Recent advances have been made in the understanding of auxin response pathways, primarily by the characterization of auxin response mutants in Arabidopsis. In addition, microRNAs (miRNAs) have been shown to be critical regulators of genes important for normal plant development and physiology. However, little is known about possible interactions between miRNAs and hormonal signaling during normal development. Here we show that an Arabidopsis microRNA, miR167, which has a complementary sequence to a portion of the A UXINRESPONSE FACTOR6 (ARF6) and ARF8 mRNAs, can cause transcript degradation for ARF8, but not for ARF6. We report phenotypic characterizations of 35S::MIR167b transgenic lines, and show that severe 35S::MIR167b transgenic lines had phenotypes similar to those of an arf6 arf8 double mutant. The transgenic phenotypes suggest that miR167 may repress ARF6 at the level of translation. We demonstrate that the transgenic plants are defective in all four whods of floral organs. In the transgenic flowers, filaments were abnormally short, anthers could not properly release pollen, and pollen grains did not germinate. Our results provide an important link between the miRNA-mediated regulatory pathway of gene expression and the auxin signaling network promoting plant reproductive development.展开更多

Homeotic conversion of lodicules and glumes into palea/lemma-like organs was observed in some flowers. Scanning electron microscopy observation showed that the size of flower meristems was maintained the same or similar until the lemma primordium started to differentiate, at which time the floral meristem became enlarged, suggesting abnormal development of the inner whorls of rice florets. Although a few quantitative trait loci associated with stigma exsertion have been fine map- ped or cloned, the underlying genetic architecture remains unclear. We performed a genome-wide associ- ation study on stigma exsertion and related floral traits using 6.5 million SNPs characterized in 533 diverse accessions of Oryza sativa. We identified 23 genomic loci that are significantly associated with stigma exsertion and related traits, three of which are co-localized with three major grain size genes GS3, GW5, and GW2. Further analyses indicated that these three genes affected the stigma exsertion by controlling the size and shape of the spikelet and stigma. Combinations of GS3 and GW5 largely defined the levels of stigma exsertion and related traits. Selections of these two genes resulted in specific distributions of floral traits among subpopulations of O. sativa. The low stigma exsertion combination gw5GS3 existed in half of the cultivated rice varieties; therefore, introducing the GW5gs3 combination into male sterile lines is of high potential for improving the seed production of hybrid rice.展开更多

We isolated an FT-homolog (GhFT1) from Gossypium hirsutum L. cultivar, Xinluzao 33 GhFT1 was predominantly expressed in stamens and sepals, and had a relatively higher expression level during the initiation stage of fiber development. GhFT1 mRNA displayed diurnal oscillations in both long-day and short-day condition, suggesting that the expression of this gene may be under the control of the circadian clock. Subcel ular analysis revealed that GhFT1 protein located in the cytoplasm and nucleus. Ectopic expression of GhFT1 in transgenic arabidopsis plants resulted in early flowering compared with wild-type plants. In addition, ectopic expression of GhFT1 in arabidopsis ft-10 mutants partial y rescued the extremely late flowering phenotype. Finally, several flowering related genes functioning downstream of AtFT were highly upregulated in the 35S::GhFT1 transgenic arabidopsis plants. This change in identity can be promoted by external signals, such as inductive photoperiod conditions or vernalization, and is accompanied by changes in expression of key developmental genes. The change in meristem identity is usually not reversible, even if the inductive signal occurs only transiently. This implies that at least some of the key genes must possess an intrinsic memory of the newly acquired expression state that ensures irreversibility of the process. In this review, we discuss different molecular scenarios that may underlie a molecular memory of gene expression.展开更多

The apricot cultivars tested were Kety, Golden Sun and Honghebao. With the development of flower buds, SCP and FP increased, which indicated that their cold resistance decreased. SCP and FP varied with different floral organs. For different apricot cultivars, it was found that, the lower SCP or FP in floral organs was, the more resistant capacity the cultivar had, and the larger the temperature interval from SCP to FP was. SCP was not a constant value, but a range. Frequency distribution of SCP in petals was more dispersing than that in stamens and pistils. Dynamism concerns different nucleosomal characteristics, including position, posttranslational modifications, and histone composition. Thus, within the nucleosome, canonical histones can be exchanged by histone variant proteins with specific functions--a process known as ‘histone replacement'. The histone variant H2A.Z has an important function in transcription and, during the last few years, its role in plant development and immune response has become evident. Compiling genetic and biochemical studies from several laboratories has revealed that plants contain a multiprotein complex, similar to the SWR1/SRCAP complex from yeast and animals, involved in H2A.Z deposition. Despite intense research in different organisms, the mechanism by which H2A.Z influences transcription is still unknown. However, recent results from Arabidopsis have shown a strong inverse correlation between H2A.Z and DNA methylation, suggesting that H2A.Z might protect genes from silencing.展开更多

OsMADS1, one of SEPALLATA-like MADS-box genes, has been unraveled to play critical roles in rice floral organ identity specification and floral meristem determinacy. However, the molecular mechanisms underlying interactions of OsMADS1 with other floral homeotic genes in regulating flower development remains largely elusive. In this work, we studied the genetic interactions of OsMADS1 with B-, C-, and D-class genes along with physical interactions among their proteins. We show that the physical and genetic interactions between OsMADS1 and OsMADS3 are essential for floral meristem activity maintenance and organ identity specification; while OsMADS1 physically and genetically interacts with OsMADS58 in regu- lating floral meristem determinacy and suppressing spikelet meristem reversion. We provided important genetic evidence to support the neofunctionalization of two rice C-class genes (OsMADS3 and OsMADS58) during flower development. Gene expression profiling and quantitative RT-PCR analyses further revealed that OsMADS1 affects the expression of many genes involved in floral identity and hormone signaling, and chromatin immunoprecipitation (ChlP)-PCR assay further demonstrated that OsMADS17 is a direct target gene of OsMADS1. Taken together, these results reveal that OsMADS1 has diversified regulatory functions in specifying rice floral organ and meristem identity, probably through its genetic and physical interactions with different floral homeotic regulators.展开更多

The results suggested that the world's mangrove plants have 84 species (including 12 varieties) in 24 genera and 16 families. Of which, true mangrove plants have 70 species (including 12 varieties) in 16 genera and 11 families, and semi-mangrove plants 14 species in eight genera and five families. The Eastern Group has 74 species (including 12 varieties) in 18 genera and 14 families, characterized by the genera Aegiceras, Osbornia, Aegialitis, Bruguiera, Ceriops, Kandelia, Scyphiphora and Nypa etc. The Western Group has only 10 species in six genera and five families, characterized by the endemic one-species family, Pelliceraceae, and the genus Laguncularia. The mangrove flora of China is composed of 26 species (including one variety) in 15 genera and 12 families, four of which are endemic. Hainan is most rich in mangrove species, making up about 96.2% of the Chinese total; Guangdong ranks second, making up about 42.3%. It has been demonstrated that Rhizophora stylosa was mistaken for R. mucronata in Taiwan by previous authors.展开更多

The developmental defects and changes of gene regulation network in reproductive tissues under drought stress are largely unknown. In this study, rice plants subjected to reproductive stage drought stress were examined for floral development and transcriptomic changes. The results showed that male fertility was dramatically affected, with differing pollen viability in flowers of the same panicle due to aberrant anther development under water stress. Examination of local starch distribution revealed that starch accumulated abnormally in terms of position and abundance in anthers of water-stressed plants. Microarray analysis using florets of different sizes identified 〉1000 drought-responsive genes, most of which were specifically regulated in only one or two particular sizes of florets, suggesting developmental stage-dependent responses to drought. Genes known to be involved in tapetum and/or microspore development, cell wall formation or expansion, and starch synthesis were found more frequently among the genes affected by drought than genome average, while meiosis and MADS-box genes were less frequently affected. In addition, pathways related to gibberellin acid signal- ing and abscisic acid catabolism were reprogrammed by drought. Our results strongly suggest interactions between reproductive development, phytohormone signaling, and carbohydrate metabolism in water-stressed plants.展开更多

Despite abundant flowering there is very low fruit and seed set,and very f...Manglietia ventii is a highly endangered plant species endemic to Yunnan province in China, where there are only five known small populations. Despite abundant flowering there is very low fruit and seed set,and very few seedlings in natural populations, indicating problems with reproduction. The causes of low fecundity in M. ventii are not known, largely because of insufficient knowledge of the species pollination ecology and breeding system. We conducted observations and pollination experiments, and analyzed floral scents to understand the pollinatoreplant interactions and the role of floral scent in this relationship, as well as the species breeding system. Like the majority of Magnoliaceae, M. ventii has protogynous and nocturnal flowers that emit a strong fragrance over two consecutive evenings. There is a closing period(the pre-staminate stage) during the process of anthesis of a flower, and we characterize the key flowering process as an "open-close-reopen" flowering rhythm with five distinct floral stages observed throughout the floral period of this species: pre-pistillate, pistillate, pre-staminate, staminate,and post-staminate. Flowers are in the pistillate stage during the first night of anthesis and enter the staminate stage the next night. During anthesis, floral scent emission occurs in the pistillate and staminate stages. The effective pollinators were weevils(Sitophilus sp.) and beetles(Anomala sp.), while the role of Rove beetles(Aleochara sp.) and thrips(Thrips sp.) in pollination of M. ventii appears to be minor or absent. The major chemical compounds of the floral scents were Limonene, b-Pinene, a-Pinene, 1,8-Cineole, Methyl-2-methylbutyrate, p-Cymene, Methyl-3-methyl-2-butenoate and 2-Methoxy-2-methyl-3-buten, and the relative proportions of these compounds varied between the pistillate and staminate stages. Production of these chemicals coincided with flower visitation by weevils and beetles.The results of pollination experiments suggest that M. ventii is pollinator-dependent, and low seed s展开更多

In order to understand the pollination system of Z. xanthoxylum, we investigate...Zygophyllum xanthoxylum, which belongs to Sarcozygium of Zygophyllaceae, is one of the ecologically important species in Northwest China. In order to understand the pollination system of Z. xanthoxylum, we investigated the following characteristics of this species in the Urat Desert-grassland Research Station in western Inner Mongolia of China: flowering dynamics, pollen viability, pollen limitation, floral visitors and breeding system. The results showed that the flowering period and flowering peak were different between the wild and managed popula- tions, being longer in the managed population. Z. xanthoxylum was pollen-limited, and pollen limitation was more intense in the wild population than in the managed population. Chalicodoma deserticola (Hymenoptera) was found to be the most frequent pollinator in the wild population, while Anthophora fulvitarsis (Hymenoptera) was the most frequent and effective visitor in the managed population. Out-crossing was dominant in the breeding system and self-pollination just played an assistant role to assure the reproduction of Z. xanthoxylum.展开更多

Endogenous IAA, DHZ+DHZR, iPA increased significantly during the floral bud formation, while Z+ZR remained changed. The levels of cytokinins, DHZ+DHZR, iPA, and Z+ZR all increased significantly during the vegetative bud formation, however IAA level was reduced during the first 7 days of culture and increased to two thirds of initial values on the day when the bud primordia were formed. The results suggested that the initiation of floral buds was associated with a high IAA/CTK ratio, whereas the induction of vegetative bud differentiation was related to a low IAA/CTK ratio.展开更多

文摘Manglietia ventii is a highly endangered plant species endemic to Yunnan province in China, where there are only five known small populations. Despite abundant flowering there is very low fruit and seed set,and very few seedlings in natural populations, indicating problems with reproduction. The causes of low fecundity in M. ventii are not known, largely because of insufficient knowledge of the species pollination ecology and breeding system. We conducted observations and pollination experiments, and analyzed floral scents to understand the pollinatoreplant interactions and the role of floral scent in this relationship, as well as the species breeding system. Like the majority of Magnoliaceae, M. ventii has protogynous and nocturnal flowers that emit a strong fragrance over two consecutive evenings. There is a closing period(the pre-staminate stage) during the process of anthesis of a flower, and we characterize the key flowering process as an "open-close-reopen" flowering rhythm with five distinct floral stages observed throughout the floral period of this species: pre-pistillate, pistillate, pre-staminate, staminate,and post-staminate. Flowers are in the pistillate stage during the first night of anthesis and enter the staminate stage the next night. During anthesis, floral scent emission occurs in the pistillate and staminate stages. The effective pollinators were weevils(Sitophilus sp.) and beetles(Anomala sp.), while the role of Rove beetles(Aleochara sp.) and thrips(Thrips sp.) in pollination of M. ventii appears to be minor or absent. The major chemical compounds of the floral scents were Limonene, b-Pinene, a-Pinene, 1,8-Cineole, Methyl-2-methylbutyrate, p-Cymene, Methyl-3-methyl-2-butenoate and 2-Methoxy-2-methyl-3-buten, and the relative proportions of these compounds varied between the pistillate and staminate stages. 文摘Zygophyllum xanthoxylum, which belongs to Sarcozygium of Zygophyllaceae, is one of the ecologically important species in Northwest China. In order to understand the pollination system of Z. xanthoxylum, we investigated the following characteristics of this species in the Urat Desert-grassland Research Station in western Inner Mongolia of China: flowering dynamics, pollen viability, pollen limitation, floral visitors and breeding system. The results showed that the flowering period and flowering peak were different between the wild and managed popula- tions, being longer in the managed population. Z. xanthoxylum was pollen-limited, and pollen limitation was more intense in the wild population than in the managed population. Chalicodoma deserticola (Hymenoptera) was found to be the most frequent pollinator in the wild population, while Anthophora fulvitarsis (Hymenoptera) was the most frequent and effective visitor in the managed population. Out-crossing was dominant in the breeding system and self-pollination just played an assistant role to assure the reproduction of Z. xanthoxylum.
文摘The sectioned thin cell layers (TCL) of flower stalk of Cichorium intybus L. were cultured in MS medium supplemented with NAA and BA or IAA and BA where floral and vegetative buds were developed from the explant. Endogenous IAA, DHZ+DHZR, iPA increased significantly during the floral bud formation, while Z+ZR remained changed. The levels of cytokinins, DHZ+DHZR, iPA, and Z+ZR all increased significantly during the vegetative bud formation, however IAA level was reduced during the first 7 days of culture and increased to two thirds of initial values on the day when the bud primordia were formed. The results suggested that the initiation of floral buds was associated with a high IAA/CTK ratio, whereas the induction of vegetative bud differentiation was related to a low IAA/CTK ratio.
