METHODS The serum TNF α, IFN γ, IL 6 and IL 8 were detected by ELISA in 66 patients with hepatitis B and 20 healthy blood donors. RESULTS TNF α and IL 6 in all types of clinical hepatitis B were significantly higher than those in healthy blood donors ( P <0 05); meanwhile the levels of TNF α, IFN γ, IL 6 and IL 8 in the patients with fulminant hepatitis B were much higher than those in the patients with acute hepatitis B ( P <0 05); the level of TNF α was positively correlated with the levels of IFN γ, Il 6 and IL 8 in all types of hepatitis B ( r IFN =0 24, r IL 6 =0 35, r IL 8 =0 44) and the TNF α, IFN γ, IL 6 and IL 8 were positively correlated with serum bilirubin ( P <0 05). Dynamic changes of these cytokines were observed in the course of acute and fulminant hepatitis. The level of IFN γ peaked in the initial period of acute hepatitis and early stage of hepatic coma in fulminant hepatitis; TNF α, IL 6 and IL 8 increased with exacerbation, and reached a peak when the liver damage was most serious, then decreased when patient conditions were improved. CONCLUSION The increased cytokines were related to the inflammation of liver cells and multiple factors may play certain roles in liver damage.展开更多

Recently a novel subset of TH cells, termed THIL-17, TH 17 or inflammatory TH (THi), has been identified as critical mediators of tissue inflammation. These cells produce IL-17 (also called IL-17A) and IL-17F, two most homologous cytokines sharing similar regulations. Here we report that when overexpressed in 293T cells, IL-17 and IL-17F form not only homodimers but also heterodimers, which we name as IL-17A/F. Fully differentiated mouse THi cells also naturally secrete IL-17A/F as well as IL-17 and IL-17F homodimeric cytokines. Recombinant IL-17A/F protein exhibits intermediate levels of potency in inducing IL-6 and KC (CXCL 1) as compared to homodimeric cytokines. IL-17A/F regulation of IL-6 and KC expression is dependent on IL-17RA and TRAF6. Thus, IL-17A/F cytokine represents another mechanism whereby T cells regulate inflammatory responses and may serve as a novel target for treating various immune-mediated diseases.展开更多

To investigate the possible role of interleukin (IL) 10 and IL 12 in the pathogenesis of psoriatic lesions and to supply theoretical basis for the gene therapy for psoriasis, the expression of IL 10 and IL 12 P35,...To investigate the possible role of interleukin (IL) 10 and IL 12 in the pathogenesis of psoriatic lesions and to supply theoretical basis for the gene therapy for psoriasis, the expression of IL 10 and IL 12 P35, P40 mRNA in 12 cases of psoriatic lesions and 6 normal skin tissues was detected by using RT PCR technique. The results showed that the expression of IL 10 mRNA in the psoriatic lesions was significantly lower than that in the normal skin tissues ( P <0.001). The expression of IL 12 P35 was positive both in the psoriatic lesions and in the normal skin tissues. IL 12 P40 mRNA was expressed positively only in the psoriatic lesions but negatively in the normal skin tissues. It was suggested that IL 12 might take an important role in the occurrence and progression of psoriasis, but IL 10 might have certain role in the regression of psoriasis.展开更多
