目的研究钙剂对血细胞分离机采集血小板过程中致患者发生枸橼酸钠中毒的预防效果。方法将400例进行机采血小板的献血者按照随机对照原则分为对照组、A、B、C四组,每组各100例,全部献血者在机采血小板前一周均未应用过阿司匹林类药物,其...目的研究钙剂对血细胞分离机采集血小板过程中致患者发生枸橼酸钠中毒的预防效果。方法将400例进行机采血小板的献血者按照随机对照原则分为对照组、A、B、C四组,每组各100例,全部献血者在机采血小板前一周均未应用过阿司匹林类药物,其中对照组在机采过程中未使用钙剂,A组在在口服10 ml 10%葡萄糖酸钙后立即进行机采血小板。B组在采集过程中如发生相关口唇麻木等症状时立即口服10 ml 10%葡萄糖酸钙。C组在每个循环机采血小板结束后,返输压积红细胞后立刻口服2 ml 10%葡萄糖酸钙,共5次,对全部献血者的枸橼酸钠中毒症状种类进行观察统计,并对每组的中毒症状发生率进行比较分析。结果全部献血者中,面部及口唇麻木发生例数位居第一,为102例(25.5%),其次由高至低依次为肢体麻木(25例,6.3%)、心悸(16例,4.0%)、胃肠道反应(6例,1.5%)、腓肠肌痉挛(4例,1.0%)。四组献血者的枸橼酸钠中毒症状发生率中,对照组的枸橼酸钠中毒症状发生率高于A、B、C三组,P<0.01,差异具有统计学意义,C组的枸橼酸钠中毒症状发生率低于对照组和A、B三组,P<0.01,差异具有统计学意义。结论小剂量多次口服钙剂能有效的减少机采血小板过程中枸橼酸钠中毒的情况。展开更多

Methods: Rat models of artery-veins bypassing thrombus formation were used. The platelets of rabbits were collected. Platelet aggregation was induced by collagen and intracellular calcium ion concentration ([Ca 2+ ]i) was assayed by Fura-2 method. Results: TFA (25, 50, 100 mg/kg) significantly and dose-dependently reduced the weight of thrombus. TFA (0.025, 0.05, 0.1 mg/ml) possessed dose-dependant inhibitory effects on rabbits' platelet aggregation induced by collagen. TFA significantly reduced the resting and CaCl 2-induced increase of free intracellular calcium concentration ([Ca 2+ ]i) in rabbit platelet in vitro . Conclusion: TFA has an antiplatelet effect via the inhibition on the influx of Ca 2+ .展开更多

But Piperol B (100, 200μmol· L-1) had no significant effect on aggregation of rabbit platelet induced by AA and ADP. Either increase of cytoplasmic free calcium[Ca2+]i or calcium influx induced by PAF was dose dependently inhibited by Piperol B (10, 100μmol· L-1). The maximum inhibition was 50. 8% and 37. 2%,respectively. But Piperol B had no effect on the increase of [Ca2+]i induced by A23187.
