This paper aims to investigate the effects of artesunate (ART) on growth and apoptosis in human osteosarcoma HOS cell line in vitro and in vivo and to explore the possible underlying mechanisms.Cell viability was meas...This paper aims to investigate the effects of artesunate (ART) on growth and apoptosis in human osteosarcoma HOS cell line in vitro and in vivo and to explore the possible underlying mechanisms.Cell viability was measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay.The induction of apoptosis was detected by light and transmission electron microscopy and flow cytometry.Western blot analysis was used to investigate the related mechanisms.Nude mice were further employed to investigate the antitumour activity of ART in vivo.MTT assay results demonstrated that ART selectively inhibits the growth of HOS cells in a dose-and time-dependent manner.Based on the findings of light and transmission electron microscopy,Hoechst 33258 staining,and fluorescein isothiocyanate (FITC)-annexin V staining,the cytotoxicity of ART in HOS cells occurs through apoptosis.With ART treatment,cytosolic cytochrome c was increased,Bax expression was gradually upregulated,Bcl-2 expression was downregulated,and caspase-9 and caspase-3 were activated.Thus,the intrinsic apoptotic pathway may be involved in ART-induced apoptosis.Cell cycle analysis by flow cytometry indicated that ART may induce cell cycle arrest at G2 /M phase.In nude mice bearing HOS xenograft tumours,ART inhibited tumour growth and regulated the expressions of cleaved caspase-3 and survivin,in agreement with in vitro observations.ART has a selective antitumour activity against human osteosarcoma HOS cells,which may be related to its effects on induction of apoptosis via the intrinsic pathway.The results suggest that ART is a promising candidate for the treatment of osteosarcoma.展开更多

Stiassnie and Shemer researched the unstable evolution process an...Owing to the Benjamin-Feir instability, the Stokes wave train experiences a modulation-demodulation process, and presents a recurrence characteristics. Stiassnie and Shemer researched the unstable evolution process and provided a theoretical formulation for the recurrence period in 1985 on the basis of the nonlinear cubic Schrodinger equation (NLS). However, NLS has limitations on the narrow band and the weak nonlinearity. The recurrence period is re-investigated in this paper by using a highly efficient High Order Spectral (HOS) method, which can be applied for the direct phase- resolved simulation of the nonlinear wave train evolution. It is found that the Stiassnie and Shemer's formula should be modified in the cases with most unstable initial conditions, which is important for such topics as the generation mechanisms of freak waves. A new recurrence period formula is presented and some new evolution characteristics of the Stokes wave train are also discussed in details.展开更多
Aim: To determine the predictive value of the hypo-osmotic swelling (HOS) test to identify viable, non-motile sperm. Methods: Semen samples from 20 men with severe asthenozoospermia underwent traditional seminal analy...Aim: To determine the predictive value of the hypo-osmotic swelling (HOS) test to identify viable, non-motile sperm. Methods: Semen samples from 20 men with severe asthenozoospermia underwent traditional seminal analysis, eosin-nigrosin (EN) staining and the HOS test. A further EN stain was then performed on a HOS pre-treated aliquot and a total of 2000 further sperm examined. Results: The median sperm density was 5.1 million/mL (IQR 4.3-13.1) and the median motility was 3.0 % (IQR 0-7). Seven samples showed complete asthenozoospermia. Initial EN staining showed 59 % viability (range 48-69) despite the poor standard parameters and 47 % (range 33-61) in the complete asthenozoospermia subgroup. The HOS test showed 49.9 % reacted overall (range 40-59) and 41.7 % (range 22-61) in the complete asthenozoospermia subgroup. The combined HOS/EN stain showed the positive predictive value of the HOS test to identify viable sperm was 84.2 % overall and 79.7 % in the complete asthenozoospermia subgroup. Conclusion: The HOS test can effectively predict sperm viability in patients with severe and complete asthenozoospermia.展开更多

This method is capable of recognizing the MIMO radar signal as well as discriminating it from single-carrier signal adopted by conventional radar. Meanwhile, the sub-carrier number of the none-coding MIMO radar signal is estimated. Extensive simulations are carried out in different operating conditions. Simulation results prove the feasibility and indicate that the recognition probability could reach over 90% when the value of SNR is above 0 dB.展开更多

To overcome the curse of dimensionality resulting from high dimensional input variables, in our developed model the features extracted from the available observations are regarded as the input variables by adopting the higher-order statistics(HOS) technique. Such a constructed model is also applied to a practical railway carriage system, simulation results indicate that the developed neurofuzzy model possesses strong identification and fault diagnosis ability.展开更多

Under the help of block-processing technology, background is reconstructed quickly. Finally, background difference is used to detect motion regions instead of adjacent frame difference. The DSP based platform tests indicate the background can be recovered losslessly in about one second, and moving regions are not influenced by moving target speeds. The Second-Order Statistics (SOS) and Higher-Order Statistics (HOS) of data are fused within a regularization framework. The steering vectors can be blindly identified by the regularized ESPRIT, from which the aim of DOA estimation can be achieved. Several variants of the regularized ESPRIT are discussed.
