Chinese kale is an important vegetable crop in China, especially in South China where the majority of varieties are grown. It originated in South China and spread throughout Southeast Asia thereafter. Chinese kale can be classified into two types according to whether the petals are white or yellow. There are also three main cultivated types based on the edible organs, including the stalk and leaf type, the stalk type,and the leaf type. Two primary types have also been defined based on stalk color, including green stalks and red stalks. They are also cultivated based on the type of stalk, including main stalk and lateral stalk types. Significant differences have also been observed in glucosinolate content among the varieties, and a 40-fold difference in neoindle-3-methyl glucosinolate was detected across the cultivars. 展开更多

ζ-Carotene desaturase(ZDS)is an important enzyme in carotenoid biosynthesis.Here,the Brassica oleracea var.alboglabra ZDS(Boa ZDS)gene was cloned from Chinese kale via reverse transcription-polymerase chain reaction(... ζ-Carotene desaturase(ZDS)is an important enzyme in carotenoid biosynthesis.Here,the Brassica oleracea var.alboglabra ZDS(Boa ZDS)gene was cloned from Chinese kale via reverse transcription-polymerase chain reaction(RT-PCR)and deposited in Gen Bank(accession number KY662297).The Boa ZDS gene contains an open reading frame of 1 686 bp that encodes a 561-amino acid protein.Sequence analysis indicates that the ZDS protein is apparently conserved during plant evolution and is most closely related to B.oleracea var.capitata and B.rapa.The promoter sequence of the Boa ZDS gene was predicted to harbor several cis-acting elements that are related to light and phytohormone responses.Semiquantitative RT-PCR analysis showed that Boa ZDS expression varied among different developmental stages and organs.Relative ZDS expression remained stable during germination and seedling stages and rapidly increased at the mature leaf stage.The leaves showed the highest ZDS expression levels compared to the other organs.ZDS expression decreased in all flower tissues during blooming.The fused protein of Boa ZDS was obtained by prokaryotic expression.Heterologous expression of Boa ZDS in Escherichia coli confirmed that Boa ZDS encodes a functionalζ-carotene desaturase that increases β-carotene accumulation in E.coli cells harboring a β-carotene-producing plasmid.The findings of the present study provide a molecular basis for the elucidation of ZDS gene function in Chinese kale. 展开更多

The proteins with glycolate oxidase activity from B.parachinensis Bailey and rice( Oryza sativa )green leaves were prepared respectively.From the second protein peak on DEAE\|Cellulose column,two glycolate oxidases,expressed as B.parachinensis Bailey GO Ⅲ(specific activity natove 13 2 U·mg -1 ·min -1 )and rice GOⅢ(specific activity 8 8 U·mg -1 ·min -1 ),could not migrate anywhere in 4%~20% native\|PAGE under a pH8.3 buffer system.GOⅢ’s p I was about pH8.3. The protein containing B.parachinensis Bailey GOⅢ showed 67±2,43±2,and 38±2 kD in SDS PAGE,band 43±2 kD was the subunit of B.parachinensis Bailey GOⅢ.From the two proteins above,another group of glycolate oxidases,expressed as B.parachinensis Beiley GOⅠ(specific activity 5 U·mg -1 ·min -1 )and rice GOⅠ(specific activity 1 2 U·mg -1 ·min -1 ),showed only one 43±2 kD band in SDS\|PAGE,and was purified on the Sepharose\|6B column which migrated towards anode in the same native\|PAGE showing the M r about 420 kD,or 460 kD and 260 kD respectively.GOⅠ’s p I was smaller than pH8.3.Antibody against B.parachinensis Bailey GOⅠ was prepared and its efficacy was about 1/1600 in ELISA.By native\|PAGE,Western blot and rocket immunoelectrophoresis,the third group of glycolate oxidases,expressed as B.parachinensis Bailey GOⅡ and rice G0Ⅱ,were confirmed in crude protein of green leaves and migrated towards cathode under the same native\|PAGE,so GOⅡ’s p I was higher than pH8.3.The M r of B.parachinensis Bailey GOⅡ was about 669 kD determined by native\|PAGE Western blot.Rice GOⅠ,rice GOⅢ and rice GOⅡ showed different quantitation under different physiological conditions.Rice GOⅡ could be induced by glycolate. 展开更多

[目的]研究花叶玉簪的组培技术。[方法]以花叶玉簪为研究对象,采用浓度75%的酒精进行消毒,并以MS为基本培养基,添加不同浓度植物生长调节剂6-BA和NAA进行增殖培养和生根培养,观察生长状况。[结果]MS+4.0 mg/L 6-BA+0.1 mg/L NAA增殖效... [目的]研究花叶玉簪的组培技术。[方法]以花叶玉簪为研究对象,采用浓度75%的酒精进行消毒,并以MS为基本培养基,添加不同浓度植物生长调节剂6-BA和NAA进行增殖培养和生根培养,观察生长状况。[结果]MS+4.0 mg/L 6-BA+0.1 mg/L NAA增殖效果最好,苗生长健壮。最佳生根培养基为:1/2MS+0.1 mg/L NAA+0.3%活性炭;20 d后平均根长可达3 cm,且根系比较粗壮,移栽后易成活。幼苗移栽时以蛭石∶珍珠岩(5∶1,W/W)、泥炭土∶珍珠岩∶蛭石(3∶1∶1,W/W/W)或腐叶土∶蛭石(3∶1,W/W)的成活率较高,小苗生长健壮。[结论]采用组培方法繁殖花叶玉簪,虽然具有繁殖系数高,时间短等优点。 展开更多

A pot experiment was conducted to study the influences of foliar application of glycine,alanine,lysine,and glutamic acid in 200 mg/kg or 500 mg/kg upon the quality and enzyme activity of flowering Chinese cabbage(Brassica parachinensis Bailey).The results showed that all the application of these four amino acids could increase the yield of flowering Chinese cabbage,significantly raise the content of soluble sugar,and reduce the accumulation of nitrate.The applications of three other amino acids except alanine can increase the content of soluble proteins and decrease the accumulation of oxalic acid.However,the application of amino acid has insignificant influences on the SPAD number of chlorophyll,and causes the decrease of Vitamin C content.Meanwhile,the application of amino acid can improve the activity of nitrate reductase(NR) and glutamate dehydrogenase(GDH) as well.It shows that the application of amino acid is beneficial to improve ammonia metabolism,reduce the accumulation of nitrate and oxalic acid,increase the content of soluble sugar and soluble proteins,and improve the quality of flowering Chinese cabbage. 展开更多

Gravel hardness effect on compaction characteristics of gravelly soil

The compaction characteristics of gravelly soil are affected by gravel hardness.To investigate the evolution and influencing mechanism of different gravel hardness on the compaction characteristics of gravelly soil,heavy compaction tests and crushing tests were conducted on gravelly soils with gravels originated from hard,soft and extremely soft rocks.According to orthogonal experiments and variance analysis,it was found that hardness has a significant impact on the maximum dry density of gravelly soil,followed by gravel content,and lastly,moisture content.For gravel compositions with an average saturated uniaxial compressive strength less than 60 MPa,the order of compacted maximum dry density is soft gravels>hard gravels>extremely soft gravels.Each type of gravelly soil has a threshold for gravel content,with 60%for hard and soft gravels and 50%for extremely soft gravels.Beyond these thresholds,the compacted dry density decreases significantly.There is a certain interaction between hardness,gravel content,and moisture content.Higher hardness increases the influence of gravel content,whereas lower hardness increases the influence of moisture content.Gravelly soils with the coarse aggregate(CA)between 0.7 and 0.8 typically achieve higher dry densities after compaction.In addition,the prediction equations for the particle breakage rate and CA ratio in the Bailey method were proposed to estimate the compaction performance of gravelly soil preliminarily.The results further revealed the compaction mechanism of different gravelly soils and can provide reference for subgrade filling construction. 展开更多

“321”贝雷装配成小跨径的连续梁结构承受荷载时,支点位置的竖腹杆是最薄弱杆件。针对施工大型临时承重设施常用的6m、9m、12m及15m四种跨度的贝雷连续梁进行分析,首先采用MIDAS Civil有限元软件进行杆单元建模计算组合应力,对比“321... “321”贝雷装配成小跨径的连续梁结构承受荷载时,支点位置的竖腹杆是最薄弱杆件。针对施工大型临时承重设施常用的6m、9m、12m及15m四种跨度的贝雷连续梁进行分析,首先采用MIDAS Civil有限元软件进行杆单元建模计算组合应力,对比“321”贝雷及三种备选优化方案的承载能力,选出最优方案。随后采用板单元建模进行局部细节分析,对比各计算对象的Mises等效应力最大值,进一步验证了优化方案的有效性。结果表明:将I8工字钢竖腹杆替换为双拼C10槽钢,使贝雷的重量增加19%,但承载力可提升50%左右。 展开更多
