However, few studies have examined the relationship between time of acupuncture and curative effect on cerebral hemorrhage. By observing the effect of acupuncture on changes in histopathology, ultrastructure, and neuroethology in a cerebral hemorrhage model of rats, we have directly examined the time-effect relationship of acupuncture. The rat model of cerebral hemorrhage was produced by slowly injecting autologous blood to the right caudate nucleus. The experimental groups were: 3-, 9-, 24-, and 48-hour model groups; and 3-, 9-, 24-, and 48-hour acupuncture groups. The sham-operation group was used for comparison. Acupuncture was performed at the Neiguan(PC6) and Renzhong(DU26) acupoints, twice a day, 6 hours apart, for 5 consecutive days. Brain tissue changes were observed by light microscopy and transmission electron microscopy. Neuroethology was assessed using Bederson and Longa scores. Our results show that compared with the sham-operation and model groups, Bederson and Longa scores were lower in each acupuncture group, with visibly improved histopathology and brain tissue ultrastructure. Further, the results were better in the 3-and 9-hour acupuncture groups than the 24-and 48-hour acupuncture groups. Our findings show that acupuncture treatment can relieve pathological and ultrastructural deterioration and neurological impairment caused by the acute phase of cerebral hemorrhage, and may protect brain tissue during this period. In addition, earlier acupuncture intervention following cerebral hemorrhage(by 3 or 9 hours) is associated with a better treatment outcome.展开更多

METHODS Human fresh surgical colonic mucosa was homogenized by adding appropriate amount of no...To observe the effect of acupuncture and moxibustion on the expression of IL 1 β and iNOSmRNA in ulcerative colitis rats. METHODS Human fresh surgical colonic mucosa was homogenized by adding appropriate amount of normal saline and centrifuged at 3000 r/min. Protein content of the supematant was measured and then mixed with Freund adguvant, and injected into the plantae of the model group rats, then into the plantea, dorsa, inguen and abdominal cavities (no Freund adjuvant for the last injection) on the 10 th, 17 th, 24 th and 31 th day respectively. When a certain titer of seurm anti colonic antibody was reached, 20 ml/L formalin and antigen fluid (no Freund adjuvant) were administered by enema to set up ulcerativecolitis rat model. The arimals were randomly divided into four groups: model control group (MC=8), electropuncture group (EP=8), herb separation moxibustion group (HPM=8) and normal control group (NC=8) HPM: Mosa cones made of refined mugwort floss were placed on the medicinal pads (medicinal pad dispensing: Radix Aconiti Praeparata, contex Cinnamomi, ectc) for Qihai (RN6) and Tianshu (ST 25, bilateral) and ignited. Two moxa cones were used for each acupuncture once a day and 14 times in all. EP: Tianshu (bilateral) and Qihai were stimulated by the intermittent pulse with 2 Hz frequency, 4 mA intensity for 20 minutes once a day and 14 times in all. After treatment, all rats were killed simultaneously. The spleen was separated and distal colon was dissected. Totai tissue RNA was isolated by the guanidinium thiocyanate phenol chloroform extraction method. RT PCR technique was used to observe the expression of IL 1 β and iNOSmRNA. no Freund adjuvant. RESULTS IL 1 β and iNOSmRNA was not all detected in the spleen and colonic mucosa of NC rats, while it was significantly expressed in that of MC rats. IL 1 β and iNOSmRNA was markedly lower in EF and HPM rats than that in MC rats. There was no significant difference in the levels of IL 1 β and iNOSmRNA between EP and HPM rats. OBJECTIVE:To investigate the effect of a decoction made from the Traditional Chinese Medicine Wumei pill,on regulatory T cells and interleukin-10(IL-10) in a rat model of ulcerative colitis induced by2,4,6-trinitroben...OBJECTIVE:To investigate the effect of a decoction made from the Traditional Chinese Medicine Wumei pill,on regulatory T cells and interleukin-10(IL-10) in a rat model of ulcerative colitis induced by2,4,6-trinitrobenzene sulfonic acid(TNBS).METHODS:Rat ulcerative colitis was induced with TNBS.All modeled rats were randomly divided into six groups:normal control group;model group;sulfasalazine suppositories treatment group;and high,moderate,and low dosage of Jiaweiwumei decoction groups(12 rats each).Colon injury index was evaluated after 14 days.After peripheral blood lymphocyte separation,CD4 + T cells and CD4+/CD25+ T cell percentage was detected by flow cytometry.The content of IL-10 in serum and intestinal mucosa tissue was detected by sandwich enzyme-linked immunosorbent assay.RESULTS:Colon injury indices in the decoction groups were effectively reduced,compared with the model group(P < 0.05).Compared with that of the control group,the CD4+/CD25+ to CD4+ T lymphocyte ratio of the model group was significantly lower,while the decoction treatment improved the CD4 +/CD25 + to CD4 + T lymphocyte ratio(P <0.05).The serum and mucosal IL-10 content of the model group was significantly lower(P < 0.05) than that in the control group,while the decoction group had significantly higher serum and intestinal mucosal IL-10 content than that in the model group(P < 0.05).The regulatory T cell content was negatively correlated with the colonic injury index(r = 0.68,P < 0.05),and positively correlated with the content of serum IL-10(r = 0.87,P < 0.05) and intestinal mucosal IL-10(r= 0.79,P < 0.05).CONCLUSION:Jiaweiwumei decoction had significant effects on regulatory T cells and IL-10 in rats with TNBS-induced ulcerative colitis.展开更多

METHODS: Twenty-eight SD rats were randomly as- signed to normal, model, electroacupuncture (EA), and acupotomy group. The last three groups were put through an operation to emulate L3 transverse process syndrome. Fourteen days after the simulation operation, EA and acupotomy treatments were applied to the respective groups. Fifty-six days afterthe simulation operation, biochemistry tests and enzyme-linked immunosorbent assay were used to measure NOS and 13-EP in the hypothalamus, spinal cord, and peripheral blood. RESULTS: Rats with the simulation operation showed significantly higher levels of NOS and II3-EP in the hypothalamus, spinal cord, and peripheral blood than those in the normal group. The EA and acupotomy groups had significantly lower levels of NOS and β-EP than those in the model group. There was no statistical difference between the EA and acupotomy groups. CONCLUSION: EA and acupotomy treatments significantly lowered NOS and β-EP levels in the hypothalamus, spinal cord, and peripheral blood and alleviated L3 transverse process syndrome.展开更多
