription quantitative polymerase chain reaction and western blotting, respectively.RESULTS: The m RNA expression levels of TGF-β1 and Smad3 in GES-1 cells were increased after Hp introduction, and these increased levels were reduced by the BXD-containing serum. The protein levels of p-Smad3, but not TGF-β1 or Smad3, were significantly increased in Hp-treated GES-1 cells,and treatment with the BXD-containing serum markedly decreased the protein levels. Smad7 expression was significantly enhanced following treatment with the BXD-containing serum at transcriptional and protein levels in a dose-dependent manner.CONCLUSION: BXD regulates the TGF-β/Smad signaling pathway by inhibiting the expression of TGF-β1 and Smad3, and increasing the expression of Smad7. 展开更多

目的:对现代半夏治疗失眠的处方用药规律进行统计分析,为临床使用半夏治疗失眠提供较为客观的参考。方法:通过数据库查阅近30年有关半夏治疗失眠的中医文献资料,遵循循证医学的原则,把符合选择标准的文献102篇录入到Excel,使用SPSS 12.... 目的:对现代半夏治疗失眠的处方用药规律进行统计分析,为临床使用半夏治疗失眠提供较为客观的参考。方法:通过数据库查阅近30年有关半夏治疗失眠的中医文献资料,遵循循证医学的原则,把符合选择标准的文献102篇录入到Excel,使用SPSS 12.0进行数据分析,统计半夏不同炮制品出现的频次、频率,剂量大小以及适用证型。结果:临床治疗失眠经常用到6种半夏炮制品,分别是生半夏、清半夏、姜半夏、法半夏、竹沥半夏和半夏曲,分别占了1.94%、21.36%、16.50%、57.28%、0.97%、1.94%。临床治疗失眠生半夏多用1~19 g;清半夏多用1~19 g,最大可用到60 g;姜半夏多用1~19 g;法半夏多用10~19 g,最大可用到60 g;竹沥半夏多用10~19 g;半夏曲多用10~19 g。半夏治疗失眠主要集中在肝火扰心(33次)、痰热扰心(62次)、心脾两虚(27次)、心肾不交(27次)、心胆气虚(26次)、痰湿内阻(8次)和其它(9次)这七大证型上,治疗多用法半夏。结论:辨证后选用安全有效剂量的不同半夏炮制品治疗失眠。 展开更多

目的:基于网络药理学方法探析经典药对“陈皮-半夏”治疗冠脉微循环障碍(coronary microvascular dysfunction,CMD)的主要活性成分、主要靶点基因、关键通路及其作用机制。方法:本研究通过TCMSP数据库获取“陈皮-半夏”药对的主要活性... 目的:基于网络药理学方法探析经典药对“陈皮-半夏”治疗冠脉微循环障碍(coronary microvascular dysfunction,CMD)的主要活性成分、主要靶点基因、关键通路及其作用机制。方法:本研究通过TCMSP数据库获取“陈皮-半夏”药对的主要活性成分及其靶点,筛选其活性成分及潜在靶点。通过Gencards和OMIM数据库获取CMD的主要靶点。运用uniprot数据库查询相对应的基因名称。利用韦恩图取交集可获得“陈皮-半夏”药对治疗冠脉微循环障碍的关键交集靶点。利用String平台进行蛋白质相互作用分析,构建PPI网络并通过Cytoscape3.7.1软件进一步挖掘网络中潜在的蛋白质功能模块。在DAVID数据库进行靶点基因(GO)功能富集分析和(KEGG)通路富集分析,使用在线绘图工具Bioinformatics网站分别绘制高级气泡图。最后采用Cytoscape3.7.1软件构建“陈皮、半夏成分-冠脉微循环障碍靶点-通路”网络。结果:构建的“陈皮、半夏成分-冠脉微循环障碍靶点-通路”网络中包括柚皮素、诺比林、黄芩素、β谷甾醇等16个药对活性成分,AKT1、VEGFA、BCL2、BAX、JUN等56个预测靶点基因,心血管系统相关通路(PI3K-AKT signaling pathway)、炎症相关通路(TNF signaling pathway)、血管内皮生长因子信号相关通路(VEGF signaling pathway)、细胞凋亡相关通路(Apoptosis)和低氧细胞应激信号通路(HIF-1 signaling pathway)等20条关键通路。结论:应用网络药理学验证了“陈皮-半夏”药对多成分、多靶点、整体调节的作用特点,推测其主要通过保护冠脉微循环内皮细胞功能、抑制细胞凋亡、影响炎性反应等途径发挥治疗冠脉微循环障碍的作用。 展开更多

BACKGROUND Helicobacter pylori(H.pylori)is the main pathogen that causes a variety of upper digestive diseases.The drug resistance rate of H.pylori is increasingly higher,and the eradication rate is increasingly lower.The antimicrobial resistance of H.pylori is an urgent global problem.It has been confirmed that Banxia Xiexin decoction(BXXXT)demonstrates the effects of treating gastrointestinal diseases,inhibiting H.pylori and protecting gastric mucosa.The purpose of the present study is to further explore the therapeutic effects of BXXXT on drug-resistant H.pylori.AIM To confirm that BXXXT demonstrates therapeutical effects in vivo and in vitro on gastritis mice with drug-resistant H.pylori and explain its mechanism to provide an experimental basis for promoting the application of BXXXT.METHODS The aqueous extract of BXXXT was gained by water decocting method.The inhibitory effect of the aqueous extract on H.pylori was detected by dilution in vitro;drug-resistant H.pylori cells were used to build an acute gastritis model in vivo.Thereafter,the model mice were treated with the aqueous extract of BXXXT.The amount of H.pylori colonization,the repair of gastric mucosal damage,changes of inflammatory factors,apoptosis,etc.,were assessed.In terms of mechanism exploration,the main medicinal compositions of BXXXT aqueous extract and the synergistic bacteriostatic effects they had demonstrated were analyzed using mass spectrometry;the immune function of peripheral blood cells such as CD3+T and CD4+T of mice with gastritis before and after treatment with BXXXT aqueous extract was detected using a flow cytometry;the H.pylori transcriptome and proteome after treatment with BXXXT aqueous extract were detected.Differently expressed genes were screened and verification was performed thereon with knockout expression.RESULTS The minimum inhibitory concentration of BXXXT aqueous extract against H.pylori was 256-512μg/mL.A dose of 28 mg/kg BXXXT aqueous extract treatment produced better therapeutical effects than the standard tri 展开更多
