目的探讨移植基质细胞衍生因子1(SDF-1)预处理的骨髓间充质干细胞(BMSC)对大鼠颈动脉狭窄的作用。方法携带增强型绿色荧光蛋白(EGFP)的质粒转染BMSC后经静脉注入大鼠。分为颈动脉损伤模型对照组、BMSC移植组、SDF-1预处理的BMSC移植组...目的探讨移植基质细胞衍生因子1(SDF-1)预处理的骨髓间充质干细胞(BMSC)对大鼠颈动脉狭窄的作用。方法携带增强型绿色荧光蛋白(EGFP)的质粒转染BMSC后经静脉注入大鼠。分为颈动脉损伤模型对照组、BMSC移植组、SDF-1预处理的BMSC移植组。细胞移植术后2周,取损伤处血管组织采用免疫荧光技术检测EGFP表达明确BMSC归巢情况;移植术后4周, Evans蓝染色观察损伤内膜内皮化程度,免疫荧光技术检测损伤血管CD31的表达情况;HE染色检测损伤颈动脉新生内膜增生程度,免疫组织化学染色法检测损伤血管增殖细胞核抗原(PCNA)的表达, Western blot法检测血管内皮生长因子(VEGF)的蛋白水平。结果移植SDF-1预处理的BMSC能有效促进干细胞向损伤血管的归巢,并促进损伤血管再内皮化程度。BMSC移植后新生内膜面积、新生内膜面积/中膜面积均低于对照组, SDF-1预处理的BMSC移植组差异更为显著。移植SDF-1预处理的BMSC可显著提高损伤内膜CD31以及VEGF的水平,并抑制PCNA的表达。结论SDF-1预处理BMSC移植可通过促进BMSC归巢至损伤处并诱导细胞分化,抑制中膜平滑肌细胞增殖来减轻血管狭窄程度。展开更多
This study was designed to determine the levels of early endothelial progenitor cells (EPCs), apelin, vascu- lar endothelial growth factor (VEGF) and stromal cell-derived growth factor-1 (SDF-1) after acute myoc...This study was designed to determine the levels of early endothelial progenitor cells (EPCs), apelin, vascu- lar endothelial growth factor (VEGF) and stromal cell-derived growth factor-1 (SDF-1) after acute myocardial infarction (AMI), and to investigate the relationships between these cytokines and early EPCs. Early EPCs, de- fined as CD133+, KDR+, and CD34~ cells, were quantified by flow cytometry. The levels of early EPCs and those cytokines in AMI patients were significantly different from those with coronary artery disease or controls (P 〈 0.05). Plasma apelin levels were inversely correlated with Gensini score and early EPCs (both P 〈 0.01). Early EPCs, VEGF and SDF-1 showed different patterns of changes in AMI patients during the first 24 h. The trend in the change of early EPCs was proportionally correlated with that of VEGF (P 〈 0.05). AMI patients exhibited in- creased early EPCs with remarkably decreased apelin levels and enhanced VEGF levels.展开更多
目的:观察独活续断汤口服和离子导入治疗肝肾亏虚型膝骨关节炎(KOA)的临床疗效及对基质细胞衍生因子-1(SDF-1)/C-X-C趋化因子受体4型(CXCR4)信号通路的影响。方法:将150例天津中医药大学第一附属医院确诊的肝肾亏虚型KOA患者随机分为对...目的:观察独活续断汤口服和离子导入治疗肝肾亏虚型膝骨关节炎(KOA)的临床疗效及对基质细胞衍生因子-1(SDF-1)/C-X-C趋化因子受体4型(CXCR4)信号通路的影响。方法:将150例天津中医药大学第一附属医院确诊的肝肾亏虚型KOA患者随机分为对照组、中药口服组和中药离子导入组,每组各50例,对照组给予硫酸氨基葡萄糖胶囊,口服0.5 g/次,2次/d口服,中药口服组给予独活续断汤150 m L/次,2次/d口服,中药离子导入组给予独活续断汤,于髋骨穴、膝关穴、膝眼穴及犊鼻穴进行离子导入,30 min/次,1次/d,三组患者均治疗4周;观察入组患者治疗前后膝关节肿胀程度及疼痛程度变化,并统计临床疗效;酶联免疫夹心吸附测定检测入组患者治疗前后膝关节液中SDF-1,CXCR4,基质金属蛋白酶-3(MMP-3)及MMP-13含量。结果:中药口服组疗效优于中药离子导入组和对照组,且复发率最低(P<0.05);与本组治疗前比较,中药口服组治疗后压痛值升高(P<0.05),中药口服组患者视觉模拟评分(VAS),膝关节肿胀评分、西安大略麦马斯特大学骨关节炎指数评分(WOMAC)及膝关节液SDF-1,CXCR4,MMP-3及MMP^-13蛋白含量均降低(P<0.05),且优于中药离子导入组和对照组(P<0.05)。结论:独活续断汤口服及离子导入治疗肝肾亏虚型KOA均有明显疗效,但口服疗效最佳,其机制可能与抑制SDF^-1/CXCR4炎症信号通路及软骨细胞降解有关。展开更多
