METHODS: A liver injury model was induced by carbon tetrachloride. Cytotoxicity was measured by MTY assay. The activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were determined with an automatic multifunction-biochemical analyzer and the levels of superoxide dismutase (SOD) and TNF-α were determined following the instructions of SOD kit and TNF radioimmunoassay kit. Uver sections were stained with hematoxylin and eosin (H&E) for histological evaluation and examined under light microscope. RESULTS: We found that GLPG can alleviate the L-02 liver cells injury induced by carbon tetrachloride (CCh) through the measurements of ALT and AST activities and the administration of GLPG to L-02 cells did not display any toxicity. Furthermore, histological analysis of mice liver injury induced by CCh with or without GLPG pretreatment indicated that GLPG can significantly suppress the toxicity induced by CCh in mice liver. We also found that GLPG reduced TNF-α level induced by CCh in the plasma of mice, whereas increased SOD activity in the rat serum. CONCLUSION: GLPG has hepatic protective activity against CCl4 induced injury both in vitro and in vivo. The possible antihepatotoxic mechanisms may be related to the suppression of TNF-α level and the free radical scavenging activity.展开更多

Therefore, it is hypothesized that melatonin combined with exercise can also al...Previous studies have demonstrated that melatonin combined with exercise can alleviate secondary damage after spinal cord injury in rats. Therefore, it is hypothesized that melatonin combined with exercise can also alleviate ischemic brain damage. In this study, adult rats were subjected to right middle cerebral artery occlusion after receiving 10 mg/kg melatonin or vehicle subcutaneously twice daily for 14 days. Forced exercise using an animal treadmill was performed at 20 m/min for 30 minutes per day for 6 days prior to middle cerebral artery occlusion. After middle cerebral artery occlusion, each rat received melatonin combined with exercise, melatonin or exercise alone equally for 7 days until sacrifice. Interestingly, rats receiving melatonin combined with exercise exhibited more severe neurological deficits than those receiving melatonin or exercise alone. Hypoxia-inducible factor la mRNA in the brain tissue was upregulated in rats receiving melatonin combined with exercise. Similarly, microtubule associated protein-2 mRNA expression was significantly upregulated in rats receiving melatonin alone. Chondroitin sulfate proteoglycan 4 (NG2) mRNA expression was significantly decreased in rats receiving melatonin combined with exercise as well as in rats receiving exercise alone. Furthermore, neural cell loss in the primary motor cortex was significantly reduced in rats receiving melatonin or exercise alone, but the change was not observed in rats receiving melatonin combined with exercise. These findings suggest that excessive intervention with melatonin, exercise or their combination may lead to negative effects on ischemia/reperfusion-induced brain damage.展开更多

文摘目的:探讨左西孟旦联合曲美他嗪对心衰(heart failure,HF)介入治疗术后患者血清半乳糖凝集素3(galectin-3)和人多配体蛋白聚糖4(syndecan-4)表达水平及心功能的影响。方法:2018年9月到2021年1月选择在本院完成介入治疗术的心衰患者108例,根据随机信封抽签原则把其分为联合组与对照组各54例。对照组给予左西孟旦治疗,联合组给予左西孟旦联合曲美他嗪治疗,两组都治疗观察1个月。结果:治疗后联合组的总有效率为98.1%,高于对照组的88.9%(P<0.05)。两组治疗后的左心室舒张末期内径(Left ventricular end diastolic diameter,LVEDD)、左心室收缩末期内径(Left ventricular end systolic diameter,LVESD)都低于治疗前(P<0.05),联合组低于对照组(P<0.05)。两组治疗后的血清galectin-3和syndecan-4含量低于治疗前(P<0.05),联合组低于对照组(P<0.05)。治疗后随访6个月,联合组的再住院率与死亡率为9.3%和3.7%,低于对照组的27.8%和14.8%(P<0.05)。结论:左西孟旦联合曲美他嗪对心衰介入治疗术后患者的应用能抑制血清galectin-3和syndecan-4表达水平,改善患者的心功能,提高治疗效果,降低患者的随访再住院率与死亡率。
