Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),has spread worldwide.Effective treatments against COVID-19 remain urgently in need although vaccination significantly reduces the incidence,hospitalization,and mortality.At present,antiviral drugs including Nirmatrelvir/Ritonavir(Paxlovid^(TM)),Remdesivir,and Molnupiravir have been authorized to treat COVID-19 and become more globally available.On the other hand,traditional Chinese medicine(TCM)has been used for the treatment of epidemic diseases for a long history.Currently,various TCM formulae against COVID-19 such as Qingfei Paidu decoction,Xuanfei Baidu granule,Huashi Baidu granule,Jinhua Qinggan granule,Lianhua Qingwen capsule,and Xuebijing injection have been widely used in clinical practice in China,which may cause potential herb-drug interactions(HDIs)in patients under treatment with antiviral drugs and affect the efficacy and safety of medicines.However,information on potential HDIs between the above anti-COVID-19 drugs and TCM formulae is lacking,and thus this work seeks to summarize and highlight potential HDIs between antiviral drugs and TCM formulae against COVID-19,and especially pharmacokinetic HDIs mediated by metabolizing enzymes and/or transporters.These well-characterized HDIs could provide useful information on clinical concomitant medicine use to maximize clinical outcomes and minimize adverse and toxic effects. 展开更多

The outbreak and spread of coronavirus disease 2019(COVID-19)highlighted the importance and urgency of the research and development of therapeutic drugs.Very early into the COVID-19 pandemic,China has begun developing drugs,with some notable progress.Herein,we summarizes the anti-COVID-19 drugs and promising drug candidates originally developed and researched in China.Furthermore,we discussed the developmental prospects,mechanisms of action,and advantages and disadvantages of the anti-COVID-19 drugs in development,with the aim to contribute to the rational use of drugs in COVID-19 treatment and more effective development of new drugs against severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and the variants.Neutralizing antibody is an effective approach to overcome COVID-19.However,drug resistance induced by rapid virus mutation will likely to challenge neutralizing antibodies.Taking into account current epidemic trends,small molecule drugs have a crucial role in fighting COVID-19 due to their significant advantage of convenient administration and affordable and broad-spectrum.Traditional Chinese medicines,including natural products and traditional Chinese medicine prescriptions,contribute to the treatment of COVID-19 due to their unique mechanism of action.Currently,the research and development of Chinese anti-COVID-19 drugs have led to some promising achievements,thus prompting us to expect even more rapidly available solutions. 展开更多

As the novel coronavirus disease 2019(COVID-19)and its variants continue to rage into the second year of a global pandemic,many success stories of applying Chinese herbal medicine(CHM)to treat COVID-19 patients continue to emerge from China and other part of the world.Herewith,from a systems medicine perspective,the authors analyze those experiences and categorize them into four major treatment principles:(1)focusing on eliminating toxins in the early stage of the disease,(2)tonifying deficiency of the body throughout the entire disease course,(3)treating the affected lung and intestine simultaneously based on visceral interactions,(4)cooling blood and removing blood stasis at the later stage,as well as interpret the rationale of these principles.This is helpful not only in reducing the complexity of promoting the CHM applications to enhance anti-COVID-19 efficacy,but also in ramping out the process of integrating traditional Chinese medicine with modern medical practices. 展开更多

The primary concern of drug scientists,virologists, and other concerned specialists in this respect is to find ready-to-use suitable and potent anticoronaviraltherapies that are broadly effective against the different species/strains of the coronaviruses in general, not only againstthe current and previous coronaviruses (e.g., the recently-appeared severe acute respiratory syndrome coronavirus 2“SARS-CoV-2”), i.e., effective antiviral agents for treatment and/or prophylaxis of any coronaviral infections, includingthose of the coming ones from the next species and strains (if any). As an expert in this field, I tried, in this up-to-dateperspective “viewpoint” article, to evaluate the suitability and applicability of using the currently-availableanticoronaviral agents for the next coronavirus diseases (COVIDs) and coronaviral pandemics, highlighting the mostimportant general guidelines that should be considered in the next pandemics from the therapeutic points of view. 展开更多

As a multifunctional fluorescent nanomaterial, carbon dots (CDs) not only have small size, stable chemical properties, excellent photoluminescence characteristics, but also exhibit good biocompatibility and low toxicity. It has attracted considerable attention in the field of nanotechnology and biological science. CDs contain abundant functional groups on the surface, which not only retain part of the properties of raw materials, but also may have new photoelectric, catalytic, biomedical, and other functions. In this review, we systematically summarize the synthesis methods, modifications, optical properties, and main biological functions of CDs in recent years. The application of functionalized modified CDs in biological detection, biological imaging, photodynamic therapy, photothermal therapy, targeted therapy, drug delivery, gene delivery, protein delivery, and other biomedical fields is introduced. The latest progress of CDs with its own biomedical function in antioxidant, anti-pathogen, and disease treatment is summarized. Finally, we discuss some problems in the practical application of CDs and look forward to the future development trend of self-functional CDs combined with surface modification to achieve multimodal treatment of diseases. 展开更多

Via an insufficient coat protein complex I(COPI)retrieval signal,the majority of SARSCo V-2 spike(S)is resident in host early secretory organelles and a tiny amount is leaked out in cell surface.Only surface-exposed S can be recognized by B cell receptor(BCR)or anti-S therapeutic monoclonal antibodies(m Abs)that is the trigger step for B cell activation after S m RNA vaccination or infected cell clearance by S m Abs.Now,a drug strategy to promote S host surface exposure is absent.Here,we first combined structural and biochemical analysis to characterize S COPI sorting signals.A potent S COPI sorting inhibitor was then invented,evidently capable of promoting S surface exposure and facilitating infected cell clearance by S antibody-dependent cellular cytotoxicity(ADCC).Importantly,with the inhibitor as a probe,we revealed Omicron BA.1 S is less cell surface exposed than prototypes because of a constellation of S folding mutations,possibly corresponding to its ER chaperone association.Our findings not only suggest COPI is a druggable target against COVID-19,but also highlight SARS-Co V-2 evolution mechanism driven by S folding and trafficking mutations. 展开更多

近两年,我国主流媒体推出多部“抗疫”纪录片,为传播中国抗疫“好故事”、构筑抗疫精神、构建国家话语体系、塑造国家形象发挥了重要作用。抗疫纪录片的相关研究备受学界重视,然而,现有的研究仍然集中在传播学领域,对抗疫精神的构建和... 近两年,我国主流媒体推出多部“抗疫”纪录片,为传播中国抗疫“好故事”、构筑抗疫精神、构建国家话语体系、塑造国家形象发挥了重要作用。抗疫纪录片的相关研究备受学界重视,然而,现有的研究仍然集中在传播学领域,对抗疫精神的构建和传播还缺乏内部话语机制的跨学科实证研究。本文以中国环球电视网推出的英文纪录片《中国战疫纪》为研究语料,依托评价理论的态度子系统(情感、判断、鉴赏)和态度意义的多模态实现框架,对片中的文字和多模态符号资源进行了系统分析。研究发现,该纪录片再现了英雄主义、乐观主义、集体主义和理想主义的美学抗疫精神,文字模态主要建构了“判断”态度意义,多模态资源主要表征了“情感”态度意义,体现了抗疫纪录片实现价值认同和情感共鸣的双重功能。在融媒体时代,其传播策略表现为“情感性”共情传播和“全媒体”精准传播两大突出特点,是我国对外话语体系构建和国家形象塑造的有益实践。 展开更多

自2020年初新冠肺炎暴发以来,科学家一直在积极寻找口服有效的防治药物。2021年11月初,默沙东的莫努匹韦在英国获批上市,并于11月30日获美国FDA紧急使用授权,这是全球首款抗新冠口服药。随后辉瑞宣布其新型口服抗新冠药帕罗韦德可将住... 自2020年初新冠肺炎暴发以来,科学家一直在积极寻找口服有效的防治药物。2021年11月初,默沙东的莫努匹韦在英国获批上市,并于11月30日获美国FDA紧急使用授权,这是全球首款抗新冠口服药。随后辉瑞宣布其新型口服抗新冠药帕罗韦德可将住院或死亡风险降低89%。本研究总结了这2个药物的合成方法:莫努匹韦的合成主要以尿苷或胞苷为起始原料,胞苷价格便宜而且路线短,操作简单高效;帕罗韦德的合成包含3个片段,其中最重要的是片段B(也是丙型肝炎药物波普瑞韦的关键中间体)的合成,主要有手性拆分以及酶催化2种方法,其中酶催化法收率高且立体选择性好。 展开更多

新冠肺炎疫情是百年来人类面临的最严重的公共卫生危机,对全球公共卫生安全构成严峻挑战。许多国家在疫情防控过程中暴露出的问题,凸显了加强全球公共卫生安全治理、加强抗疫合作的重要性和紧迫性。各国在加强国内疫情防控的同时,要重... 新冠肺炎疫情是百年来人类面临的最严重的公共卫生危机,对全球公共卫生安全构成严峻挑战。许多国家在疫情防控过程中暴露出的问题,凸显了加强全球公共卫生安全治理、加强抗疫合作的重要性和紧迫性。各国在加强国内疫情防控的同时,要重视加强双边合作在内的国际抗疫合作,特别要加强同邻国间的合作,以有效遏制疫情向周边地区扩散。疫情发生以来,中老两国始终坚持人类命运共同体理念,在抗疫过程中守望相助,密切沟通协作,积极开展公共卫生外交,全面加强抗疫双边合作,极大地丰富了全球公共卫生安全治理的内涵,为全球抗疫斗争作出了积极贡献。双方通过信心支持、技术交流、物资援助和边境管控等,不断深化双边抗疫合作,取得了良好的抗疫效果,充分展现了中老命运共同体和社会主义制度的显著优势,为流行病的全球治理提供了可供借鉴的宝贵经验。中老抗疫合作的实践表明,抗疫合作需要提升到政治外交的高度加以重视。以人类命运共同体为理论依据、植根于中老命运共同体建设的中老抗疫合作,在推进过程中受益于两国密切的双边关系并作用于双边关系的发展,特别是中老命运共同体建设。中老抗疫合作中展现出的人民至上、命运与共精神,对构建人类命运共同体具有一定的示范效应,在今后全球公共卫生安全治理体系重构中理应得到坚持和发扬。 展开更多

本文旨在运用互信理论阐释以色列和哈马斯在面对共同威胁时达成的有限合作,进而推导暂时性合作对塑造互信从而消除彼此间持续性冲突的可能性。2020年新型冠状病毒性肺炎疫情迫使各国政府在面临重大公共卫生危机时实施防疫隔离措施,同时... 本文旨在运用互信理论阐释以色列和哈马斯在面对共同威胁时达成的有限合作,进而推导暂时性合作对塑造互信从而消除彼此间持续性冲突的可能性。2020年新型冠状病毒性肺炎疫情迫使各国政府在面临重大公共卫生危机时实施防疫隔离措施,同时在疫情共同威胁下也寻求多形式的抗疫合作。以色列和巴勒斯坦在疫情暴发之初就宣布放弃冲突并寻求合作,然而,加沙地带的哈马斯却在合作态度上存在波动。疫情初期,哈马斯拒绝与以色列合作抗疫,双方间的军事冲突也没有停歇。随着疫情的加剧,哈马斯陆续释放与以色列开展抗疫合作的信号,以色列继而修改了合作条件,彼此间达成了合作的第一阶段计划。哈马斯与以色列之间的合作非常罕见,双方抗疫合作能否拓展到其他领域,进而达成促进区域和平之目的,仍有待观察。 展开更多
