Aim: To find an in vitro system for the measurement of the androgenic effects of different extracts of Hibiscus macranthus (Malvaceae) and Basella alba (Basellaceae). Methods: The production of testosterone from testes slices incubated in two media, either Krebs-Henseleit buffer containing 0.5% Bovine serum albumin (BSA) or Dubecco's Modified Eagle's medium-F12 Ham nutrient mixture (DME/Ham F12), under a mixture of 5% CO2 in 95% air was determined either in the presence or absence of cofactors and Hibiscus macranthus plus Basella alba (HMBA) extracts. Results: The testosterone production was increased in testes slices incubated in DME/Ham F12 medium in response to the cofactors (49%) and aqueous extracts (34%-60% according to dilutions). Under the same atmospheric conditions, there was no positive response of the testes slices to either cofactor or HMBA extract stimulation in Krebs-Henseleit buffer containing 0.5% BSA, In further investigations related to the effect of HMBA, the DME/ Ham F12 medium was used. The results obtained from the in vitro test showed that the activity was present mainly in methylene chloride and methanol, since these extracts induced an increase in testosterone production by testes slices. Conclusion: The testes slice system is suitable to be used for further in vitro investigations of the isolation of androgenic bioactive components of plants. Antimicrobial resistance (AMR) is one of the major significant concerns in developing nations due to either over-use or inadequate use of antimicrobial agents. The purpose of this study was to emphasize the potential antimicrobial activity of Nepeta cataria and Basella alba against nosocomial diseases caused by Acinetobacter baumannii. Nepeta cataria and Basella alba leaves were collected from Oldonyo Sabuk National Park and Botanical garden JKUAT respectively. The leaves were dried under shade at room temperature for thirty days. After methanolic and aqueous plant extraction, the percentage yield was determined, and the extracts were subjected to toxicity testing using brine shrimp eggs and preliminary phytochemical screening. The results recorded from this study demonstrated that 70% methanolic extract of Nepeta cataria and Basella alba had greater antimicrobial effect against Acinetobacter baumannii than aqueous extracts. Nepeta cataria extracts showed greater antimicrobial effect than Basella alba extracts. The methanolic plant extracts had MIC values 60 mg/ml when tested against Acinetobacter baumannii. The phytochemical screening showed the presence of various phytochemical constituents from the methanolic and aqueous extraction of Nepeta cataria and Basella alba. The methanolic plant extracts revealed more phytochemicals as compared to aqueous extracts. More phytochemicals were detected from Nepeta cataria than Basella alba plant extracts. 展开更多

A pot experiment was carried out to determine the effect of lime and phosphorus on the growth and nutrient uptake by Indian spinach (Basella alba L.) on an acidic soil. The experimental soil was amended with four levels of lime (0, 500, 1000 and 2000 kg CaCO3 ha-1) and phosphorus (P) (0, 50, 100, and150 kgP ha-1) and their combinations. The results showed that lime and P applied separately or in combination had significant (P < 0.001) effects on growth parameters (height and number of leaves), fresh and dry weight of shoot and root and N, K and Ca uptake by Indian spinach. Combined application of lime and P gave a better result than the separate application of lime and P. Application of 2000 kg·ha-1 lime plus 150 kg P ha-1 had higher uptake of N, K and Ca and better morphological characters that eventually resulted in greater yield compared with other treatments. The results revealed that lime and phosphorus could be used in combination to improve growth performance and nutrient uptake when plants grown in an acidic soil. 展开更多

Aim: To determine the androgenic effects of Basella alba and Hibiscus macranthus extracts in the rat and the bull, and to develop a novel in vitro test system using Leydig cells from bull testes. Methods: The effect of methanol extracts from both plants on testosterone production in isolated Leydig cells from the rat and the bull was analyzed using ^125I-radioimmunoassay (^125I-RIA). Rat Leydig cells were obtained by common methods, whereas a novel technique was used to purify Leydig cells from bull testes. Results: Bull testes from the slaughter house were a cheap source of pure Leydig cells. In culture, these cells produced testosterone for 5-6 days, which can be stimulated by human chorionic gonadotrophin (hCG). Basella alba extracts significantly enhanced testosterone production in bull and rat Leydig cells in a concentration-dependent manner. Hibiscus macranthus showed no androgenic effect but was shown to inhibit testosterone production at higher concentrations. Conclusion: Leydig cells purified from bull testes can be used as an alternative tool in experimental animal research. Certain fractions of Basella alba extract demonstrated androgenic potential whereas Hibiscus macranthus extracts did not. 展开更多

To isolate and identify the cultivable leaf endophytic fungi of Basella alba L.and B.rubra L.,healthy and fresh leaves were chosen and collected from Sri Devaraj Urs Academy of Higher Education and Research college botanical garden.Authenticated materials were made into small bits,surface sterilized by applying the standard protocol and cultured on a potato dextrose agar plate supplemented with appropriate antibiotics,incubated for 7 to10 days at 28˚C.Seed borne fungal endophytes were also identified by keeping scarified seeds on sterilized agarose.A phylogenetic tree was constructed for the fungi of both the plants.A total of 26 and 19 endophytic cultivable fungi were isolated from B.alba and B.rubra respectively.Majority of the endophytes belongs to the genus Alternaria and several genera in class Dothideomycetes.Our preliminary observation indicated that the vertically transmitted endophytic fungi are the dominant endophytes of the selected plants.We conclude that Alternaria is predominant in both the plants.The mechanism of transition of the microbe from being non-pathogenic in its host to being pathogenic in other plants needs to be studied and elucidated. 展开更多

Objective:Stress is becoming an unavoidable threat in recent times,there has been increasing interest by researchers in the use of naturally occurring biologically active compounds with medicinal value to cure body ailments.The present work was carried out to investigate the effect of methanol extract of Basella alba leaves on stress in Wistar rats(Rattus norvegicus).Methods:A total of 35 male rats were used in this study.They were grouped into seven groups of five rats each.Group 1(normal control)was received 10 m L/kg normal saline.Group 2 contained restraint stress rats only.Group 3 contained forced swim stress rats only.Group 4 and 5 were treated with 60 mg/kg of B.alba extract(BAE)thereafter subjected to restraint and forced swim stresses respectively.Group 6 and 7 were treated with 120 mg/kg of BAE thereafter subjected to restraint and forced swim stresses respectively.Stress procedures were carried out at the end of first and third weeks.Results:In the stressed rats,there were significant increases(P<0.05)in fasting blood glucose and white blood cell count while there were significant decreases in superoxide dismutase activity and glutathione concentration when compared to group 1.There were significant decreases(P<0.05)in blood glucose and white blood cell count and significant increases in superoxide dismutase and glutathione concentrations in BAE treated rats when compared to group 2 and 3.Some of the significant differences were either dose or duration dependent.Conclusion:In conclusion,results from this research suggest that BAE alleviates hyperglycaemia,chronic activation of immune system and generation of free radicals due to stress in Wistar rats. 展开更多

落葵色素是由落葵果实提取的天然色素,色泽鲜艳,是很好的着色剂。通过试验探索,确定了最佳提取工艺:以落葵干果为原料,采用溶剂法进行提取,最佳溶剂为pH=4的弱酸性水溶液。再用大孔吸附树脂吸附,以无水乙醇为洗脱液洗脱,最后在低于50... 落葵色素是由落葵果实提取的天然色素,色泽鲜艳,是很好的着色剂。通过试验探索,确定了最佳提取工艺:以落葵干果为原料,采用溶剂法进行提取,最佳溶剂为pH=4的弱酸性水溶液。再用大孔吸附树脂吸附,以无水乙醇为洗脱液洗脱,最后在低于50℃的条件下真空浓缩得到色素胶质,进一步干燥即可获得粉末状产品。同时,对落葵色素的稳定性进行了研究:落葵色素在pH=3~7时比较稳定,适宜于弱酸性和中性条件下使用;落葵色素的耐光、耐热性较差,应低温下避光贮藏,生产过程中的浓缩温度应控制在50℃以下;氧化剂、还原剂、柠檬酸对该色素的稳定性有一定的影响;Fe^3+、Cu^2+对该色素的影响较大,其它金属离子基本上对其稳定性没有影响。落葵来源广泛,色素含量高,提取工艺简单,且安全无毒,可广泛应用于食品、化妆品的着色中,因此落葵色素是一种值得开发和利用的天然色素资源。 展开更多

通过正交试验确定提取落葵色素的最佳工艺条件,研究色素的溶解性、光谱特性、热稳定性、光稳定性、复原性以及色价等理化性质,并考查pH值、金属离子和常用食品添加剂对色素的影响。结果表明,0.01mol/L盐酸溶液为提取剂、提取时间40min... 通过正交试验确定提取落葵色素的最佳工艺条件,研究色素的溶解性、光谱特性、热稳定性、光稳定性、复原性以及色价等理化性质,并考查pH值、金属离子和常用食品添加剂对色素的影响。结果表明,0.01mol/L盐酸溶液为提取剂、提取时间40min、提取温度45℃是落葵色素的最佳提取工艺;落葵色素的水溶性强,复原性好,平均色价为11.39,在60℃以下,pH2~10稳定性较好,但光照会加速其分解;K+、Na+、Ca2+、Mg2+、Zn2+等不影响色素的稳定性,Fe2+、Fe3+和Cu2+则有影响;落葵色素对NaCl、VC、葡萄糖、苯甲酸钠、柠檬酸和Na2SO3稳定,对H2O2不稳定。落葵色素在一定的条件下具有良好的稳定性。 展开更多

用FTIR对500 mg U·kg-1土壤胁迫下空心莲子草、落葵和菊苣茎叶和根系的物质成分进行测定,用半定量法分析3种植物茎叶和根系红外光谱的差异,探讨高浓度U胁迫对植物物质成分的影响.结果表明:(1)3种植物茎叶和根系吸收峰峰形未发生较... 用FTIR对500 mg U·kg-1土壤胁迫下空心莲子草、落葵和菊苣茎叶和根系的物质成分进行测定,用半定量法分析3种植物茎叶和根系红外光谱的差异,探讨高浓度U胁迫对植物物质成分的影响.结果表明:(1)3种植物茎叶和根系吸收峰峰形未发生较大改变,但吸光度变化明显,说明该浓度U对这3种植物的物质成分改变较小,含量改变较大.(2)羟基、羧基和酰胺基吸收峰发生明显位移,说明U胁迫对这些基团有明显影响,它们可能与U的吸收、络合、运输密切相关.(3)半定量分析表明,该浓度U使3种植物羟基、羧基等基团及蛋白质和糖类物质有明显的数量变化,不同植物变化不同,这可能由植物耐受U能力不同所致.因此,FTIR能快速、灵敏地检测植物在高浓度U胁迫下物质成分的变化,能解释植物吸收积累U的影响机理,可作为一种有效手段应用于U等核素对植物效应的研究. 展开更多

落葵叶绿体基因组特征分析

【目的】落葵(Basella alba)为落葵科落葵属缠绕茎生草本植物,原产于中国和印度。植物叶绿体基因组遗传稳定,常被用于植物的鉴定及系统发育研究,旨在对落葵的叶绿体基因组进行特征分析。【方法】采用15对叶绿体基因组通用引物对落葵新... 【目的】落葵(Basella alba)为落葵科落葵属缠绕茎生草本植物,原产于中国和印度。植物叶绿体基因组遗传稳定,常被用于植物的鉴定及系统发育研究,旨在对落葵的叶绿体基因组进行特征分析。【方法】采用15对叶绿体基因组通用引物对落葵新鲜叶片提取的DNA进行扩增,并利用Illumina HiSeq X Ten平台进行测序,获得其叶绿体基因组序列。【结果】落葵叶绿体基因组全长156 533 bp,具有典型的四分体结构,包含83个编码蛋白基因、28个tRNA基因以及4个rRNA基因。检测到基因组中45个重复序列和263个简单重复序列,二核苷酸序列均为碱基AT/AT重复。密码子分析表明落葵叶绿体基因组密码子第三位碱基偏好使用A或U,综合中性绘图分析、ENC标准曲线分析、PR2-plot分析发现落葵叶绿体基因组密码子偏好性受自然选择因素为主的多种因素共同影响,符合落葵为双子叶植物的形态学分类结果。【结论】落葵与落葵薯叶绿体基因组高度相似,比较基因组学分析显示落葵与其他石竹目物种在反向重复区的收缩与扩张上存在差异。基于落葵在内的20种不同植物叶绿体基因组matK基因序列进行系统发育分析,结果表明落葵与其他落葵科植物处于同一进化分支,但其属间进化关系与形态学分类存在一定差异。研究结果为落葵科的资源分析及利用研究提供了理论基础。 展开更多
