METHODS: Transplantable murine hepatoma22 model was used to evaluate the anti-tumor activity of resveratrol (RES) alone or in combination with 5-FU in vivo. H22 cell cycles were analyzed with flow cytometry. RESULTS: Resveratrol could inhibit the growth of murine hepatoma22, after the mice bearing H22 tumor were treated with 10 mg/kg or 15 mg/kg resveratrol for ten days, and the inhibition rates were 36.3% (n = 10) and 49.3% (n = 9), respectively, which increased obviously compared with that in control group (85+22 vs 68+17, P<0.01). RES could induce the S phase arrest of H22 cells, and increase the persentage of cells in S phase from 59.1% (n = 9) to 73.5% (n = 9) in a dose-dependent manner (P<0.05). The enhanced inhibition of tumor growth by 5-FU was also observed in hepatoma22 bearing mice when 5-FU was administered in combination with 10 mg/kg resveratrol. The inhibition ratesfor 20 mg/kg or 10 mg/kg 5-FU in combination with 10 mg/kg resveratrol were 77.4% and 72.4%, respectively, compared with the group of 20 mg/kg or 10 mg/kg 5-FU alone, in which the inhibition rates were 53.4% and 43.8%, respectively (n = 8). There was a statistical significance between the combination group and 5-FU group. CONCLUSION: RES could induce the S phase arrest of H22 cells and enhance the anti-tumor effect of 5-FU on murine hepatoma22 and antagonize its toxicity markedly. These results suggest that resveratrol, as a biochemical modulator to enhance the therapeutic effects of 5-FU, may be potentially useful in cancer chemotherapy.展开更多

After treatment with various concentrations of mifepristone (2.5, 5, 10,20μmol/L) at various time intervals, the ultrastructural changes, cell proliferation, cell-cycle phase distribution, and the expression of caspase-3 and Bcl-XL were analyzed using transmission electron microscopy (TEM), tetrazolium blue (MTT) assay, ^3H-TdR incorporation, flow cytometry, and reverse transcription-polymerase chain reaction (RT-PCR).RESULTS: Mifepristone markedly induced apoptosis and inhibited cell proliferation of PR- positive SGC-7 901 cells revealed by TEM, MTT assay and ^3H-TdR incorporation, in a dose- and time-dependent manner. The inhibitory rate was increased from 8.98% to 51.29%. Flow cytometric analysis showed mifepristone dose-dependently decreased cells in S and G2/M phases, increased cells in G0/G1 phase,reduced the proliferative index from 57.75% to 22.83%.In addition, mifepristone up-regulated the expression of caspase-3, and down- regulated the Bcl-XL expression,dose-dependently.CONCLUSION: Mifepristone effectively inhibited the proliferation of PR-positive human gastric adenocarcinoma cell line SGC-7 901 in vilrothrough multiple mechanisms, and may be a beneficial agent against human adenocarcinoma.展开更多

Methods: To examine the expressed leve...Objective: To investigate the relationship between P-glycoprotein (P-gp) and adhesion molecule CD44 expression as well as their clinical significance in esophageal carcinoma. Methods: To examine the expressed level of P-gp and CD44 by flow cytometry (FCM) in the operated samples of 70 cases with esophageal carcinoma and their normal mucosa of esophageal incision, and to evaluate their relationship with clinicopathological factors. Results: Among the 70 cases with esophageal carcinoma, the expression of P-gp in the 27 cases (38.6%) was negative (positive cells 〈25%); 11 cases (15.7%) were 25%-40% expression of P-gp positive cells; 14 cases (20%) were 41%-60% expression of P-gp positive cells; 18 cases (25.7%) were the high expression (positive cells 〉60%) of P-gp. Of the cases with the tumor sizes being more than 4 cm, the expression of CD44 showed a significant difference (P〈0.05) in 25 cases with P-gp positive, compared with 19 cases with P-gp negative. Of the cases with high-mild differentiated esophageal carcinoma, the expression of CD44 showed a significant difference (P〈0.05) in 22 cases with P-gp positive, compared with 17 cases with P-gp negative. Of the cases with clinical Ⅲ-Ⅳ stage, the expression of CD44 showed a significant difference (P〈0.05) in 26 cases with P-gp positive, compared with 10 cases with P-gp negative. Of the cases with lymph node metastasis, the CD44 expression showed a significant difference (P=0.050) in 27 cases with P-gp positive, compared with 11 cases with P-gp negative. Of the cases of the patients' age being more than 56 years, the expression of CD44 showed a significant difference (P〈0.01) in 27 cases with P-gp positive, compared with 12 cases with P-gp negative. When the P-gp and CD44 expression were positive, the clinical Ⅱ stage and Ⅲ-Ⅳ stage in esophageal carcinoma was showed a significant difference (P〈0.05). The paper, using microscopical counting method, studies the relations...This paper aims at providing a scientific basis for unifying the normal reference value standards of red blood cell count of Chinese presenile men. The paper, using microscopical counting method, studies the relationship between the normal reference values of 38,061 samples of red blood cell count of presenile men and eight geographical factors in 297 units in China. It is found that the correlation of geographical factors and the normal reference value of red blood cell count of presenile men is quite significant (F=303.00, P=-0.000). By using the method of stepwise regression analysis, one regression equation is inferred. It is concluded that if geographical data are obtained in a certain area, the normal reference value of red blood cell count of presenile men in this area can be reckoned by using the regression analysis. Furthermore, according to the geographical factors, China can be divided into eight regions: Northeast China Region, North China Region, Shanxi-Shaanxi-Irmer Mongolia Region, Middle and Lower Reaches of the Changjiang River Region, Southeast China Region, Northwest China Region, Southwest China Region and Qinghai-Tibet Plateau Region.展开更多
AIM: To asses the expression of myeloid dendritic cells (CD11c+) subset during acute HCV hepatitis and its possible involvement in natural history of the infection.METHODS: We enrolled 11 patients with acute hepa...AIM: To asses the expression of myeloid dendritic cells (CD11c+) subset during acute HCV hepatitis and its possible involvement in natural history of the infection.METHODS: We enrolled 11 patients with acute hepatitis C (AHC) (Group A), 10 patients with acute hepatitis A (AHA) (as infective control-Group B) and 10 healthy donors (group C) in this study. All patients underwent selective flow cytometry gating strategies to assess the peripheral number of the myeloid dendritic cells (mDCs) to understand the possible role and differences during acute hepatitis.RESULTS: Eight of 11 patients with acute HCV hepatitis did not show any increase of mDCs compared to healthy individuals, while a significant decrease of mDCs was found in absolute cell count (z=-2.37, P〈0.05) and percentage (z=-2.30; P〈 0.05) as compared with AHA. On the contrary, The remaining three patients of the group A had a higher mDCs number and percentage as occur in group B. Interestingly, after six months, those patients did not show any increase of mDCs subset were chronically infected, while the three subjects with an increase of peripheral mDCs, as in HAV acute infection, resolved the illness.CONCLUSION: The lack of increase of mDCs during acute hepatitis C might be an important factor involved in chronicization of the infection.展开更多

RR was calculated in NOD/SCID...The aim of this study was to evaluate the relationship between the embryo resorption rate (RR) and the status of local immunity at the feto-maternal interface in pregnant NOD/SCID mice. RR was calculated in NOD/SCID x NOD/SCID mice and compared with non-immunodeficiency BALB/c × BALB/c mice on day 13.5 of gestation. Intraeellular detection of cytokine expression was performed with four-color flow cytometry to identify the functional subsets of lymphocytes in NOD/SCID mice before being bred or during pregnancy. No statistically supported difference in RRs was observed between NOD/SCID × NOD/SCID and control BALB/c × BALB/c mice. Accordingly, although multiple immunodeficits were confirmed in NOD/SCID mice, the percentages of several functional cell subsets were spontaneously altered during pregnancy, and this may be correlated with the roughly normal pregnancy outcomes observed in these mice. The spontaneous alteration of cell percentages at the feto-maternal interfaee in NOD/SCID mice may be of benefit to the pregnancy outcomes.展开更多
A simple in vivo bioassay suitable for the routine quality control testing of a new erythropoiesis stimulating protein was developed.Subcutaneous administration of the new erythropoiesis stimulating protein to Balb/c ...A simple in vivo bioassay suitable for the routine quality control testing of a new erythropoiesis stimulating protein was developed.Subcutaneous administration of the new erythropoiesis stimulating protein to Balb/c mice in a single dose resulted in a dose-dependent increase in the number of circulating reticulocytes.Within the erythropoiesis stimulating protein dose range of 3.125 to 200 ng per mouse,there is a strong linear relationship between the dose and reticulocyte counts in the treated mice.This linear relationship allows us to determine the biological potency of the testing erythropoiesis stimulating protein preparation relative to a reference standard using parallel line assay.Accuracy,precision,dose variation and blood collection time of this method were analyzed in order to choose doses in the linear range that are suitable for setting up a useful,precise,and economical bioassay.展开更多
