Background miR-338-3p is a recently discovered miRNA and is involved in cell differentiation.However,few data are yet available on the aberrant expression of miR-338-3p in human colorectal carcinoma (CRC).This work aimed to investigate the relationship between miR-338-3p expression pattern and clinicopathological features of human CRC and the possible regulative mechanisms.Methods The 40 CRC,adjacent nontumorous tissues and 2 human CRC-derived cell lines (SW-480 and SW-620) were collected,respectively,and the total RNA and protein were isolated routinely.The miR-338-3p expression pattern was detected by real-time reverse transcription-polymerase chain reaction (RT-PCR) and Northern blotting.Smoothened (SMO,possible target of miR-338-3p) mRNA and corresponding protein expression pattern were detected by semiquantitative RT-PCR and Western blotting.miR-338-3p expression patterns were compared between nontumor mucosa and CRC samples,graded by progression-related factors.Disease outcome was calculated by Kaplan-Meier survival analysis to determine whether miR-338-3p was related to disease-free survival (DFS) and overall survival (OS) of patients.Moreover,SMO 3'-UTR fragment was PCR amplified from genome DNA of human colon and inserted into a luciferase reporter plasmid.The luciferase reporter plasmid construct was then transfected into CRC cells together with pre-miR-338-3p or anti-miR-338-3p and the luciferase activity in the transfected cells was detected.Results The expression of miR-338-3p was significantly downregulated in CRCs than those in the adjacent nontumorous tissues,and the value was negatively related to advanced TNM stage and local invasion (P <0.01).Furthermore,miR338-3p value was decreased markedly in SW-620 cell line relative to SW-480 (P <0.01).Low expression of miR-338-3p was associated with unfavorable outcome in DFS but not in OS independent of clinical covariates.Moreover,RT-PCR and Western blotting analysis demonstrated that there was no significant differ 展开更多

目的:探讨赤芍总苷(total paeony glucosides,TPG)通过调控miR-338-3p/胶质细胞成熟因子β(glia maturation factorβ,GMFB)表达对结直肠癌细胞增殖、凋亡、迁移的影响。方法:体外培养人结直肠癌SW-480细胞株,使用不同浓度的TPG处理细胞... 目的:探讨赤芍总苷(total paeony glucosides,TPG)通过调控miR-338-3p/胶质细胞成熟因子β(glia maturation factorβ,GMFB)表达对结直肠癌细胞增殖、凋亡、迁移的影响。方法:体外培养人结直肠癌SW-480细胞株,使用不同浓度的TPG处理细胞,平板克隆实验检测细胞增殖,流式细胞仪检测细胞凋亡,细胞划痕实验检测细胞迁移,Transwell实验检测细胞侵袭,qRT-PCR检测细胞中miR-338-3p和GMFB的表达,双荧光素酶报告基因检测miR-338-3p和GMFB的靶向关系。结果:与对照组比较,TPG低剂量组、中剂量组和高剂量组的克隆形成数量、细胞愈合率、迁移数量明显降低(P<0.001),其中TPG高剂量组的克隆形成数量、细胞愈合率和迁移数量降低最明显;与对照组比较,TPG低剂量组、中剂量组和高剂量组的细胞凋亡率显著增加(P<0.001),其中TPG高剂量组的细胞凋亡率增加最明显;与对照组比较,TPG低剂量组、中剂量组和高剂量组的miR-338-3p表达明显升高(P<0.001),其中TPG高剂量组的miR-338-3p表达升高最明显;与对照组比较,TPG低剂量组、中剂量组和高剂量组的GMFB mRNA表达显著降低(P<0.001),其中TPG高剂量组的GMFB mRNA表达降低最明显;与miR-con组比较,anti-miR-338-3p组的克隆形成数量、细胞愈合率和迁移数量显著增加,细胞凋亡率明显降低(P<0.001);miR-338-3p显著降低GMFB-WT的荧光素酶活性(P<0.05),但对GMFB-MUT的荧光素酶活性没有产生影响(P>0.05);与TPG高剂量+miR-con组比较,TPG高剂量+anti-miR-338-3p组的克隆形成数量、细胞愈合率和迁移数量明显升高,细胞凋亡率显著下降(P<0.001)。结论:TPG可能通过调控miR-338-3p/GMFB抑制结直肠癌细胞的增殖、凋亡、迁移。 展开更多

目的基于miRNA-219a-5p、miRNA-338-3p探讨健脾合剂干预腹泻型肠易激综合征(Diarrhea-predominant irritable bowel syndrome,IBS-D)大鼠的作用机制。方法采用急-慢应激法联合番泻叶灌胃法制备IBS-D大鼠模型,分组后给予健脾合剂干预。... 目的基于miRNA-219a-5p、miRNA-338-3p探讨健脾合剂干预腹泻型肠易激综合征(Diarrhea-predominant irritable bowel syndrome,IBS-D)大鼠的作用机制。方法采用急-慢应激法联合番泻叶灌胃法制备IBS-D大鼠模型,分组后给予健脾合剂干预。实验过程中及治疗完成后,记录大鼠一般情况、体质量及粪便性状;采用直肠扩张下腹壁回缩反射(Abdominal withdrawal reflex,AWR)评分检测大鼠的疼痛阈值,评估IBS-D大鼠的内脏敏感性;采用显色基质鲎试剂检测大鼠血液中的细菌内毒素含量,观察其肠道通透性变化;观察大鼠结肠黏膜组织病理,对其肠黏膜状态进行评估;并使用RT-PCR法检测大鼠结肠黏膜miR-219a-5p、miR-338-3p的含量。结果粪便性状评分、内脏敏感性测定、组织病理结果表明IBS-D大鼠模型制备成功。与空白组比较,模型组大鼠大便稀溏甚则水样,体质量下降;健脾合剂治疗后,低剂量组及高剂量组大鼠大便成形,粪便Bristol评分显著降低,体质量有所增加。内脏敏感性结果显示,IBS-D大鼠内脏疼痛阈值下降;健脾合剂干预后,低剂量组及高剂量组大鼠内脏疼痛阈值有所升高,内脏敏感性降低。显色基质鲎试剂检测结果显示,IBS-D大鼠血清内毒素含量升高;健脾合剂干预后,低剂量组及高剂量组大鼠内毒素含量较前下降,肠道通透性降低。大鼠结肠粘膜病理结果表明,IBS-D大鼠结肠组织黏膜上皮轻度破损,局部可见水肿。健脾合剂治疗后,低剂量组及高剂量组大鼠结肠黏膜上述情况得到有效改善。Realtime PCR法检测大鼠结肠粘膜结果显示,IBS-D大鼠结肠黏膜miR-219a-5p、miR-338-3p水平显著降低,健脾合剂治疗后,高剂量组大鼠miR-219a-5p、miR-338-3p含量有所升高。结论健脾合剂对改善IBS-D症状具有一定的作用,健脾合剂能够增加IBS-D大鼠结肠黏膜miR-219a-5p、miR-338-3p水平,提示其可能通过对miR-219a-5p、miR-338-3p的调控降低IBS-D大鼠 展开更多

AIM:To investigate the regulative effect of miRNA-3383p(miR-338-3p) on cell growth in colorectal carcinoma(CRC).METHODS:The lentiviral vector pLV-THM-miR-338-3p and pLV-THM-miR-338-3p-inhibitor were constructed.The recombinant viral vector encoding the pre-miR338-3p or miR-338-3p-inhibitor and the two packaging plasmids psPAX2 and pMD2.G were cotransfected into human embryonic kidney 293T cells to package lentivirus.The supernatant containing the lentivirus particles was harvested to determine the viral titer,and this supernatant was then used to transduce CRCderived cell line,SW-620.Flow cytometry was utilized for sorting the green fluorescent protein(GFP) + cells to establish the SW-620 cell line stably expressing premiR-338-3p or miR-338-3p-inhibitor.Moreover,the expression of miR-338-3p was determined by real-time reverse transcriptase polymerase chain reaction,andWestern blotting was used to detect the expression of the smoothened(SMO,the possible target of miR-3383p) protein in SW-620 cells.Furthermore,the status of CRC cell proliferation and apoptosis were detected by 3-(4,5-dimethyl-2 thiazoyl)-2,5-diphenyl-2H-tetrazolium bromide assay and flow cytometry,respectively.RESULTS:Restriction enzyme digestion and DNA sequencing demonstrated that the lentiviral vector pLVTHM-miR-338-3p and pLV-THM-miR-338-3p-inhibitor were constructed successfully.GFP was expressed after the SW-620 cells were transduced by the lentivirus.Expression of miR-338-3p in SW-620 cells transduced with the lentivirus pLV-THM-miR-338-3p was significantly increased(relative expression 3.91 ± 0.51 vs 2.36 ± 0.44,P < 0.01).Furthermore,overexpression of miR-338-3p inhibited the expression of SMO protein in SW-620 cells,which showed obviously suppressed proliferation ability [cellular proliferation inhibition rate(CPIR) 61.9% ± 5.2% vs 41.6% ± 4.8%,P < 0.01].Expression of miR-338-3p in SW-620 cells transduced with the lentivirus pLV-THM-miR-338-3p-inhibitor was significantly decreased(relative expression 0.92 ± 0.29 vs 2.36 ± 0.44,P < 0. 展开更多

目的:分析未破裂颅内动脉瘤(UIAs)病人血清微小RNA-338-3p(miR-338-3p)的表达情况及其临床意义。方法:选取住院的UIAs病人105例作为观察组,另选取同期无脑动脉瘤的常规体检者100名作为对照组。UIAs病人入院后1 d、对照组体检当日的清晨... 目的:分析未破裂颅内动脉瘤(UIAs)病人血清微小RNA-338-3p(miR-338-3p)的表达情况及其临床意义。方法:选取住院的UIAs病人105例作为观察组,另选取同期无脑动脉瘤的常规体检者100名作为对照组。UIAs病人入院后1 d、对照组体检当日的清晨空腹肘静脉血,检测血清miR-338-3p、血清基质金属蛋白酶2(MMP-2)水平,分析血清miR-338-3p、MMP-2表达与UIAs病人临床病理特征的关系,采用Pearson相关分析UIAs病人血清miR-338-3p与MMP-2的相关性。结果:观察组血清miR-338-3p、MMP-2水平均高于对照组(P<0.01和P<0.05)。不同动脉瘤直径及动脉瘤数目病人血清miR-338-3p、MMP-2水平差异均有统计学意义(P<0.05~P<0.01)。Pearson相关分析显示,UIAs病人血清miR-338-3p与MMP-2呈显著正相关关系(P<0.01)。结论:UIAs病人血清miR-338-3p、MMP-2水平均高于对照组,二者呈正相关,且均与病人动脉瘤直径和数目有关。 展开更多
