The thickness of coats can be used not only as a good age indicator for the geomorphic surfaces but also coats themselves can be directly used as dating materials in the ESR method. Through measuring the thickness of carbonate coats on stones in soils on the alluvial terraces in the mouth of the Heishanxia gorge of the Yellow River and ESR dating of layers separated from the carbonate coats, the average of accumulation rates of the thickness of carbonate coats on stones since 1.57 Ma was calculated to be 0.10 mm/ka in the studied area, and a regression equation between carbonate coat thickness and age was also generated. From these research results, ages of T2-T11 terraces of the Yellow River in the studied area were systematically determined, and their values are in turn 18 ka, 94 ka, 139 ka, 215 ka, 305 ka, 410 ka, 495 ka, 742 ka,展开更多

In this study, we applied lipid chromatography-electrospray ionization mass spec- trometry (LC-ESI-MSn ) to compare flavonoids in developing seeds of natural black-seeded B. na- pus and yellow-seeded introgression lines selected from progenies of B. napus–Sinapis alba so- matic hybrids. Aside from the most abundant phenolic compounds (sinapine and sinapic acid) and 1, 2-disinapoylglucose, 16 different flavonoids were identified and quantified, including (-)- epicatechin, five monocharged oligomers of (-)-epicatechin ([DP 2]- , [DP 3]- , [DP 4] [DP 2]- B2 and [DP 2]- B5), quercetin, kaempferol, isorhamnetin-dihexoside, kaempferol-sinapoyl-trihexoside, isorhamnetin- sinapoyl-trihexoside, isorhamnetin-hexoside-sulfate, and isorhamnetin-3-O-glucoside. Most of the flavonoids accumulated with seed development, whereas some rapidly decreased during maturation. The content of these flavonoids was lower in the yellow-seeded materials than in the black seeds. In addition, variations of insoluble procyanidin oligomers and soluble phenolic acids were observed among both rapeseed varieties. Transcriptome changes of genes participating in the flavonoid pathway were discovered by quantitative reverse transcription polymerase chain reaction analysis. Consistent with flavonoid changes identified by high performance liquid chromatography analysis, the expression of most genes in the flavonoid biosynthetic pathway was also downregulated.展开更多
Leaf color mutants in higher plants are considered to be ideal materials for studying the chlorophyll biosynthesis,photosynthesis mechanism and chloroplast development.Herein,we identified a spontaneous mutant,yc412,i...Leaf color mutants in higher plants are considered to be ideal materials for studying the chlorophyll biosynthesis,photosynthesis mechanism and chloroplast development.Herein,we identified a spontaneous mutant,yc412,in cultivated cucumber that exhibited yellow cotyledons.The yellow-lethal mutant was diagnosed with an abnormal chloroplast ultrastructure,and reduced photosynthetic capacity and pigment content.Through bulked segregant analysis-based whole-genome sequencing and fine genetic mapping,we narrowed the yellow cotyledons (yc) locus to a 96.8 kb interval on chromosome 3.By resequencing and molecular cloning,we showed that Csyc is a potential candidate gene,which encodes a yellow stripe-like (YSL) transporter.The T to C mutation in the promoter region of Csyc caused the yellow cotyledon phenotype in yc412.Compared to YZU027A (WT),the expression of Csyc was significantly downregulated in the cotyledons of yc412.Silencing of Csyc in cucumber via virus-induced gene silencing resulted in chlorotic leaves,mainly by suppressing the chlorophyll content.Furthermore,a comparative transcriptome analysis revealed that chloroplast-related genes and chlorophyll biosynthesis genes were significantly downregulated in yc412 cotyledons.Our results provide new insights into the molecular function of the YSL transporter in plant chloroplast development and chlorophyll synthesis.展开更多
